Phenotypic Changes in Macrophage Activation in a Model of Nonalcoholic Fatty Liver Disease using Microminipigs.

AIM: Non-alcoholic fatty liver disease (NAFLD) is one of the most common chronic liver disorders associated with metabolic syndrome, and its prevalence has been on the rise. The pathogenesis of NAFLD has not yet been sufficiently elucidated due to the multifactorial nature of the disease, although the activation of macrophages/Kupffer cells is considered to be involved. We previously reported an animal model of NAFLD using MicrominipigsTM (µMPs) fed high-fat diets containing cholesterol with or without cholic acid. The aim of this study was to investigate the phenotypic changes of macrophages

Influence of fat-rich versus carbohydrate-rich diets on bile acid kinetics, biliary lipids, and net steroid balance in hyperlipidemic subjects.

Altogether, 14 patients with hyperlipoproteinemia type IIA (n = 3), IIb (n = 4), or IV (n = 7) were maintained on diets in which 60% of the energy was supplied as fat or carbohydrates. The switch from the fat-rich to the carbohydrate-rich diet resulted in elevation of the plasma triglyceride levels. Although not consistent in all instances, the change of diet also resulted in an enhanced formation of both cholic acid and chenodeoxycholic acid and a higher contribution of deoxycholic acid in duodenal bile. Due to a reciprocal decrease in the excretion of neutral steroids in feces, the mean ster

Fasting serum bile acid levels in the primary hyperlipoproteinemias.

A newly developed radioimmunoassay specific for conjugates of cholic acid (CCA) was used to measure the fasting serum levels of these bile acids in 233 patients with primary hyperlipoproteinemia classified as type IIa, IIb, III, IV, or V hyperlipoproteinemia as well as in 80 healthy control subjects. Subjects with type IIa hyperlipoproteinemia had significantly lower levels of CCA (0.9 plus or minus 0.06 muM, mean plus or minus SE) than did healthy controls (0.50 plus or minus 0.08 muM). Patients with type IIb, III, IV, and V hyperlipoproteinemia had values similar to those of the control grou

Ursodeoxycholic acid corrects defective natural killer activity by inhibiting prostaglandin E2 production in primary biliary cirrhosis.

We evaluated the effect of ursodeoxycholic acid on the defective natural killer activity in primary biliary cirrhosis. Administration of ursodeoxycholic acid (600 mg daily) for one month significantly increased natural killer activity in patients with primary biliary cirrhosis (P < 0.05). Ursodeoxycholic acid also enhanced the in vitro natural killer activity of lymphocytes from healthy volunteers, while other hydrophobic bile acids depressed it. Furthermore, ursodeoxycholic acid reduced the prostaglandin E2 concentration in culture supernatants of lymphocytes from healthy volunteers to a lowe

Clinical grading of intrahepatic cholestasis pregnancy.

Intrahepatic cholestasis of pregnancy (ICP) is the pregnancy induced liver disorder causing intense itching of palm, sole or even whole body especially in the evening at late second and third trimester. This disease is categorized into mild and severe ICP according to raised level of LFT including serum level of bile acid and cholic acid. ICP has less morbidity to mother but significant risk to fetus in uterus. The predisposing element to cause intense pruritus is because of high amount of bile acid in maternal serum. Toxic bile acids affect fetal cardiomyocytes retained in fetus body causing

[Bile acids in liver diseases--current indications].

Ursodeoxycholic acid (UDCA) has beneficial effects in cholestatic diseases such as primary biliary cirrhosis, primary sclerosing cholangitis and cholestasis of pregnancy. In chronic hepatitis and alcoholic hepatitis, an effect of UDCA is uncertain. After organ transplantation (liver, bone, heart), favorable effects of UDCA which await confirmation were observed. UDCA very likely has a beneficial effect in children with Byler's and Alagille's syndrome, extrahepatic biliary atresia after the Kasai procedure and in cholestasis of cystic fibrosis. Unclear is the effect of UDCA in benign intermitte

Pylorus and antroseromuscular flap-preserving gastrectomy--a new type of reconstruction after subtotal gastrectomy for treatment of gastroduodenal ulcer: clinical and experimental study.

