Thermal perturbation studies of membrane-bound acetylcholine receptor from Torpedo: effects of cholinergic ligands and membrane perturbants.

Thermal perturbation techniques have been used to probe structural features of the nicotinic acetylcholine receptor (AcChR). The information obtained from differential scanning calorimetry (DSC) of AcChR membranes (M.C. Farach and M. Martinez-Carrion (1983) J. Biol. Chem. 258, 4176) in the absence and in the presence of cholinergic ligands and local anesthetics, is comparable to that obtained from a simpler technique of heat inactivation of the alpha-bungarotoxin (alpha-Bgt) binding sites on the AcChR protein in similar samples. When AcChR membranes are heated at approximately 1 degree C/min,

Labelled bile salt excretion and diarrhoea in coeliac disease.

Faecal excretion of labelled bile salts after intravenous injection of carboxyl 14C-cholic acid was normal or low in eleven patients with untreated coeliac disease without diarrhoea. Five other coeliac patients with evident diarrhoea had increased excretion of labelled bile salts, suggesting that bile salt malabsorption may contribute to diarrhoea in coeliac disease.

Metabolomic profiling reveals the step-wise alteration of bile acid metabolism in patients with diabetic kidney disease.

BACKGROUND: Diabetic kidney disease (DKD) is the major complication of diabetes concomitant with gut dysbiosis and glycometabolic disorder, which are strongly associated with bile acid (BA) metabolism. Yet studies investigating the BA metabolism involving in DKD pathogenesis are limited. This study aimed to explore the metabolomic profiling of BAs in DKD and analyze its association with DKD progression. METHODS: An ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method was established to quantify BAs in the plasma, fecal and urine samples of patients with DKD or

The Potential Role of Bile Acids in Acquired Laryngotracheal Stenosis.

OBJECTIVE: Gastroesophageal reflux is thought to be a risk factor for laryngotracheal stenosis. Bile acids are a component of gastric refluxate and have previously been implicated in the development of fibrosis in other airway subsites. There is clear evidence that bile acids reflux into the upper airway. We therefore investigated the potential role of bile acids in the pathophysiology of laryngotracheal fibrosis and stenosis, specifically investigating the highly conserved process of epithelial-mesenchymal transition (EMT). STUDY DESIGN: Translational research study. METHODS: Human primary

A computer study of the risk of cholesterol gallstone associated with obesity and normal weight.

Obese people differ from the people of normal weight in gall bladder motility and have a higher risk of cholesterol stone formation. In this study, using a mathematical model of cholesterol homeostasis, which also considers the enterohepatic circulation of bile as well as cholesterol, we investigated the risk of cholesterol stone formation in obese and normal-weight groups who had normal blood cholesterol levels. We associated the risk of stone formation with the amount of cholesterol released into bile and the amount of de novo-synthesized cholic acid. For both groups, we determined the condi

Progressive familial intrahepatic cholestasis and inborn errors of bile acid synthesis.

Progressive familial intrahepatic cholestasis (PFIC), types 1, 2 and 3, are due to defects in genes involved in bile secretion (FIC1, BSEP, MDR3). PFIC and inborn errors of bile acid synthesis (IEBAS) often present in infancy with cholestasis. The distinctive feature of PFIC 1 and 2 and IEBAS is a normal level of GGT, while IEBAS are suspected in patients with low plasma bile acids concentration. Molecular testing, urinary bile acid analysis (IEBAS), liver biopsy and immuno-staining are used for the diagnosis. Some patients with PFIC can be successfully treated with ursodeoxycholic acid or par

Bile acid indices as biomarkers for liver diseases I: Diagnostic markers.

