Apicobasal polarity plays key roles in airway epithelial maturation and regeneration. Once polarized, the airway epithelium functions as a barrier against opportunistic pathogens. Yet, the mechanisms that temporally regulate polarization, a process which is disrupted in cystic fibrosis (CF), remain unclear. Here, we demonstrate the functional importance of connexin 43 (Cx43), a gap junction protein, which is normally repressed during regeneration of the airway epithelium. We show that prolonged post-transcriptional stabilization of Cx43 leads to altered collective cell orientation and compromi

Rapid assessment of tumor proliferation and malignancy is critical for intraoperative margin guidance. However, conventional Hematoxylin and Eosin (H&E)-stained images and fluorescence imaging require extensive sample preparation, limiting real-time applications. Here, we propose a label-free deep ultraviolet polarization photoacoustic imaging technique for identifying mitotic states and assessing tumor aggressiveness through sensitivity to microstructural anisotropy. Following theoretical modeling and system development, cell and tissue experiments demonstrated distinct polarization-dependent

Hemangiosarcoma (HSA) is a malignant tumor derived from endothelial cells. Tumor-associated macrophages are one of the major components of tumor microenvironment and crucial for cancer development. The presence and function of macrophages in HSA have not been studied because there is no syngeneic model for HSA. In this study, we evaluated two mouse HSA cell lines and one immortalized mouse endothelial cell line for their usefulness as syngeneic models for canine HSA. Our results showed that the ISOS-1 cell line developed tumors with similar morphology to canine HSA. ISOS-1 cells highly express

T helper 1 (Th1) immunity is typically viewed as a critical adaptation by vertebrates against intracellular pathogens. Identifying novel targets to enhance Th1 cell differentiation and function is increasingly important for anti-infection immunity. Here, through small-molecule screening focusing on epigenetic modifiers during the in vitro Th1 cell differentiation process, we identified that the selective histone deacetylase 6 (HDAC6) inhibitors ricolinostat and nexturastat A (Nex A) promoted Th1 cell differentiation. HDAC6-depleted mice exhibit elevation of Th1 cell differentiation, and decrea

Efficient motility requires polarized cells, with pseudopods at the front and a retracting rear. Polarization is maintained by restricting the pseudopod catalyst, active Rac, to the front. Here, we show that the actin nucleation-promoting factor Wiskott-Aldrich syndrome protein (WASP) contributes to maintenance of front-rear polarity by controlling localization and cellular levels of active Rac. Dictyostelium cells lacking WASP inappropriately activate Rac at the rear, which affects their polarity and speed. WASP's Cdc42 and Rac interacting binding ("CRIB") motif has been thought to be essenti

Herein, we report a 25-year-old male polar bear suffering from a pancreatic islet cell tumor. The aim of this report is to present a case of this rare tumor in a captive polar bear. The implication of potential risk factors such as high carbohydrate diet or the presence of amyloid fibril deposits was assessed. Necropsy examination revealed several other changes, including nodules observed in the liver, spleen, pancreas, intestine, and thyroid glands that were submitted for histopathologic analysis. Interestingly, the multiple neoplastic nodules were unrelated and included a pancreatic islet ce

Atherosclerosis is a chronic inflammatory disease characterized by T lymphocyte infiltration into the atherosclerotic plaque. Assessments of T cell subtypes have demonstrated a predominance of CD4(+) T helper (Th) cells, implicated Th1 and Th17 immunity in both human and mouse atherogenesis, and provided some evidence suggesting protective roles of Th2 and T regulatory cells. Observations that certain inbred mouse strains have an inherent T helper bias suggest a genetic predisposition toward developing a particular T helper phenotype. This review summarizes our current understanding of mechani

The bacterial pathogen Salmonella penetrates the intestinal epithelium by inducing its own phagocytosis into epithelial cells. The dramatic reorganization of the actin cytoskeleton required for internalization is driven by bacterial manipulation of host signaling pathways, including activation of the Rho family GTPase Rac1 and subsequent activation of the Arp2/3 complex. However, the mechanisms linking these two events remain poorly understood. Rac1 is thought to promote activation of the Arp2/3 complex through its interaction with suppressor of cAMP receptor/WASP family verprolin-homologous (

Two biosynthetic pathways exist for delivery of membrane proteins to the apical surface of epithelial cells, direct transport from the trans-Golgi network (TGN) and transcytosis from the basolateral membrane. Different epithelial cells vary in the expression of these mechanisms. Two extremes are MDCK cells, that use predominantly the direct route and hepatocytes, which deliver all apical proteins via the basolateral membrane. To determine how epithelial cells establish a particular targeting phenotype, we studied the apical delivery of endogenous dipeptidyl peptidase IV (DPPIV) at early and la