To avoid motility disturbances after Billroth gastrectomy, the authors designed pylorus and antroseromuscular flap-preserving subtotal gastrectomy (PAFPG). Results showed that gastric motility and emptying time of dogs after PAFPG were close to normal. PAFPG was applied to 125 consecutive patients with gastroduodenal ulcers (gastric ulcer, 15 patients; duodenal ulcer, 94 patients; and combined ulcers, 16 patients) confirmed by barium examination and fibro-gastroendoscopy. All patients recovered smoothly, none of them had postoperative complication. Gastric acid output reduction rates were as f

Fatty acid profiles in various lipid fractions in the female epidermis. Does the body site and age matter?

BACKGROUND: The epidermis forms the barrier between an organism and its external environment. Although one of the major functional elements of the epidermis is the lipid-enriched extracellular matrix, containing mainly ceramides, cholesterol (CHOL) and free fatty acids, the data are limited regarding the lipid profile in the epidermis. The aim of the study was to determine the whole profile of fatty acids (FAs) in the epidermis and to examine any dependence according to the age of the subject and the site on the epidermis. MATERIALS AND METHODS: Epidermis extracts obtained from 10 adults and

Steroid pattern of bile and feces in response to a fruit-enriched diet in hypercholesterolemic hamsters.

The effect of a fruit (apple)-enriched diet on bile secretion and on fecal steroid excretion was studied in two strains of hamster: normal hamsters with normal cholesterolemia and spontaneous hypercholesterolemic hamsters with high-level hepatic cholesterol esters (FEC hamsters). Quantitative and qualitative alterations in the steroid composition in bile and feces were accompanied by changes in intestinal morphology. The fruit diet displayed a choleretic effect and increased the output of bile acids from liver in FEC hamsters. In addition, bile collected continuously from cannulated apple-fed

Characterization of bumetanide transport in isolated skate hepatocytes.

The uptake of [3H]bumetanide was studied in isolated skate hepatocytes in an albumin-free elasmobranch Ringer solution and compared with the uptake of bile acids in the presence of other cholephilic organic anions. [3H]bumetanide uptake was energy dependent, temperature sensitive, and exhibited saturation kinetics. In contrast to taurocholate and cholate, which are transported only by Na(+)-independent mechanisms, removal of Na+ reduced the maximal uptake rate (Vmax) for bumetanide from 404 +/- 80 to 230 +/- 47 pmol.mg-1 x min-1 without a change in the apparent Michaelis constant (Km). Carboxylic submetabolome-driven signature characterization of COVID-19 asymptomatic infection.

Asymptomatic infection of COVID-19 is a global threat for public health. Unfortunately, the study about metabolic dysregulation of asymptomatic infection is barely investigated. Here, we performed carboxylic submetabolome profiling of serum from 62 asymptomatic and 122 control individuals, by a highly sensitive chemical isotope labelling method. Twenty-one discriminative carboxylic features, including 12-hydroxyeicosatetraenoic acid, cholic acid, glycoursodeoxycholic acid and 15,16-dihydroxyoctadeca-9,12-dienoic acid were discovered to be dysregulated in asymptomatic patients. This panel conta

Serum glycine-conjugated bile acids in pediatric hepatobiliary disorders.

Measurements of serum bile acids (glycine conjugates of cholic, chenodeoxycholic, deoxycholic, and lithocholic acids) by radioimmunoassay in a variety of pediatric hepatobiliary disorders showed elevations in neonatal hepatitis syndromes, cholestasis, and hepatitis of extrahepatic or intrahepatic origin. Measurements of individual serum bile acids failed to differentiate between the various neonatal hepatitis syndromes. In one patient with cholestasis, the increased levels of bile acids observed returned to normal following therapy with cholestyramine and phenobarbital. In chronic active hepat

Lactobacillus gasseri LG-G12 Alleviates High-Fat Diet-induced Obesity through Gut Microbiota-Bile Acid Axis.

