Functional expression of heteromeric insect/human nicotinic acetylcholine receptor chimeras without accessory proteins.

Nicotinic acetylcholine receptors (nAChRs) are primary targets of insecticides. While most mammalian nAChRs form functional channels in heterologous systems, insect nAChRs remain notoriously difficult to reconstitute. Here, we co-expressed insect/human subunit chimeras to enable functional analysis of heteromeric insect nAChR ligand‑binding sites in Xenopus laevis oocytes. First, EGFP-tagged human/cockroach α7 chimeras (Hα7/Pα7) were generated by exchanging the signal peptide (SP), extracellular (ECD), transmembrane (TMD), and intracellular (L3-4) domains. While all constructs were translated …

Circular Bivalent Aptamer Chimeras Leveraging LDLR-Mediated Lysosomal Shuttling for Targeted Protein Degradation.

Dysregulation of membrane proteins underlies various human diseases, with their overexpression or mutation frequently being associated with cancer progression. Although targeted protein degradation technologies such as proteolysis-targeting chimeras and lysosome-targeting chimeras represent promising therapeutic strategies, their efficacy is often limited by scarce targeting ligands, complex preparation procedures, potential immunogenicity, and other factors. Here, we present a lysosome-targeting degradation platform based on circular bivalent aptamer chimeras (CBACs), which simultaneously eng…

Systematic Optimization of Proteolysis-Targeting Chimeras for PIN1 Enables Selective Degradation and Antitumor Activity In Vivo.

Background: The peptidyl-prolyl cis-trans isomerase PIN1 regulates multiple oncogenic and tumor-suppressive pathways and is frequently overexpressed in human cancers. Although pharmacological inhibition of PIN1 has shown antitumor potential, existing PIN1-targeting degraders lack systematic structure-activity relationship (SAR) analyses and display inconsistent cellular efficacy, leaving the therapeutic relevance of PIN1 degradation unclear. Methods: Two series of PIN1-targeting PROTACs were designed using the covalent inhibitor sulfopin as the PIN1 binder and ligands for either cereblon (CRBN

PROteolysis TArgeting Chimeras (PROTACs) and beyond: targeted degradation as a new path to fight microbial pathogens.

Targeted degradation is emerging as a new therapeutic approach in the treatment of different diseases. It allows hijacking the cellular pathways deputed to protein or nucleic acid homeostasis to degrade a target macromolecule of interest involved in a pathogenic process. In the last decades, targeted protein degradation has been widely applied for the treatment of cancer or neurodegenerative disorders and some of such therapies are already in clinical use. More recently, therapeutic degraders such as PROTACs, LYTACs, HyTs, BacPROTACs, and others have also been explored in the field of antimicr

A Dual-Targeted Molecule for Disease-Activatable Proteolysis Targeting Chimeras and Targeted Radionuclide Therapy of Cancer.

Proteolysis targeting chimeras (PROTACs) represent a cutting-edge approach for targeted protein degradation in cancer therapy, yet they face challenges such as poor pharmacokinetics and specificity issues, leading to undesirable off-target effects and limited antitumor potency. To address these issues, we introduce dual-targeted unimolecular theranostic probes (e.g., radioactive 177Lu-P-A and its cold counterpart natLu-P-A) for disease-activatable PROTACs in combination with targeted radionuclide therapy (TRT) against prostate cancer with high specificity and effectiveness. The probes achieve …

Targeting METTL3 protein by proteolysis-targeting chimeras: A novel therapeutic approach for acute myeloid leukemia.

Despite numerous studies suggesting that RNA m6A transferase core complex including METTL3 and METTL14 play essential roles in both the initiation and maintenance of acute myeloid leukemia (AML), effective pharmacological targeting of these two proteins remains elusive. Here, we report the development and evaluation of a novel METTL3 degrader, ZW27941, designed to induce METTL3 degradation via the VHL-mediated proteasomal degradation pathway. ZW27941 exhibited potent and selective degradation of METTL3 and its binding partner METTL14, leading to significant anti-leukemic activity in AML cell l

Botulinum Neurotoxin Chimeras Suppress Stimulation by Capsaicin of Rat Trigeminal Sensory Neurons In Vivo and In Vitro.

