Distinct HLA Associations for Antibody Multireactivity With Citrulline-Containing Type II Collagen Epitopes Versus More Limited Antibody Reactivity With Citrulline-Containing IgG Epitopes in Rheumatoid Arthritis.

OBJECTIVE: Anticitrullinated protein antibodies (ACPAs) in rheumatoid arthritis (RA) can be promiscuous, with cross-reactive binding to many antigens containing short motifs, or private with little cross-reactivity. Also, ACPA reactivity patterns differ among patients with RA, including for motif-containing epitopes in important self-antigens like collagen and IgG (bound by RA-associated rheumatoid factors [RFs]), with limited understanding of the underlying mechanism. The objective of this study was to determine if HLA alleles associate with ACPA reactivity patterns. METHODS: For 100 ACPA+RF

T cell repertoire to citrullinated self-peptides in healthy humans is not confined to the HLA-DR SE alleles; Targeting of citrullinated self-peptides presented by HLA-DP4 for tumour therapy.

Post-translational modifications are induced in stressed cells which cause them to be recognised by the system. One such modification is citrullination where the positive charged arginine is modified to a neutral citrulline. We demonstrate most healthy donors show an oligoclonal CD4 response in vitro to at least one citrullinated vimentin or enolase peptide. Unlike rheumatoid arthritis patients, these T cell responses were not restricted by HLA-DRB1 shared epitope (SE) alleles, suggesting they could be presented by other MHC class II alleles. As HLA-DP is less polymorphic than HLA-DR, we inves…

Comparative analysis of autoantibodies targeting peptidylarginine deiminase type 4, mutated citrullinated vimentin and cyclic citrullinated peptides in rheumatoid arthritis: associations with cytokine profiles, clinical and genetic features.

Antibodies against cyclic citrullinated peptides (anti-CCP) are widely used for diagnosis of rheumatoid arthritis (RA). We performed a comparative analysis of antibodies targeting the citrullinating enzyme peptidylarginine deiminase type 4 (anti-PAD4) and mutated citrullinated vimentin (anti-MCV) with anti-CCP autoantibodies in RA patients and examined their relationships with clinical parameters, cytokine profiles and the PADI4 gene. Autoantibodies were examined by enzyme-linked immunosorbent assay (ELISA) in sera of 170 RA patients and 103 controls. Cytokine profiles were measured using a mu

Beyond natural amino acids: extending immunogenicity risk assessment to modified peptides through chemical feature encoding. A case study on citrullination.

INTRODUCTION: Peptide therapeutics are increasingly explored to treat challenging diseases, but immunogenicity risks limit their clinical success. In silico tools enable immunogenicity screening through prediction of peptide-MHCII binding, yet current methods do not account for chemical properties of non-natural amino acids routinely incorporated to improve peptide properties. METHODS: Here, we present a machine learning approach combining chemical fingerprints with sequence information to predict MHC class II binding for peptides including both natural (NAA) and non-natural (NNAA) amino acid

Increased citrullination and expression of peptidylarginine deiminases independently of P. gingivalis and A. actinomycetemcomitans in gingival tissue of patients with periodontitis.

BACKGROUND: A relationship between rheumatoid arthritis (RA) and periodontitis has been suggested from findings that individuals with RA are … teins or PAD enzymes. CONCLUSION: Chronic gingival inflammation is associated with increased local citrullination and PAD2 and PAD4 expression in periodontitis. The increased citrullination and PAD2 and PAD4 expression in periodontitis were, however, independent of the presence of periodontal pathogen P. gingivalis and A. actinomycetemcomitans leukotoxin.

Reduced TCR-dependent activation through citrullination of a T-cell epitope enhances Th17 development by disruption of the STAT3/5 balance.

Citrullination is a post-translational modification of arginine that commonly occurs in inflammatory tissues. Because T-cell receptor (TCR) signal quantity and quality can regulate T-cell differentiation, citrullination within a T-cell epitope has potential implications for T-cell effector function. Here, we investigated how citrullination of an immunedominant T-cell epitope affected Th17 development. Murine naïve CD4(+) T cells with a transgenic TCR recognising p89-103 of the G1 domain of aggrecan (agg) were co-cultured with syngeneic bone marrow-derived dendritic cells (BMDC) presenting the …

PAD4-generated citrullinated histones are triggers for autoinflammation in cryopyrin-associated periodic syndrome.

