Modulation of Extrinsic and Intrinsic Signaling Together with Neuronal Activation Enhances Forelimb Motor Recovery after Cervical Spinal Cord Injury.

Singular strategies for promoting axon regeneration and motor recovery after spinal cord injury (SCI) have been attempted with limited success. For instance, the deletion of RhoA and phosphatase and tensin homolog (Pten) (an extrinsic and intrinsic modulating factor, respectively) in corticospinal neurons (CSNs) promotes axon sprouting after thoracic SCI; however, it is unable to restore motor function. Here, we examine the effects of combining RhoA/Pten deletion in CSNs with chemogenetic neuronal stimulation on axonal growth and motor recovery after SCI in mice. We find that this combinatoria

Sodium Leak Channel in Glutamatergic Neurons of the Lateral Parabrachial Nucleus Helps to Maintain Respiratory Frequency Under Sevoflurane Anesthesia.

The lateral parabrachial nucleus (PBL) is implicated in the regulation of respiratory activity. Sodium leak channel (NALCN) mutations disrupt the respiratory rhythm and influence anesthetic sensitivity in both rodents and humans. Here, we investigated whether the NALCN in PBL glutamatergic neurons maintains respiratory function under general anesthesia. Our results showed that chemogenetic activation of PBL glutamatergic neurons increased the respiratory frequency (RF) in mice; whereas chemogenetic inhibition suppressed RF. NALCN knockdown in PBL glutamatergic neurons but not GABAergic neurons

Circuit mechanism for suppression of frontal cortical ignition during NREM sleep.

Conscious perception is greatly diminished during sleep, but the underlying circuit mechanism is poorly understood. We show that cortical ignition-a brain process shown to be associated with conscious awareness in humans and non-human primates-is strongly suppressed during non-rapid-eye-movement (NREM) sleep in mice due to reduced cholinergic modulation and rapid inhibition of cortical responses. Brain-wide functional ultrasound imaging and cell-type-specific calcium imaging combined with optogenetics showed that activity propagation from visual to frontal cortex is markedly reduced during NRE

Activation of ventral tegmental area dopaminergic neurons ameliorates anxiety-like behaviors in single prolonged stress-induced PTSD model rats.

Post-traumatic stress disorder (PTSD) is a debilitating psychiatric condition that arises after extremely traumatic events, with clinically significant and lasting impacts on both physical and psychological health. The present study examined the role of ventral tegmental area (VTA) dopaminergic signaling in anxiety-like behaviors and the underlying mechanisms in PTSD model rats. Chemogenetic technology was employed to specifically activate VTA dopamine (DA) neurons in rats subjected to single prolonged stress (SPS), and open field and elevated plus maze tests were applied to evaluate the anxie

C1q and immunoglobulins mediate activity-dependent synapse loss in the adult brain.

Complement component 1q (C1q), the initiator of the classical complement cascade, mediates synaptic elimination in development and disease, yet the triggers for its deposition on synapses remain unclear. Using in vivo chemogenetics, we demonstrate that neuronal hyperactivity induces region-specific, C1q-dependent synapse loss in the adult hippocampus. Suppressing perforant pathway hyperactivity in a mouse model of Alzheimer's disease reduced local amyloid-β amounts and C1q deposition and partially rescued synapse loss. Combining spatial transcriptomics, live cell tracking, and super-resolution…

The dentate gyrus in depression.

Extensive preclinical research has been conducted in recent years to reveal the cell types, neuronal circuits and molecular and morphological changes implicated in the function of the dentate gyrus in depression. This was profoundly facilitated by the emergence of methods such as gene targeting, neuronal cell activity manipulation, including optogenetics and chemogenetics, and the development of novel RNA sequencing technology and powerful MRI imagers that were used in clinical studies. These advancements provided researchers with the precise skills needed to evaluate the changes in the dentat…

Complementary lateral hypothalamic populations resist hunger pressure to balance nutritional and social needs.

Animals continuously weigh hunger and thirst against competing needs, such as social contact and mating, according to state and opportunity. Yet neuronal mechanisms of sensing and ranking nutritional needs remain poorly understood. Here, combining calcium imaging in freely behaving mice, optogenetics, and chemogenetics, we show that two neuronal populations of the lateral hypothalamus (LH) guide increasingly hungry animals through behavioral choices between nutritional and social rewards. While increased food consumption was marked by increasing inhibition of a leptin receptor-expressing (LepR…

A Neural Circuit from the Pedunculopontine Tegmental Nucleus to the Central Medial Thalamic Nucleus for the Regulation of Nociceptive and Anxiety-like Behaviors in Male Mice with Chronic Inflammatory Pain.

BACKGROUND: Chronic pain and anxiety exhibit a high degree of comorbidity, in which these conditions can mutually exacerbate each other. The authors investigated whether glutamatergic neurons in the pedunculopontine tegmental nucleus (PPTg Glu ) and their projections to the central medial thalamic nucleus (CM) regulate nociceptive and anxiety-like behaviors in a complete Freund's adjuvant (CFA)-induced chronic pain model. METHODS: The authors used c-Fos staining, chemogenetics, and optogenetics to assess the role of PPTg Glu and the PPTg Glu -CM pathway in regulating nociceptive and anxiety-l…

Relationships between lateral hypothalamic orexin circuits and electroacupuncture-induced mitigation of anxiety in a post-traumatic stress disorder model.

