Cardiovascular and temperature adverse actions of stimulants.

The vast majority of illicit stimulants act at monoaminergic systems, causing both psychostimulant and adverse effects. Stimulants can interact as substrates or antagonists at the nerve terminal monoamine transporter that mediates the reuptake of monoamines across the nerve synaptic membrane and at the vesicular monoamine transporter (VMAT-2) that mediates storage of monoamines in vesicles. Stimulants can act directly at presynaptic or postsynaptic receptors for monoamines or have indirect monoamine-mimetic actions due to the release of monoamines. Cocaine and other stimulants can acutely incr

A unique porin meditates iron-selective transport through cyanobacterial outer membranes.

Cyanobacteria are globally important primary producers and nitrogen fixers with high iron demands. Low ambient dissolved iron concentrations in many aquatic environments mean that these organisms must maintain sufficient and selective transport of iron into the cell. However, the nature of iron transport pathways through the cyanobacterial outer membrane remains obscure. Here we present multiple lines of experimental evidence that collectively support the existence of a novel class of substrate-selective iron porin, Slr1908, in the outer membrane of the cyanobacterium Synechocystis sp. PCC 680

Contributions of Drug Transporters to Blood-Brain Barriers.

Blood-brain interfaces comprise the cerebral microvessel endothelium forming the blood-brain barrier (BBB) and the epithelium of the choroid plexuses forming the blood-cerebrospinal fluid barrier (BCSFB). Their main functions are to impede free diffusion between brain fluids and blood; to provide transport processes for essential nutrients, ions, and metabolic waste products; and to regulate the homeostasis of central nervous system (CNS), all of which are attributed to absent fenestrations, high expression of tight junction proteins at cell-cell contacts, and expression of multiple transporte

Metabolite transporters in trypanosomatid parasites: promising therapeutic targets but... How to deal with them?

Infections caused by protozoan parasites are one of the most important public health problems in developing countries. One approach to design new drugs for these parasitic diseases relies on metabolic and molecular features which are ideally absent in mammalian hosts. Out of them, nutrient transporters play an important role since they were subjected to millions of years of adaptation to parasitism, in which this protozoan replaced many biosynthetic routes for transport systems. Here we address the current knowledge of trypanosomatids transport systems and the molecules related to such process

Amphetamines, new psychoactive drugs and the monoamine transporter cycle.

In monoaminergic neurons, the vesicular transporters and the plasma membrane transporters operate in a relay. Amphetamine and its congeners target this relay to elicit their actions: most amphetamines are substrates, which pervert the relay to elicit efflux of monoamines into the synaptic cleft. However, some amphetamines act as transporter inhibitors. Both compound classes elicit profound psychostimulant effects, which render them liable to recreational abuse. Currently, a surge of new psychoactive substances occurs on a global scale. Chemists bypass drug bans by ingenuous structural variatio

Na+, K+, and Cl- transport in resting pancreatic acinar cells.

To understand the role of Na+, K+, and Cl- transporters in fluid and electrolyte secretion by pancreatic acinar cells, we studied the relationship between them in resting and stimulated cells. Measurements of [Cl-]i in resting cells showed that in HCO3(-)-buffered medium [Cl-]i and Cl- fluxes are dominated by the Cl-/HCO3- exchanger. In the absence of HCO3-, [Cl-]i is regulated by NaCl and NaK2Cl cotransport systems. Measurements of [Na+]i showed that the Na(+)-coupled Cl- transporters contributed to the regulation of [Na+]i, but the major Na+ influx pathway in resting pancreatic acinar cells

Active membrane transport and receptor proteins from bacteria.

A general strategy for the expression of bacterial membrane transport and receptor genes in Escherichia coli is described. Expression is amplified so that the encoded proteins comprise 5-35% of E. coli inner membrane protein. Depending upon their topology, proteins are produced with RGSH6 or a Strep tag at the C-terminus. These enable purification in mg quantities for crystallization and NMR studies. Examples of one nutrient uptake and one multidrug extrusion protein from Helicobacter pylori are described. This strategy is successful for membrane proteins from H. pylori, E. coli, Enterococcus

Crosslinking-Dependent Structure-Transport-Function Relationships in Gelatin-Hyaluronic Acid Hydrogels for Wound-Fluid Management.

Effective wound-fluid management is essential in medical applications, as dressings must absorb, retain, and regulate excess blood, exudate, and other biological fluids. Gelatin-hyaluronic acid (Gel/HA) hydrogels are promising dressing materials because they provide a hydrated, tissue-mimetic environment and combine the biological functions of both polymers. Their performance depends strongly on the crosslinking strategy, which regulates network architecture, fluid transport, and mechanical integrity. Three Gel/HA networks were prepared by physical association (Network I), glutaraldehyde-media

Diversity of human choline transporters: substrate specificity, kinetic properties, and inhibitor sensitivity.

Carrier-mediated uptake of choline is rate-limiting for acetylcholine biosynthesis and various other biological processes. To date, 16 solute carrier (SLC) proteins have been identified that may facilitate choline permeation across the outer cell membrane. However, their biochemical functions have not yet been experimentally compared. We overexpressed 16 SLC proteins with known choline-transporting capacity and compared their choline transport kinetics. Additionally, we evaluated their capacity to transport choline analogues as well as metabolites involved in its biosynthesis and degradation o

Rosmarinic Acid Is an hCtr1 Inhibitor that Interferes with Copper Transport Activity.

