In vivo assessing flavonols in white grape berries (Vitis vinifera L. cv. Pinot Blanc) of different degrees of ripeness using chlorophyll fluorescence imaging.

Exposed and non-exposed halves of field-grown berries of the white grapevine Vitis vinifera L. cv. Pinot Blanc at various stages of ripeness were analysed using chlorophyll fluorescence imaging. The stage of ripeness was classified by the total sugar concentration which ranged between 120 and 300 g L-1 for the different berries but was similar in the exposed and the non-exposed half of individual berries. Fluorescence was excited in the UV-A and the blue spectral region and detected at red as well as far-red wavelengths. At both emission ranges, UV-excited fluorescence was weak and required co

Advancing real-time coastal data monitoring: Bio-optical property analysis (chlorophyll-a and TSM) in the Northern Bay of Bengal using Sentinel-3 OLCI, IRS Oceansat-3, and artificial neural networks.

Monitoring bio-optical attributes in estuarine environments is crucial for understanding ecosystem productivity, sediment dynamics, and climate-induced hydrological changes. This research introduces an integrated multi-sensor and machine learning approach to assess the spatiotemporal variability of Chl-a and TSM within the Northern Bay of Bengal. In-situ data gathered during pre-monsoon, monsoon, and post-monsoon seasons were integrated with satellite observations from Sentinel-3 OLCI and Oceansat-3 OCM to investigate seasonal trends and to train ANN models for enhanced parameter retrieval. The performance of directional area scattering factor proxies for correcting canopy structural and soil effects on leaf chlorophyll content estimation varies with canopy conditions in agricultural systems.

Accurate leaf chlorophyll content (LCC) estimation from canopy reflectance is essential for agricultural monitoring but remains challenging due to confounding effects of canopy structure and soil background. While spectral invariants theory, implemented through directional area scattering factor (DASF) proxies, offers a framework to decouple these effects from canopy bidirectional reflectance factor (BRF), their comparative performance across diverse canopy conditions remains unexamined. In this study, we comprehensively evaluated four DASF proxies, practical DASF (pDASF), improved DASF (iDASF

Up-regulation of nitrogen metabolism and chlorophyll biosynthesis by hydrogen sulfide improved photosystem photochemistry and gas exchange in chromium-contaminated bean (Phaseolus vulgaris L.) plants.

Hydrogen sulfide (H₂S) is considered as plant growth promoter under heavy metal stress, though its specific effects on photosynthesis are rarely explored. This study investigates the protective effects of exogenous H2S donor sodium hydrosulfide (NaHS) on chlorophyll metabolism and photosystem II (PSII) function in 24-day-old bean plants exposed to 10 μM chromium (Cr) stress. Sodium hydrosulfide (100 μM) reduced Cr accumulation in both roots and leaves, leading to restored plant growth. Concomitantly, H₂S mitigated Cr-induced oxidative damages by decreasing reactive oxygen species levels and fu

Ethanolic guava leaf extracts with different chlorophyll removal processes: Anti-melanosis, antibacterial properties and the impact on qualities of Pacific white shrimp during refrigerated storage.

Ethanolic guava leaf extract (EGLE) without chlorophyll removal (GLE-C) and those with chlorophyll removal using sedimentation process (GLE-S) or dechlorophyllization using chloroform (GLE-Ch) were prepared. Antibacterial and anti-melanosis properties of all extracts were examined. All extracts showed promising antibacterial properties, polyphenoloxidase inhibitory activity and copper chelating activity. These activities were highest in GLE-S (P < 0.05). Piceatannol 4'-galloylglucoside, epicatechin, 8-hydroxyluteolin 8-sulfate, quercetin 3-(2''-galloyl-alpha-l-arabinopyranoside), and aclurin 3

Ectopic expression of GA 2-oxidase 6 from rapeseed (Brassica napus L.) causes dwarfism, late flowering and enhanced chlorophyll accumulation in Arabidopsis thaliana.

Gibberellins (GAs) are endogenous hormones that play an important role in higher plant growth and development. GA2-oxidase (GA2ox) promotes … effect on seed development. Additionally, BnGA2ox6 overexpression enhanced chlorophyll b and total chlorophyll accumulation, and downregulated mRNA expression levels of the CHL1 and RCCR genes, which are involved in the chlorophyll degradation. These findings suggest that BnGA2ox6 regulates plant hight, silique development, flowering and chlorophyll accumulati …

Chlorophylls and β-carotene in Spirulina platensis Dietary Supplements: Nano Liquid Chromatography Versus Diffuse Reflectance Spectroscopy for Establishing Composition, Quality Control, and Comparison of Commercial Products.

