Microgravity and the intervertebral disc: The impact of space conditions on the biomechanics of the spine.

The environmental conditions to which astronauts and other military pilots are subjected represent a unique example for understanding and studying the biomechanical events that regulate the functioning of the human body. In particular, microgravity has shown a significant impact on various biological systems, such as the cardiovascular system, immune system, endocrine system, and, last but not least, musculoskeletal system. Among the potential risks of flying, low back pain (LBP) has a high incidence among astronauts and military pilots, and it is often associated with intervertebral disc dege

Bacilli in the International Space Station.

Astronauts remote from Earth, not least those who will inhabit the Moon or Mars, are vulnerable to disease due to their reduced immunity, isolation from clinical support, and the disconnect from any buffering capacity provided by the Earth. Here, we explore potential risks for astronaut health, focusing on key aspects of the biology of Bacillus anthracis and other anthrax-like bacilli. We examine aspects of Bacillus cereus group genetics in relation to their evolutionary biology and pathogenicity; a new clade of the Bacillus cereus group, close related to B. anthracis, has colonized the Intern

Bone height measurements around a dental implant after a 6-month space flight: a case report.

PURPOSE: In space, astronauts are subject to microgravity, which reduces skeletal loading and osteoblast function and can cause bone resorption and a decrease in bone density. No known research to date has studied the effect of microgravity on dental implants. This study evaluated peri-implant bone changes around a dental implant placed in a French astronaut who spent 6 months in Russia's Mir Space Station. MATERIALS AND METHODS: Measurements were performed by 2 examiners before the flight (baseline), after the flight (stage 1), and following a recovery period (stage 2). Standardized periapic

Translating genetic findings to epigenetics: identifying the mechanisms associated with aging after high-radiation exposure on earth and in space.

PURPOSE: Exposure to radiation is a health concern within and beyond the Earth's atmosphere for aircrew and astronauts in their respective austere environments. The biological effects of radiation exposure from a multiomics standpoint are relatively unexplored and stand to shed light on tailored monitoring and treatment for those in these career fields. To establish a reference variable for genetic damage, biological age seems to be closely associated with the effect of radiation. Following a genetic-based study, this study explores the epigenetic landscape of radiation exposure along with its

Women in space: A review of known physiological adaptations and health perspectives.

Exposure to the spaceflight environment causes adaptations in most human physiological systems, many of which are thought to affect women differently from men. Since only 11.5% of astronauts worldwide have been female, these issues are largely understudied. The physiological nuances affecting the female body in the spaceflight environment remain inadequately defined since the last thorough published review on the subject. A PubMed literature search yielded over 2200 publications. Using NASA's 2014 review series 'The effects of sex and gender on adaptation to space' as a benchmark, we identifie

Anemia and Spaceflight Associated Neuro-Ocular Syndrome (SANS).

Spaceflight associated neuro-ocular syndrome (SANS) is a collection of distinct findings seen in some astronauts following prolonged spaceflight and is characterized by: optic disc edema, globe flattening, and choroidal folds. In this manuscript, we describe the potential mechanisms linking anemia and SANS. Future research aimed at understanding the relationship between these conditions may help to develop countermeasures and mitigation efforts for SANS.

Probabilistic Risk Assessment of Prophylactic Surgery Before Extended-Duration Spaceflight.

Introduction. Prophylactic surgery before spaceflight may eliminate the risk of appendicitis and cholecystitis in astronauts on deep space missions. However, even minimally invasive surgery increases the risk of small bowel obstruction (SBO). Probabilistic risk assessment (PRA) is a method that can be used to estimate the benefits and risks of prophylactic surgery. Methods. Risks of appendicitis and cholecystitis during a 2.5-year Mars mission are compared to the risk of SBO after laparoscopic removal of the appendix, gallbladder, or both. A PRA model using Monte Carlo methodology was used to

Interrelationships between pulse arrival time and arterial blood pressure during postural transitions before and after spaceflight.

We tested the hypothesis that acute changes in arterial blood pressure (BP) when astronauts moved between supine and standing posture before and after spaceflight can be tracked by beat-to-beat changes in pulse arrival time (PAT). Nine male crewmembers (45 ± 7 yr of age; mean mission length: 165 ± 13 days) participated in a standardized supine-to-sit-to-stand test (5 min-30 s-3 min) before flight and 1 day following return to Earth with continuous monitoring of ECG and finger arterial BP. PAT was determined from the R-wave of the ECG to the foot of the BP waveform. On average, modest cardiovas

Urea and Minerals Monitoring in Space Missions by Spot Samples of Saliva and Urine.

BACKGROUND: Microgravity induces redistribution of body fluids and reductions in muscle and bone mass. These effects correlate with changes in lab test results, including urea and bone minerals. Difficulties with collecting blood and urine during space missions limit the available data. This pilot study investigated metabolic changes during a space mission using untimed spot samples of urine and saliva. Untimed spot urine was used for urinalysis with data normalization per creatinine concentration. Saliva was proven useful as an index of serum urea and phosphorus.METHODS: Two astronauts collec

Anorexia in space and possible etiologies: an overview.

Space travelers experience a flight duration-dependent loss in weight and body mass while in a microgravity environment, despite the absence of increased energy expenditure. Anorexia in space can lead to in-flight caloric deficits of 1330 kcal per 70 kg astronaut per day in the presence of abundant food and has a critical effect on endurance and performance. Microgravity, alterations in the light-and-dark cycle, and exposure to radiation energy are the environmental stresses believed to influence appetite, food intake, and gastrointestinal function during space flight. Review of data and recen

The relationship between space sickness and preflight diet.