BACKGROUND: Hepatobiliary diseases result in the accumulation of toxic bile acids (BA) in the liver, blood, and other tissues which may contribute to an unfavorable prognosis. AIM: To discover and validate diagnostic biomarkers of cholestatic liver diseases based on the urinary BA profile. METHODS: We analyzed urine samples by liquid chromatography-tandem mass spectrometry and compared the urinary BA profile between 300 patients with hepatobiliary diseases vs 103 healthy controls by statistical analysis. The BA profile was characterized using BA indices, which quantifies the composition, met

Variability in dissolution rates of cholesterol pellets in bile acid solutions.

Large variations in dissolution rate behavior of cholesterol monohydrate pellets may result from small changes in experimental procedures. For example, when cholesterol monohydrate pellets were stored overnight prior to a dissolution run, the initial dissolution rates varied by more than a factor of 2. It is well known that cholesterol monohydrate is converted to anhydrous cholesterol; cholesterol may be unstable toward light. heat, and other radiation in the presence of air, leading to its decomposition. To determine the cause of the variable dissolution rates, experiments were conducted with

Antral mucosal bile acids in two types of chronic atrophic gastritis.

Bile acids may damage the gastric mucosa, and they are cocarcinogenic in experimental colonic and gastric cancer. Chronic atrophic gastritis (CAG) and chronic atrophic gastritis with intestinal metaplasia (CAGIM) are associated with gastric carcinoma. We, therefore, analysed bile acids in the antral mucosa in controls (n = 10), in patients with CAG (n = 12) and CAGIM (n = 20). In both forms of chronic antral gastritis, total mucosal bile acid concentrations drop, caused mainly by lower primary bile acids. The proportions of secondary bile acids rise, in particular of toxic lithocholic acid. Th

Effect of glycoursodeoxycholate on precipitation of calcium carbonate.

The potential role of bile salts in preventing calcium carbonate precipitation was investigated by studying their interaction of Ca2+ and their inhibitory effects on calcium carbonate formation. Glycochenodeoxycholate micelles bound more calcium than did glycocholate. At bile salt concentrations exceeding 12.5 mM, glycoursodeoxycholate bound calcium as well as glycochenodexycholate did. Similar results for calcium binding were observed in mixed micelles of bile salts and lecithin. In bicarbonate (25 or 50 mM) and CaCl2 (10 mM) solutions, calcium carbonate formation was inhibited by the bile sa

Green tea polyphenol (epigallocatechin-3-gallate) improves gut dysbiosis and serum bile acids dysregulation in high-fat diet-fed mice.

Gut microbiota have profound effects on bile acid metabolism by promoting deconjugation, dehydrogenation, and dehydroxylation of primary bile acids in the distal small intestine and colon. High-fat diet-induced dysbiosis of gut microbiota and bile acid dysregulation may be involved in the pathology of steatosis in patients with non-alcoholic fatty liver disease. Epigallocatechin-3-gallate (EGCG), the most abundant polyphenolic catechin in green tea, has been widely investigated for its inhibitory or preventive effects against fatty liver. The aim of the present study was to investigate the eff

Fecal steroids in diarrhea. I. Acute shigellosis.

Fecal bile acid and neutral sterol patterns of five healthy adult male volunteers, who were challenged by a virulent Shigella flexneri 2a (M42-43) strain and developed dysentery were studied. It was observed that cholic acid was increased from 1.9 +/- 0.4% of total bile acid in the feces before infection to 14.5 +/- 2.1% during diarrhea (P less than 0.001). Chenodeoxycholic acid also was increased from 3.2 +/- 0.7 to 8.7 +/- 3.2% in diarrhea but the difference was not significant statistically. Deoxycholic and lithocholic acids constituted 34.1 +/- 4.1 and 40.5 +/- 2.8%, respectively, of total…

Identifying novel metabolites in children with attention-deficit hyperactivity disorder through metabolome profiling.

