Tympanic membrane perforations.

The recent literature on the healing of tympanic membrane perforations is reviewed. Research on the migratory phenomena, epithelial migration and keratin dispersion on top of the tympanic membrane, is discussed. The etiology and pathogenesis of tympanic membrane perforations is then briefly described. The literature on the healing process of tympanic membrane on animal experiments as well as observations in humans is amply discussed. Finally different non invasive and or office procedures for tympanic membrane repair are reported.

Asymmetric Gene Flow Maintains Range Edges in a Marine Invertebrate.

AbstractSpecies are restricted to finite ranges, and when they are in demographic equilibrium with the environment, this implies a failure to adapt at the range periphery. There are several competing evolutionary hypotheses for the maintenance and persistence of range edges. One hypothesis is gene swamping of edge populations, in which migrants from the center carrying alleles adapted to the range center but maladaptive along the edge hamper local adaptation. A competing explanation is dispersal limitation, in which reduced migration to the periphery keeps edge populations small, causing drift

Aganglionosis in rodents.

The etiology of aganglionosis of the bowel remains controversial. Initial embryological studies in chicks, mice, and humans suggested the defect was in the migratory capacity of the vagal neural crest cells. This traditional theory has recently been challenged by the demonstration of a defect in the local microenvironment and the suggestion that neural crest cells migrate normally until they reach the terminal defective segment of bowel which then excludes them. To contribute to this debate we studied three rodent animal models using histological, "in vivo" (kidney capsule), and "in vitro" (ti

Nanohydroxyapatite-Stabilized Pickering-Emulsion-Incorporated Hydrogel Prevents Postoperative Osteosarcoma Recurrence and Promotes Bone Repair.

Osteosarcoma (OS) is a highly aggressive malignancy. Current therapies offer limited efficacy, necessitating innovative strategies. This study introduces a novel injectable hydrogel integrated with nanohydroxyapatite (nHAP)-stabilized Pickering emulsions for the local delivery of receptor activator of nuclear factor κB ligand inhibitor. We assessed the system's efficacy through cell viability, apoptosis, and migration assays in osteosarcoma cells and evaluated tumor suppression and bone regeneration in a critical-sized skull defect model in mice. Our results indicate that the system significan

Fingolimod reduces salivary infiltrates and increases salivary secretion in a murine Sjögren's model.

Sjögren's Syndrome (SjS) is a chronic, systemic autoimmune disease causing xerostomia, xerophthalmia, and systemic symptoms. The principal pathological finding in SjS is the accumulation of lymphocytes in exocrine glandular tissue and elsewhere, leading to secretory dysfunction and other abnormalities. A rational therapeutic approach might be to interfere with lymphocyte migration to the periphery from central lymphoid tissues. We thus examined in an animal model of SjS the effects of Fingolimod (FTY720, Gilenya™), which interferes with migration of lymphocytes to peripheral sites. Fingolimod

Preclinical evaluation of the InCraft" aortic endograft in a sheep model.

Animal models remain the gold standard for the preclinical evaluation of tissue response, sealing and integrity of aortic endografts. Preclinical testing of the InCraft® device was performed to evaluate these attributes. Through the femoral arteries of eight male crossbred sheep, 22 mm diameter InCraft® Aortic Bifurcate devices were deployed in the abdominal aortas, and shortened 13 mm diameter iliac limbs were deployed in the right iliac arteries. Vessels were excised for radiographic and histopathologic assessment at six months. There were no instances of graft thrombosis, type I endoleak or

First report about a cerebrospinal nematode infection in an alpaca (Vicugna pacos).

The number of New World camelids in European farms is rising and thus, the need for veterinary care towards these animals arises. However, veterinary care requires sophisticated knowledge on disease and pathogen occurrence within New World camelids. Here, an alpaca cria with neurological signs was admitted to the veterinary clinic. Although the animal was treated with antibiotics, vitamins and dexamethason, it refused to drink milk and the clinical status worsened. After euthanasia, necropsy and histopathological examination were carried out and revealed intracerebral nematode larvae. The morp

Distribution of mesenchymal stem cells in the area of tissue inflammation after transplantation of the cell material via different routes.

