Neural regulation of cancer: from microenvironmental hijacking to systemic circuit co-option.

The nervous system is increasingly recognized as an active regulator of tumor biology rather than a passive mediator of cancer-associated symptoms. Growing evidence supports a hierarchical framework of neuro-tumor interactions encompassing chemical, structural, and electrical levels, through which bidirectional communication influences tumor initiation, progression, metastasis, and therapeutic response. At the chemical level, neurotransmitters and neuropeptides regulate tumor cell proliferation, invasion, immune modulation, and stemness. At the structural level, neurogenesis and tumor-nerve in

Preparation and characterization of neural stem cell-loaded conductive hydrogel cochlear implant electrode coatings.

Sensorineural deafness is a hearing impairment resulting from damage to the auditory nerve or inner ear hair cells. Currently, cochlear implants (CIs) are widely used as hearing aids for sensorineural deafness patients. A fundamental limitation of cochlear implants (CIs) is that spiral ganglion neurons (SGNs) cannot be replenished. This greatly restricts the rehabilitation of sensorineural deafness. Additionally, the insertion of CIs can cause secondary cochlear damage, worsening the condition of the patients' cochlear. Therefore, a new type of neural stem cells (NSCs) loaded graphene oxide-po

The sympathetic innervation of the heart: Important new insights.

Autonomic control of the heart has a significant influence over development of life threatening arrhythmias that can lead to sudden cardiac death. Sympathetic activity is known to be upregulated during these conditions and hence the sympathetic nerves present a target for treatment. However, a better understanding of the anatomy and physiology of cardiac sympathetic nerves is required for the progression of clinical interventions. This review explores the organization of the cardiac sympathetic nerves, from the preganglionic origin to the postganglionic innervations, and provides an overview o

Effects of direct current stimulation on synaptic plasticity in a single neuron.

BACKGROUND: Transcranial direct current stimulation (DCS) has lasting effects that may be explained by a boost in synaptic long-term potentiation (LTP). We hypothesized that this boost is the result of a modulation of somatic spiking in the postsynaptic neuron, as opposed to indirect network effects. To test this directly we record somatic spiking in a postsynaptic neuron during LTP induction with concurrent DCS. METHODS: We performed rodent in-vitro patch-clamp recordings at the soma of individual CA1 pyramidal neurons. LTP was induced with theta-burst stimulation (TBS) applied concurrently

Current and Future Neuropharmacological Options for the Treatment of Essential Tremor.

BACKGROUND: Essential Tremor (ET) is likely the most frequent movement disorder. In this review, we have summarized the current pharmacological options for the treatment of this disorder and discussed several future options derived from drugs tested in experimental models of ET or from neuropathological data. METHODS: A literature search was performed on the pharmacology of essential tremors using PubMed Database from 1966 to July 31, 2019. RESULTS: To date, the beta-blocker propranolol and the antiepileptic drug primidone are the drugs that have shown higher efficacy in the treatment of ET.

Inhibitory modulation of long-term potentiation: evidence for a postsynaptic locus of control.

Long-term potentiation (LTP) of the excitatory synapses of the perforant path onto the granule cells of the fascia dentata was prevented, or greatly reduced in amount, by stimulation of the contralateral hilus, a source of the commissural afferents and an indirect source of granule cell inhibition. The LTP was reduced only when the onset of contralateral stimulation fell within 50 ms before and 1 ms after the onset of the perforant path conditioning train. Over this time interval, the ability of the volley from the contralateral hilus to block granule cell discharge (population spike) caused b

Neurokinin A mimics the slow excitatory postsynaptic current in submucous plexus neurons of the guinea-pig caecum.

Single microelectrode voltage-clamp recordings were made from submucous neurons of the guinea-pig caecum. The slow excitatory postsynaptic current was compared with the currents induced by neurokinin A and substance P. The current induced by neurokinin A (100-300 nM) was associated with a decreased membrane conductance and reversed in polarity between -90 and -100 mV. The neurokinin A current was reduced by Co2+ (1-2 mM), but was not affected by Cs+ (1-2 mM), Ba2+ (10-100 microM) or low Cl- (20-40 mM) solutions. In about 80% of the neurons, the current induced by substance P (100-300 nM) was a

Inputs and outputs of giant neurons B1 and B2 in the buccal ganglia of Helix pomatia: an electrophysiological and morphological study.

The identified giant neurons B1 and B2 in the buccal ganglia of Helix pomatia were studied with electrophysiological and morphological techniques in order to establish a baseline for the study of neuronal communication in a relatively simple nervous system. Different synaptic inputs to neurons B1 and B2 were found to come from neurons of the buccal ganglia and to come via buccal nerves. In the epithelium of the pharynx, fibers of bipolar peripheral neurons could be stained, the central fibers of which probably contribute to the synaptic inputs of neurons B1 and B2. The dendrites of neurons B1

Prolonged time course for vibratory suppression of stretch reflex in the decerebrate cat.

We studied the effects of longitudinal tendon vibration on the stretch reflex of the soleus and gastrocnemius muscles in 11 decerebrate cats. Vibration was applied at amplitudes (40-80 micrometer) and frequencies (120-250 HZ) sufficient to provide a strong tonic vibration reflex. In keeping with previous reports, we found that during an established tonic vibration reflex, the force and emg response to superimposed ramp and hold stretch are largely suppressed. This suppression is most obvious during the dynamic phase of stretch where it gives rise to a complex force response resembling that of

[Heterosynaptic interactions in fields CA1 and CA3 of the hippocampus in vitro].

