Structure-enabled discovery of carboxylate esters prodrugs as potential potent inhibitors targeting Glutaminase C.

The role of glutaminase C (GAC) in regulating glutaminolysis has established it as a promising therapeutic target for cancer treatment, driving increased interest in GAC inhibitors in recent years. The mitochondrial localization of GAC presents a particular challenge for balancing high target binding affinity with effective delivery to its subcellular site. Our previous study identified a basic hotspot within the allosteric pocket of GAC, which could be leveraged to enhance binding affinity. Herein, a series of acidic derivatives targeting the basic hotspot were designed based on the scaffold

The kappa-opioid agonist U69,593 blocks cocaine-induced enhancement of brain stimulation reward.

BACKGROUND: Increasing evidence indicates that brain kappa-opioid receptors (KORs) are involved in regulation of mood states. In animal models often used to study psychiatric illness, KOR agonists produce depressive-like effects (e.g., anhedonia), whereas KOR antagonists produce antidepressant- and anxiolytic-like effects. The ability of KOR agonists to produce anhedonia-like signs in laboratory animals raises the possibility that this class of drugs might be useful to ameliorate states characterized by excess reward or motivation, such as mania or stimulant intoxication. METHODS: We examined

In vitro T-cell responses to beta-lactam drugs in immediate and nonimmediate allergic reactions.

BACKGROUND: beta-Lactam drugs may induce both cellular and humoral allergic reactions, and there is evidence that T cells play an important role in the pathogenesis of these reactions. The aim of this work was to assess the sensitivity and specificity of the lymphocyte transformation test (LTT) as an in vitro diagnostic tool, in patients with either an immediate or a nonimmediate reaction to penicillin G and/or amoxicillin. METHODS: Fifty patients with a well-documented history of allergic reactions to beta-lactams (31 immediate and 19 nonimmediate) were studied by means of skin tests (prick

Effects of U-50,488H on scopolamine-, mecamylamine- and dizocilpine-induced learning and memory impairment in rats.

The role of kappa opioid receptor agonists in learning and memory is controversial. In the present study, the effects of U-50,488H on scopolamine-, mecamylamine- and dizocilpine-induced learning and memory impairments in rats were investigated. Scopolamine (3.3 mumol/kg s.c.), a muscarinic cholinergic antagonist, and mecamylamine (40 mumol/kg s.c.), a nicotinic cholinergic antagonist, significantly impaired learning and memory in rats in a step-through type passive avoidance test. Administration of U-50,488H (0.17 or 0.51 mumol/kg s.c.) 25 min before the acquisition trial reversed the impairme

Activation of kappa opioid receptors by U50488H and morphine enhances the release of substance P from rat trigeminal nucleus slices.

The modulation of the release of substance P (SP) from sensory primary afferents by activation of kappa opioid receptors is not only equivocal, but also contradictory. Thus, in the present study, we have determined the effect of nanomolar concentrations of the highly selective kappa opioid receptor agonist trans-(+)-3,4-dichloro-N-methyl-N-[2-91- (pyrrolidinyl)cyclohexyl]benzacetamide methane sulphonate (U50488H), as well as micromolar concentrations of moderately mu-selective agonist morphine, on K(+)-evoked SP release from rat trigeminal nucleus caudalis slices. Production of hypothermia in the guinea pig by a kappa-agonist opioid alone and in combination with chlorpromazine.

In rats, kappa opioids decrease body temperature and the combination of a selective kappa agonist with chlorpromazine induces a profound hypothermia. Because of the greater density of kappa-opioid receptors and increased ratio of kappa to mu in the guinea pig, the actions of these drugs on body temperature were compared in this species. Groups of young adult, male Hartley guinea pigs were injected SC with trans-3,4-dichloro-N-methyl-N-(2-(1-pyrrolidinyl)-cyclohexyl)-benzeneace tamide methanesulfonate, hydrate (U50, 488H; 20-80 mg/kg) and chlorpromazine (2.5-5 mg/kg), either alone or in combina

Novel stable micellar formulation for ocular delivery of nepafenac: optimization, characterization and in-vitro evaluation.

