Increased hydrogen and ethanol production in transformants of the filamentous cyanobacterium Phormidium lacuna.

Research on biofuel production by cyanobacteria has largely focused on unicellular species. To date, there are no reports on engineered filamentous cyanobacteria for bioethanol or biohydrogen production. However, biofilm formation in filamentous cyanobacteria could provide an advantage for biotechnological applications. We used natural transformation and homologous recombination to establish recombinant expression at the psaD locus of the filamentous cyanobacterium Phormidium lacuna. A similar approach has already been successful in the unicellular cyanobacterium Synechocystis sp. PCC 6803 (Ap…

Raman spectroscopic analysis of a desert cyanobacterium Nostoc sp. in response to UVB radiation.

Cyanobacteria are capable of tolerating environmental extremes. To survive in extreme environments, cyanobacteria have developed the capability to adapt to a variety of stresses. For example, cyanobacteria have adopted a number of strategies with which to survive UV stress, including expression of UV-screening pigments and antioxidant systems. We have previously shown that several antioxidants are significantly expressed in Nostoc sp. by UVB irradiation. We report here that the content of UV-responsive biomarkers such as β-carotene and scytonemin can be easily detected by Fourier transform Ram…

Automatic detection of harmful cyanobacterial genera using deep CNN models and artemisinin optimization.

Concerns over the spread of Cyanobacteria, which can lead to dangerous blooms that harm drinking water quality and, therefore, the health of plants and animals, are being raised by global warming. Traditional methods for assessing the amount of toxic species in water samples are often time-consuming, require intensive manual effort, are prone to subjective errors, and can lead to delays in necessary water management interventions. This emphasizes the pressing need for a quick and precise automated method. Both aquatic and terrestrial environments include cyanobacteria, and under some circumsta…

Driving Factors of Geosmin Appearance in a Mediterranean River Basin: The Ter River Case.

In recent decades, human activity coupled with climate change has led to a deterioration in the quality of surface freshwater. This has been related to an increase in the appearance of algal blooms, which can produce organic compounds that can be toxic or can affect the organoleptic characteristics of the water, such as its taste and odor. Among these latter compounds is geosmin, a metabolite produced by certain cyanobacteria that confers an earthy taste to water and which can be detected by humans at very low concentrations (nanogram per liter). The difficulty and cost of both monitoring the …

Okara and okara modified and functionalized with iron oxide nanoparticles for the removal of Microcystis aeruginosa and cyanotoxin.

Eutrophicating compounds promote the growth of cyanobacteria, which has the potential of releasing toxic compounds. Alternative raw materials, such as residues, have been used in efficient adsorption systems in water treatment. The aim of the present study was to apply the residue Okara in its original form and modified by hydrolysis with immobilization of magnetic nanoparticles as an adsorbent. For the removal, the cyanobacteria Microcystis aeruginosa was chosen, as well as its secondary metabolites, L-amino acids leucine and arginine (MC-LR microcystin), from aqueous solutions. The adsorbent…

Cyanobacterial blooms: statistical models describing risk factors for national-scale lake assessment and lake management.

Cyanobacterial toxins constitute one of the most high risk categories of waterborne toxic biological substances. For this reason there is a clear need to know which freshwater environments are most susceptible to the development of large populations of cyanobacteria. Phytoplankton data from 134 UK lakes were used to develop a series of Generalised Additive Models and Generalised Additive Mixed Models to describe which kinds of lakes may be susceptible to cyanobacterial blooms using widely available explanatory variables. Models were developed for log cyanobacterial biovolume. Water colour and …

Efficient utilization of xylose requires CO2 fixation in Synechococcus elongatus PCC 7942.

Cyanobacteria show great promise as autotrophic hosts for the renewable biosynthesis of useful chemicals from CO2 and light. While they can efficiently fix CO2, cyanobacteria are generally outperformed by heterotrophic production hosts in terms of productivity and titer. Photomixotrophy, or co-utilization of sugars and CO2 as carbon feedstocks, has been implemented in cyanobacteria to greatly improve productivity and titers of several chemical products. We introduced xylose photomixotrophy to a 2,3-butanediol producing strain of Synechococcus elongatus PCC 7942 and characterized the effect of …

Prediction of Antibiotic Resistance Genes in Cyanobacterial Strains by Whole Genome Sequencing.

