Bioactive Peptides for Oral Diseases: Biomaterial-Assisted Therapeutics and Translational Applications.

Dental caries, periodontitis, oral mucosal inflammation, peri-implant infections, and oral tissue defects remain major clinical burdens. Conventional treatments, including antimicrobials, antibiotics, anti-inflammatory agents, and regenerative materials, are frequently limited by rapid clearance from saliva-exposed oral surfaces, insufficient discrimination between pathogenic and commensal microorganisms or between diseased and healthy tissues, antimicrobial resistance in biofilm-associated infections, and inadequate support for functional tissue repair. In this context, bioactive peptides hav

Bacterially grown living materials with resistant and on-demand functionality.

Inspired by naturally occurring biomaterials, autonomously grown engineered living materials (ELMs) feature cell-driven growth and programmable biological functions. However, the "livingness" of cells poses a short life span and low tolerance to harsh conditions, limiting the practical use of such materials. Here, we developed materials with programmable and dormant functionalities, grown from a mixture of Komagataeibacter rhaeticus and Bacillus endospores under engineered medium conditions. K. rhaeticus produces the bacterial cellulose (BC) matrix that integrates Bacillus spores within, where

Soft biocompatible polymer optical fiber tapers for implantable neural devices.

Optical fibers are between the most common implantable devices for delivering light in the nervous system for optogenetics and infrared neural stimulation applications. Tapered optical fibers, in particular, can offer homogeneous light delivery to a large volume and spatially resolved illumination compared to standard flat-cleaved fibers while being minimally invasive. However, the use of tapers for neural applications has up to now been limited to silica optical fibers, whose large Young's modulus can cause detrimental foreign body response in chronic settings. Here, we present the fabricatio

Boolean logic-gated protein presentation through autonomously compiled molecular topology.

Stimulus-responsive materials have enabled advanced applications in biosensing, tissue engineering and therapeutic delivery. Although controlled molecular topology has been demonstrated as an effective route toward creating materials that respond to prespecified input combinations, prior efforts suffer from a reliance on complicated and low-yielding multistep organic syntheses that dramatically limit their utility. Harnessing the power of recombinant expression, we integrate emerging chemical biology tools to create topologically specified protein cargos that can be site-specifically tethered

Cardiovascular Ischemia: Advancement and Potential Use of Collagen-Based Therapeutic Strategies.

Cardiac ischemia causes a shortage of blood flow and oxygen to the heart muscle (i.e., myocardial infarction, MI), causing cell damage/death that initiates a cascade of healing processes. Every year, more than one million people in the United States die as a result of MI. After MI, approximately 40% of patients develop maladaptive myocardial remodeling that associates with decreasing cardiac function levels. Collagen treatment, particularly the use of collagen-based biomaterials as well as collagen supplementation, has shown promise in treating cardiac ischemia by supporting processes involved

Recent progress on polySarcosine as an alternative to PEGylation: Synthesis and biomedical applications.

Biotherapeutic PEGylation to prolong action of medications has gained popularity over the last decades. Various hydrophilic natural polymers have been developed to tackle the drawbacks of PEGylation, such as its accelerated blood clearance and non-biodegradability. Polypeptoides, such as polysarcosine (pSar), have been explored as hydrophilic substitutes for PEG. pSar has PEG-like physicochemical characteristics such as water solubility and no reported cytotoxicity and immunogenicity. This review discusses pSar derivatives, synthesis, characterization approaches, biomedical applications, in ad

Sustainable Immunomodulatory via Macrophage P2Y12 Inhibition Mediated Bioactive Patche for Peritendinous Antiadhesion.

Persistent anti-inflammatory responses are critical for the prevention of peritendinous adhesion. Although modified anti-adhesion barriers have been studied extensively, the immune response induced by the implants and the unclear mechanism limits their application. In this research, the advantage of the multi-functionalities of CA (caffeic acid) is taken to synthesize biodegradable poly (ester urethane) urea elastomers with ester- and carbamate-bonded CA (PEUU-CA). PEUU-CA is electrospun into bioactive patches that can uniquely present a sustained CA niche, referred to as BPSN. In the early st

Physiomimetic biocompatibility evaluation of directly printed degradable porous iron implants using various cell types.

Additively manufactured (AM) degradable porous metallic biomaterials offer unique opportunities for satisfying the design requirements of an ideal bone substitute. Among the currently available biodegradable metals, iron has the highest elastic modulus, meaning that it would benefit the most from porous design. Given the successful preclinical applications of such biomaterials for the treatment of cardiovascular diseases, the moderate compatibility of AM porous iron with osteoblast-like cells, reported in earlier studies, has been surprising. This may be because, as opposed to static in vitro

Solute diffusion and partitioning in multi-arm poly(ethylene glycol) hydrogels.

Controlling solute transport in hydrogels is critical for numerous chemical separation applications, tissue engineering, and drug delivery systems. In previous review work, we have pointed out that proposed theoretical models and associated experiments tend to oversimplify the influence of the hydrogel structure on solute transport by addressing only the effects of the polymer volume fraction and mesh size of the networks on solute transport. Here, we reexamine these models by experimenting with a library of multi-arm poly(ethylene glycol) (PEG) hydrogels with simultaneous variations in four i

Leveraging Microgels Prepared from Poly(ethylene glycol) Bisepoxide and Polyetheramine for Versatile Surface Structuring of Agarose Hydrogels.

We demonstrate a macromer-type bisepoxide, poly(ethylene glycol) diglycidyl ether, polymerizing readily with a trifunctional polyetheramine Jeffamine T-403 in water to facilitate the development of a series of microgels abbreviated as PMG. Simply by varying the concentration of the as-prepared thermoresponsive intermediate prepolymer from 1 to 2 and 4%, hydrodynamic sizes of the resulting P1MG, P2MG, and P4MG are easily tuned in the submicrometer to micrometer range shown by the dynamic light scattering results. Besides size difference, these microgels also deform differently, where the drying

A Balance between Pro-Inflammatory and Pro-Reparative Macrophages is Observed in Regenerative D-MAPS.

Microporous annealed particle scaffolds (MAPS) are a new class of granular materials generated through the interlinking of tunable microgels, which produce an interconnected network of void space. These microgel building blocks can be designed with different mechanical or bio-active parameters to facilitate cell infiltration and modulate host response. Previously, changing the chirality of the microgel crosslinking peptides from L- to D-amino acids led to significant tissue regeneration and functional recovery in D-MAPS-treated cutaneous wounds. In this study, the immunomodulatory effect of D-

PEG-Based Hydrogel Coatings: Design Tools for Biomedical Applications.

Device failure due to undesired biological responses remains a substantial roadblock in the development and translation of new devices into clinical care. Polyethylene glycol (PEG)-based hydrogel coatings can be used to confer antifouling properties to medical devices-enabling minimization of biological responses such as bacterial infection, thrombosis, and foreign body reactions. Application of hydrogel coatings to diverse substrates requires careful consideration of multiple material factors. Herein, we report a systematic investigation of two coating methods: (1) traditional photoinitiated
