Convergent multidimensional neural phenotypes in human patients and schizophrenia-relevant mouse models identify reduced recurrent excitation in cortical circuits.

Objective. NMDA-receptor hypofunction has been implicated as a core pathophysiological mechanism in schizophrenia, but conflicting evidence exists regarding the locus of the hypofunction-i.e. predominantly affecting receptors on pyramidal neurons or interneurons. To investigate the locus of NMDA-receptor hypofunction we integrated imaging data from humans and mice with biophysical model simulations. Methods. Translational imaging measures-intrinsic neural timescales (INT) and spontaneous co-activation pattern similarity-were investigated in three schizophrenia-relevant mouse models (chronic ke

Comparison of Cortical Autonomic Network-Linked Sympathetic Excitation by Mueller Maneuvers and Breath-Holds in Subjects With and Without Obstructive Sleep Apnea.

In healthy young volunteers, acquisition of blood oxygen level-dependent (BOLD) magnetic resonance (MR) and muscle sympathetic nerve (MSNA) signals during simulation of obstructive or central sleep apnea identified cortical cardiovascular autonomic regions in which the BOLD signal changed synchronously with acute noradrenergic excitation. In the present work, we tested the hypothesis that such Mueller maneuvers (MM) and breath-holds (BH) would elicit greater concomitant changes in mean efferent nerve firing and BOLD signal intensity in patients with moderate to severe obstructive sleep apnea (

Cortical excitation does not drive changes in skin sympathetic nerve activity during single-pulse transcranial magnetic stimulation in humans.

We have shown that single-pulse transcranial magnetic stimulation (TMS) of the dorsolateral prefrontal cortex (dlPFC) inhibits muscle sympathetic nerve activity. However, this was likely due to arousal caused by the TMS pulses themselves, rather than altering the underlying neuronal circuitry. In extension, we have aimed to explore the effects of single-pulse TMS on skin sympathetic nerve activity (SSNA), which is more sensitive to arousal. It was hypothesised that TMS-evoked arousal would increase SSNA but would not generate de novo bursts from the dlPFC. Microneurographic recordings were tak

G Protein-Coupled Receptor 32 Contributes to Inflammation Resolution and Neuronal Excitability Dysfunction in Patients With Focal Cortical Dysplasia IIb and Tuberous Sclerosis Complex.

BACKGROUND: Focal cortical dysplasia IIb (FCDIIb) and tuberous sclerosis complex (TSC) show persistent neuroinflammation that promotes epileptogenesis and epilepsy progression, suggesting that endogenous resolution of inflammation is inadequate to relieve neuronal network hyperexcitability. G-protein-coupled receptor 32 (GPR32) is a key regulator of inflammation resolution and we aimed to explore the roles of GPR32 in cortical lesions of patients with FCDIIb and TSC. METHOD: We examined the expression and distribution of GPR32 in patients with FCDIIb and TSC and its effects on human microglia

Modulation of Human Motor Cortical Excitability and Plasticity by Opuntia Ficus Indica Fruit Consumption: Evidence from a Preliminary Study through Non-Invasive Brain Stimulation.

Indicaxanthin (IX) from Opuntia Ficus Indica (OFI) has been shown to exert numerous biological effects both in vitro and in vivo, such as antioxidant, anti-inflammatory, neuro-modulatory activity in rodent models. Our goal was to investigate the eventual neuro-active role of orally assumed fruits containing high levels of IX at nutritionally-relevant amounts in healthy subjects, exploring cortical excitability and plasticity in the human motor cortex (M1). To this purpose, we applied paired-pulse transcranial magnetic stimulation and anodal transcranial direct current stimulation (a-tDCS) in b…

Effect of transcranial direct current stimulation on paroxysmal sympathetic hyperexcitability with acquired brain injury and cortical excitability: a randomized, double-blind, sham-controlled pilot study.

Paroxysmal sympathetic hyperexcitation (PSH) refers to a clinical syndrome characterized by a sudden increase in sympathetic excitability ca … efficacy compared to medication therapy alone. Electrophysiological results also indicated enhanced cortical excitability. Therefore, this single-center pilot study suggests that multi-target, multi-session tDCS combined with medication may be an effective treatment protocol for PSH.

Glutamatergic modulation of transcranial direct current stimulation over the premotor cortex combined with peripheral nerve stimulation promotes observation-execution-related cortical excitability and motor performance.

Action observation and execution, transcranial direct current stimulation (tDCS) and peripheral nerve stimulation (PNS) enhance motor performance. This study aimed to investigate the effect of concurrent tDCS over the premotor cortex (PMC) and PNS during action observation on motor cortex excitability and motor performance, and its underlying mechanisms. One hundred and twenty-eight healthy adults were randomly assigned to tDCS + PNS combined with presentation of a landscape picture (tDCS + PNSlandscape), and tDCS + PNS, tDCSsham+ PNSsham, tDCSsham + PNS and tDCS + PNSsham combined with action

Acute pain drives different effects on local and global cortical excitability in motor and prefrontal areas: insights into interregional and interpersonal differences in pain processing.

Pain-related depression of corticomotor excitability has been explored using transcranial magnetic stimulation-elicited motor-evoked potentials. Transcranial magnetic stimulation-electroencephalography now enables non-motor area cortical excitability assessments, offering novel insights into cortical excitability changes during pain states. Here, pain-related cortical excitability changes were explored in the dorsolateral prefrontal cortex and primary motor cortex (M1). Cortical excitability was recorded in 24 healthy participants before (Baseline), during painful heat (Acute Pain), and non-no…

Chronic Stress Alters Synaptic Inhibition/Excitation Balance of Pyramidal Neurons But Not PV Interneurons in the Infralimbic and Prelimbic Cortices of C57BL/6J Mice.

