Assessment of fluoride-infused calcium phosphate resin composites as effective remineralisation agents for human dental pulp stem cells.

OBJECTIVES: This study aimed at developing specific experimental pulp-protection resin-based materials containing bioactive FDCP fillers and assessing their influence on essential functions of human Dental Pulp Stem Cells (hDPSCs), including cytotoxicity, stemness, and cell migration, as well as their activation of mineralisation processes. METHODS: Experimental FDCPCs were synthesized incorporating varying concentrations of calcium and sodium fluoride salts (5, 10 and 20 wt%). A free FDCP filler (R-VS0F) served as the control group. The hDPSCs were exposed to eluates from these materials to

Maturogenesis of an Early Erupted Immature Permanent Tooth: A Case Report With 7-Year Follow-Up.

The main objective of treatment of an undeveloped tooth (immature) is to provide vital pulp therapy to allow continued development of root dentin. A case report is presented that demonstrates the use of calcium hydroxide (CaOH₂) as an indirect pulp-copping material for the purpose of continued maturogenesis of an early-erupted permanent tooth with severe mobility and almost-begun root formation. Seven-year radiographic and clinical follow-up demonstrated a vital pulp and physiologic root development without any endodontic failure clinically or radiographicaly.

Dental-Dedicated MRI in the Assessment of Pulp Vitality and Apical Periodontitis: A Feasibility Study.

INTRODUCTION: This feasibility study presents the diagnostic accuracy of a novel dental-dedicated magnetic resonance imaging (ddMRI) system for assessing pulp vitality and diagnosing apical periodontitis (AP), as an adjunct to clinical assessment and cone beam CT (CBCT) imaging. METHODS: Ten consecutive patients with possible endodontic problems were screened, and ultimately 18 teeth in nine patients (10 healthy teeth, five necrotic teeth with suspected AP and three necrotic or root canal treated teeth with no suspicion of AP) with a recent CBCT were included. The teeth were tested for vitali

A clinical study of direct pulp capping applied to carious-exposed pulps.

Direct pulp capping of carious-exposed pulp was performed on 44 teeth. We evaluated the success rates of these cases, and analyzed the relationships between the success rates and their clinical findings. Furthermore, we examined the length of time necessary for adequate postoperative follow-up. The success rate in this study was 81.8%. Age of the patients, type of teeth, responses to thermal stimuli and percussion, and the diameter of pulpal exposure had no bearing on the success rate. However, the degree of bleeding on pulpal exposure was related to the success rate (p = 0.042). The success r

Characterisation of Roseomonas mucosa isolated from the root canal of an infected tooth.

OBJECTIVE: The genus Roseomonas comprises a group of pink-pigmented, slow-growing, aerobic, non-fermentative Gram-negative bacteria, which have been isolated from environmental sources such as water and soil, but are also associated with human infections. In the study presented here, Roseomonas mucosa was identified for the first time as part of the endodontic microbiota of an infected root canal and characterised in respect to growth, antibiotic susceptibility and biofilm formation. RESULTS: The isolated R. mucosa strain showed strong slime formation and was resistant to most β-lactam antibi

Effect of vasoconstriction by dexmedetomidine in the mandible of rat.

BACKGROUND: In recent years, dexmedetomidine (DEX) has been proposed as a useful vasoconstrictor for local anesthesia because it is less effective in circulation than clonidine of antihypertensive drugs. In addition, DEX is expected to act as a vasoconstrictor during local anesthesia. However, histomorphometric studies demonstrating that DEX exerts vasoconstrictive effects are lacking. This study aimed to clarify whether DEX exerts a histomorphologically vasoconstrictive effect on blood vessels in the mandible of rats. METHODS: A total of 12 male Wistar rats were used. General anesthesia was

Detection of 400-year-old Yersinia pestis DNA in human dental pulp: an approach to the diagnosis of ancient septicemia.

Ancient septicemic plague epidemics were reported to have killed millions of people for 2 millenniums. However, confident diagnosis of ancient septicemia solely on the basis of historical clinical observations is not possible. The lack of suitable infected material has prevented direct demonstration of ancient septicemia; thus, the history of most infections such as plague remains hypothetical. The durability of dental pulp, together with its natural sterility, makes it a suitable material on which to base such research. We hypothesized that it would be a lasting refuge for Yersinia pestis, th

Electrical Pulp Testing: Sources of Error.

INTRODUCTION: Accurate identification and assessment of an inflamed or necrotic tooth is essentialfor endodontic treatment. The purpose of this research was to investigate possible sources of error associated with the use of the electric pulp tester (EPT). METHODS: Forty-six intact teeth (23 tooth pairs) in 22 patients were evaluated in vivo. For the tooth pairs, one tooth had to have been previously endodontically treated and restored with a class II amalgam restoration. The restoration was required to have proximal contact with a class II amalgam of another vital posterior tooth. EPT was pe

IL-1 alpha and TNF-alpha expression in rat periapical lesions and dental pulp after unilateral sympathectomy.

OBJECTIVES: Apical periodontitis is an inflammatory disease characterized by bone resorption, and sympathetic nerves are known to modulate bone resorption and bone remodeling. Higher numbers of osteoclasts and larger periapical lesions have been observed after sympathectomy in rats, but the mechanisms underlying the inhibitory effect of sympathetic nerves on osteoclasts are unknown. This study aimed to test the hypothesis that sympathetic nerves inhibit the production of the bone-resorbing pro-inflammatory cytokines IL-1 alpha and TNF-alpha in rat periapical lesions. METHODS: Rats were unilat

A Miniature Swine Model for Stem Cell-Based De Novo Regeneration of Dental Pulp and Dentin-Like Tissue.

The goal of this study was to establish mini-swine as a large animal model for stem cell-based pulp regeneration studies. Swine dental pulp stem cells (sDPSCs) were isolated from mini-swine and characterized in vitro. For in vivo studies, we first employed both ectopic and semi-orthotopic study models using severe combined immunodeficiency mice. One is hydroxyapatite-tricalcium phosphate (HA/TCP) model for pulp-dentin complex formation, and the other is tooth fragment model for complete pulp regeneration with new dentin depositing along the canal walls. We found that sDPSCs are similar to thei

Restoring Cell-Cell Junctions in DSPP-Deficient Odontoblasts Through Nanofibrous Topography and Wnt5a-Cdc42 Activation: A Laboratory Investigation.

AIM: Biomaterial-based strategies offer therapeutic potential for inherited disorders such as Dentinogenesis imperfecta type II (DGI-II). DGI-II, a hereditary dental disorder caused by dentine sialophosphoprotein (Dspp) gene mutations, results in fragile, discoloured teeth susceptible to wear and decay. This study investigates the structural alterations in the dentine matrix induced by mutant DSPP and the underlying molecular mechanisms. Furthermore, we evaluated a therapeutic strategy combining nanofibrous substrates with molecular activation of the Wnt5a-Cdc42 pathway, aiming to mitigate the