In experiments on male Wistar-Kyoto rats we studied the distribution of mesenchymal stem cells in intact body and in the presence of a focus of acute tissue inflammation. In healthy animals mesenchymal stem cells were transplanted intravenously. In the second case we used various routes of transplantation of mesenchymal stem cells: intravenous, into tissue adjacent to the inflammation focus, and into intact lobe of the damaged organ (prostate gland). Three weeks after transplantation, mesenchymal stem cells labeled with a fluorescent dye were detected in the bone marrow and intestine of intact

A critical role of erythropoietin receptor in neurogenesis and post-stroke recovery.

Erythropoietin (EPO) is the principal growth factor regulating the production of red blood cells. Recent studies demonstrated that exogenous EPO acts as a neuroprotectant and regulates neurogenesis. Using a genetic approach, we evaluate the roles of endogenous EPO and its classical receptor (EPOR) in mammalian neurogenesis. We demonstrate severe and identical embryonic neurogenesis defects in animals null for either the Epo or EpoR gene, suggesting that the classical EPOR is essential for EPO action during embryonic neurogenesis. Furthermore, by generating conditional EpoR knock-down animals,

Schistosoma mansoni: migration and attrition of irradiated and challenge schistosomula in the mouse.

The fate of irradiated, immunizing cercariae and challenge schistosomula was investigated in mice using a quantitative, histological technique which would appear to be more efficient in estimating parasite numbers in skin and lungs than does the tissue mincing and incubation recovery technique used previously by other workers. There was evidence for slight retardation of irradiated (20 krad.) parasite migration in skin, but death of schistosomula in the skin appeared negligible. It was observed that the majority of irradiated parasites remained in the lungs until at least day 21 after infectio

Immune responses to thyroglobulin in experimental allergic thyroiditis.

Guinea-pigs immunized with homologous thyroglobulin in complete Freund's adjuvant were observed for lesions in the thyroid gland, and for development of specific delayed type skin hypersensitivity. Their blood leucocytes were examined for specific sensitivity in vitro by determining the inhibition of migration in the presence of thyroglobulin. Serum was tested for humoral antibodies by the passive haemagglutination technique. There was a significant correlation between the development of thyroiditis and the intensity of the skin reactions. No relationship was observed between the presence of t

The aetiology and pathogenesis of craniofacial deformity.

Craniofacial malformations have been recorded since time immemorial. While observational studies have assisted in the recognition of syndromes, little light has been shed on the causal mechanisms which interfere with craniofacial development. Animal studies in which malformations occur spontaneously or have been induced by teratogenic agents have permitted step-by-step investigation of such common deformities as cleft lip and palate. The role of the ectomesenchymal cells of the neural crest and the possible phenomenon of disorganized spontaneous cell death are described in relation to lip clef

Legumain inhibitor prevents breast cancer bone metastasis by attenuating osteoclast differentiation and function.

Breast cancer is the main lethal disease among females, and metastasis to lung and bone poses a serious threat to patients' life. Therefore, identification of novel molecular mediators that can potentially be exploited as therapeutic targets for treating osteolytic bone metastases is needed. A murine model of breast cancer bone metastasis was developed by injection of 4 T1.2 cells into the left ventricle and hence directly into the arterial system leading to bone. AEP (Asparagine endopeptidase) inhibitor combined with epirubicin or epirubicin alone was administered by intraperitoneal injection

Roles of carbonic anhydrase 8 in neuronal cells and zebrafish.

BACKGROUND: Carbonic anhydrase 8 (CA8) is an isozyme of α-carbonic anhydrases (CAs). Previous studies showed that CA8 can be detected in human adult brain, with more intense expression in the cerebellum. Single mutations in CA8 were reported to cause novel syndromes like ataxia, mild mental retardation or the predisposition to quadrupedal gait. METHODS: In the present study, we examine the functions of CA8 in neuronal cell lines, mouse cerebellar granule neurons and zebrafish. RESULTS AND CONCLUSIONS: We demonstrated that overexpression of CA8 in neuronal cells significantly decreased cell d

Aberrant extradural spinal migration of Spirocerca lupi: four dogs.

Spirocerca lupi is a nematode mainly affecting dogs but has been found in other animals, particularly carnivores. Anatomical areas of typical and aberrant migration vary. This report describes four cases of Spirocerca lupi causing neurological symptoms, similar to thoracolumbar disc syndrome, as a result of aberrant migration of the nematode into the spinal canal. In two of the cases, the nematode could be demonstrated intraoperatively. The third was discovered on post-mortem examination, while the fourth case displayed compelling evidence of Spirocerca lupi involvement. Surgical removal of th
