Cold Saline Perfusion before Ischemia-Reperfusion Is Harmful to the Kidney and Is Associated with the Loss of Ezrin, a Cytoskeletal Protein, in Rats.

BACKGROUND: Organ protection for transplantation is perfusion with ice-cold preservation solutions, although saline is also used in animal experiments and living donor transplantations. However, ice-cold perfusion can contribute to initial graft injury. Our aim was to test if cytoskeletal damage of parenchymal cells is caused by saline itself or by the ice-cold solution. METHODS: F344 rat kidneys were flushed with cold (4 °C) saline, ischemic and sham kidneys were not perfused. In a separate set, F344 kidneys were flushed with saline or preservation solution at 4 or 15 °C. Ischemia time was 3

Finger and toe temperatures on exposure to cold water and cold air.

INTRODUCTION: Subjects with a weak cold-induced vasodilatation response (CIVD) to experimental cold-water immersion of the fingers in a laboratory setting have been shown to have a higher risk for local cold injuries when exposed to cold in real life. Most of the cold injuries in real life, however, occur in the foot in cold air rather than in the hand in cold water. Therefore, an experiment was conducted to investigate the within-subject relation between CIVD in the fingers and toes exposed to cold water and cold air. METHODS: In 4 experimental sessions, 11 healthy male subjects immersed the

Rapid cold hardening improves tissue viability and facilitates recovery of activity following nonlethal cold exposure in Drosophila melanogaster.

Low temperature is a significant stressor for small-bodied ectotherms like insects, and they employ a range of physiological mechanisms to m … incurred at nonlethal temperatures, but studies on the ability of RCH to protect against sublethal cold injury are limited, despite nonlethal cold exposure being more common in nature. Here, we test the hypothesis that RCH protects against sublethal cold injury in the model species Drosophila melanogaster. Specifically, we exposed flies to a nonlethal cold s …

Exogenous Melatonin Enhances the Cold Tolerance of Harvested Peach (Prunus persica Batsch) Fruit by Elevating SAMDC and Decreasing ACC Synthase Activities.

Exogenous melatonin can alleviate cold injury in post-harvested fruit during fruit refrigeration. The mechanism of melatonin-mediated cold tolerance is increasingly being focused on in studies. However, in cold + melatonin-treated fruit, the changes in the activities of S-adenosylmethionine decarboxylase (SAMDC) and 1-aminocyclopropane-1-carboxylic acid (ACC) synthase, the key enzymes that catalyze polyamine and ethylene biosynthesis, respectively, are still unclear. In this study, the typical symptoms of cold injury, the contents of polyamines and ethylene, and the activities of SAMDC and ACC…

Prevalence and severity of cold intolerance in patients after hand fracture.

Cold intolerance is a well-known phenomenon that develops in the first months after hand injury and generally does not decrease over time. In this study, we evaluated the prevalence and severity of cold intolerance after hand fracture in 129 patients using the Cold Intolerance Symptom Severity (CISS) questionnaire. Patients with nerve and/or vascular injuries were excluded. The response rate was 59%. The mean CISS score was 23. Pathological cold intolerance, defined as a CISS score over 30, was experienced by 38% of the patients. Cold intolerance is common after hand fractures and can be sever

Determination of Maximum Tolerable Cold Ischemia Time in a Mouse Model of Cervical Heterotopic Uterus Transplantation.

BACKGROUND: Uterus transplantation (UTx) is an emerging treatment for uterine factor infertility. Determining the maximum tolerable cold ischemia time is crucial for successful UTx. However, the limit for cold ischemia in the uterus is unclear. This study aimed to examine cold ischemia's effects on mouse uteri and identify the maximum cold ischemia duration that uteri can endure. METHODS: We systematically assessed the tolerance of mouse uteri to extended cold ischemia, 24 h, 36 h, and 48 h, using the cervical heterotopic UTx model. Multiple indicators were used to evaluate ischemia-reperfusi

Ameliorative potential of Ocimum sanctum in chronic constriction injury-induced neuropathic pain in rats.

The present study was designed to investigate the ameliorative potential of Ocimum sanctum and its saponin rich fraction in chronic constriction injury-induced neuropathic pain in rats. The chronic constriction injury was induced by placing four loose ligatures around the sciatic nerve, proximal to its trifurcation. The mechanical hyperalgesia, cold allodynia, paw heat hyperalgesia and cold tail hyperalgesia were assessed by performing the pinprick, acetone, hot plate and cold tail immersion tests, respectively. Biochemically, the tissue thio-barbituric acid reactive species, super-oxide anion

Modality-Specific Modulation of Temperature Representations in the Spinal Cord after Injury.

Different types of tissue injury, such as inflammatory and neuropathic conditions, cause modality-specific alternations on temperature perception. There are profound changes in peripheral sensory neurons after injury, but how patterned neuronal activities in the CNS encode injury-induced sensitization to temperature stimuli is largely unknown. Using in vivo calcium imaging and mouse genetics, we show that formalin- and prostaglandin E2-induced inflammation dramatically increase spinal responses to heating and decrease responses to cooling in male and female mice. The reduction of cold response

Hypothermic machine perfusion of the liver and the critical balance between perfusion pressures and endothelial injury.

Hypothermic machine perfusion (HMP) provides better protection against cold ischemic injury than cold storage in marginal donor kidneys. Also, in liver transplantation a switch from static cold storage to HMP could be beneficial as it would allow longer preservation times and the use of marginal donors. A critical question concerning application of HMP in liver preservation is the crucial balance between perfusion pressure and occurrence of endothelial injury. Rat livers were cold-perfused for 24 hours to study perfusion pressures for both hepatic artery and portal vein. Cold storage served as

An engineered poly(A) tail attenuates gut ischemia/reperfusion-induced acute lung injury.

