Comparative serum biochemical changes induced by experimental infection of T. brucei and T. congolense in pigs.

A comparative evaluation of the serum biochemical parameters was carried out in groups of young pigs aged 3-5 months experimentally infected with single infection of Trypanosoma brucei, Trypanosoma congolense, and a mixed infection of the two species. All the parameters studied (alanine amino transferase, aspartate amino transferase, albumin, globulin, cholesterol and creatinine) with the exception of total protein and urea varied significantly (p<0.05) between the infected groups and uninfected control group. Serum concentrations of alanine amino transferase, aspartate amino transferase, crea

Increased erythrophagocytic activity of macrophages in dogs with Babesia gibsoni infection.

To elucidate the mechanism of anemia caused by Babesia gibsoni infection in dogs, the erythrophagocytic activity of macrophages in infected dogs was investigated in vitro. In the present study, macrophages obtained from peripheral blood (PB-macrophages) and bone marrow (BM-macrophages) of splenectomized dogs with chronic B. gibsoni infection were examined. The BM-macrophages in the splenectomized dogs with chronic babesiosis exhibited an increased erythrophagocytic activity compared with those from splenectomized, non-infected dogs. In the infected dogs, erythrophagocytic activities of macroph

Trypanocidal action of tea tree oil (Melaleuca alternifolia) against Trypanosoma evansi in vitro and in vivo used mice as experimental model.

This study aimed to evaluate the Trypanosoma evansi susceptibility to tea tree oil (TTO - Melaleuca alternifolia) and tea tree oil nanocapsules (TTO nanocapsules) in vitro and in vivo tests. In vitro, we observed a mortality curve of trypomastigotes proportional to dose, i.e., the TTO and TTO nanocapsules have trypanocidal effect. Treatment with TTO in vivo was assessed in experiments (I and II). For Experiment I, T. evansi infected mice were treated with TTO and/or combinations of essential oil with chemotherapy (diminazene aceturate - D.A.). Treatment with TTO at a dose of 1mLkg(-1) was able

Babesiosis in the Greyhound.

Babesiosis was diagnosed in five 11- to 18-day old Greyhound pups. In 3 pups, Babesia canis organisms were identified by examination of a Wright's-Giemsa-stained smear of blood. In 2 pups, the diagnosis was established by examination of a splenic impression smear obtained at necropsy. Findings in the 3 clinical cases included depression, weakness, anorexia, pallor, anemia, and thrombocytopenia. Subcutaneous administration of diminazene aceturate resulted in rapid clinical recovery in these cases. In the 2 pups that were necropsied, variable numbers of erythrocytes containing Babesia organisms

Antitrypanosomal potential of Salvia officinalis terpenoids-rich fraction in Trypanosoma evansi-infected rat model.

UNLABELLED: BACKGROUND TRYPANOSOMA EVANSI : (T. evansi) is a major protozoan disease that affects animals, including camels, and causes substantial economic detriments. The failure to control T. evansi infections is due to the unavailability of vaccines and the development of resistance to existing chemical drugs. In this study, we evaluated the effect of Salvia officinalis terpenoids-rich fraction on the degree of parasitemia and associated pathological alterations in rats experimentally infected with T. evansi. METHOD: Eighty adult male rats were equally divided into 4 groups. The first gro

In vitro screening of novel anti-Babesia gibsoni drugs from natural products.

Babesia gibsoni is a tick-transmitted intraerythrocytic apicomplexan parasite that causes babesiosis in dogs. Due to the strong side effects and lack of efficacy of current drugs, novel drugs against B. gibsoni are urgently needed. Natural products as a source for new drugs is a good choice for screening drugs against B. gibsoni. The current study focuses on identifying novel potential drugs from natural products against B. gibsoniin vitro. Parasite inhibition was verified using a SYBR green I-based fluorescence assay. A total of 502 natural product compounds were screened for anti-B. gibsoni

Trypanosomiasis 'risk' or 'challenge': a review.

Definitions of the term 'challenge' as applied to the African trypanosomiases are reviewed. Data from one West and one East African site show simple linear relationships between the incidence of trypanosomiasis in both humans and animals, and either the amount of man-tsetse contact, or the Apparent Density of flies. Data from a number of East African sites are analysed and show a linear relationship between the mean Berenil Index of cattle and the logarithm of the challenge, where challenge is the simple product of Apparent Density and mean fly infection rate. Apparent Density is a more variab

Novel flavanones with anti-trypanosomal activity isolated from Zambian and Tanzanian propolis samples.

A bioassay-guided phytochemical investigation of propolis samples from Tanzania and Zambia that screened for activity against Trypanosoma brucei has led to the isolation of two novel flavanones with promising antitrypanosomal activity. The compounds were characterized based on their spectral and physical data and identified as 6-(1,1-dimethylallyl) pinocembrin and 5-hydroxy-4″,4″-dimethyl-5″-methyl-5″-H-dihydrofuranol [2″,3″,6,7] flavanone. The two compounds, together with the propolis extracts and fractions, were assayed against a standard drug-sensitive strain of T. b. brucei (s427 wild-type

A luciferase based viability assay for ATP detection in 384-well format for high throughput whole cell screening of Trypanosoma brucei brucei bloodstream form strain 427.

BACKGROUND: Human African Trypanosomiasis (HAT) is caused by two trypanosome species, Trypanosoma brucei rhodesiense and Trypanosoma brucei gambiense. Current drugs available for the treatment of HAT have significant issues related to toxicity, administration regimes with limited effectiveness across species and disease stages, thus there is a considerable need to find alternative drugs. A well recognised approach to identify new drug candidates is high throughput screening (HTS) of large compound library collections. RESULTS: We describe here the development of a luciferase based viability a

Drug delivery systems in the treatment of African trypanosomiasis infections.

INTRODUCTION: Animal African trypanosomiasis (AT) is treated and controlled with homidium, isometamidium and diminazene, whereas human AT is treated with suramin, pentamidine, melarsoprol and eflornithine (DFMO), or a combination of DFMO and Nifurtimox. Monotherapy can present serious side effects, for example, melarsoprol, the more frequently used drug that is effective for both hemolymphatic and meningoencephalic stages of the disease, is so toxic that it kills 5% of treated patients. These treatments are poorly efficient, have a narrow safety index and drug resistance is a growing concern.

Genotypic and phenotypic characterization of Trypanosoma brucei gambiense isolates from Ibba, South Sudan, an area of high melarsoprol treatment failure rate.

Resistance of trypanosomes to melarsoprol is ascribed to reduced uptake of the drug via the P2 nucleoside transporter. The aim of this study was to look for evidence of drug resistance in Trypanosoma brucei gambiense isolates from sleeping sickness patients in Ibba, South Sudan, an area of high melarsoprol failure rate. Eighteen T. b. gambiense stocks were phenotypically and only 10 strains genotypically characterized. In vitro, all isolates were sensitive to melarsoprol, melarsen oxide, and diminazene. Infected mice were cured with a 4 day treatment of 2.5mg/kg bwt melarsoprol, confirming tha

The impact of canine African trypanosomiasis in anti-poaching dogs in the Republic of Zambia.

Zambia introduced canine units in 2014 as an anti-poaching tool. Working in challenging environments, these dogs face several health threats, one being the protozoal, vector-borne canine African trypanosomiasis (CAT). This report aims to investigate the incidence, management and impact of CAT on Zambia's canine units. Case data during 2019-2023 were collated to investigate incidence and clinical trends. A questionnaire examining mitigation methods and perceived impact of the disease was distributed. Participants, conservation organisations with canine units, were identified using non-probabili
