Chitosan-ricobendazole complex: Synthesis, characterization and anthelmintic activity.

Synthesis, characterization and assessment of therapeutic efficacy of chitosan-ricobendazole complex were carried out for the first time in this work. Study of physico-chemical properties revealed the optimal ratio of chitosan: ricobendazole (30:4). Quantum chemical modeling set the optimal parameters for the formation of the chitosan-ricobendazole molecular system (E = -3765.26 kcal/mol, η = 0.127 eV), which was confirmed by Fourier-transform infrared spectroscopy. Scanning electron microscopy showed spherical particles of chitosan-ricobendazole complex ranging in size from 100 to 200 μm. Stu

Repurposing methuosis-inducing anticancer drugs for anthelmintic therapy.

Drug-resistant parasitic nematodes pose a grave threat to plants, animals, and humans. An innovative paradigm for treating parasitic nematodes is emphasized in this opinion. This approach relies on repurposing methuosis (a death characterized by accumulation of large vacuoles) inducing anticancer drugs as anthelmintics. We review drugs/chemicals that have shown to kill nematodes or cancerous cells by inducing multiple vacuoles that eventually coalesce and rupture. This perspective additionally offers a succinct summary on Structure-Activity Relationship (SAR) of methuosis-inducing small molecu…

Multiple anthelmintic drug resistant Ancylostoma caninum in foxhounds.

Ancylostoma caninum is the most common and important gastrointestinal nematode of dogs in the United States. Despite recent reports of A. caninum isolates resistant to all classes of anthelmintics, little is known about the frequency and extent of this anthelmintic resistance. The study aim was to evaluate the efficacy of three commercial anthelmintic products in the treatment of foxhound dogs with a history of persistent A. caninum infections. In the first phase of this study, 35 foxhounds were randomly divided into three treatment groups: moxidectin/imidacloprid (MI), pyrantel pamoate/febant…

Anthelmintic resistance of horse strongyle nematodes to fenbendazole in Lithuania.

BACKGROUND: Control of strongyle infections presents a global challenge for horse practitioners due to the development of anthelmintic resistance (AR), however comprehensive information on AR in Lithuania is still lacking. The aim of this study was to assess the current situation of fenbendazole (FBZ) AR in horses at stable level in Lithuania. RESULTS: Faecal samples from 121 horses from six stables were examined using the Mini-FLOTAC method. Of these, 89 horses met the inclusion criteria that included strongyle faecal egg counts (FEC) exceeding 200 eggs per gram (EPG). Faecal egg count reduc

Understanding the role of wild ruminants in anthelmintic resistance in livestock.

Wild ruminants are susceptible to infection from generalist helminth species, which can also infect domestic ruminants. A better understanding is required of the conditions under which wild ruminants can act as a source of helminths (including anthelmintic-resistant genotypes) for domestic ruminants, and vice versa, with the added possibility that wildlife could act as refugia for drug-susceptible genotypes and hence buffer the spread and development of resistance. Helminth infections cause significant productivity losses in domestic ruminants and a growing resistance to all classes of anthelm

Behavioral fingerprints predict insecticide and anthelmintic mode of action.

Novel invertebrate-killing compounds are required in agriculture and medicine to overcome resistance to existing treatments. Because insecticides and anthelmintics are discovered in phenotypic screens, a crucial step in the discovery process is determining the mode of action of hits. Visible whole-organism symptoms are combined with molecular and physiological data to determine mode of action. However, manual symptomology is laborious and requires symptoms that are strong enough to see by eye. Here, we use high-throughput imaging and quantitative phenotyping to measure Caenorhabditis elegans b…

Anthelmintic resistance in commercial sheep farms in Costa Rica.

Forty-eight commercial sheep farms were sampled between the years 2017 and 2019 to evaluate the percentage of reduction of the three familie … rmectin, and the presence of strains of Haemonchus spp. and Trichostrongylus sp. resistant to the 3 anthelmintics tested. The results of this study demonstrate that anthelmintic resistance is widespread, particularly in Haemonchus spp., and that the major drugs available in the country have lost much of their effectiveness against sheep nematodes.

The anthelmintic drug praziquantel promotes human Tr1 differentiation.

Praziquantel (PZQ) is an anthelminthic human and veterinary drug used to treat trematode and cestode worms. Changes in immune responses have been demonstrated in humans following curative PZQ treatment of schistosome infections. These changes have been attributed to the removal of immunosupressive worms and immune responses to parasite antigens exposed from dying worms. To date, there has been no study investigating the potential direct effect of PZQ on the host immune cells. Herein, we analyzed the effect of PZQ on human CD4+ T cells classically costimulated by CD3/CD28 or costimulated by the

Repositioning salicylanilide anthelmintic drugs to treat adenovirus infections.