BACKGROUND: Gut ischemia/reperfusion causes the release of damage-associated molecular patterns, leading to acute lung injury and high mortality. Cold-inducible ribonucleic acid-binding protein is a ribonucleic acid chaperon that binds the polyadenylation tail of messenger ribonucleic acid intracellularly. Upon cell stress, cold-inducible ribonucleic acid-binding protein is released, and extracellular cold-inducible ribonucleic acid-binding protein acts as a damage-associated molecular pattern, worsening inflammation. To inhibit extracellular cold-inducible ribonucleic acid-binding protein, we

Transient receptor potential melastatin 8 contributes to the interleukin-33-mediated cold allodynia in a mouse model of neuropathic pain.

Cold allodynia is a common complaint of patients suffering from neuropathic pain initiated by peripheral nerve injury. However, the mechanisms that drive neuropathic cold pain remain elusive. In this study, we show that the interleukin (IL)-33/ST2 signaling in the dorsal root ganglion (DRG) is a critical contributor to neuropathic cold pain by interacting with the cold sensor transient receptor potential melastatin 8 (TRPM8). By using the St2-/- mice, we demonstrate that ST2 is required for the generation of nociceptor hyperexcitability and cold allodynia in a mouse model of spared nerve injur

Influence of Osteochondral Allograft Harvest Irrigation Technique and Graft Characteristics on Temperature-Related Viability Changes.

BACKGROUND: Long-term clinical success and graft integration after osteochondral allograft (OCA) transplantation rely on chondrocyte viability, which can be compromised by thermal injury during graft harvest. HYPOTHESIS: It was hypothesized that handheld cold saline irrigation and cold saline submersion would reduce thermal injury during OCA harvest and preserve chondrocyte viability compared with dry harvest, and that submersion would provide superior protection than handheld irrigation. STUDY DESIGN: Controlled laboratory study. METHODS: Ten fresh human femoral condyles (from 7 donors) we

Inflammatory and neuropathic cold allodynia are selectively mediated by the neurotrophic factor receptor GFRα3.

Tissue injury prompts the release of a number of proalgesic molecules that induce acute and chronic pain by sensitizing pain-sensing neurons (nociceptors) to heat and mechanical stimuli. In contrast, many proalgesics have no effect on cold sensitivity or can inhibit cold-sensitive neurons and diminish cooling-mediated pain relief (analgesia). Nonetheless, cold pain (allodynia) is prevalent in many inflammatory and neuropathic pain settings, with little known of the mechanisms promoting pain vs. those dampening analgesia. Here, we show that cold allodynia induced by inflammation, nerve injury,

Protective mechanisms of end-ischemic cold machine perfusion in DCD liver grafts.

BACKGROUND & AIMS: The aim of this study was to identify protective mechanisms of cold machine perfusion in liver grafts donated after cardiac death. METHODS: Pig livers exposed to 60-min warm ischemia were cold stored for 7 h or treated after 6-h cold storage with 1-h hypothermic oxygenated perfusion (HOPE) through the portal vein. Different physical (perfusion pressure) and chemical (oxygen, mitochondrial transition pore inhibition) parameters were analyzed during machine perfusion to dissect key steps of mechanism. RESULTS: HOPE treatment led to a significant slowdown of mitochondrial res

Treatment of frostbites of the upper extremities with prolonged blockade of axillary plexus.

Encouraged by the beneficial effect in the treatment of cold injury of the lower extremities, the authors applied permanent plexus anesthesia for treatment of the cold injury of the upper extremities too. Of the total of 10 in-patients treated with this method, amputation had to be made only in 2 cases due to the lack of compliance of the patients retarding the commencement of therapy. 7 patients with the same severity of cold injury admitted from other hospitals where they had been treated with traditional methods have undergone multiple phalangectomies. The method had beneficial effect only …

Empagliflozin Supplementation in Cardioplegic Solution Improves Donor Heart Preservation by Maintaining Mitochondrial Homeostasis.

BACKGROUND: Prolonged cold ischemia and subsequent reperfusion are major causes of primary graft dysfunction in heart transplantation. Empagliflozin, a sodium-glucose cotransporter 2 (SGLT2) inhibitor, has shown cardioprotective effects in ischemia-reperfusion injury, but its role in donor heart preservation and the underlying mechanisms remain unexplored. METHODS: We evaluated the effects of empagliflozin supplementation in University of Wisconsin (UW) preservation solution using both in vitro and in vivo models. HL-1 cardiomyocytes were subjected to cold hypoxia/reoxygenation injury to simu

Combat Casualty Care in Arctic or Extreme Cold Environments: Considerations for the Army Health System.

The Department of the Army's 2021 strategy to "Regain Arctic Dominance" highlights the operational necessity of sustaining combat readiness in Arctic or extreme cold environments (AoECE). Presented are five scenarios inspired by casualties that illustrate capability gaps and best practices as the Army Health System applies tactical combat casualty care (TCCC) in austere, frigid climates: (1) adapting TCCC to the AoECE as a unique environment; (2) technological limitations in the prevention and treatment of hypothermia; (3) freezing cold injury (FCI) and non-freezing cold injury (NFCI) treatmen…

Development of interactive guidance for cold exposure using a thermoregulatory model.

For decades, the Wind Chill Temperature Index (WCT) and its various iterations have been used to assess the risk of frostbite on unclothed b … pothermia and ensures that a selected clothing ensemble will provide adequate protection to prevent cold injury. CoWEDA predictions provide not only estimates of frostbite risk similar to WCT tables but also hypothermia times and clothing required to prevent cold injuries. In addition, a CoWEDA model variant can predict survivability and clothing requirements …
