Identifying clinical correlates of drinking clusters during treatment for alcohol use disorder.

BACKGROUND AND OBJECTIVES: Despite the availability of treatments for alcohol use disorder (AUD), relapse prevalence and health-related consequences associated with AUD remains high. Using data-driven approaches that enhance generalizability can help elucidate relationships between treatment outcomes and alcohol consumption, aiding in the discovery of novel treatment targets for AUD subtypes. METHODS: We merged data (n = 2045) across four Phase 2 randomized clinical trials affiliated with the NIAAA Clinical Investigations Group and a Phase 3 trial (NIAAA Sponsored). Participants were clustere

Increased alcohol drinking and seizure susceptibility in female humanized ApoE4 knockin rats.

Epidemiological analyses suggest that the Ɛ4 allele of apolipoprotein E (ApoE) genes may influence the effects of alcohol on cognitive and executive function and dementia risk compared to the Ɛ3 allele. Here, we investigated this question in female rats given that women are more vulnerable than men to the Ɛ4 genotype effects on various diseases. Experiment 1 examined the effects of alcohol drinking on performance in a Barnes maze and an operant strategy set-shifting (OSS) task during abstinence in wildtype (WT) and homozygous ApoE4 knock-in (E4) rats. Experiment 2 repeated the behavioral asses

Aquaporin 9 regulates acetaldehyde uptake, alcohol-induced liver injury, and drinking behavior.

BACKGROUND: Acetaldehyde (AcH), a highly reactive metabolite of ethanol, plays a pivotal role in the pathogenesis of alcohol-associated liver disease (ALD) and alcohol use disorder (AUD). Post alcohol consumption, AcH generated in hepatocytes is further metabolized into acetate by aldehyde dehydrogenase 2 (ALDH2) or excreted into circulation and bile via the basolateral and apical membranes, respectively. Our previous studies have demonstrated that aquaporin 8 (AQP8), which is a water channel and mainly expressed on the apical membrane, facilitates AcH excretion into bile. AQP9 is predominantl

KCa2 channel positive modulation reduces alcohol drinking in female C57BL/6J mice.

Although men have historically exhibited higher levels of alcohol use disorder (AUD) diagnosis, the gap between men and women has been diminishing quickly. Preclinical screening for pharmacological treatments for AUD has typically focused solely on males, ignoring the possibility that males and females may differ mechanistically for the same behavioral phenotype. To ensure the efficacy of treatment targets across the sexes, it is crucial to study the pharmacological effects of AUD treatments in males and females. While positive KCa2 channel modulation can reduce ethanol consumption and seeking

Noradrenaline Modulates Central Amygdala GABA Transmission and Alcohol Drinking in Female Rats.

BACKGROUND: Alcohol use disorder (AUD) is a chronic relapsing disorder and a leading preventable cause of death worldwide. The central nucleus of the amygdala (CeA) is a hub for stress and AUD. Norepinephrine (NE), also known as noradrenaline, regulates the brain's response to stress and alcohol. We previously reported that α1-adrenergic receptors drive moderate alcohol intake, while β receptors contribute to excessive drinking associated with dependence in male rats. METHODS: Here, we determined that alcohol dependence and withdrawal alter the CeA noradrenergic system in female rats using ex

Swiss Webster mice as a model for excessive alcohol binge drinking consumption.

Binge drinking (BD) is a widespread pattern of excessive alcohol consumption among adolescents and young adults with detrimental consequences for brain development. Animal models are essential for investigating the neurobiological mechanisms underlying BD, but selecting an appropriate model is critical to ensure relevance to human behavior. This study aims to validate a murine model of (BD) using Swiss Webster mice. To achieve this, both adolescent and adult mice were exposed to either a single binge (SB) or multiple binge (MB) of BD through intraperitoneal ethanol injections. The findings rev

Examining Alcohol-Related Blackouts and Drinking Motives Over Time Among College Women.

OBJECTIVE: Excessive alcohol consumption and its consequences among college women continues despite prevention efforts. One common consequence, alcohol-related blackouts (ARBs), are periods of alcohol-activated anterograde amnesia. The purpose of the current project is to extend the ARB and drinking motive literature by examining their relationship over time. METHOD: A sample of 424 women (88.9% White) completed online surveys assessing their ARBs and drinking motives weekly for 10 weeks. A series of hierarchical generalized linear models were estimated to examine the between-person and withi

Studies using macaque monkeys to address excessive alcohol drinking and stress interactions.

