In drug discovery, assessing and stratifying drug efficacy on cells is vital. To demonstrate the efficacy of drugs on cells in different cell cycle phases, it is essential not only to analyze multidimensional protein data on the single-cell level but also to establish a methodology for extracting key molecules from subtle differences in cellular states in response to the drug. Here we introduce a single cell-PLOM-CON analysis method by combining multiplex immunofluorescence and image-based covariation network analysis and reveal several changes of proteins in early cell state changes across di

Cancer represents a pathological manifestation of uncontrolled cell division; therefore, it has long been anticipated that our understanding of the basic principles of cell cycle control would result in effective cancer therapies. In particular, cyclin-dependent kinases (CDKs) that promote transition through the cell cycle were expected to be key therapeutic targets because many tumorigenic events ultimately drive proliferation by impinging on CDK4 or CDK6 complexes in the G1 phase of the cell cycle. Moreover, perturbations in chromosomal stability and aspects of S phase and G2/M control media

Liver injury from any number of causes (e.g. chemical material, drugs and diet, viral infection) is a global health problem, and its mechanism is not clearly understood. MicroRNAs (miRNAs) expression profiling is gaining popularity because miRNAs, as key regulators in gene expression networks, can influence many biological processes and have also shown promise as biomarkers for disease. Previous studies reported the regulation effects of miRNAs in liver injury, whereas function and molecular mechanisms of miR-322-5p were still unclear. Therefore, our study focused on the biological role of miR

Twenty-six novel derivatives of paeonol chalcone were synthesized by introducing various hydrophilic aminoalkyl to the 2'-hydroxy of paeonol. Their antiproliferative activity was evaluated by MTT assay against five human cancer cell lines (HeLa, HepG2, U2OS, HCT-116 and A549). The majority of these compounds demonstrated remarkable cytotoxicity against the tested cancer cells. Among them, compound 4k exhibited the most potent antiproliferative activity (IC50 values ranging from 1.31 μM to 7.56 μM for the tested cancer cells), stronger than 5-FU (IC50 = 6.29-14.56 μM). Notably, it effectively i

High-grade serous ovarian carcinoma (HGSOC) remains a lethal malignancy with few effective therapeutic options. In this study, we systematically evaluated the anti-tumor effect of Bi2536, an inhibitor of Polo-like kinase 1 (PLK1), in HGSOC, and clarified its mechanism. Bi2536 inactivates PLK1, leading to the subsequent inactivation of cyclin-dependent kinase 1 (CDK1). This disruption triggers a cascade of antitumor effects, including G2/M phase arrest, induction of mitochondrial apoptosis, and suppression of cell migration and invasion. Furthermore, we identified circadian oscillations in PLK1

In this study, emodin was used as the starting material to synthesize two novel ligands, L1 and L2, containing bipyridine or phenanthroline moieties. Seven metal complexes (1-7) were obtained through coordination with Co, Rh, Ru, and Pt ions. Their structures were confirmed via NMR, HRMS, UV-Vis, IR, HPLC and SC-XRD analyses. The MTT assay showed that L1 and complex 2 selectively inhibited HepG-2 cells, with IC50 values of 8.77 μM and 6.10 μM, respectively. Both compounds exhibited stronger activity than cisplatin (9.05 μM) and low toxicity toward normal 293T cells. Mechanistic studies demonst

Cell division cycle 25 (CDC25) phosphatases are critical activators of cyclin-dependent kinases (CDKs) and guardians of genome integrity, making them attractive anticancer targets. Building on the quinoline-5,8-dione scaffold NSC663284 (6a), we synthesized derivatives with diverse C-6/C-7 alkylamino side chains. Structure-activity studies identified D3a/D3b (2-(4-methylpiperidin-1-yl)ethylamino) and D11a/D11b (2-(dimethylamino)ethylamino) as the most potent, exhibiting low-submicromolar inhibition of CDC25. In cell-based assays, these compounds suppressed leukemia (IC50 0.21-1.22 μM) and color

Mantle cell lymphoma (MCL) is typically an aggressive and rare form of non-Hodgkin lymphoma (NHL) with a poor prognosis despite recent advances in immunochemotherapy and targeted therapeutics against NHL. New therapeutic agents are needed for MCL. In this study, we generated a potent inhibitor of cyclin-dependent kinase 7 (CDK7), designated QS1189, and confirmed its anti-cancer effects towards MCL and other lymphomas. QS1189 was highly selective for CDK7 and showed potent anticancer effects in MCL compared to other targeted therapeutic agents, such as ibrutinib and venetoclax. Consistent with

