Rheumatoid arthritis (RA) is a disease characterized by symmetrical polyarthritis of the large and small joints, and in the majority of patients, there is a presence of the rheumatoid factor and erosions in the X-ray of the joints. More recently, the presence of anti-cyclic citrullinated peptide antibodies (anti-CCP) in this disease has been described, with diagnostic and prognostic value. Nevertheless, these antibodies have also been described in infectious diseases. The aim of the present study was to make a systematic review of the presence of antibodies against citrullinated peptides in in

BACKGROUND: Immunological abnormalities in rheumatoid arthritis (RA) imply several antibodies, among which anti-cyclic cytrullinated peptide antibodies (anti-CCP) have the highest sensitivity and specificity. Their diagnostic and prognostic value in RA is well known, although their value as markers of the disease activity has not been established yet.
OBJECTIVES: The aim of this study is to evaluate the correlation between anti-CCP antibodies and RA activity which eventually leads to the best treatment of choice.
PATIENTS AND METHODS: 217 consecutive patients hospitalized in the Department o

BACKGROUND: The presence of rheumatoid factor (RF) is one of the classification criteria of the American College of Rheumatology (ACR) for rheumatoid arthritis (RA), but it has a limitation of low specificity. We compared the diagnostic utility of anti-cyclic citrullinated peptide (CCP) antibodies analyzed by an automated immunoassay system with that measured by a 96 well plate ELISA method.
METHODS: The RF and anti-CCP antibodies were determined in 172 serum samples: 52 RA patients, 73 disease controls (systemic lupus, Sjogren's syndrome, palindromic rheumatism), and 47 healthy controls. Ant

The pathogenesis of MS is unknown. In our studies, we have demonstrated an important role for citrullinated myelin basic protein (MBP). The accompanying loss of positive charge compromises the ability of MBP to interact with the lipid bilayer. The conversion of arginine to citrulline in brain is carried out by an enzyme peptidyl arginine deiminase (PAD) 2. The amount of PAD 2 in brain was increased in MS normal-appearing white matter. The mechanism responsible for this increase involved hypomethylation of the promoter region in the PAD 2 gene in MS, but no change (compared to normal) was found

Antibodies directed against citrullinated vimentin are members of the family of autoantibodies reactive with citrullinated proteins and are among the most specific serological markers for the diagnosis of rheumatoid arthritis (RA). This study was performed to test the diagnostic value of a newly developed enzyme-linked immunosorbent assay (ELISA) for the detection of antibodies against a genetically modified citrullinated vimentin (anti-MCV) in comparison with a second-generation anti-cyclic citrullinated peptides (anti-CCP2) ELISA test system. Protein arginine deiminase-4 (PAD4) catalyzes hydrolysis of arginine to citrulline in proteins that promote widespread cellular changes that can induce innate immunity and promote cancer. Hyperactivity of PAD4 leads to a form of cell death called NETosis, releasing PAD4 to the extracellular space to promote various autoimmune diseases through the generation of anticitrulline protein antibodies (ACPAs). Little is known about the specific citrullinated substrates that lead to autoimmunity, but there is growing evidence that PAD4 is localized to the cell surface in response to inflammation. Here,

Cancer cells must maintain molecular mechanisms relying on an energy trade-off between resistance and key functions to survive. PADI4 (peptidyl arginine deiminase 4) is an enzyme implicated in the conversion of arginine to citrulline (citrullination), that has been related to the development of several types of cancers and in numerous inflammatory routes. Salvianolic acid A (SAA) is a stilbenoid, obtained from the radix of Salvia miltiorrhiza, with several anti-cancer and anti-inflammatory features. In this work, we report and characterize the interaction between PADI4 and SAA. The binding rea

Rheumatoid Arthritis (RA) is a prevalent form of inflammatory arthritis in which post-translational modifications are known to play a major role in the loss of tolerance. This review summarizes current knowledge of the role that such modifications play in the disease. Most individuals with RA have antibody responses directed against citrullinated and/or carbamylated self-antigens, but additional modifications are under active investigation. Post-translational modifications can alter the primary protein sequence, increasing binding to HLA molecules and recognition by T cells, and can generate n