The repositioning of drugs already approved by regulatory agencies for other indications is an emerging alternative for the development of new antimicrobial therapies. The repositioning process involves lower risks and costs than the de novo development of novel antimicrobial drugs. Currently, infections by adenovirus show a steady increment with a high clinical impact in immunosuppressed and immunocompetent patients. The lack of a safe and efficacious drug to treat these infections supports the search for new antiviral drugs. Here we evaluated the anti-adenovirus activity of niclosanide, oxyc

Anthelmintic efficacy against equine strongyles in the United States.

Equine strongyle parasites are ubiquitous in grazing equids across the world. Anthelmintic resistance is widely developed in cyathostomin populations, but very few surveys have evaluated anthelmintic efficacy in equine populations in the United States, and most of these are over 15 years old. The present study was carried out as part of the National Animal Health Monitoring Systems (NAHMS) Equine 2015-2016 study. The aims were to investigate anthelmintic treatment efficacy by means of the fecal egg count reduction test (FECRT) and identify parameters associated with decreased efficacy. Data we

Species differences in hepatic biotransformation of the anthelmintic drug flubendazole.

Flubendazole (FLBZ) is a broad-spectrum benzimidazole anthelmintic used in pigs, poultry, and humans. It has been proposed as a candidate for development for use in elimination programmes for lymphatic filariasis and onchocerciasis in humans. Moreover, FLBZ has shown promise in cancer chemotherapy, particularly for neuroblastoma. This work investigated the hepatic carbonyl-reducing pathway of FLBZ in different species, including humans. Microsomal and cytosolic fractions were obtained from sheep, cattle, pig, hen, rat, and human liver. Both subcellular fractions of each species converted FLBZ

Gastrointestinal nematodes and anthelmintic resistance in Danish goat herds.

The prevalence of gastrointestinal parasites in Danish goats and the presence of anthelmintic resistance (AR) in 10 selected herds were investigated during April-September 2012. All Danish herds (n = 137) with 10 or more adult goats were invited to participate, and of these 27 herds met the inclusion criterion of more than 10 young kids never treated with anthelmintics. Questionnaire data on management were collected, and faecal samples from 252 kids were analysed by the McMaster technique. From all herds with a mean faecal egg count (FEC) above 300 eggs per g of faeces, pooled samples were st…

Anthelmintic efficacy against cyathostomins in horses in Southern England.

Cyathostomins are considered to be the most important group of helminths to affect equids due to their high prevalence, potential pathogenicity and ability to develop anthelmintic resistance. Their control relies almost exclusively on frequent anthelmintic use. Currently, fenbendazole (FBZ), pyrantel embonate (PYR), ivermectin (IVM) and moxidectin (MOX) are licensed for use in horses in the UK. With no new anthelmintics likely to be licensed in the near future, it is essential that investigations into the efficacy of current anthelmintics in different locations are performed to help inform con…

Lipid-like properties and pharmacology of the anthelmintic macrocyclic lactones.

INTRODUCTION: Broad-spectrum antiparasitic macrocyclic lactones (MLs) are the principal nematicides used today, but drug resistance compromises the efficiency of ML-based therapy. Drug action in the parasite is essential for the effectiveness. Thus, to meet the needs of sustainable control of nematodes, the challenge is to maintain an effective drug concentration in the host tissues where parasites locate. This requires knowledge of the site of action of the drug, and processes that govern the pharmacokinetics of MLs in the host and in the parasites. These processes are primarily biotransforma

Improvement of the oral praziquantel anthelmintic effect by cyclodextrin complexation.

Schistosomiasis is a parasitic disease which kills a half million people per year, all over the world. Praziquantel (PZQ) is the drug-of-choice for schistosomiasis because of its effectiveness, ease of administration, and low cost. However, poor solubility restricts its delivery, especially via the oral route. In this study, we describe beta-cyclodextrin (beta-CD) complexation as an alternative to improve the PZQ bioavailability. Physicochemical analysis were performed to characterize the inclusion complex formed between PZQ and beta-CD. Differential scanning calorimetry (DSC) thermograms and

Ivermectin resistance and overview of the Consortium for Anthelmintic Resistance SNPs.

Ivermectin (IVM) has transformed nematode parasite control in veterinary medicine and the control of some nematode infections in humans, such as onchocerciasais, lymphatic filariasis in Africa and strongyloidiasis. Unfortunately, IVM resistance is now a serious problem for parasite control in livestock and there is a concern about resistance development and spread in nematode parasites of humans. IVM is believed to act by opening glutamate-gated chloride channels and GABA-gated channels in invertebrate neurons or muscle cells, leading to hyperpolarisation of the cells and to an inhibitory para

Growth of Enterobius vermicularis in a chimpanzee after anthelmintic treatment.

The growth of Enterobius vermicularis in a chimpanzee was investigated by observing worms discharged in feces after administration of pyrantel pamoate. Just after the final molting, immature adult male pinworms developed to a fully mature stage in 2 wk, after passing through a stage corresponding to the so-called Enterobius gregorii, which was surmised to be a younger adult form of E. vermicularis. The frequency distribution of body length forms 2 peaks in both male and female pinworms, with a depression in the transitional forms from the immature to the fully mature stage. This depression see