Human copper transporter 1 (hCtr1) is a transport protein essential for cellular copper uptake and homeostasis, yet excess copper influx is toxic to cells. Currently, no inhibitors of hCtr1 are available to modulate intracellular copper levels. Here, we discovered that the natural phenolic compound rosmarinic acid (RA), as a moderate-strength copper chelator, can directly interact with hCtr1 and inhibit its copper-transport function. RA binds to the N-terminal copper-binding domain of hCtr1, lowering the Cu(II)-binding affinity of hCtr1. More crucially, RA and hCtr1 form a ternary complex with

Solving the puzzle of copper trafficking in Trypanosoma cruzi: candidate genes that can balance uptake and toxicity.

Trypanosoma cruzi, the causative agent of Chagas disease, depends on acquiring nutrients and cofactors, such as copper (Cu), from different hosts. Cu is essential for aerobic organisms, but it can also be toxic, and so its transport and storage must be regulated. In the present study, we characterized the effects of changes in Cu availability on growth behavior, intracellular ion content and oxygen consumption. Our results show that copper is essential for epimastigote proliferation and for the metacyclogenesis process. On the other hand, intracellular amastigotes suffered copper stress during

HDL and Lipid Metabolism.

Mediating reverse cholesterol transport (RCT) is the most classic function of HDL. HDL and HDL-C participate in the entire process of RCT, including cholesterol removal from cells, cholesterol transport in circulation, and cholesterol excretion. As cholesterol is a component of lipid rafts and lipid droplets in cells, HDL and RCT can influence cell activity. HDL has also been shown to be related to the metabolism of some other biological lipids, such as S1P and ox-PL. Here we will introduce in detail the molecular mechanism of HDL participation in RCT and its significance.

Transport Across the Blood-Brain Barrier.

The blood-brain barrier (BBB) is a dynamic barrier essential for maintaining the microenvironment of the brain. Although the special anatomical features of the BBB determine its protective role for the central nervous system (CNS) from blood-borne neurotoxins, the BBB extremely limits the therapeutic efficacy of drugs into the CNS, which greatly hinders the treatment of major brain diseases. In addition to describing the unique structures of the BBB, demonstrating a variety of in vivo and in vitro experimental methods for quantification of the transport properties of the BBB, presenting the ma

Mesoscopic Insights Into Low-Density Lipoprotein Transport: A Dissipative Particle Dynamics Study.

This study employs dissipative particle dynamics (DPD) to investigate low-density lipoprotein (LDL) transport across the endothelium under varying conditions of endothelial leakiness size, LDL concentration, and blood pressure. The endothelial gap size was found to strongly influence LDL transport efficiency: a 32.24 nm gap permitted approximately 1.2 times more LDL particles to cross than a 28.69 nm endothelial leakiness. Moreover, higher LDL concentrations significantly promoted transendothelial movement, with tenfold elevations in LDL levels increasing transport by about 1.6 times compared

Extracellular Vesicles in Hepatobiliary Health and Disease.

Extracellular vesicles (EVs) are membrane-bound nanoparticles released by cells and are an important means of intercellular communication in physiological and pathological states. We provide an overview of recent advances in the understanding of EV biogenesis, cargo selection, recipient cell effects, and key considerations in isolation and characterization techniques. Studies on the physiological role of EVs have relied on cell-based model systems due to technical limitations of studying endogenous nanoparticles in vivo . Several recent studies have elucidated the mechanistic role of EVs in li

Tracheal branching in ants is area-decreasing, violating a central assumption of network transport models.

The structure of tubular transport networks is thought to underlie much of biological regularity, from individuals to ecosystems. A core assumption of transport network models is either area-preserving or area-increasing branching, such that the summed cross-sectional area of all child branches is equal to or greater than the cross-sectional area of their respective parent branch. For insects, the most diverse group of animals, the assumption of area-preserving branching of tracheae is, however, based on measurements of a single individual and an assumption of gas exchange by diffusion. Here w

Colloidal-Bound Polyphosphates and Organic Phosphates Are Bioavailable: A Nutrient Solution Study.

Colloidal forms of Fe(III) minerals can be stabilized in solution by coatings of organic or poly-phosphate (P), which reduce the zeta-potential. This opens up a route toward the development of nanoforms of P fertilizers. However, it is unclear if such P forms are bioavailable. To address this question, spinach (Spinacia oleracea) was grown in nutrient solutions, at equal total P, using three different forms of P (orthophosphate = Pi; hexametaphosphate = HMP; myo-inositol hexaphosphate = IHP), free or bound to goethite/ferrihydrite colloids. After 10 days, P uptake was determined with a dose-re

Computational model for oxygen transport and consumption in human vitreous.

PURPOSE: Previous studies that measured liquefaction and oxygen content in human vitreous suggested that exposure of the lens to excess oxygen causes nuclear cataracts. Here, we developed a computational model that reproduced available experimental oxygen distributions for intact and degraded human vitreous in physiologic and environmentally perturbed conditions. After validation, the model was used to estimate how age-related changes in vitreous physiology and structure alter oxygen levels at the lens. METHODS: A finite-element model for oxygen transport and consumption in the human vitreous