Cryopyrin-associated periodic syndromes (CAPS) are autoinflammatory disorders caused by gain-of-function NLRP3 variants. Although NLRP3 inflammasomes mediate IL-1β secretion through Gasdermin D (GSDMD), we show that GSDMD deletion did not prevent autoinflammation in mice ubiquitously expressing the Nlrp3A350V variant. Inflamed skin of Nlrp3A350V-expressing GSDMD-deficient mice displayed citrullinated histone 3-containing neutrophil extracellular traps (CitH3-NETs). CitH3-NETs induced IL-1β secretion from murine Nlrp3A350V-expressing GSDMD-deficient macrophages as well as from human CAPS patien

Microglial GLUL loss worsens TBI outcomes by amplifying the arginine-citrulline pathway.

Microglial hyperactivation-induced neuroinflammation is a central driver of neurological dysfunction after traumatic brain injury (TBI). Metabolic reprogramming is essential for microglial activation, but the specific metabolic alterations following TBI and their causal relationship with activation remain poorly defined. This study investigated the role and underlying mechanisms of glutamate-ammonia ligase (GLUL) in regulating microglial activation after TBI. A murine TBI model was established using a controlled cortical impact device, and brain injury severity, neuroinflammation, and behavior

Impact of L-arginine and L-citrulline supplementation on macrophage responses to Mycobacterium tuberculosis.

L-arginine (ARG) availability is a critical determinant of macrophage antimicrobial capacity, as it fuels nitric oxide production and other immune effector pathways essential for restricting Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis (TB). L-citrulline (CIT), a precursor in the ARG regeneration cycle, can replenish intracellular ARG pools when transport is limited. However, the comparative and combined effects of exogenous ARG and/or CIT on intracellular Mtb control across macrophage lineages and activation states remain insufficiently defined. This study investigate

Effects of Quercetin and Citrulline on Nitric Oxide Metabolites and Antioxidant Biomarkers in Trained Cyclists.

BACKGROUND: Quercetin (QCT) and citrulline (CIT) have been independently associated with improved antioxidant capacity and nitric oxide (NO) production, potentially enhancing cardiovascular function and exercise performance. This study aimed to evaluate the combined and independent effects of QCT and CIT supplementation on NO metabolites and antioxidant biomarkers in 50 trained cyclists undergoing a 20 km cycling time trial (TT). METHODS: In a randomized, double-blind, placebo-controlled design, forty-two male and eight female trained cyclists were assigned to QCT + CIT, QCT, CIT, or placebo

Evaluation of Diagnostic Efficacy of Anticyclic Citrullinated Peptide Antibody in Diagnosis of Rheumatoid Arthritis.

BACKGROUND: The prevalent diagnostic tests available for rheumatoid arthritis (RA), i.e. rheumatoid factor, C-reactive protein (CRP), and erythrocyte sedimentation rate (ESR), are nonindicative in the initial stages of disease as well as nonspecific. The latest marker introduced for detection of RA is anticyclic citrullinated peptide (anti-CCP), which can help in early diagnosis and check the progression of severity of RA. AIM: The present study was designed to evaluate the role of anti-CCP antibody in the diagnosis of RA in early stage of disease as compared to ESR and CRP. MATERIALS AND ME

SNP associations in the L-citrulline metabolic pathway and vascular aging in the Japanese population.

Decreased nitric oxide (NO) production from the vascular endothelium is a major factor for vascular aging. Because vascular aging has few specific subjective symptoms, assessing the susceptibility to vascular aging is beneficial for its early detection and improvement. Therefore, this study evaluated the associations between single nucleotide polymorphisms (SNPs) on L-citrulline (Cit) pathways essential for NO production and the health characteristics involved in vascular aging using a candidate gene approach. Associations with a significance level included those between the KCNMB4 rs17108108

Citrulline and ADI-PEG20 reduce inflammation in a juvenile porcine model of acute endotoxemia.