The use of non-human primates (NHPs) in studies of volitional, oral self-administration of alcohol can help address the complex interplay between stress and excessive alcohol consumption. There are aspects to brain, endocrine and behavior of NHPs, particularly macaques, that provide a critical translational link towards understanding the risks and consequences of alcohol use disorders (AUDs) in humans. These include wide individual differences in escalating daily alcohol intake, accurate measures of hypothalamic-pituitary-adrenal (HPA) axis hormonal interactions, neuroanatomical specificity of

Social anxiety and alcohol-related impairment: The mediational impact of solitary drinking.

Social anxiety disorder more than quadruples the risk of developing an alcohol use disorder, yet it is inconsistently linked to drinking frequency. Inconsistent findings may be at least partially due to lack of attention to drinking context - it may be that socially anxious individuals are especially vulnerable to drinking more often in specific contexts that increase their risk for alcohol-related problems. For instance, socially anxious persons may drink more often while alone, before social situations for "liquid courage" and/or after social situations to manage negative thoughts about thei

College Status, Perceived Drinking Norms, and Alcohol Use among Sexual Minority Women.

Lesbian, gay, and bisexual young adults are at elevated risk for drinking compared to heterosexual young adults, and this discrepancy is particularly striking for sexual minority women (SMW). Perceived social norms are strong predictors of young adult alcohol use, especially among college students. The college environment is often one where increases in alcohol use are seen, but the impact of college status on SMW's drinking has been understudied. The present study explored patterns of alcohol use and consequences among SMW and the extent to which social norms relate to use and consequences. P

Paternal drinking, intimate relationship quality, and alcohol consumption in pregnant Ukrainian women.

OBJECTIVE: Maternal alcohol consumption during pregnancy and fetal alcohol spectrum disorders (FASDs) represent a significant public health problem. The influence of the male partner's alcohol consumption patterns and the quality of the partner's intimate relationship might be important factors to consider in the design of successful FASD prevention programs. METHOD: As part of the Collaborative Initiative on Fetal Alcohol Spectrum Disorders, 166 pregnant women in two regions in Ukraine participated in an in-person interview at an average gestational age of 18-19 weeks. Subjects were classifi

Family history of alcohol abuse associated with problematic drinking among college students.

Studies examining family history of alcohol abuse among college students are not only conflicting, but have suffered various limitations. The current report investigates family history of alcohol abuse (FH+) and its relationship with alcohol expectancies, consumption, and consequences. In the current study, 3753 student participants (35% FH+), completed online assessments. Compared to FH- same-sex peers, FH+ males and FH+ females endorsed greater overall positive expectancies, consumed more drinks per week, and experienced more alcohol-related negative consequences. Further, FH+ females evalua

Drinking alcohol during pregnancy: evidence from Canadian Community Health Survey 2007/2008.

BACKGROUND: Drinking alcohol during pregnancy may cause many health problems for the child, one of which is fetal alcohol spectrum disorder (FASD). Since FASD is incurable, actions meant to prevent the occurrence of the disability by targeting drinking women become more important. Epidemiological data on drinking among pregnant women, including prevalence and determinants/risk factors, is essential for designing and evaluating prevention programs. OBJECTIVES: To estimate the prevalence of drinking alcohol during pregnancy and examine the determinants of this behaviour. METHODS: Using the 200

Brief intervention for prenatal alcohol use: the role of drinking goal selection.

The behavioral problems and cognitive deficits resulting from even small amounts of prenatal alcohol exposure can be significant and enduring but completely preventable. The purpose of this study was to examine the impact of a prenatal drinking goal selected during a brief intervention for 115 pregnant women and their partners on subsequent consumption. Higher proportions of women having their first pregnancy chose abstinence as a goal over drinking reduction. Goal selection was highly predictive of subsequent drinking behavior. Interestingly, the participants who were abstinent at enrollment

Naltrexone alters subjective and psychomotor responses to alcohol in heavy drinking subjects.

Preclinical studies support endogenous opioid system involvement in alcohol reinforcement and consumption; however, recent clinical trials and human laboratory studies have provided mixed findings of the effects of naltrexone (a non-selective opioid antagonist) on alcohol responses. This study used a within-subject design (n = 23) to investigate naltrexone effects (0, 50 and 100 mg qd) on subjective and psychomotor responses to alcohol (none, moderate, high) in heavy drinkers. Before alcohol administration, subjects reported decreased desire to drink alcohol when maintained on 50 mg compared w
