High richness of ungulate Sarcocystis species in intestines of the grey wolf (Canis lupus) from Lithuania.

There is a lack of studies on the richness of Sarcocystis species in naturally infected grey wolves that serve as definitive hosts of these parasites. In this work, we used a nested PCR approach to examine the intestines of 13 grey wolves from Lithuania for the presence of Sarcocystis species that use canids as definitive host and ungulates as intermediate host. Under a light microscope, Sarcocystis spp. sporocysts/oocysts were found in 12 animals (92.3%). Based on comparisons of newly acquired cytochrome c oxidase subunit I (cox1) gene nucleotide sequences, we identified a total of 12 differe

Molecular Identification of Onchocerciasis Vectors (Diptera: Simuliidae) from the Central Himalayan Landscape of India: A DNA Barcode Approach.

Background: Black flies (Diptera: Simuliidae) are a notorious group of blood-sucking insects acting as vectors of various diseases in humans and other animals, most notable being Onchocerciasis. Due to its medical and veterinary significance, accurate and quick species identification is of utmost importance in the field of black fly research. DNA barcoding is one such taxonomic tool, aiding in quick and efficient species identification using molecular methods. Despite sporadic reports of ocular and cutaneous Onchocerciasis, especially from North-East India, Indian Simuliidae has been understud

Molecular phylogeny of Psoroptes ovis isolated from ear canker in a domestic rabbit from South India.

BACKGROUND: Ear canker in domestic rabbits is caused by infestations of non-burrowing parasitic mites, Psoroptes spp., but the specific species responsible for these infestations remains unclear. This study reports the clinical signs and performs the molecular characterization and phylogenetic analysis of Psoroptes ovis isolated from the ear canal of a domestic rabbit in South India. METHODS AND RESULTS: A one-year-old rabbit from a farm that housed both goats and rabbits presented with thick, grayish-white debris in the ear canal for the past week. Clinical examination revealed crusty otitis

Screening System of Cannabis sativa Extracts Based on Their Mitochondrial Safety Profile Using Cytochrome c Oxidase Activity as a Biomarker.

The development of Cannabis sativa strains with high cannabidiol (CBD) and low tetrahydrocannabinol (THC) content is a growing field of research, both for medical and recreational use. However, the mechanisms behind clinical actions of cannabinoids are still under investigation, although there is growing evidence that mitochondria play an important role in many of them. Numerous studies have described that cannabinoids modulate mitochondrial activity both through activation of mitochondrial cannabinoid receptors and through direct action on other proteins such as mitochondrial complexes involv

Autochthonous transmission of the Indomalayan parasite, Transversotrema patialense, in the Caribbean: Molecular, morphological, and experimental evidence.

The Asian freshwater snail Melanoides tuberculata has been established since the 1960s in the Americas, where it transmits cercariae of a small number of digenetic trematode species from its native range. In 2021-2022, 24 M. tuberculata were discovered shedding transversotrematid cercariae in Puerto Rico, where parasites of this snail have not been previously studied. Adult transversotrematids (in some cases, gravid) were found on field-caught fish and on fish exposed to shedding snails, including on fish species native to Puerto Rico. Adults and cercariae were identified as Transversotrema pa

A Novel MTTK Gene Variant m.8315A>C as a Cause of MERRF Syndrome.

In this study, we report on a novel heteroplasmic pathogenic variant in mitochondrial DNA (mtDNA). The studied patient had myoclonus, epilepsy, muscle weakness, and hearing impairment and harbored a heteroplasmic m.8315A>C variant in the MTTK gene with a mutation load ranging from 71% to >96% in tested tissues. In muscle mitochondria, markedly decreased activities of respiratory chain complex I + III and complex IV were observed together with mildly reduced amounts of complex I and complex V (with the detection of V*- and free F1-subcomplexes) and a diminished level of complex IV holoenzyme. Assessing modulators of cytochrome c oxidase activity in Galleria mellonella larvae.

Caterpillars of the greater wax moth, Galleria mellonella, are shown to be a useful invertebrate organism for examining mitochondrial toxicants (inhibitors of electron transport) and testing putative antidotes. Administration of sodium azide, sodium cyanide, or sodium (hydro)sulfide by intra-haemocoel injection (through a proleg) results in a dose-dependent paralysed state in the larvae lasting from <1 to ~40 min. The duration of paralysis is easily monitored, because if turned onto their backs, the larvae right themselves onto their prolegs once they are able to move again. The efficacy of pu

Molecular phylogeny of Anopheles hyrcanus group (Diptera: Culicidae) based on mtDNA COI.

BACKGROUND: The Anopheles hyrcanus group, which includes at least 25 species, is widely distributed in the Oriental and Palearctic regions. Some group members have been incriminated as vectors of malaria and other mosquito-borne diseases. It is difficult to identify Hyrcanus Group members by morphological features. Thus, molecular phylogeny has been proposed as an important complementary method to traditional morphological taxonomy. METHODS: Based on the GenBank database and our original study data, we used 466 mitochondrial DNA COI sequences belonging to 18 species to reconstruct the molecul

Pseudomonas fluorescens CHA0 can kill subterranean termite Odontotermes obesus by inhibiting cytochrome c oxidase of the termite respiratory chain.

Pseudomonas fluorescens CHA0 has been shown to suppress the growth of a wide range of microbial plant pathogens as well as invertebrate pests such as root nematodes. Hydrogen cyanide, a secondary metabolite produced by the bacterium, has been credited as being one of the determinants of its biocontrol ability. The use of biocontrol agents against social insect pests such as termite Odontotermes obesus has limitations because of behavioural adaptations that include (1) removal of the pathogen when grooming by the termites and (2) isolation of infested members of the colony. In this study, we sh

On the plausibility of a UV transparent biochemistry.

Some molecules, particularly aromatics, have high molar extinction coefficients at wavelengths in the damaging ultraviolet radiation region of the spectrum between 200 and 400 nm. Thus, under a UV radiation flux in which these wavelengths are represented, it could be argued that a selection pressure would exist for a UV transparent biochemistry in which they were not represented. This hypothesis is explored using data made available from proteomics, focusing particularly on tryptophan, against which a selection pressure could exist on present-day Earth as a result of its absorbance shoulder at

Thermal acclimation, growth, and burst swimming of threespine stickleback: enzymatic correlates and influence of photoperiod.

Threespine sticklebacks (Gasterosteus aculeatus) that had been reared in the laboratory under natural photoperiods were acclimated to 23 degrees and 8 degrees C in late spring under increasing day lengths and again in late fall under decreasing day lengths. The parents of these fish were from the anadromous Isle Verte population. In the spring, cold- and warm-acclimated fish grew at the same rates and attained similar condition factors (mass L(-3)), although food intake was considerably higher at 23 degrees C. As both groups had similar increases in mass and condition, the higher axial muscle