BACKGROUND: Arginine is a conditionally essential amino acid that is depleted in critically ill or surgical patients. In pediatric and adult patients, sepsis results in an arginine-deficient state, and the depletion of plasma arginine is associated with greater mortality. However, direct supplementation of arginine can result in the excessive production of nitric oxide (NO), which can contribute to the hypotension and macrovascular hypo-reactivity observed in septic shock. Pegylated arginine deiminase (ADI-PEG20, pegargiminase) reduces plasma arginine and generates citrulline that can be trans

L-citrulline attenuates lipopolysaccharide-induced inflammatory lung injury in neonatal rats.

BACKGROUND: Prenatal or postnatal lung inflammation and oxidative stress disrupt alveolo-vascular development leading to bronchopulmonary dysplasia (BPD) with and without pulmonary hypertension. L-citrulline (L-CIT), a nonessential amino acid, alleviates inflammatory and hyperoxic lung injury in preclinical models of BPD. L-CIT modulates signaling pathways mediating inflammation, oxidative stress, and mitochondrial biogenesis-processes operative in the development of BPD. We hypothesize that L-CIT will attenuate lipopolysaccharide (LPS)-induced inflammation and oxidative stress in our rat mode

Accuracy of citrulline, I-FABP and D-lactate in the diagnosis of acute mesenteric ischemia.

Early diagnosis of acute mesenteric ischemia (AMI) remains a clinical challenge, and no biomarker has been consistently validated. We aimed to assess the accuracy of three promising circulating biomarkers for diagnosing AMI-citrulline, intestinal fatty acid-binding protein (I-FABP), and D-lactate. A cross-sectional diagnostic study enrolled AMI patients admitted to the intestinal stroke center and controls with acute abdominal pain of another origin. We included 129 patients-50 AMI and 79 controls. Plasma citrulline concentrations were significantly lower in AMI patients compared to the contro

Complementary Effects of Carbamylated and Citrullinated LL37 in Autoimmunity and Inflammation in Systemic Lupus Erythematosus.

LL37 acts as T-cell/B-cell autoantigen in Systemic lupus erythematosus (SLE) and psoriatic disease. Moreover, when bound to "self" nucleic acids, LL37 acts as "danger signal," leading to type I interferon (IFN-I)/pro-inflammatory factors production. T-cell epitopes derived from citrullinated-LL37 act as better antigens than unmodified LL37 epitopes in SLE, at least in selected HLA-backgrounds, included the SLE-associated HLA-DRB1*1501/HLA-DRB5*0101 backgrounds. Remarkably, while "fully-citrullinated" LL37 acts as better T-cell-stimulator, it loses DNA-binding ability and the associated "adjuva

Current view on the pathogenic role of anti-citrullinated protein antibodies in rheumatoid arthritis.

Epidemiological findings suggest a potential role for anti-citrullinated protein antibodies (ACPAs) in rheumatoid arthritis (RA) pathogenesis. ACPA-positive RA is associated with unique genetical and environmental risk factors, in contrast to seronegative RA. ACPA-positive healthy individuals are at risk of developing RA and can develop joint pain and bone loss already before disease onset. ACPA injection triggered bone loss and pain-like behaviour in mice and, in the presence of additional arthritis inducers, exacerbated joint inflammation. In cell culture experiments, ACPAs could bind to and

Facile synthesis of a high efficiency and durability L-citrulline flame retardant for cotton.

A novel flame retardant (FR), the ammonium salt of citrulline-penta (methylphosphonic acid) (ACPMPA) based on L-citrulline was synthesized, and its structure was characterized by 13C, 1H, and 31P nuclear magnetic resonance (NMR) spectroscopy. The ACPMPA flame retardant molecule contains five ammonium salts of phosphorus acid and one ammonium salt of carboxylic acid, which allowed the covalent attachment of ACPMPA onto cellulose via -P=O(-O-C) and -COOC bonds. The results showed that the treated cotton fabrics had very high flame retardance and excellent durability. The limiting oxygen index (L