Chimeras of botulinum neurotoxin (BoNT) serotype A (/A) combined with /E protease might possess improved analgesic properties relative to either parent, due to inheriting the sensory neurotropism of the former with more extensive disabling of SNAP-25 from the latter. Hence, fusions of /E protease light chain (LC) to whole BoNT/A (LC/E-BoNT/A), and of the LC plus translocation domain (HN) of /E with the neuronal acceptor binding moiety (HC) of /A (BoNT/EA), created previously by gene recombination and expression in E. coli., were used. LC/E-BoNT/A (75 units/kg) injected into the whisker pad of …

Characterization of hepatitis C virus intra- and intergenotypic chimeras reveals a role of the glycoproteins in virus envelopment.

Hepatitis C virus (HCV) is highly variable and associated with chronic liver disease. Viral isolates are grouped into seven genotypes (GTs). Accumulating evidence indicates that viral determinants in the core to NS2 proteins modulate the efficiency of virus production. However, the role of the glycoproteins E1 and E2 in this process is currently poorly defined. Therefore, we constructed chimeric viral genomes to explore the role of E1 and E2 in HCV assembly. Comparison of the kinetics and efficiency of particle production by intragenotypic chimeras highlighted core and p7 as crucial determinan…

Purification and characterization of Yersinia enterocolitica and Yersinia pestis LcrV-cholera toxin A(2)/B chimeras.

Yersinia pestis is a virulent human pathogen and potential biological weapon. Despite a long history of research on this organism, there is no licensed vaccine to protect against pneumonic forms of Y. pestis disease. In the present study, plasmids were constructed to express cholera toxin A(2)/B chimeric molecules containing the LcrV protective antigen from Yersinia enterocolitica and Y. pestis. These chimeras were expressed and purified to high yields from the supernatant of transformed Escherichia coli. Western and GM(1) ELISA assays were used to characterize the composition, receptor-bindin…

Production of germ-line chimeras in zebrafish by cell transplants from genetically pigmented to albino embryos.

To determine whether embryonic cells transplanted from one zebrafish embryo to another can contribute to the germ line of the recipient, and to determine whether pigmentation can be used as a dominant visible marker to monitor cell transplants, we introduced cells from genetically pigmented (donor) embryos to albino recipients at midblastula stage. By 48 hr many of the resulting chimeras expressed dark pigment in their eyes and bodies, characteristics of donor but not albino embryos. By 4-6 weeks of age pigmentation was observed on the body of 23 of 70 chimeras. In contrast to fully pigmented …

The development of handed asymmetry in aggregation chimeras of situs inversus mutant and wild-type mouse embryo.

Mutant iv/iv mice develop as if they have no sense of left and right, so the development of asymmetry is random: half normal, half as a mirror-image of normal, situs inversus. We have made aggregation chimeras of 8-cell stage iv/iv and +/+ embryos, transferred them into pseudopregnant mice, and examined their phenotype on day 10 of gestation. The contribution of mutant and wild-type cells to tissues of the embryo was estimated by strain-specific isozyme (GPI-1) analysis. We have also performed reciprocal embryo transfers, iv/iv blastocysts into +/+ mice, and vice versa. These transfers show th…

Development of High-Efficiency Mitochondrial Protease Targeting Chimeras (MtPTACs) for Targeted Degradation of POLRMT and Beyond.

Despite the broad success of targeted protein degradation (TPD), achieving efficient degradation within mitochondria remains a challenge. Herein, we report a highly active degrader targeting human mitochondrial RNA polymerase (POLRMT) based on the Mitochondrial Protease Targeting Chimeras (MtPTACs) strategy. The lead compound, MTP-B1, is a highly potent MtPTAC to achieve essentially complete target degradation (D max > 98%) within 24 h at sub-micromolar concentrations. Mechanistically, MTP-B1 circumvents the compensatory transcriptional upregulation induced by POLRMT inhibitors and sustains do…

In Situ Transformable Photothermal Targeting Chimeras for Spatiotemporally Controllable Protein Degradation and Combination Immunotherapy for Cancer.