Spirulina is considered a superfood due to its chlorophylls. Two new methods for the determination of chlorophylls and β-carotene were developed here, one based on in-tube solid-phase microextraction (IT-SPME) coupled online to nanoliquid chromatography (nanoLC) with diode array detection (DAD), and the other on ultraviolet-visible diffuse reflectance spectroscopy (UV-vis DRS). A protocol to extract the pigments from spirulina was proposed using ethanol (1.5 mL). The aim is to provide fast and environmentally friendly techniques for both the extraction and measurement of pigments, generating l

Participation of the inositol phospholipid signaling pathway in the increase in cytosolic calcium induced by tributyltin chloride intoxication of chlorophyllous protozoa Euglena gracilis Z and its achlorophyllous mutant SM-ZK.

Exposure of tri-n-butyl tin chloride (TBTCl) as a stressor to Euglena gracilis Z causes rapid alteration of cell morphology followed by deflagellation. The present study was undertaken to reveal the mechanism of the cell response at a molecular level. Chlamydomonas reinhardtii, in this study E. gracilis Z and its achlorophyllous mutant SM-ZK, gave similar results when subject to the same stressor. Indeed, similar results were obtained with both strains. Next, assuming that the morphological alteration caused by TBTCl is mediated by the inositide phosphate-lipid signaling pathway, the effects o

Effects of 4-month Fe deficiency exposure on Fe reduction mechanism, photosynthetic gas exchange, chlorophyll fluorescence and antioxidant defense in two peach rootstocks differing in Fe deficiency tolerance.

Fe deficiency was imposed by omission of Fe (-Fe), or by inclusion of bicarbonate (supplied as 20 mM NaHCO3) in the nutrient solution in two contrasting peach rootstocks (GF-677; tolerant to Fe deficiency and Cadaman; sensitive to Fe deficiency) for 4 months. In the Fe-deprived leaves and roots, and especially in those treated with bicarbonate, a decrease in Fe concentrations was recorded. Omission of Fe resulted in an increase of the activity of root Fe(III)-chelate reductase (FCR) in both rootstocks, whereas FCR activity decreased in the bicarbonate-treated roots of Cadaman. The results obta

Self-aggregation of synthetic zinc chlorophyll derivatives possessing 3(1)-hydroxy or methoxy group and 13(1)-mono- or dicyanomethylene moiety in nonpolar organic solvents as models of chlorosomal bacteriochlorophyll-d aggregates.

Methyl 13(1)-(di)cyanomethylene-pyropheophorbides were synthesized by Knoevenagel reactions of the corresponding 13(1)-oxo-chlorins prepared from modifying chlorophyll-a with malononitrile or cyanoacetic acid. Alternatively, methyl 13(1)-cyanomethylene-pyropheophorbides were produced by Wittig reactions of 13(1)-oxo-chlorins with Ph3P=CHCN. Self-aggregation of zinc complexes of the semi-synthetic chlorophyll derivatives possessing a hydroxy or methoxy group at the 3(1)-position was examined in 1%(v/v) tetrahydrofuran or dichloromethane and hexane by electronic absorption and circular dichroism

SOME REMARKS ON THE PHOTOSYNTHESIS OF GREEN PLANTS.

1. It is suggested that in the assimilation process of green plants the reduction of the CO(2) takes place with the help of Fe(++) ions (present in the chloroplast) under the influence of light, which is absorbed by a sensitizing chlorophyll-CO(2)-complex. 2. It seems that the chlorophyll has to fulfill two different functions depending on its situation in the chloroplast. The chlorophyll molecules on the surface of the lipoid phase (in contact with an aqueous phase containing Fe(++)) combine with CO(2) to form a light absorbing chlorophyll-CO(2)-complex and in this way take part in the reduct

Satellite Remote Sensing of Space-Time Plankton Variability in the Bay of Bengal: Connections to Cholera Outbreaks.

Cholera bacteria exhibit strong association with coastal plankton. Characterization of space-time variability of chlorophyll, a surrogate for plankton abundance, in Northern Bay of Bengal is an essential first step to develop any methodology for predicting cholera outbreaks in the Bengal Delta region using remote sensing. This study quantifies the space-time distribution of chlorophyll, using data from SeaWiFS, in the Bay of Bengal region using ten years of satellite data. Variability of chlorophyll at daily scale, irrespective of spatial averaging, resembles white noise. At a monthly scale, c…

Physiological and transcriptomic responses to magnesium deficiency in Neolamarckia Cadamba.

Magnesium (Mg2+) is a critical component of chlorophyll and enzymes involved in various physiological and biochemical processes essential for plant growth, biomass accumulation, and photosynthesis. Mg2+ deficiency (MgD) is common in hot and rainy subtropical areas due to its easy loss from soil. Neolamarckia cadamba, an important tropical tree in South Asia, faces severe effects of MgD, however, the responses of N. cadamba to MgD stress remain unclear. In here, effects of N. cadamba under MgD stress were investigated. The study revealed that MgD had lower plant biomass, fresh and dry weight, r…

Impact of Drought Stress on Photosynthetic Electron Transport in Rice Seedlings.