Cyanobacteria are ubiquitous in freshwater environments, but their role in aquatic resistome remains unclear. In this work, we performed whole genome sequencing on 43 cyanobacterial strains isolated from Portuguese fresh/wastewaters. From 43 available non-axenic unicyanoabacterial cultures (containing only one cyanobacterial strain and their co-occurring bacteria), it was possible to recover 41 cyanobacterial genomes from the genomic assemblies using a genome binning software, 26 of which were classified as high-quality based on completeness, contamination, N50 and contig number thresholds. By…

Synechococcus sp. Strain PCC7002 Uses Sulfide:Quinone Oxidoreductase To Detoxify Exogenous Sulfide and To Convert Endogenous Sulfide to Cellular Sulfane Sulfur.

Eutrophication and deoxygenation possibly occur in coastal waters due to excessive nutrients from agricultural and aquacultural activities, leading to sulfide accumulation. Cyanobacteria, as photosynthetic prokaryotes, play significant roles in carbon fixation in the ocean. Although some cyanobacteria can use sulfide as the electron donor for photosynthesis under anaerobic conditions, little is known on how they interact with sulfide under aerobic conditions. In this study, we report that Synechococcus sp. strain PCC7002 (PCC7002), harboring an sqr gene encoding sulfide:quinone oxidoreductase …

Characterization of phosphorus forms in a Eutrophic Lake, China.

In this study, molecular compositions of cyanobacteria, suspended matter, and surface sediments in the Dianchi Lake, a highly eutrophic lake, were investigated by solution and solid-state 31P nuclear magnetic resonance (NMR) spectroscopy. A solution-state 31P NMR spectral analysis of NaOH-EDTA-extracted samples revealed the presence of orthophosphate (ortho-P, 48.5%-91.2% of the total extracted phosphate), orthophosphate monoester (mono-P, 7.3%-43.9%), orthophosphate diester (diester-P, 0.9%-3.9%), and pyrophosphate (pyro-P, 0.7%-5.5%). The organic phosphorus (Po) distribution in suspended mat…

Effects of phosphorus sources on volatile organic compound emissions from Microcystis flos-aquae and their toxic effects on Chlamydomonas reinhardtii.

There is diverse phosphorus (P) in eutrophicated waters, but it is considered as a crucial nutrient for cyanobacteria growth due to its easy precipitation as insoluble salts. To uncover the effects of complex P nutrients on the emission of volatile organic compounds (VOCs) from cyanobacteria and their toxic effects on other algae, the VOCs from Microcystis flos-aquae supplied with different types and amount of P nutrients were analyzed, and the effects of VOCs and their two main compounds on Chlamydomonas reinhardtii growth were investigated. When M. flos-aquae cells were supplied with K2HPO4,…

The facilitating role of phycospheric heterotrophic bacteria in cyanobacterial phosphonate availability and Microcystis bloom maintenance.

BACKGROUND: Phosphonates are the main components in the global phosphorus redox cycle. Little is known about phosphonate metabolism in freshwater ecosystems, although rapid consumption of phosphonates has been observed frequently. Cyanobacteria are often the dominant primary producers in freshwaters; yet, only a few strains of cyanobacteria encode phosphonate-degrading (C-P lyase) gene clusters. The phycosphere is defined as the microenvironment in which extensive phytoplankton and heterotrophic bacteria interactions occur. It has been demonstrated that phytoplankton may recruit phycospheric b…

Optimized extraction and analytical protocols for guanitoxin: an organophosphate cyanobacterial neurotoxin of environmental concern.