T-cell acute lymphoblastic leukemia (T-ALL) is a heterogeneous disease of the blood affecting children, adolescents and adults. Although current treatment protocols for T-ALL have improved overall survival, a portion of T-ALL patients still experiences treatment failure. Thus, the development of novel therapies is needed. In this study, we used several patient-derived T-ALL cell lines to screen for an effective drug for T-ALL. Using a panel of 378 inhibitors against different kinases, we identified the CDK inhibitor dinaciclib as a potential drug for T-ALL. Dinaciclib treatment significantly r

Hepatocellular carcinoma (HCC) accounts for approximately 90% of all cases of primary liver cancer, and the majority of patients with HCC are deprived of effective curative methods. Osthole is a Chinese herbal medicine which has been reported to possess various pharmacological functions, including hepatocellular protection. In the present study, we investigated the anticancer activity of osthole using HCC cell lines. We found that osthole inhibited HCC cell proliferation, induced cell cycle arrest, triggered DNA damage and suppressed migration in HCC cell lines. Furthermore, we demonstrated th

Intracellular lipid droplets (LDs) are ubiquitous in several cell types. LDs store neutral lipids such as triacylglycerol and cholesteryl ester and play important roles in energy metabolism, signaling, and stress responses. Although LDs are recognized as intracellular organelles, their behavior during cell cycle progression remains unclear. During mitosis, several organelles, including mitochondria and the Golgi apparatus, undergo extensive remodeling and subsequent reassembly, raising the question whether LDs also undergo cell cycle-dependent changes. Since LDs can serve as energy sources and

Cell cycle-dependent maintenance of centromere protein A (CENP-A) levels and its spatiotemporal assembly are essential for centromere propagation. CENP-A synthesis peaks in late G2, while its assembly occurs during late telophase/early G1. We have previously shown that phosphorylation of CENP-A at Ser68 by CDK1-cyclin B during mitosis impairs its binding to holiday junction recognition protein (HJURP) and facilitates DCAF11-dependent polyubiquitination and degradation. However, the mechanisms governing CENP-ApS68 stability remain elusive. Here, we demonstrate that spindlin interactor and repre

Aging is a major contributor to the increasing incidence of acute kidney injury (AKI) and chronic kidney disease (CKD), however the molecular events explaining aging-related nephropathies remains elusive. Here, we found that the RAS signaling adapter protein Erbin (ErbB2-interacting protein) is abundantly expressed in renal tubular cells, but its levels are decreased in murine AKI models and human CKD patient samples. Tubule-specific deletion of Erbin rendered mice highly susceptible to acute kidney injury, and exacerbated renal fibrosis. In contrast, restoring Erbin protected against AKI and

Premature ovarian failure (POF) is closely linked to ovarian granulosa cell (OGC) senescence and cell cycle arrest. This study explored the role of the miR-322/Klotho axis in OGC aging. Using a cyclophosphamide-induced mouse model and cultured OGCs, we found Klotho significantly downregulated in POF, inversely correlating with elevated miR-322 and senescence markers p16, p21, and p53. Luciferase assays confirmed Klotho as a direct miR-322 target. miR-322 overexpression induced G0/G1 and G2/M phase arrest and OGC senescence by silencing Klotho, while miR-322 inhibition partially rescued the POF

Given the profound multiple myeloma (MM) heterogeneity in clonal proliferation of malignant plasma cells (PCs) and anti-MM therapeutic potential of nanotherapies, it is inevitable to develop treatment plan for patients with MM. Two composite nanoparticles (NPs), As4S4/Fe3O4 (4:1) and As4S4/Fe3O4 (1:1) demonstrated effective anti-MM activity in in vitro, ex vivo, and in vivo in xenograft mouse model. Composite NPs triggered activation of p-ERK1/2/p-JNK, and downregulation of c-Myc, p-PI3K, p-4E-BP1; G2/M cell cycle arrest with increase in cyclin B1, histones H2AX/H3, activation of p-ATR, p-Chk1

A series of novel 9-N-substituted-13-alkylberberine derivatives from Chinese medicine were designed and synthesized with improved anti-hepatocellular carcinoma (HCC) activities. The optimal compound 4d showed strong activities against HepG2, Sk-Hep-1, Huh-7 and Hep3B cells with IC50 values of 0.58-1.15 μM, which were superior to positive reference cisplatin. Interestingly, 4d exhibited over 40-fold more potent activity against cisplatin-resistant HepG2/DPP cells while showing lower cytotoxicity in normal LX-2 cells. The mechanism studies revealed 4d greatly stabilized G-quadruplex DNA leading