To investigate photosynthetic physiological response mechanisms of rice (Oryza sativa L.) seedling leaves under drought stress, a pot experiment with controlled watering was conducted. Four treatments were applied-control (CK), mild drought (LD), moderate drought (MD), and severe drought (SD)-to four cultivars (H17B7, HY3015, LH1H, WNSM) at seedling stage. The results showed that, after drought stress, the morphological indicators and chlorophyll content of the four rice cultivars all showed decreased trend, but a compensatory effect appeared in some varieties under the LD treatment. As drough

CabHLH18-like Gene Promotes Leaf Yellowing and Directly Activates CaCLH1 in Pepper.

Yellow leaf phenotypes represent an important trait in pepper and enrich genetic germplasm resources. Previous studies have primarily examined leaf yellowing caused by defects in chlorophyll biosynthesis, whereas the functions of chlorophyll degradation-related genes remain less well characterized. In this study, a yellow leaf phenotype (NY) was identified. Transmission electron microscopy and RNA-seq showed that differentially expressed genes were concentrated in photosynthesis- and chlorophyll-related pathways. CaCLH1 was then demonstrated to promote chlorophyll degradation. Yeast one-hybrid

Stable albinism induced without mutagenesis: a model for ribosome-free plastid inheritance.

Maternally inherited chlorophyll deficiency, or albinism, is a standard marker in plant cytoplasmic genetics. Its stability is consistent with mutations in the plastid genome. Nuclear mutations inducing plastid ribosome deficiency (PRD) also lead to maternally inherited chlorophyll deficiency. Here we report that stable chlorophyll deficiency can be efficiently generated in cruciferous plants without mutagenesis by a short exposure to spectinomycin, an inhibitor of plastid protein synthesis. We show that the chlorophyll-deficient phenotype results from a deficiency in plastid ribosomes and pla

Expression of BoNOL and BoHCAR genes during postharvest senescence of broccoli heads.

BACKGROUND: Chlorophyll is the most abundant pigment on Earth, essential for the capture of light energy during photosynthesis. During senescence, chlorophyll degradation is highly regulated in order to diminish toxicity of the free chlorophyll molecule due to its photoactivity. The first step in the chlorophyll degradation pathway is the conversion of chlorophyll b to chlorophyll a by means of two consecutive reactions catalyzed by enzymes coded by NYC1 (NON-YELLOW COLORING 1), NOL (NYC1-LIKE) and HCAR. RESULTS: In this work, we studied the expression of NOL and HCAR genes during postharvest…

LCAA, a novel factor required for magnesium protoporphyrin monomethylester cyclase accumulation and feedback control of aminolevulinic acid biosynthesis in tobacco.

Low Chlorophyll Accumulation A (LCAA) antisense plants were obtained from a screen for genes whose partial down-regulation results in a strong chlorophyll deficiency in tobacco (Nicotiana tabacum). The LCAA mutants are affected in a plastid-localized protein of unknown function, which is conserved in cyanobacteria and all photosynthetic eukaryotes. They suffer from drastically reduced light-harvesting complex (LHC) contents, while the accumulation of all other photosynthetic complexes per leaf area is less affected. As the disturbed accumulation of LHC proteins could be either attributable to …

Comparative transcriptome analysis of differentially expressed genes related to the physiological changes of yellow-green leaf mutant of maize.

Chlorophylls, green pigments in chloroplasts, are essential for photosynthesis. Reduction in chlorophyll content may result in retarded growth, dwarfism, and sterility. In this study, a yellow-green leaf mutant of maize, indicative of abnormity in chlorophyll content, was identified. The physiological parameters of this mutant were measured. Next, global gene expression of this mutant was determined using transcriptome analysis and compared to that of wild-type maize plants. The yellow-green leaf mutant of maize was found to contain lower contents of chlorophyll a, chlorophyll b and carotenoid…

Floodplain inundation and lateral connectivity promote productivity in a managed river ecosystem.

River-floodplain ecosystems near urban centers are heavily engineered for flood protection and water delivery, which has led to a loss of lateral hydrologic connectivity between rivers and their floodplains. This study has two objectives: (1) Does increased lateral connectivity resulting from floodplain inundation increase chlorophyll a biomass? (2) Does that bump in chlorophyll a get transported downstream? The San Francisco Estuary in California, USA, has a robust and long-term monitoring network for water quality. We integrated water temperature, chlorophyll a, flow, and floodplain inundati…