The medial prefrontal cortex (mPFC) plays a pivotal role in regulating working memory, executive function, and self-regulatory behaviors. Dysfunction in the mPFC circuits is a characteristic feature of several neuropsychiatric disorders including schizophrenia, depression, and post-traumatic stress disorder. Chronic stress (CS) is widely recognized as a major triggering factor for the onset of these disorders. Although evidence suggests synaptic dysfunction in mPFC circuits following CS exposure, it remains unclear how different neuronal populations in the infralimbic (IL) and prelimbic (PL) c

Rescuing prefrontal cortical excitation-inhibition imbalance with early-life microglial ablation ameliorates ASD-like behaviors in male BTBR mice.

Autism spectrum disorder (ASD) is a neurodevelopmental condition characterized by deficits in social communication and interaction, alongside the presence of restricted and repetitive behaviors. Microglia, the resident immune cells of the central nervous system, play a crucial role in the development of ASD by regulating synaptic development and plasticity. In this study, we investigate the pharmacological effects and underlying cellular mechanisms of PLX3397 (PLX), a selective inhibitor of the colony-stimulating factor 1 receptor (CSF1R), for the pharmacological ablation of microglia in the c

Dynorphin / kappa-opioid receptor regulation of excitation-inhibition balance toggles afferent control of prefrontal cortical circuits in a pathway-specific manner.

The medial prefrontal cortex (mPFC) controls behavior via connections with limbic excitatory afferents that engage various inhibitory motifs to shape mPFC circuit function. The dynorphin (Dyn) / kappa-opioid receptor (KOR) system is highly enriched in the mPFC, and its dysregulation is implicated in neuropsychiatric disorders. However, it is unclear how the Dyn / KOR system modulates excitatory and inhibitory circuits that are integral for mPFC information processing and behavioral control. Here, we provide a circuit-based framework wherein mPFC Dyn / KOR signaling regulates excitation-inhibit

Prone to excitement: adolescent females with Non-suicidal self-injury (NSSI) show altered cortical pattern to emotional and NSS-related material.

Emotion-regulation difficulties have been identified as one of the core components in Non-suicidal self-injury (NSSI), a behaviour often beginning in adolescence. This pilot study evaluated differences in emotion processing between 18 female adolescents with and without NSSI by using verbal responses and functional magnetic resonance imaging (fMRI). Responses to pictures taken from the International Affective Picture System and slides with reference to NSSI were recorded both by verbal rating of valence and arousal and by fMRI. The NSSI group rated pictures with self-injurious reference as sig

Disrupted ERK signaling during cortical development leads to abnormal progenitor proliferation, neuronal and network excitability and behavior, modeling human neuro-cardio-facial-cutaneous and related syndromes.

Genetic disorders arising from copy number variations in the ERK (extracellular signal-regulated kinase) MAP (mitogen-activated protein) kinases or mutations in their upstream regulators that result in neuro-cardio-facial-cutaneous syndromes are associated with developmental abnormalities, cognitive deficits, and autism. We developed murine models of these disorders by deleting the ERKs at the beginning of neurogenesis and report disrupted cortical progenitor generation and proliferation, which leads to altered cytoarchitecture of the postnatal brain in a gene-dose-dependent manner. We show th

Preliminary observations in cortical excitability changes using transcranial magnetic stimulation (TMS), it's correlation with diffusion tensor imaging (DTI) in subjects with neuromyelitis optica spectrum disorder (NMOSD).

BACKGROUND: Neuromyelitis optica spectrum disorder (NMOSD) is a relapsing central nervous system disease most commonly associated with aquaporin-4 antibodies (AQP4-Ab) and Myelin oligodendrocyte glycoprotein (MOG) antibodies. These demyelinating disorders influence cortical excitability, which has been studied using advanced imaging techniques and transcranial magnetic stimulation (TMS) in our study. METHODS: This is a prospective study of 30 subjects. Ten subjects, each of AQP4, MOGAD, and dual seronegative (SN)-NMOSD, were recruited and compared to 30 healthy controls. All the subjects unde…

Synergistic effects of action observation and object affordance on cortical excitability during BCI-triggered motor imagery in stroke: a randomized, within-subject, 2 × 2 factorial study protocol.

BACKGROUND: Brain-computer interfaces (BCI) using motor imagery (MI) offer promising rehabilitation potential for upper limb function in stroke survivors. Conventional paradigms rely on abstract visual feedback, which may lack ecological validity and could limit neural recruitment. Action observation (AO) and object affordance (OA) independently modulate motor cortical excitability via the mirror neuron system and canonical neural pathways. However, their potential synergistic effects in enhancing BCI performance have not been systematically investigated. We propose a novel "Phygital" interven…

Transcranial Direct Current Stimulation Over the Prefrontal Cortex in Depression Modulates Cortical Excitability in Emotion Regulation Regions as Measured by Concurrent Functional Magnetic Resonance Imaging: An Exploratory Study.

BACKGROUND: A well-established impaired top-down network for effortful emotion regulation (ER) in major depressive disorder (MDD) includes the dorsal and ventromedial prefrontal cortex (PFC) and the amygdala. Transcranial direct current stimulation (tDCS) is a noninvasive neuromodulation method that has been used successfully to induce mood changes in MDD. Despite reliable findings, little is known regarding the precise effects of tDCS on cortical excitability in vivo in depression and how such changes relate to ER. Here, we addressed this question by combining-for the first time in a psychiat…