Heterosynaptic interactions between the perforant path and Schaffer's collaterals in the field CA1 and between the perforant path and dentate mossy fibres in the field CA3 were investigated in guinea pig hippocampal slices. Using the method of paired stimuli, space-time summation (for 20--50 ms) was observed in both systems with stimuli sub-threshold for spike generation. Spike responses of the neurons to testing stimulation of afferents synapsing upon the terminal parts of apical dendrites (perforant path) were depressed after spike discharge to conditioning stimulation of proximal afferents

Stable Synapse-Like Memory Switching in N-Heterocyclic Carbene Monolayers.

We report a robust redox-active N-heterocyclic carbene (NHC) monolayer that exhibits synapse-like behavior driven by proton-coupled electron transfer (PCET). Our quinone-functionalized NHC (Rex-NHC) forms densely packed, upright self-assembled monolayers (SAMs) on Au, confirmed by cyclic voltammetry, x- ray photoelectron spectroscopy, sum-frequency generation spectroscopy, and infrared reflection absorption spectroscopy. Molecular junctions built as Au-Rex-NHC//Ga2O3/EGaIn operate over ± 2 V and can withstand electric fields up to 3.3 GV/m. Bias-induced PCET toggles between quinone (off) and h

Carotid body gap junctions: secretion of transmitters and possible electric coupling between glomus cells and nerve terminals.

It is proposed that intercellular coupling between glomus cells and carotid nerve terminals form an integral part of the chemoreceptor process. Coupling is possible because gap junctions occur between these elements. At rest, most glomus cells would be coupled. Stimuli uncouple (or reduce coupling) most glomus cells that extrude their contents toward the nerve terminals. However, other glomus cells do not secrete but recharge and intercellular coupling increases. These phenomena would allow for sustained chemoreceptor activity during prolonged stimulation. Coupling between glomus and sustentac

The interpretation of electromyographic responses to electrical stimulation of the motor cortex in diseases of the upper motor neurone.

The complexities of interpreting results of electrical stimulation of the motor cortex in pathological states are discussed and illustrated by reference to results from a variety of patients with diseases affecting the upper motor neurone (multiple sclerosis, cervical spondylosis and myelopathy, motor neurone disease, hemiparesis due to cerebral infarction, and hereditary spastic paraplegia). The abnormalities of the electromyographic (EMG) responses after anodal cortical stimulation consisted of delay in the latency to onset, dispersion or reduction in response size or even absence of EMG res

Fiber-type-specific architecture and pathophysiology of the neuromuscular junction.

The neuromuscular junction (NMJ) is a specialized synapse essential for translating neuronal signals into muscle contraction. This review examines the complex structural, functional, and molecular differences in NMJs that innervate fast- and slow-twitch skeletal muscle fibers. Fast-twitch fibers, optimized for rapid and powerful contractions, possess elaborate NMJs with deep folds, high neurotransmitter turnover, and greater vulnerability to synaptic fatigue and degeneration. In contrast, slow-twitch fiber NMJs exhibit simpler but more stable architectures that support sustained, fatigue-resis

The Quantum Tunneling of Ions Model Can Explain the Pathophysiology of Tinnitus.

Tinnitus is a well-known pathological entity in clinical practice. However, the pathophysiological mechanisms behind tinnitus seem to be elusive and cannot provide a comprehensive understanding of its pathogenesis and clinical manifestations. Hence, in the present study, we explore the mathematical model of ions' quantum tunneling to propose an original pathophysiological mechanism for the sensation of tinnitus. The present model focuses on two major aspects: The first aspect is the ability of ions, including sodium, potassium, and calcium, to depolarize the membrane potential of inner hair ce

Effects of Focal Ionizing Radiation of the Squid Stellate Ganglion on Synaptic and Axonal Transmission in the Giant-Fiber Pathway.

Ionizing radiation is clinically used to treat neurological problems and reduce pathological levels of neural activity in the brain, but its cellular-level mechanisms are not well understood. Although spontaneous and stimulated synaptic activity has been produced in rodents by clinically and environmentally relevant doses of radiation, the effects on basic excitability properties of neurons have seldom been reported. This study examined the effects of focused ionizing radiation on synaptic transmission and action potential generation in the squid giant-fiber system, which includes the giant sy

Integrative systems and functional analyses reveal a role of dopaminergic signaling in myelin pathogenesis.

BACKGROUND: Myelin sheaths surrounding axons are critical for electrical signal transmission in the central nervous system (CNS). Diseases with myelin defects such as multiple sclerosis (MS) are devastating neurological conditions for which few effective treatments are available. Dysfunction of the dopaminergic system has been observed in multiple neurological disorders. Its role in myelin pathogenesis, however, is unclear. METHODS: This work used a combination of literature curation, bioinformatics, pharmacological and genetic manipulation, as well as confocal imaging techniques. Literature

The role of reward pathways in the development of drug dependence.

In commenting on the discovery of "opiate" receptors, Goldstein (1976) said: "It seemed unlikely, a priori, that such highly stereospecific receptors should have been developed by nature to interact with alkaloids from the opium poppy" (p. 1081). Endogenous opioid peptides and opioid receptor systems have now been identified in invertebrates that are unlikely to have had ancestors exposed to opium poppies (Kavaliers et al., 1983; Kream et al., 1980; Leung and Stefano, 1984; Stefano et al., 1980). Moreover, endogenous opioids play a role in stress-induced feeding in the slug (Kavaliers and Hirs