Maintaining a healthy cellular microenvironment, characterized by optimal polarity, viscosity, pH, and temperature, is critical for normal physiological function. Disruptions in these parameters are often associated with disease onset, oxidative stress, as well as metabolic disorders. Fluorescent molecular probes capable of simultaneously monitoring multiple environmental factors offer powerful tools for understanding cellular dynamics. In this context, the design of multiresponsive solvatochromic probes is particularly attractive for real-time, non-invasive tracking of microenvironmental chan

The loss of polarity and cell cycle arrest of renal tubular epithelial cells (RTECs) represent important mechanisms of acute kidney injury (AKI). Endoplasmic reticulum stress (ER stress) often occurs in the kidneys during AKI, regulating protein translation and post-translational modification. E-cadherin, a key regulator of cell polarity, undergoes O-glycosylated during ER stress. However, the role of E-cadherin O-glycosylation in kidney injury and repair remains to be elucidated. Cisplatin (CP)/lipopolysaccharide (LPS)-induced AKI mouse models and cell models were established to evaluate ER s…

Studies of the planar cell polarity (PCP) protein complexes Fat1/Fjx1 and Dchs/Fjx1 that form heterotypic interacting bridges of Fat1-Dchs between adjacent cells to confer PCP, as noted in Drosophila, are also found in mammalian cells and tissues as orthologs, such as in Sertoli cells and condensed spermatids in the seminiferous epithelium of the testis. Recent studies have shown that these two interacting PCP protein complexes are also crucial regulators of microtubule and actin dynamics, modulating the polymerization of both microtubules and actin filaments in the testis. In this review, we …

Helicobacter pylori (Hp.) infection is one of the high-risk factors for gastric carcinogenesis (GC). However, the underlying mechanism remains largely unclear. In this study, we uncover an essential role of Hp. infection in mediating tumor suppressor gene silencing in gastric epithelial cells through promoter DNA hypermethylation. Hepatocyte nuclear factor HNF4A was downregulated in GC and predicted poor survival. The in vitro and in vivo assays together confirmed that HNF4A plays a tumor suppressive role in GC. Single-cell analysis showed that HNF4A was selectively expressed in gastric epithe

INTRODUCTION: Tall cell carcinoma of the breast with reverse polarity (TCCRP) is a recently described rare entity with low potential for malignancy which exhibits morphological features with tall cell variant of papillary thyroid carcinoma. Immunohistochemical and molecular studies help establish the diagnosis.
CASE REPORT: We report the case of a 45-year-old woman with no significant medical or surgical history who presented with mastodynia. Physical examination was negative. Screening by bilateral mammography revealed a mass of the right breast which was classified as 4B. The mass was surgi

BACKGROUND: According to previous studies, tall cell carcinoma with reversed polarity can be easily distinguished from ductal carcinoma in situ based on the absence of myoepithelium and a typical histologic feature. However, to the best of our knowledge, no cases of papillary ductal carcinoma exhibiting tall cell and reversed polarity features with intact myoepithelium have been reported, and thus its diagnosis and prognosis remain unclear.
CASE PRESENTATION: A 54-year-old female with a palpable lump in her right breast for 3 years. There were no other symptoms found. A preoperative ultrasoun

During neurogenesis in Xenopus, apicobasally polarised superficial and non-polar deep cells take up different fates: deep cells become primary neurons while superficial cells stay as progenitors. It is not known whether the proteins that affect cell polarity also affect cell fate and how membrane polarity information may be transmitted to the nucleus. Here, we examine the role of the polarity components, apically enriched aPKC and basolateral Lgl2, in primary neurogenesis. We report that a membrane-tethered form of aPKC (aPKC-CAAX) suppresses primary neurogenesis and promotes cell proliferatio…

Leprosy, caused by Mycobacterium leprae, presents a clinical spectrum reflecting distinct host immune responses. Tuberculoid/borderline tuberculoid (TT/BT) patients exhibit Th1-skewed granulomatous immunity with restricted bacillary spread, whereas lepromatous/borderline lepromatous (LL/BL) patients display Th2 dominant responses with disseminated disease and high bacterial load. While Th1/Th2 polarization is well established, the contribution of Natural Killer T (NKT) cells to disease immunopathogenesis is not fully understood. NKT cells, identified by CD3 + CD161+ phenotype and semi-invarian
