Identification, Virulence, and Molecular Characterization of a Recombinant Isolate of Grass Carp Reovirus Genotype I.

The hemorrhagic disease of grass carp (HDGC) caused by grass carp reovirus (GCRV) still poses a great threat to the grass carp industry. Isolation and identification of the GCRV genotype I (GCRV-I) has been rarely reported in the past decade. In this study, a new GCRV was isolated from diseased fish with severe symptoms of enteritis and mild hemorrhages on the body surface. The isolate was further identified by cell culture, transmission electron, indirect immunofluorescence, and SDS-PAGE electrophoretic pattern analysis of genomic RNA. The results were consistent with the new isolate as a GCR

Streptococcus dysgalactiae: A Pathogen of Feral Populations of Silver Carp from a Fish Kill Event.

In August 2018, a series of large fish kills involving only Silver Carp Hypophthalmichthys molitrix occurred on the Mississippi River in northern Louisiana. Clinical signs observed in moribund animals included erratic swimming behavior, such as spiraling and spinning at the surface. A moribund specimen was captured by dip net near the surface at Lake Providence Landing in East Carroll Parish, northern Louisiana, and was submitted for analysis. An aseptic necropsy was performed, and diagnostic procedures, including bacteriology, parasitology, histopathology, virology, and electron microscopy, r

Structure and function analysis of various brain subregions and pituitary in grass carp (Ctenopharyngodon idellus).

It has been generally acknowledged that environment could alter the morphology and functional differentiation of vertebrate brain. However, as the largest group of all vertebrates, studies about the structures and functions of various brain subregions in teleost are still scarce. In this study, using grass carp as a model, histology method and RNA-sequencing were recruited to examine the microstructure and transcript levels among different brain subregions and pituitary. Histological results showed that the grass carp brain was composed of six parts, including olfactory bulb, telencephalon, hy

Nuclear and Cellular Abnormalities of Erythrocytes in Response to Thermal Stress in Common Carp Cyprinus carpio.

As a consequence of global warming, increase of water temperature is likely to alter physiological functions of fish. Hence, we examined the effects of high temperature on blood glucose, hematological parameters [hemoglobin (Hb), red blood cell (RBC), and white blood cell (WBC)], and nuclear and cellular structure of blood cells of common carp (Cyprinus carpio) after exposure to three temperature regimes (27, 31, and 35°C) for 14 days. Fish were sacrificed on 3, 7, and 14 days of exposure. The blood glucose level increased significantly in the fish exposed to 35°C compared to 27 and 31°C. The

Functional characterization of grass carp (Ctenopharyngodon idella) interleukin-2 in head kidney leukocytes.

Interleukin (IL)-2 belongs to the four-helix bundle cytokine family and plays key roles in growth, survival, activation-induced cell death and differentiation of the immune cells. In cyprinid fish, only common carp interleukin-2 (il2) has been cloned because of relatively low sequence homology between carp Il-2 and its homologs in other fish species. In the present study, the coding sequence of grass carp Il-2 (gcIl-2) was cloned and its identity was verified via bioinformatic analysis. Tissue distribution study showed that grass carp il2 (gcil2) mRNA was expressed in thymus, head kidney and g

Evaluation of genotoxicity and hematological effects in common carp (Cyprinus carpio) induced by disinfection by-products.

Disinfection is intended to improve drinking water quality and human health. Although disinfectants may transform organic matter and form disinfection by-products (DBPs), many are branded as cyto- and genotoxic. Traditionally, research focuses on the effects of DBPs on human health, but cytogenic impacts on aquatic organisms still remain ill defined. The current study examines the potential toxic effect of chloroform and iodoform (DBPs) on Cyprinus carpio, selected as a model organism. Fish specimens were exposed to various concentrations of DBPs primarily based on LD50 values, where acute tox

NK3R Mediates the EGF-Induced SLα Secretion and mRNA Expression in Grass Carp Pituitary.

Epidermal growth factor (EGF) is a potent regulator of cell function in many cell types. In mammals, the EGF/EGFR system played an important role in both pituitary physiology and pathology. However, it is not clear about the pituitary action of EGF in lower vertebrates. In this study, using grass carp as a model, we found that EGF could stimulate NK3R mRNA and protein expression through pituitary ErbB1 and ErbB2 coupled to MEK/ERK and PI3K/Akt/mTOR pathways. In addition, EGF could also induce pituitary somatolactin α (SLα) secretion and mRNA expression in a dose- and time-dependent manner in v

Evidence that ghrelin may be associated with the food intake of gibel carp (Carassius auratus gibelio).

Ghrelin, a non-amidated peptide hormone, is a potent anorectic neuropeptide implicated in feeding regulation in mammals and non-mammalian vertebrates. However, the involvement of ghrelin in the feeding behavior of teleosts has not been well understood. To better understand the role of ghrelin in the regulation of appetite in fish, in this study, we cloned the cDNAs encoding ghrelin and investigated their mRNA distributions in gibel carp tissues. We also assessed the effects of different nutritional status on ghrelin mRNA abundance. Ghrelin mRNAs were ubiquitously expressed in ten tissues (inte

Molecular cloning, characterization and expression of cathepsin D from grass carp (Ctenopharyngodon idella).