Targeted protein degradation (TPD) technology has been extensively exploited for cancer therapy. However, current TPD approaches suffer from two major limitations that the lack of tissue and subcellular specificity for precise localization, and insufficient postdegradation benefits due to their reliance on single traditional pathway regulation. To achieve tumor-specific degradation of immune-related membrane and intracellular proteins of interest, we rationally engineered a set of photothermal-targeting chimeras (PTTACs) by integrating a photothermal agent and a protein ligand. The PTTACs disp…

Mapping Determinants of Hepatitis C Virus E1/E2 Transmembrane Interactions Using Intergenotypic Chimeras.

Hepatitis C virus (HCV) infection remains a major global health burden, and no vaccine preventing chronic infection is available. The envelope glycoproteins, E1 and E2, form a complex essential for viral entry; however, the mechanisms governing E1/E2 assembly and stability remain incompletely defined. Here, we investigated the role of the E1/E2 transmembrane (TM) regions in HCV infectivity using chimeras of JFH1-based recombinants with isolate-specific Core-NS2 sequences in which the C-terminal TM domains of E1 (TME1), E2 (TME2), or both (TME1E2) from the H77 isolate (genotype 1a) replaced tho…

Development of glucagon-like Peptide-1 lysosomal targeting chimeras for degradation of extracellular and membrane proteins.

Lysosome-targeting chimeras (LYTAC) represent a revolutionary technology for targeted protein degradation. However, the scarcity of highly efficient lysosomal transport receptors poses a major bottleneck to LYTAC advancement. In previous studies, we identified the glucagon-like peptide-1 receptor (GLP-1R) as a novel lysosomal transport receptor that promotes the degradation of membrane proteins. Here, leveraging the stability of semaglutide, we developed GLP-1R-mediated lysosomal-targeting chimeras (g-LYTAC), including antibody-peptide conjugate-based chimeras (APCTACs) targeting epidermal gro…

Chimeras co-targeting antigens and FcγRIIb trigger degradation of extracellular soluble proteins and pathological aggregates.

While the clinical utility of conventional antibody therapies is undeniable, their therapeutic potential is often constrained high antigen loads and the recycling of antibody-antigen complexes via neonatal Fc receptor (FcRn). Here, we present a platform, based on a design similar to bispecific antibodies, FcγRIIb-Targeting Chimeras (FcRTAC). These constructs recognise antigens with one arm and bind FcγRIIb with the other arm to harness the unique endocytic properties of FcγRIIb to direct the recognized pathogenic antigens to lysosomes for irreversible degradation. The FcRTAC platform demonstra…

An NTCP-mediated liver-targeting chimeras (NTLiverTac) design strategy and its application to hepatoblastoma models.

Hepatoblastoma (HB) has attracted much attention due to its high recurrence rate, and its first- and second-line chemotherapeutic agents have severe neurotoxicity, cardiotoxicity, and hepatorenal dysfunction. To enhance the liver-targeting efficiency, we proposed an NTCP-mediated liver-targeting chimeras (NTLiverTac) design strategy, which utilized the PROTAC molecule to couple with the Na+-taurocholate co-transporting polypeptide (NTCP) ligand to enhance cell selectivity through NTCP-mediated endocytosis. As a proof-of-concept, we selected cholic acid as its ligand, conjugated it with sorafen…

In vitro adaptation and characterization of attenuated hypervariable region 1 swap chimeras of hepatitis C virus.

Hepatitis C virus (HCV) chronically infects 70 million people worldwide with an estimated annual disease-related mortality of 400,000. A vaccine could prevent spread of this pervasive human pathogen, but has proven difficult to develop, partly due to neutralizing antibody evasion mechanisms that are inherent features of the virus envelope glycoproteins, E1 and E2. A central actor is the E2 motif, hypervariable region 1 (HVR1), which protects several non-overlapping neutralization epitopes through an incompletely understood mechanism. Here, we show that introducing different HVR1-isolate sequen