BACKGROUND: Post-traumatic stress disorder (PTSD) is a distressing condition characterized by persistent negative affective states. Electroacupuncture (EA) is a clinically recognized, safe, and efficacious treatment for managing negative emotions associated with PTSD. However, the neural circuits involved in the effects of EA on these emotional disturbances remain unclear. METHODS: A modified single prolonged stress (MSPS) procedure was used to develop a mouse model presenting PTSD-like anxiety-related behaviors (ALBs). Adeno-associated viral tracing showed excitatory synaptic transmission fr

Trigeminal nerve stimulation modulates dopaminergic circuits and neuroinflammation to alleviate depression.

BACKGROUND: Trigeminal nerve stimulation (TNS) is a promising noninvasive neuromodulation therapy for depression; however, its mechanisms remain unclear. OBJECTIVE: This study investigated antidepressant-like effects of TNS and its underlying mechanisms in post-traumatic brain injury depression (PTD). METHODS: The antidepressant efficacy of TNS was evaluated in multiple depression models using standardized behavioral assays. Peripheral mechanisms were probed by chemogenetic activation or inhibition of trigeminal ganglion (TG) neuronal subpopulations in Cre-line mice. Centrally, pathway-selec

Paraventricular thalamus controls consciousness transitions during propofol anaesthesia in mice.

BACKGROUND: The neuronal mechanisms underlying propofol-induced modulation of consciousness are poorly understood. Neuroimaging studies suggest a potential role for non-specific thalamic nuclei in propofol-induced loss of consciousness. We investigated the contribution of the paraventricular thalamus (PVT), a midline thalamic nucleus that has been implicated in arousal control and general anaesthesia with inhaled anaesthetics, to loss and recovery of consciousness during propofol anaesthesia. METHODS: Polysomnographic recordings and righting reflex test were used to determine the transitions

Activation of the VpdmVGLUT1-VPM pathway contributes to anxiety-like behaviors induced by malocclusion.

Occlusal disharmony has a negative impact on emotion. The mesencephalic trigeminal nucleus (Vme) neurons are the primary afferent nuclei that convey proprioceptive information from proprioceptors and low-threshold mechanoreceptors in the periodontal ligament and jaw muscles in the cranio-oro-facial regions. The dorsomedial part of the principal sensory trigeminal nucleus (Vpdm) and the ventral posteromedial nucleus (VPM) of thalamus have been proven to be crucial relay stations in ascending pathway of proprioception. The VPM sends numerous projections to primary somatosensory areas (SI), which

Anterior cingulate cortex glutamatergic projection neurons modulate motion sickness susceptibility via a cortico-vestibular circuit.

Motion sickness (MS) is a prevalent and debilitating vestibular disorder with limited therapeutic options, yet the cortical mechanisms governing individual susceptibility remain elusive. Using a biaxial rotation mouse model, we investigated the role of the anterior cingulate cortex (ACC) in MS by integrating morphology, calcium imaging, electrophysiology, cell ablation, chemogenetics, and optogenetics. Biaxial rotation induced hypothermia, anorexia, taste avoidance, locomotor hypoactivity, and gait incoordination. ACC pyramidal neurons exhibited enhanced activation and increased intrinsic exci…

Genetic identification of mouse trigeminal afferents responsible for mechanical allodynia.

The identities of primary afferents transducing mechanical allodynia following nerve injury remain unclear. We genetically label brushing-activated (BA) trigeminal ganglia (TG) neurons using FosCreER mice with trigeminal nerve injury (TNI). BA TG neurons are largely medium-sized. Many express neurofilament200 and Ntrk3, markers for low-threshold mechanoreceptors, with lower co-localization with nociceptor markers such as Calca or Trpv1. Chemogenetic inhibition of BA TG neurons reduces mechanical allodynia, whereas their chemogenetic activation increases spontaneous face wiping after TNI. Brush

Transcranial direct current stimulation restores addictive behavior via prefrontal-striatal circuit.

Dependence on methamphetamine (METH) is a severe brain disorder characterized by high relapse rates and cognitive decline following detoxification. Recent research suggests that transcranial direct current stimulation (tDCS) may treat addiction, but the underlying neural mechanisms remain unknown. Here, we employed METH-conditioned place preference (CPP) paradigm integrated with fMRI, electrophysiology, chemogenetics, in vivo fiber photometry recordings and a novel rodent tDCS model to examine the neural circuit underlying tDCS modulation on METH-induced addictive behavior. We demonstrated tha…

Mechanosensation of the heart and gut elicits hypometabolism and vigilance in mice.

Interoception broadly refers to awareness of one's internal milieu. Although the importance of the body-to-brain communication that underlies interoception is implicit, the vagal afferent signalling and corresponding brain circuits that shape perception of the viscera are not entirely clear. Here, we use mice to parse neural circuits subserving interoception of the heart and gut. We determine that vagal sensory neurons expressing the oxytocin receptor (Oxtr), referred to as NGOxtr, send projections to cardiovascular or gastrointestinal tissues and exhibit molecular and structural features indi

Spinal lumbar Urocortin 3-expressing neurons are associated with both scratching and Compound 48/80-induced sensations.

Urocortin 3 is a neuropeptide that belongs to the corticotropin-releasing hormone family and is involved in mechanosensation and stress regulation. In this study, we show that Urocortin 3 marks a population of excitatory neurons in the mouse spinal cord, divided into 2 nonoverlapping subpopulations expressing protein kinase C gamma or calretinin/calbindin 2, populations previously associated with mechanosensation. Electrophysiological experiments demonstrated that lumbar spinal Urocortin 3 neurons receive both glycinergic and GABAergic local tonic inhibition, and monosynaptic inputs from both