OBJECTIVE: The objective of this study was to develop a stable clear ophthalmic nano-micellar formulation of Nepafenac. SIGNIFICANCE: The current marketed suspension formulation of nepafenac indicated for post-surgical uveitis is associated with higher manufacturing complexity and cost due to the need for sterile drug substance and specialized aseptic processing inherent to suspension-based ophthalmic products. It also exhibits limitations in patient compliance due to relatively high viscosity, lacrimation, and foreign body sensation following instillation. METHODS: Pre-formulation study was

Efficacy of Dual Inhibition of Glycolysis and Glutaminolysis for Therapy of Renal Lesions in Tsc2+/- Mice.

Tuberous sclerosis is caused by mutations in the TSC1 or TSC2 gene and characterized by development of tumors in multiple organs including the kidneys. TSC-associated tumors exhibit somatic loss of the second allele of the TSC genes, leading to aberrant activation of the mechanistic target of rapamycin (mTOR) signaling pathway. Activation of mTOR complex 1 (mTORC1) causes addiction to glucose and glutamine in Tsc1-/-or Tsc2-/- mouse embryonic fibroblasts (MEFs). Blocking of glutamine anaplerosis in combination with glycolytic inhibition causes significant cell death in Tsc2-/- but not Tsc2+/+

Clinical experience with penicillin skin testing in a large inner-city STD clinic.

OBJECTIVE: To establish (1) the prevalence of positive penicillin skin tests among outpatients with well-defined but variable history of penicillin allergy and (2) the reproducibility, safety, and negative predictive value of skin testing with benzylpenicilloyl polylysine (PPL) and a minor-determinant mixture (MDM). DESIGN: Serial consenting outpatients with current indications for penicillin therapy were skin-tested in duplicate with PPL and MDM. Subjects with negative skin tests (93% of those positive by history and 95% of those negative by history) received therapeutic courses of benzylpen

The influence of conditioned fear-induced stress on the opioid systems in the rat.

In this study the rats were repeatedly placed in a conditioning box, and 30 min later were subjected to a mild foot-shock. Anticipation of painful stimuli resulted in development of antinociception before a painful stimulus was applied. This conditioned fear-induced antinociception was antagonized by naloxone (1 mg/kg IP), as well as by ipsapirone (10 mg/kg IP), as measured by a tail-flick test. Stressed rats were hypersensitive to the analgesic action of morphine (1 mg/kg SC), but not to the specific kappa agonist U69,593 (0.1 mg/kg SC). In order to determine the involvement of the proopiomel

Differential cross-tolerance to opioids in squirrel monkeys responding under a shock titration schedule.

The opioid agonists morphine, l-methadone, ethylketazocine, U50,488 and bremazocine were examined in squirrel monkeys responding under a schedule of shock titration before, during and after chronic morphine administration. Before chronic morphine administration, all drugs produced dose-dependent increases in median shock level (the intensity below which monkeys maintained shock 50% of the time). After approximately 6 weeks of daily morphine administration, dose-effect curves for the mu agonist morphine shifted to the right approximately 3/4 log unit. Dose-effect curves for the mu agonist l-met

Nalfurafine is Aversive at Antinociceptive Doses in Mice.

Nalfurafine is the only clinically approved kappa opioid receptor (KOPr) agonist that can cross the blood-brain barrier and exert CNS effects. Because its clinical use is not associated with dysphoria, it is widely believed to have an atypical pharmacological profile. Nalfurafine's atypical properties are proposed to result from its G-protein-biased KOPr agonist property, leading to the widespread use of nalfurafine as a nonaversive KOPr agonist in preclinical research. The validity of nonaversive claims for nalfurafine was investigated in mice by comparing its antinociceptive and aversive eff

Physical presence of nor-binaltorphimine in mouse brain over 21 days after a single administration corresponds to its long-lasting antagonistic effect on κ-opioid receptors.

In the mouse 55°C warm-water tail-withdrawal assay, a single administration of nor-binaltorphimine (nor-BNI; 10 mg/kg i.p.) antagonized κ-opioid receptor (KOR) agonist-induced antinociception up to 14 days, whereas naloxone (10 mg/kg i.p.)-mediated antagonism lasted less than 1 day. In saturation binding experiments, mouse brain membranes isolated and washed 1 or 7 (but not 14) days after nor-BNI administration demonstrated a significant time-dependent decrease in maximal KOR agonist [(3)H]U69,593 binding. To determine whether brain concentrations of nor-BNI were sufficient to explain the anta