Cathepsin D is a lysosomal aspartic proteinase which participates in various degradation functions within the cell. In this current study, we cloned and characterized the complete cDNA of grass carp cathepsin D through 5'- and 3'-RACE. The cathepsin D contained a 56 bp 5' terminal untranslated region (5'-UTR), a 1197 bp open reading frame encoding 398 amino acids, and a 394 bp 3'-UTR. Grass carp cathepsin D shared high similarity with those from other species, and showed the highest amino acid identity of 91% to Danio rerio. Unlike many other organisms, the grass carp cathepsin D contains only

The mechanisms of Zn2+ effects on Ca2+-permeable AMPA receptors on carp retinal horizontal cells.

It was demonstrated in our recent study that Zn(2+) had dual effects on Ca(2+)-permeable AMPA receptors on carp retinal horizontal cells. However, the possible mechanism(s) underlying the Zn(2+) effects is still unclear. In the present study, we examined the Zn(2+) effects in the presence of cyclothiazide (CTZ), an allosteric potentiator of AMPA receptors, which could attenuate the receptor desensitization. It was shown that the potentiation effect that low concentration Zn(2+) (10 microM) exerted on the amplitude of the current mediated by Ca(2+)-permeable AMPA receptors was more remarkable i

Expression of genes involved in excitatory neurotransmission in anoxic crucian carp (Carassius carassius) brain.

The crucian carp, Carassius carassius, survives months without oxygen. During anoxia it needs to keep energy expenditure low, particularly in the brain, with its high rate of ATP use related to neuronal activity. This could be accomplished by reducing neuronal excitability through altered expression of genes involved in excitatory neurotransmission. Through cloning and the use of a recently developed real-time RT-PCR approach, with an external RNA control for normalization, we investigated the effect of 1 and 7 days of anoxia (12 degrees C) on the expression of 29 genes, including 8 3-hydroxy-

The roles of alpha- and beta-adrenoceptors in the chronotropic responses to norepinephrine in carp heart (Cyprinus carpio).

1. The chronotropic effect of norepinephrine was studied in isolated spontaneously beating atrial preparations of carp (Cyprinus carpio) heart. 2. Norepinephrine, 0.1 microM, caused a positive chronotropic effect, while at 1 microM it caused either a positive or a negative chronotropic effect. The positive chronotropic effect, observed in 13 preparations, was potentiated by phentolamine and almost completely blocked by propranolol. 3. The negative chronotropic effect observed in the other 5 preparations was greater in the presence of propranolol, reduced by phentolamine and not affected by atr

Unveiling Carbonic Anhydrase VIII, X, XI Expression in Cancer and Neurological Diseases Through Integrated Bioinformatics Approaches.

Carbonic anhydrases are metalloenzymes found both in vertebrates and invertebrates. The CAs catalyze the reversible hydration of CO2 to bicarbonate and H+ ions and play a significant role in respiration, transport of CO2, pH homeostasis, electrolyte secretion, and biosynthetic reactions. The enzymatic activity of CAs is due to the coordination of Zn2+ in the active site by three histidine residues; however, in humans, there are three CAs known as CA-related proteins (CARPs) that are catalytically inactive due to the absence of one or more of the three histidine residues required for the coordi…

A multi-omics investigation reveals the hepatic response to salinity stress in grass carp (Ctenopharyngodon idella).

In the context of increasing freshwater scarcity and the effects of climate change on aquatic environments, developing salt-tolerant fish strains has become a critical strategy for sustainable aquaculture. The limited availability of suitable species for saline-water aquaculture poses a significant challenge, severely impacting the development of the fishery economy. This study investigated the effects of 24 h exposure to freshwater, and seawater at salinities of 4, 9 ppt and 12 ppt, on juvenile grass carp (Ctenopharyngodon idella). Following these exposures, liver tissues were collected to as

Comparative genomics of crucian carp-isolated Flavobacterium psychrophilum: toward the foundations of overwintering mortality syndrome.

Flavobacterium psychrophilum is a Gram-negative bacterium that acts as a primary etiological agent of bacterial cold-water disease (BCWD) in salmonid fish. In the past 5 years, this pathogen has also been isolated from multiple freshwater fish species in China, including crucian carp (Carassius auratus gibelio), grass carp (Ctenopharyngodon idellus) and silver carp (Hypophthalmichthys molitrix), and identified as the causal agent of overwintering mortality syndrome. We report the whole-genome sequencing and analysis of F. psychrophilum strain NJ01, the first isolate from crucian carp, a cyprin

Protective Efficacy of Vaccines Against Grass Carp Hemorrhagic Disease: A Systematic Review and Meta-Analysis.

Grass carp hemorrhagic disease, mainly caused by grass carp reovirus (GCRV), is a major threat to grass carp aquaculture, and vaccination is considered a sustainable strategy for disease prevention and control. However, the protective efficacy of GCRV vaccines and the factors influencing it have not been systematically synthesised. In this study, literature on GCRV vaccines was retrieved from CNKI, ScienceDirect, and Web of Science in accordance with the PRISMA 2020 statement, covering database inception to March 10, 2026, followed by meta-analysis. After screening, 44 studies were included, o

Optimizing Feeding Regimes and Vitamin Delivery Methods in Microdiet for Improving Survival and Growth of Carp Larvae.

In pond carp production, there is growing interest in producing larvae out of season to shorten the production cycle and increase profitability. For this reason, carp fries are produced in indoor recirculation systems, where larvae are fed Artemia salina. Due to the limited ability of fish larvae to digest conventional microdiets-attributable to their short, agastric intestines-live feed remains essential. Therefore, the encapsulation of micronutrients, such as vitamins, may improve delivery efficiency. To address these challenges, three trials were conducted with the following aims: (i) to de