This study evaluated cell growth, chlorophyll-a production, and guanitoxin (GNT) synthesis by the cyanobacterium Sphaerospermopsis torques-reginae (strain ITEP-24) in different culture media over 52 days. The maximum total GNT production was observed in Z8 medium, peaking on the 45th day, indicating potential for biotechnological studies. BG-11 with 5% nitrogen offered a good balance between yield and stability, while ASM-1 with 5% nitrogen showed higher GNT production efficiency per cell, suggesting suitability for metabolic studies. An optimized extraction protocol using 70% methanol with 0.

Harmful and beneficial properties of cyanotoxins: Two sides of the same coin.

Cyanotoxins are by definition "harmful agents" produced by cyanobacteria. Their toxicity has been extensively studied and reviewed over the years. Cyanotoxins have been commonly classified, based on their poisonous effects on mammals, into three main classes, neurotoxins, hepatotoxins and dermatotoxins, and, considering their chemical features, mainly identified as peptides, alkaloids and lipopolysaccharides. Here we propose a broader subdivision of cyanotoxins into eight distinct classes, taking into account their molecular structures, biosynthesis and modes of action: alkaloids, non-ribosoma…

Deletion of the gene for a cyanobacterial ribosome-associated protein affects the carbon/nitrogen metabolism.

UNLABELLED: In contrast to their important structural and regulatory functions, such as in the metabolism of cyanobacteria, genes encoding small proteins are often not well characterized. Cyanobacteria use redox equivalents and energy from oxygenic photosynthesis to produce organic carbon compounds from inorganic carbon (C i ) and organic nitrogen compounds from inorganic nitrogen sources. Therefore, the assimilation and metabolism of carbon and nitrogen are coordinated at multiple levels in cyanobacteria. Here, we analyzed the Synechocystis sp. PCC 6803 gene ssr3189 encoding a 55 amino acids …

Adjunctive surgical procedures enhancing treatment outcome - A literature review.

BACKGROUND: Brasilonema is a cyanobacterial genus found on the surface of mineral substrates and plants such as bromeliads, orchids and eucalyptus. B. octagenarum stands out among cyanobacteria due to causing damage to the leaves of its host in an interaction not yet observed in other cyanobacteria. Previous studies revealed that B. octagenaum UFV-E1 is capable of leading eucalyptus leaves to suffer internal tissue damage and necrosis by unknown mechanisms. This work aimed to investigate the effects of B. octagenarum UFV-E1 inoculation on Eucalyptus urograndis and to uncover molecular mechanis…

Critical review of actually available chemical compounds for prevention and management of cyanobacterial blooms.

Cyanobacteria proliferation is among the most threatening consequences of freshwater pollution. Health risks from human and other-organism exposure to cyanobacteria have led to an effort to find practical methods for cyanobacterial water-bloom reduction. Hence, methods and techniques have been developed in order to reduce the amount of phosphorus or to decrease the abundance of nuisance phytoplankton species directly in the water bodies (in-lake measures). Although these "acute" methods do not solve the problem of catchment area eutrophication, they are cheaper, easier to manage, and for some …

Insights into cyanobacterial blooms through the lens of omics.

Cyanobacteria are oxygen-producing photosynthetic bacteria that convert carbon dioxide into biomass upon exposure to sunlight. However, favorable conditions cause harmful cyanobacterial blooms (HCBs), which are the dense accumulation of biomass at the water surface or subsurface, posing threats to freshwater ecosystems and human health. Understanding the mechanisms underlying cyanobacterial bloom formation is crucial for effective management. In this regard, recent advancements in omics technologies have provided valuable insights into HCBs, which have raised expectations to develop more effec…

Molecular insights into the elevator-type mechanism of the cyanobacterial bicarbonate transporter BicA.

Cyanobacteria are responsible for up to 80% of aquatic carbon dioxide fixation and have evolved a specialized carbon concentrating mechanism to increase photosynthetic yield. As such, cyanobacteria are attractive targets for synthetic biology and engineering approaches to address the demands of global energy security, food production, and climate change for an increasing world's population. The bicarbonate transporter BicA is a sodium-dependent, low-affinity, high-flux bicarbonate symporter expressed in the plasma membrane of cyanobacteria. Despite extensive biochemical characterization of Bic…
