Secondary metabolite arsenal of an opportunistic pathogenic fungus.

Aspergillus fumigatus is a versatile fungus able to successfully exploit diverse environments from mammalian lungs to agricultural waste products. Among its many fitness attributes are dozens of genetic loci containing biosynthetic gene clusters (BGCs) producing bioactive small molecules (often referred to as secondary metabolites or natural products) that provide growth advantages to the fungus dependent on environment. Here we summarize the current knowledge of these BGCs-18 of which can be named to product-their expression profiles in vivo, and which BGCs may enhance virulence of this oppor

Effect of aging on bioaccessibility of arsenic and lead in soils.

The effect of aging on the bioaccessibility of As and Pb in three soils spiked with As (40 or 400 mg kg(-1)), Pb (150 or 1500 mg kg(-1)) or As + Pb (40 mg kg(-1) As and 150 mg kg(-1) Pb) were investigated using the physiologically based extraction test (PBET). Prolonged aging in soils resulted in a decrease in As/Pb bioaccessibility, especially within the first month. After 76 weeks, As/Pb bioaccessibility in soils decreased to a stable level, with 48-84% and 8-34% for bioaccessible As and Pb respectively in the intestinal phase, illustrating that As in spiked soils was much more bioaccessible

An arsenal of magnetic nanoparticles; perspectives in the treatment of cancer.

Nanomedicine is an emerging field, which constitutes a new direction in the treatment of cancer. Magnetic nanoparticles (MNPs) can circumvent vascular tissue to concentrate at the site of the tumor. Under the influence of an external, alternating magnetic field, MNPs generate high temperatures within the tumor and ablate malignant cells while inflicting minimal damage to healthy host tissue. Due to their theranostic properties, they constitute a promising candidate for the treatment of cancer. A critical review of the type, size and therapeutic effect of different MNPs is presented, following

The Biochemical Toxin Arsenal from Ant Venoms.

Ants (Formicidae) represent a taxonomically diverse group of hymenopterans with over 13,000 extant species, the majority of which inject or spray secretions from a venom gland. The evolutionary success of ants is mostly due to their unique eusociality that has permitted them to develop complex collaborative strategies, partly involving their venom secretions, to defend their nest against predators, microbial pathogens, ant competitors, and to hunt prey. Activities of ant venom include paralytic, cytolytic, haemolytic, allergenic, pro-inflammatory, insecticidal, antimicrobial, and pain-producin

Arsenic and manganese alter lead deposition in the rat.

Lead (Pb) continues to be a major toxic metal in the environment. Pb exposure frequently occurs in the presence of other metals, such as arsenic (As) and manganese (Mn). Continued exposure to low levels of these metals may lead to long-term toxic effects due to their accumulation in several organs. Despite the recognition that metals in a mixture may alter each other's toxicity by affecting deposition, there is dearth of information on their interactions in vivo. In this work, we investigated the effect of As and Mn on Pb tissue deposition, focusing on the kidney, brain, and liver. Wistar rats…

The chemical arsenal of Burkholderia pseudomallei is essential for pathogenicity.

Increasing evidence has shown that small-molecule chemistry in microbes (i.e., secondary metabolism) can modulate the microbe-host response in infection and pathogenicity. The bacterial disease melioidosis is conferred by the highly virulent, antibiotic-resistant pathogen Burkholderia pseudomallei (BP). Whereas some macromolecular structures have been shown to influence BP virulence (e.g., secretion systems, cellular capsule, pili), the role of the large cryptic secondary metabolome encoded within its genome has been largely unexplored for its importance to virulence. Herein we demonstrate tha

K2ScSn(AsO4)3: an arsenate-containing langbeinite.

The title compound, dipotassium tri-mu-arsenato-scandium(III)tin(IV), is the first arsenate-containing langbeinite to be characterized by single-crystal methods and crystallizes in the aristotype P2(1)3 cubic symmetry for this structure type in which the K(+) ions and the octahedral scandium and tin cations lie on crystallographic threefold axes. The Sc(III) and Sn(IV) ions show a slight segregation over the two octahedral sites, with Sc/Sn populations of 0.582 (5):0.418 (5) on one site and 0.418 (5):0.582 (5) on the other. Bond-valence-sum calculations indicate that the K(+) ions are signific

Understanding arsenic carcinogenicity by the use of animal models.