Metabolomics research offers promising potential for identifying key metabolites and exploring the pathophysiological underpinnings of attention-deficit hyperactivity disorder (ADHD). However, serum metabolomics in ADHD remains largely uncharted. Our study aimed to search for metabolomic biomarkers in children with ADHD. 70 drug-naïve children diagnosed with ADHD according to DSM-5 criteria and 70 sex-, age-, IQ-matched healthy controls were recruited from the National Taiwan University Hospital. All participants were assessed for clinical and ADHD symptoms using the Clinical Global Impression

Ursodeoxycholic acid for treatment of primary sclerosing cholangitis: a placebo-controlled trial.

The efficacy and safety of ursodeoxycholic acid for the treatment of primary sclerosing cholangitis were evaluated in a prospective, randomized, double-blind, placebo-controlled trial. Fourteen patients with primary sclerosing cholangitis documented by cholestatic serum enzyme pattern, liver histological appearance and endoscopic retrograde cholangiography were included in the trial. Six patients received ursodeoxycholic acid (13 to 15 mg/kg body wt/day), and eight patients received placebo. Two patients had to be withdrawn from the study, one because of UDCA-related diarrhea and the other bec

Hydrolysis of 1-palmitoyl-2-[6-(pyren-1-yl)]hexanoyl-sn-glycero- 3-phospholipids by phospholipase A2: effect of the polar head-group.

The effect of the phospholipid polar head-group on the porcine pancreatic phospholipase A2 (phosphatidylcholine 2-acylhydrolase, EC 3.1.1.4) reaction was studied using 1-palmitoyl-2-[6-(pyren-1-yl)]hexanoyl-sn-glycero-3- phosphatidylcholine, -ethanolamine, -glycerol, -monomethylester and -serine as substrates. Except for the monomethylester analogue, which was maximally activated by 3.5 mM CaCl2, maximal enhancement of hydrolysis of the other pyrenephospholipids was obtained at 2 mM Ca2+. Sodium cholate inhibited hydrolysis of the ethanolamine and serine lipids, whereas a slight (1.4-2.0-fold)

The effect of weight reduction on biliary lipid metabolism in patients with hyperlipoproteinemia type IV.

Nine slightly obese patients with hyperlipoproteinemia type IV were studied, before and after a mean weight reduction of about 15 kg, with respect to bile acid kinetics, cholesterol balance and biliary lipid composition. The bile acid pool size was not consistently changed. The synthesis of cholic acid and chenodeoxycholic acid was decreased by about 65% and 50%, respectively. The net steroid balance, calculated as bile acid synthesis plus fecal excretion of neutral steroids minus dietary cholesterol intake, was reduced by about 50%. In all but one of the patients bile was supersaturated with

Structural Characteristics of Low-Molecular-Weight Fucoidan and Its Protective Effects on DSS-Induced Colitis and Associated Behavioral Abnormalities by Improving Intestinal Homeostasis.

This study characterized low-molecular-weight fucoidan (Degradation Laminaria japonica fucoidan (DLJF)) and evaluated its protective effects against dextran sulfate sodium (DSS)-induced colitis. Structural characterization revealed that DLJF had a molecular weight of 40.64 kDa and was a fucose-rich sulfated heteropolysaccharide predominantly composed of sulfated α-L-Fucp residues, with minor α-D-Manp and α-D-Glcp units. DLJF administration markedly alleviated colitis and suppressed pro-inflammatory factors, possibly through the modulation of the Toll-like receptor 4/NF-κB pathway. It also impr

Sustained hyperuricemia disrupts enterohepatic circulation through 12α-hydroxy BA and gut microbiota.

Hyperuricemia (HUA) has become a worldwide health issue, drawing increasing public attention. This study aimed to elucidate the roles of bile acids (BAs) and their associations with gut microbiota in a HUA rat model. We established normal control (N) and HUA rat groups, then characterized the BA profiles in liver, serum, and ileum contents using a targeted metabolomics approach. Additionally, gut microbiota composition was analyzed through 16S rDNA gene sequencing. The results were that hyperuricemia induced elevated levels of cholic acid (CA) in liver and elevated levels of taurocholic acid (