Peptidylarginine deiminase (PAD) catalyzes the post-translational conversion of positively charged arginine residues into neutral citrulline. This modification, known as citrullination, is implicated in various pathological conditions, including those affecting the central nervous system. This study investigated the pro-inflammatory role of PAD in microglia using a middle cerebral artery occlusion (MCAO) animal model of ischemic stroke. Pharmacological inhibition of PAD via BB-Cl-amidine (BBCA; a pan-PAD inhibitor) exerted robust anti-inflammatory and neuroprotective effects in the post-ischem

Proteins affect most physiological functions in cells and play a key role in maintaining cellular homeostasis. Protein function is tuned at multiple levels, including synthesis and degradation, intracellular localization, and posttranslational modifications (PTMs). PTMs are a key chemical modification for regulating protein biological activity, including protein-folding relevant shifts in electrical charge, protein-protein interactions, and hydrogen bond formation. Such chemical changes can also result in protein unfolding or denaturation. PTMs can target proteins across multiple cellular comp

HIV-1 infection establishes a reservoir of long-lived cells with integrated proviral DNA that can persist despite antiretroviral therapy (ART). Certain reservoir cells can be reactivated to reinitiate infection. The mechanisms governing proviral latency and transcriptional regulation of the provirus are complex. Here, we identify a role for histone H3 citrullination, a post-translational modification catalyzed by protein-arginine deiminase type-4 (PADI4), in HIV-1 transcription and latency. PADI4 inhibition by the small molecule inhibitor GSK484 reduces HIV-1 transcription after T cell activat

BACKGROUND: The DNA-binding peptide LL37 is a suspected autoantigen in psoriasis. It can be found in neutrophil extracellular traps (NETs) which have been suggested to play a role in the pathogenesis of the disease. Citrullination, the conversion of peptidyl-arginine into peptidyl-citrulline, can be implicated in the formation of NETs. We hypothesized that citrullination increases LL37 immunogenicity and that NETs are a source of LL37.
OBJECTIVES: We aimed to characterize cytokine responses of B cells and T cells to native and citrullinated LL37 (citLL37) and determine the prevalence and comp

The immune mechanisms that mediate synovitis and joint destruction in rheumatoid arthritis (RA) remain poorly defined. Although increased levels of CD8+ T cells have been described in RA, their function in pathogenesis remains unclear. Here we perform single cell transcriptome and T cell receptor (TCR) sequencing of CD8+ T cells derived from anti-citrullinated protein antibodies (ACPA)+ RA blood. We identify GZMB+CD8+ subpopulations containing large clonal lineage expansions that express cytotoxic and tissue homing transcriptional programs, while a GZMK+CD8+ memory subpopulation comprises smal

BACKGROUND: Citrullinated fibrinogen (C-Fbg) has been detected in rheumatoid arthritis; however, few studies have reported the role of C-Fbg in other inflammatory diseases. This study aimed to clarify the changes in serum C-Fbg associated with the bacteremia phase.
METHODS: We measured serum C-Fbg concentration in bacteremia patients. C-Fbg levels at each phase of bacteremia, classified by white blood cell (WBC) count and neutrophil left shift change, were compared with those of healthy control (HC). The correlation between C-Fbg concentration and certain inflammatory markers, or citrullinate

Proteins are well-known to undergo a variety of post-translational modifications (PTMs). One such PTM is citrullination, an arginine modification that is catalyzed by a group of hydrolases called protein arginine deiminases (PADs). Hundreds of proteins are known to be citrullinated and hypercitrullination is associated with autoimmune diseases including rheumatoid arthritis (RA), lupus, ulcerative colitis (UC), Alzheimer's disease, multiple sclerosis (MS), and certain cancers. In this Account, we summarize our efforts to understand the structure and mechanism of the PADs and to develop small m