Although numerous epidemiological studies have indicated that human arsenic exposure is associated with increased incidences of bladder, liver, skin, and lung cancers, limited attempts have been made to understand mechanisms of carcinogenicity using animal models. Dimethylarsinic acid (DMA), an organic arsenic compound, is a major metabolite of ingested inorganic arsenics in mammals. Recent in vitro studies have proven DMA to be a potent clastogenic agent, capable of inducing DNA damage including double strand breaks and cross-link formation. In our attempts to clarify DMA carcinogenicity, we …

Arsenic trioxide in the management of acute promyelocytic leukaemia.

Arsenical compounds were used as early as 2000 BC, both as medicines as well as poisons. Arsenicals gained importance in the beginning of the last century as the primary mode of treating syphilis. In 1931, Folkner and Scott used an arsenical preparation called Fowler's solution in the treatment of chronic myeloid leukaemia. This continued to be used until the introduction of busulphan in 1953. In the 1970s, arsenic trioxide was introduced for the treatment of acute promyelocytic leukaemia in China and was found to be extremely effective in treating this condition. Since then, numerous in vitro…

Heterogeneity of the DNA damage provoked by antimony and arsenic.

Data on the mechanism of antimony genotoxicity is scarce. Arsenic and antimony are proposed to share some toxicological features. Thus comparative and combined experiments with As(III) and Sb(III) were performed to gain a deeper knowledge of the mechanism of antimony genotoxicity. Trivalent arsenic proved to be five times more cytotoxic and one order of magnitude more potent in induction of micronuclei in human lymphocytes in vitro than was antimony. Significantly increased micronucleus frequencies were achieved with As(III) at a dose of 0.5 microM and with Sb(III) at a dose of 5 microM. Neith…

Lead arsenate poisoning in a herd of beef cattle.

Lead arsenate poisoning was diagnosed in 2 beef heifers and was suspected in 6 other cattle from the same herd that had died previously and were not examined. Clinical signs in affected cattle included staggering, dehydration, hemorrhage, acidemia, and shock. Diagnosis was by arsenic and lead analysis of urine samples and kidney and liver tissue digests. Both examined heifers died within 4 days of onset of clinical signs. These cattle had been moved from an area with poor grazing conditions to a pasture with abundant forage. This pasture had an open shed that contained an open sack of lead ars…

Arsenic toxicity in Hawaii: a case report and review.

As mentioned at the beginning of this article, many questions were raised in our one particular case including the problem of verifying true arsenic toxicity and in determining the source of the exposure. In our case, there was a markedly elevated concentration of arsenic in samples of pubic hair and in the sample of urine. While arsenic toxicity can present with GI symptoms, we felt that in this particular case the association of the abdominal pain with arsenic toxicity was unlikely. For one, the patient's symptoms persisted despite apparent adequate treatment for arsenic toxicity. Also, the …

The mitochondrial activation of sulfate and arsenate and their role in carcinogenesis.

Sulfate substitutes for phosphate in the transitory uncoupling of rat liver mitochondria induced by hydrazine when beta-hydroxybutyrate is the substrate. A high level of sulfate in the absence of added phosphate induces a pseudo state three of the mitochondria. Uncoupling is inhibited by rutamycin. Thus sulfate is activated by the mechanism usually utilized by phosphate, and the target for hydrazine is the bond holding electrophilic sulfate. ATP, ADP, PPi, and Mg++ protect against hydrazine, presumably by causing a conformational change of the phosphorylating enzymes which participate in oxida

Mortality in a region surrounding an arsenic emitting plant.

The purpose of the investigation is to study whether an increased mortality from certain causes exists in an area around the Rönnskärsverken smelter works in northern Sweden. Founded in 1928, this metallurgical plant processing mainly nonferrous metals has since its initial operations been using ore with a high arsenic content. This has resulted in environmental pollution to air and water of arsenic, as well as other metals and sulfur dioxide. The causes of death for the population of two parishes in the vicinity of the plant were listed from the National Sedish Register on Death Causes. A ref…