The role of probiotics in weight management remains controversial. In this study, we found that Lactobacillus gasseri LG-G12 exhibits gut colonization capacity and metabolic regulation potential in vitro. In vivo, LG-G12 alleviated high-fat diet-induced weight gain and visceral fat accumulation in mice. Fecal targeted metabolomics identified bile acid metabolism as a key pathway. Furthermore, LG-G12 might exhibit bile salt hydrolase and hydroxysteroid dehydrogenase activities, contributing to a bile acid profile characterized by low cholic acid and high ursodeoxycholic acid levels. Taurocholic acid metabolism by gut microbes and colon cancer.

Colorectal cancer (CRC) is one of the most frequent causes of cancer death worldwide and is associated with adoption of a diet high in animal protein and saturated fat. Saturated fat induces increased bile secretion into the intestine. Increased bile secretion selects for populations of gut microbes capable of altering the bile acid pool, generating tumor-promoting secondary bile acids such as deoxycholic acid and lithocholic acid. Epidemiological evidence suggests CRC is associated with increased levels of DCA in serum, bile, and stool. Mechanisms by which secondary bile acids promote CRC are

[The liver-protective properties of the pediatric drug form of propolis in animals of different age groups].

The pharmacological activity of a pediatric formulation of the phenolic hydrophobic drug propolis was studied in the experiments on albino rats of various age with toxic liver damages of various duration and in acute hepatic ischemia. In all models of hepatic abnormalities, the drug was found to show antioxidative properties which were moderate (30-60%). In addition, there were improvements in hepatic secretion of bile, cholic acids, and cholesterol. On the other hand, the membrane-stabilizing effect of the drug was exerted in not all the tested models of hepatic damage.

Endogenously modified multi-bioresponsive silk fibroin nanoparticles enhancing the cellular delivery and anti-inflammatory activity of curcumin.

Hydrophobic bioactives such as curcumin suffer from poor aqueous solubility, low bioaccessibility, and targeting capability. In this study, silk fibroin, a natural macromolecular protein, was used to form self-assembled nanoparticles. These nanoparticles were surface-grafted with bile acids to modulate interfacial properties and exploit endogenous bile acid transport pathways for ileum-targeted curcumin delivery. Cholic acid, deoxycholic acid, and glycocholic acid were covalently grafted onto silk fibroin nanoparticles to tailor surface hydrophobicity and charge. Spectroscopic and physicochemi

LC/MS-Based Global Metabolomic Identification of Serum Biomarkers Differentiating Hepatocellular Carcinoma from Chronic Hepatitis B and Liver Cirrhosis.

Chronic hepatitis B virus (CHB) infection is one of the primary risk factors associated with the development of hepatocellular carcinoma (HCC). Despite having been extensively studied, diagnosing early-stage HCC remains challenging, and diagnosed patients have a poor (3-5%) survival rate. Identifying new approaches to detect changes in the serum metabolic profiles of patients with CHB and liver cirrhosis (LC) may provide a valuable approach to better detect HCC at an early stage when it is still amenable to treatment, thereby improving patient prognosis and survival. In the present study, we,

Differential response of the liver to bile acid treatment in a mouse model of Niemann-Pick disease type C.

Niemann-Pick disease type C (NPC) disease is a neurodegenerative lysosomal storage disease caused by mutations in the NPC1 or NPC2 genes. Liver disease is also a common feature of NPC that can present as cholestatic jaundice in the neonatal period. Liver enzymes can remain elevated above the normal range in some patients as they age. We recently reported suppression of the P450 detoxification system in a mouse model of NPC disease and in post-mortem liver from NPC patients. As bile acids regulate the P450 system, we tested bile acid treatment using ursodeoxycholic acid (UDCA; 3α, 7β-dihydroxy-
