Biotransformation of sodium nitroprusside into dinitrosyl iron complexes in tissue of ascites tumors of mice.

We have found that treatment of various types of murine ascites tumors with sodium nitroprusside (SNP) both in vitro (1.5 mM) and in vivo (25 mg/kg; i.p.) results in formation of EPR-detectable dinitrosyl nonheme iron complexes (DNIC) with RS- groups of proteins (g perpendicular = 2,037; g parallel = 2,012). The DNIC were mainly localized in the ascitic extracellular fluid. The appearance of DNIC was unaffected by an inhibitor of nitric oxide synthase, N omega-nitro-L-arginine (1 mM), but abolished with a SH-blockator, p-chloromercuribenzoate (0.1 mM). No DNIC formation was observed when ascit

Regioselective biotransformation of midazolam by members of the human cytochrome P450 3A (CYP3A) subfamily.

The capabilities of cytochrome P4503A4 (CYP3A4), CYP3A5, and fetal hepatic microsomes containing CYP3A7 to metabolize midazolam were investigated using human hepatic microsomes and purified CYP3A4 and CYP3A5. Under initial rate conditions and high substrate concentration (400 microM midazolam), variability among eighteen human liver microsomal samples was 30- and 16- fold for 1'- and 4-hydroxylation of midazolam, respectively. Exclusion of two samples isolated from patients previously administered barbiturates reduced the inter-individual variability to 10.5- and 6.0-fold for 1'- and 4-hydroxy

Microcosm and in situ field studies of enhanced biotransformation of trichloroethylene by phenol-utilizing microorganisms.

The ability of different aerobic groundwater microorganisms to cometabolically degrade trichloroethylene (TCE), 1,2-cis-dichloroethylene (c-DCE), and 1,2-trans-dichloroethylene (t-DCE) was evaluated both in groundwater-fed microcosms and in situ in a shallow aquifer. Microcosms amended with phenol or toulene were equally effective in removing c-DCE (> 90%) followed by TCE (60 to 70%), while the microcosm fed methane was most effective in removing t-DCE (> 90%). The microcosm fed ammonia was the least effective. None of the microcosms effectively degraded 1,1,1-trichloroethane. At the Moffett F

Covalent binding of oxidative biotransformation intermediates is associated with halothane hepatotoxicity in guinea pigs.

In vivo covalent binding of halothane biotransformation-reactive intermediates to hepatic protein and lipid was examined in association with the subsequent development of hepatic necrosis in the guinea pig. Oxidative halothane biotransformation was inhibited by the use of deuterated halothane, whereas reductive metabolism was enhanced by low inspired oxygen concentrations. Male outbred Hartley guinea pigs (n = 8) were exposed to either 1% (v/v) halothane or deuterated halothane--with a fractional inspired O2 concentration (FIO2) of 0.40 or 0.10--for 4 h. Livers removed from half of the animals…

Bioconcentration, biotransformation, oxidative stress and neurochemical response of environmental concentration diazepam in yellowbelly pufferfish.

The benzodiazepine diazepam is an emerging environmental contaminant that poses a potential threat to ecosystems and human health. However, there are knowledge gaps regarding tissue-specific accumulation and biotransformation, and the potential toxicity of diazepam in economically important fish at environmental concentrations. This study explored the tissue distribution and accumulation of diazepam and its metabolites in juvenile yellowbelly pufferfish after 35 days of exposure to environmentally relevant concentrations (2, 20, and 100 ng/L). Additionally, changes in oxidative stress markers …

Pyochelin biotransformation by Staphylococcus aureus shapes bacterial competition with Pseudomonas aeruginosa in polymicrobial infections.

Pseudomonas aeruginosa and Staphylococcus aureus are among the most frequently isolated bacterial species from polymicrobial infections of patients with cystic fibrosis and chronic wounds. We apply mass spectrometry guided interaction studies to determine how chemical interaction shapes the fitness and community structure during co-infection of these two pathogens. We demonstrate that S. aureus is equipped with an elegant mechanism to inactivate pyochelin via the yet uncharacterized methyltransferase Spm (staphylococcal pyochelin methyltransferase). Methylation of pyochelin abolishes the sider

Molecular density regulating electron transfer efficiency of S. oneidensis MR-1 mediated roxarsone biotransformation.

Efficient extracellular electron transport is a key for sufficient bioremediation of organoarsenic pollutants such as 4-hydroxy-3-nitrobenzenearsonic acid (roxarsone). The related apparent kinetics characteristics are essential for engineering practice of bioremediation activities and for full understanding the environmental fate of roxarsone, yet remains poorly understood. We report, to our knowledge, the first study of the electron transfer characteristics between roxarsone and participating S. oneidensis MR-1. The electron transfer rate during roxarsone biotransformation was estimated up to…

Biotransformation of selected iodinated X-ray contrast media and characterization of microbial transformation pathways.

Iodinated X-ray contrast media (ICM) are commonly detected in the aquatic environment at concentrations up to the low microgram per liter range. In this study, the biotransformation of selected ICM (diatrizoate, iohexol, iomeprol, and iopamidol) in aerobic soil-water and river sediment-water batch systems was investigated. In addition, microbial transformation pathways were proposed. Diatrizoate, an ionic ICM, was not biotransformed, while three nonionic ICM were transformed into several biotransformation products (TPs) at neutral pH. Iohexol and iomeprol were biotransformed to eleven TPs and …

Enzymatic biotransformation of polystyrene: Interfacial oxidative modification by a serine hydrolase from Pseudomonas aeruginosa.

Polystyrene (PS) is highly resistant to biodegradation, and the enzymatic processes capable of initiating its environmental transformation remain poorly defined. This study investigates the previously underexplored association between a purified serine hydrolase (SH) from Pseudomonas aeruginosa, a bacterium isolated from the gut microbiome of PS-fed Zophobas atratus, and the early-stage oxidative modification of PS at the polymer-water interface. Under aqueous conditions, SH-treated high-molecular-weight PS films exhibited pronounced nanoscale surface alterations and progressive oxygen functio

Production of Anti-melanogenic Glucosides via Molecular Docking-guided Biotransformation of p-hydroxyphenethyl anisate.

p-Hydroxyphenethyl anisate (HP) has been identified as a natural compound with whitening activity in melanoma cells and zebrafish. Considering that glycosylation can enhance the solubility, stability, and bioactivity of natural products, we investigated whether HP could serve as a substrate for glycosylation enzymes. Molecular docking predicted favorable interactions between HP and amylosucrase as well as glycosyltransferases, suggesting the potential formation of α- and β-glucosides. Enzymatic reactions confirmed the production of phenethylanisate-4'-O-α-glucoside (HP-α-glc) and phenethylanis