The nine astronauts who flew three Skylab missions during 1973 and 1974 were on carefully planned diets. Each astronaut selected his preferences from a list of approximately 70 food items, from which dietitians developed a rotating sequence of six daily menus. In addition to strict control of the inflight diet, each crewman consumed his planned diet for 21 days preflight and for 18 days postflight. While the amount consumed at each particular meal was largely discretionary, all deviations from the planned meals (which were few) were fully documented. Diets were controlled to provide adequate c

MAPK/PMK-1 innate immune signaling protects the nematode Caenorhabditis elegans from increased intestinal colonization in an animal host-pathogen model in space.

With the recent rise in the numbers and diversity of astronauts and space travelers, health and prevention of illness in space are of primary importance. Changes in immune function among astronauts during spaceflight have been reported, but gaps remain in understanding how this may translate to increases in an in-flight risk of infection. To understand how immunity and infection are affected by microgravity, we used the nematode Caenorhabditis elegans as an animal host-pathogen model. Worms exposed to either space or simulated microgravity for several days exhibited increased Enterobacter gut

Intermediate risk factors in ionising radiation cataractogenesis.

Recent reports indicate a new concept of low-dose ionising radiation cataractogenesis. In 2020, a two-stage aetiology of initiation and maturation was proposed by Richardson and colleagues for posterior subcapsular cataract (PSC) and perhaps cortical cataract development. The mechanisms involve various oxidative stress and biochemical factors, some of which are relevant to A-bomb survivors, such as ocular ion imbalance, inflammation, and maybe oxygen level changes. An example of a known, specific cataractogen is hypoparathyroidism and associated hypocalcaemia. However, a recent report indicate

Neck-vein thrombosis during spaceflight.

Spaceflight imposes unique environmental challenges, including weightlessness, increased radiation exposure and confinement, which can lead to unexpected health effects. One such example is neck-vein thrombosis, a condition rarely seen on Earth without predisposing factors such as venous catheters or infections. Although spaceflight has not previously been associated with thromboembolic events, recent cases of neck-vein thrombosis in astronauts aboard the International Space Station have raised concern. These events were detected incidentally and showed no obvious symptoms, complicating diagno

Evaluating the Suitability of Perfusion-Based PD Probes for Use in Altered Gravity Environments.

Measurable changes in electrophysiology have been documented in spaceflight, creating a pathway for disease genesis and progression in astronauts. These electrophysiology changes can be measured using potential difference (PD). A probe to measure PD was developed and is used clinically on Earth; this probe relies on fluid perfusion to establish an electrical connection to make PD measurements. The changes to fluid behavior in microgravity and partial gravity (including lunar and Martian gravity) drives the need to test this probe in a spaceflight environment. Here, we test the PD probe in a no

Exposure to low (10 cGy) doses of 4He ions leads to an apparent increase in risk taking propensity in female rats.

The planned missions to the Moon and Mars will present more significant health challenges to astronauts compared to low earth orbit missions. During deep space missions, astronauts will be constantly exposed to Space radiation (SR). Multiple rodent studies suggest that < 25 cGy of SR impairs performance in executive functions, which play a key role in advanced cognitive processes, but also regulate response inhibition and impulse control. There is the possibility that SR exposure may exacerbate aberrant behaviors evoked by psychological stress related to exposure to isolated and confined (ICE)

Ground-based training for the stimulus rearrangement encountered during spaceflight.

Approximately 65-70% of the crew members now experience motion sickness of some degree during the first 72 h of orbital flight on the Space Shuttle. Lack of congruence among signals from spatial orientation systems leads to sensory conflict, which appears to be the basic cause of space motion sickness. A project to develop training devices and procedures to preadapt astronauts to the stimulus rearrangements of microgravity is currently being pursued. The preflight adaptation trainers (PATs) are intended to: demonstrate sensory phenomena likely to be experienced in flight, allow astronauts to t

The effective intensity of Coriolis, cross-coupling stimulation is gravitoinertial force dependent: implications for space motion sickness.

Coriolis, cross-coupled angular acceleration stimulation readily induces motion sickness under terrestrial conditions. Nevertheless, the Skylab astronauts, when tested with such stimulation in-flight, were insusceptible even though each had been susceptible pre-flight. It is unclear whether this decreased susceptibility was the consequence of in-flight adaptation or in part the result of immediate changes in sensory-motor and vestibulo-motor function that occur during exposure to microgravity conditions. To evaluate this issue, we have tested individuals both in the high and low force phases o

Synergistic effect of IL-4 and simulated microgravity on osteogenic differentiation of periosteal stem cells.

In space, astronauts' cortical bone demonstrates a more stable maintenance of bone mass compared to the trabecular bone. In our previous research, enhanced periosteal bone formation, along with increased osteogenic differentiation capacity of periosteal stem cells (PSCs) under simulated microgravity (SMG) by clinostat was demonstrated. This phenomenon may contribute to the relative stability of astronauts' cortical bone under microgravity, whereas the underlying mechanism remains unclear. In this study, the contribution of Interleukin-4 (IL-4) and its synergistic effect with simulated microgra

Integration of Proteomic and Metabolomic Data Reveals the Lipid Metabolism Disorder in the Liver of Rats Exposed to Simulated Microgravity.

Long-term exposure to microgravity is considered to cause liver lipid accumulation, thereby increasing the risk of non-alcoholic fatty liver disease (NAFLD) among astronauts. However, the reasons for this persistence of symptoms remain insufficiently investigated. In this study, we used tandem mass tag (TMT)-based quantitative proteomics techniques, as well as non-targeted metabolomics techniques based on liquid chromatography-tandem mass spectrometry (LC-MS/MS), to comprehensively analyse the relative expression levels of proteins and the abundance of metabolites associated with lipid accumul
