Cryptic stage of sleeping-sickness trypanosome developing in choroid plexus epithelial cells.

Electronmicrographs of the choroid plexus from rats infected with Trypanosoma brucei rhodesiense showed that trypomastigotes from the perivascular spaces may penetrate and undergo multiple division in the ependymal cells which locally constitute the blood-brain barrier. Progressive degeneration of the ependymal cell liberates trypomastigotes back into the perivascular space, from which re-entry into the blood may occur. Re-entry to the blood does not take place from any tissues other than the brain and its membranes. These findings suggest that the ependymal cells of the choroid plexus are the…

Microsurgical Anterior Transcallosal Resection of Colloid Cysts of the Third Ventricle.

Colloid cysts of the third ventricle are benign and rare lesions of the central nervous system. Although gross total resection is the standa … rgeon over 37 years have been studied and the long-term outcomes have been presented. Additionally, ependyma, choroid plexus, and colloid cyst specimen have been compared using immunohistological staining with cytokeratin, S100, and PAX-7.The long-term outcomes with comparison with the literature show that microsurgical gross total resection using anterior …

The Role of Aquaporins in Spinal Cord Injury.

Edema formation following traumatic spinal cord injury (SCI) exacerbates secondary injury, and the severity of edema correlates with worse neurological outcome in human patients. To date, there are no effective treatments to directly resolve edema within the spinal cord. The aquaporin-4 (AQP4) water channel is found on plasma membranes of astrocytic endfeet in direct contact with blood vessels, the glia limitans in contact with the cerebrospinal fluid, and ependyma around the central canal. Local expression at these tissue-fluid interfaces allows AQP4 channels to play an important role in the …

AAV gene transfer delays disease onset in a TPP1-deficient canine model of the late infantile form of Batten disease.

The most common form of the childhood neurodegenerative disease late infantile neuronal ceroid lipofuscinosis (also called Batten disease) is caused by deficiency of the soluble lysosomal enzyme tripeptidyl peptidase 1 (TPP1) resulting from mutations in the TPP1 gene. We tested whether TPP1 gene transfer to the ependyma, the epithelial lining of the brain ventricular system, in TPP1-deficient dogs would be therapeutically beneficial. A one-time administration of recombinant adeno-associated virus (rAAV) expressing canine TPP1 (rAAV.caTPP1) resulted in high expression of TPP1 predominantly in e…

Subependymal seeding of low-grade oligodendroglial neoplasms: a case series.

The CSF dissemination of low-grade glial tumors is a known albeit rare entity. Few cases have been reported in the literature. We describe a … this finding represents dissemination of the supratentorial low-grade glioma along the ventricular ependyma or through the ventricular CSF. Although the small size of our series does not allow us to draw statistically significant conclusions, this abnormality correlates with progression of the supratentorial disease with or without features of a higher gr …

Localization and functional analyses of the MLC1 protein involved in megalencephalic leukoencephalopathy with subcortical cysts.

Mutations in the MLC1 gene are responsible for one form of the neurological disorder megalencephalic leukoencephalopathy with subcortical cy … d vessels and in astrocyte-astrocyte membrane contact regions. Other neuronal barriers, such as the ependyma and the pia mater, were also positive for MLC1 expression. MLC1 was detected in vivo and in heterologous systems at the plasma membrane. MLC mutations impaired folding, and the defect was corrected in vitro by addition of curcumin, a Ca(2+)-ATPase i …

D-aspartate localizations imply neuronal and neuroendocrine roles.

Though L-amino acids predominate in living organisms, substantial levels of free D-serine and D-aspartate occur in mammals, especially in ne … eurons of the hippocampus, cerebral cortex, and olfactory epithelium, as well as choroid plexus and ependyma. Localizations of D-aspartate oxidase are reciprocal to D-aspartate, suggesting that the enzyme depletes endogenous stores of the amino acid and might inactivate synaptically released D-aspartate.

Persistence of FoxJ1+ Pax6+ Sox2+ ependymal cells throughout life in the human spinal cord.

Ependymal cells lining the central canal of the spinal cord play a crucial role in providing a physical barrier and in the circulation of cerebrospinal fluid. These cells express the FOXJ1 and SOX2 transcription factors in mice and are derived from various neural tube populations, including embryonic roof and floor plate cells. They exhibit a dorsal-ventral expression pattern of spinal cord developmental transcription factors (such as MSX1, PAX6, ARX, and FOXA2), resembling an embryonic-like organization. Although this ependymal region is present in young humans, it appears to be lost with age…

Ependymal tissue in microphthalmia with cyst.

PURPOSE: To report the nature of the contents of a cyst associated with microphthalmia and optic nerve coloboma in a male infant. DESIGN: C … ll of the cyst that was adherent to the optic nerve. Histological evaluation of the tissue revealed ependymal cells. DISCUSSION: The presence of CSF in microphthalmic cysts does not necessarily imply a connection to the CNS, but may be due to local CSF production by ectopic ependymal cells.

Early ependymal changes in experimental hydrocephalus after mumps virus inoculation in hamsters.

To elucidate the pathogenesis of early ventricular dilatation in hydrocephalus, we examined early morphological changes in ependymal layers at the lateral ventricles in suckling hamsters without aqueductal stenosis 5 days after the intracerebral inoculation of mumps virus. Mumps virus antigen was detectable in all ependymal cells. The ependymal cilia had almost disappeared and only the microvilli remained. A number of supraependymal cells were also observed on the surface of the lateral ventricles. Transmission electron microscopy revealed intracytoplasmic viral-like inclusions in the infected…

An ultrastructural study of ependymal cell differentiation during lizard (Gallotia galloti) midbrain development.

Ependymal cell differentiation was examined in the lizard Gallotia galloti from E31 to adult. From E31 to E34 only one type of cell could be identified making up the pseudostratified columnar neuroepithelium but by E35 to E37 three types of ependymal cell were present. The first type was a narrow, elongated, columnar cell containing rough endoplasmic reticulum filled with an amorphous ground substance similar to that of astrocytes. The second type was broader with the nucleus close to the ventricular surface with numerous lipid droplets of varying sizes in the cytoplasm. The third type had an …
