Proteomic-based prediction of functional bioactive peptides in proteins extracted from Torreya grandis using choline chloride-based deep eutectic solvents.

Five deep eutectic solvents were used to extract T. grandis nut protein, with water as comparison. ChCl:Urea was the most effective, preserving integrity and functionality of extracted proteins. Cold ethanol purification yielded highest protein recovery rate 6.89 % ± 0.12 %. Time, temperature, and solid-liquid ratio, were optimized using response surface methodology and a Box-Behnken design. Extracted proteins were characterized via SDS-PAGE to assess digestibility and functional properties. Proteins extracted by DES were further analyzed via LC-MS/MS. De novo sequencing identified 600 peptide…

Establishing the constitutive relationship between deep eutectic solvents pre-treatment extent, cellulose structure characteristics, and dissolution efficiency in lyocell processing.

Conventional dissolution of cellulose fibers in N-methylmorpholine-N-oxide (NMMO) solution necessitates harsh conditions including high temperature and prolonged dissolution time, frequently resulting in inhomogeneous dissolution and inferior product characteristics. Herein, a facile pre-treatment strategy was developed to remarkably improve the dissolution efficiency by multiple concentration gradients of ZnCl₂/H3PO4 solvents. The novelty of this work lies in the establishment of the constitutive relationship between deep eutectic solvents (DES) pre-treatment extent, cellulose structure chara…

Spontaneous formation of highly concentrated phenylacetylcarbinol via in situ phase separation in deep eutectic solvents using Candida magnoliae whole cells.

Deep eutectic solvents (DESs) are promising green media for biocatalysis, yet their application in pyruvate decarboxylase (PDC)-mediated phenylacetylcarbinol (PAC) biotransformation with product separation remains limited. This study screened nine DESs using frozen-thawed whole cells (FT-WHC) of Candida magnoliae (C. mag.), followed by evaluation of water content and DES to phosphate buffer ratios. Glycerol-based DES resulted in significantly higher (p ≤ 0.05) volumetric PDC activity and PAC productivity than other DESs. Choline chloride: glycerol (ChCl: Gly (1:2) 20% (v/v) H2O) achieved the h…

Hydrophobic natural deep eutectic solvents as a green desorption media for parabens from solid sorbents: A proof-of-concept study.

A sustainable sample preparation method was developed to determine the concentrations of four parabens (methyl, ethyl, n-propyl, and n‑butyl paraben) in human urine. Initial experiments evaluated three sorbents, C18, PSA, and Florisil®, combined with different methanol and water (MeOH:H₂O) elution strategies. The best performance was achieved using 50 mg of C18 with 500 µL of MeOH:H₂O (80:20, v/v), which was selected as the reference protocol. The method was then redesigned to incorporate a hydrophobic natural deep eutectic solvent (H-NADES) composed of DL-menthol and acetic acid (MAc) in a 1:

Enantioselective Catalytic Aldol Reactions in the Presence of Knoevenagel Nucleophiles: A Chemoselective Switch Optimized in Deep Eutectic Solvents Using Mechanochemistry.

In the presence of different nucleophilic Knoevenagel competitors, cyclic and acyclic ketones have been shown to undergo highly chemoselective aldol reactions with aldehydes. In doing so, the substrate breadth for this emerging methodology has been significantly broadened. The method is also no longer beholden to proline-based catalyst templates, e.g., commercially available O-t-Bu-L-threonine is advantageous for acyclic ketones. The key insight was to exploit water-based mediums under conventional (in-water) and non-conventional (deep eutectic solvents) conditions. With few exceptions, high a…

Recent advances in AI-driven deep eutectic solvent design for biocatalysis: Co-design of extremozyme and reaction medium.

Deep eutectic solvents (DES) have emerged as sustainable, highly tunable alternatives to conventional organic media for non-aqueous biocatalysis. DES dynamically interact with enzymes to shift reaction equilibria, enhance thermostability and tailor stereoselectivity. However, the vast combinatorial space and complex physicochemical properties of DES make traditional trial-and-error solvent engineering highly inefficient. This review systematically examines recent advancements in DES-driven biocatalysis and highlights the role of artificial intelligence (AI) for rational DES design. Specificall…
