The effect of ethanol on heme and porphyrin synthesis.

Despite the inherent difficulties in interpreting in vivo data on the effects of alcohol, it appears quite evident that ethanol itself alters the metabolism of the porphyrins and in so doing directly influences heme and hemoglobin synthesis. The mechanism of ethanol-induced iron overload remains elusive but, through observing the effects of phlebotomies and rechallenge with iron, the relationships of iron and alcohol to porphyria cutanea tarda are being more clearly defined. The control mechanisms of porphyrin synthesis by endogenous factors may be part of a more general red blood cell regulat…

Effect of ethanol and nicotine in the pregnant rat.

Treatment of pregnant Sprague-Dawley rats on day 9 of gestation with both ethanol (0.02 ml/gm of a 12.5% or 25% solution, i.p.) and nicotine (5 mg/kg, s.c.) resulted in no significant adverse effects on embryonic development and fetal viability.

Acute metabolic interaction of ethanol and drugs.

Addition of ethanol in vitro was found to inhibit the microsomal metabolism of a variety of drugs such as meprobamate, aminopyrine, pentobarbital and zoxazolamine. In all cases, a mixed type of inhibition was obtained. When the concentration of alcohols of different chain lengths required to inhibit 50% of the metabolism of drugs was plotted against their corresponding octanol-water partition coefficients (Po/w) it was found that the inhibitory potency of alcohols is linearly related to the partition coefficients, with a slope of 0.98. In vivo acute administration of ethanol also resulted in d…

The toxicity of ethanol. A tentative risk evaluation.

A substantial knowledge has been collected on the dose-dependent toxic effects of ethanol. Whereas ethanol probably is the chemical substance exerting most toxic effects in many populations, a quantitative estimate of the upper limit of long term safe consumption from a toxicological and public health standpoint has not earlier been performed. It may be summarized that the toxic range of ethanol consumption is 35-200 g per day. Based on the available data we would propose an acceptable daily intake (ADI) for ethanol of 7 g per day. The practical consequences are discussed.

Association of methanol and ethanol with heme proteins.

The behavior of ferrihemoglobin and ferrimyoglobin in widely varying concentrations of the lowest four alcohols has been studied by optical and electron paramagnetic resonance absorption spectroscopy. Methanol and ethanol, at concentrations too low to cause general conformational destabilization of the protein, produce both optical and electron paramagnetic resonance absorption spectral changes in ferrihemoglobin. These changes arise from equilibrium associations, characterized by dissociation constants at 25 degrees C of about 40 and 200 mM, respectively, for the methanol-ferrihemoglobin and …

Ethanol dependence in the rat: a parametric analysis.

Rats were maintained on liquid diets as their sole source of calories and fluid for 10, 15, 20, and 30 days. The diets consisted 35-40% of total calories in the form of ethanol. This procedure resulted in substantial ethanol intake leading to behavioral intoxication. Blood ethanol concentrations were found to be elevated throughout the day with a peak during the dark phase of the light cycle. The removal of ethanol resulted in evidence of physiological dependence, including behavioral manifestation of autonomic and somatic dysfunction and an increased susceptibility to audiogenic convulsions. …

Ethanol alleviates glucose stress from Euglena gracilis.

Euglena gracilis, a microalga, can be cultured under photoautotrophic, heterotrophic, or mixotrophic conditions. This species can produce pr … concentrations of glucose became hypertrophied rather than dehydrated and shrunken. The addition of ethanol (concentration of 0.5%) improved the proliferation of E. gracilis under a high glucose concentration of 400 mM. The addition of ethanol resulted in spindle-shaped cells similar to those observed under a no-glucose condition. These results indicate t …

Ethanol-related transcriptomic changes in mouse testes.

BACKGROUND: Alcohol consumption is widely known to have detrimental effects on various organs and tissues. The effects of ethanol on male reproduction have been studied at the physiological and cellular levels, but no systematic study has examined the effects of ethanol on male reproduction-related gene expression. RESULTS: We employed a model of chronic ethanol administration using the Lieber-DeCarli diet. Ethanol-fed mice showed normal testicular and epididymal integrity, and sperm morphology, but decreased sperm count. Total RNA sequencing analysis of testes from ethanol-fed mice showed th…

Endogenous ethanol production in health and disease.

The gut microbiome exerts metabolic actions on distal tissues and organs outside the intestine, partly through microbial metabolites that diffuse into the circulation. The disruption of gut homeostasis results in changes to microbial metabolites, and more than half of the variance in the plasma metabolome can be explained by the gut microbiome. Ethanol is a major microbial metabolite that is produced in the intestine of nearly all individuals; however, elevated ethanol production is associated with pathological conditions such as metabolic dysfunction-associated steatotic liver disease and aut…

Oleanolic acid protects ethanol-induced memory impairments.

A moderate amount of ethanol (EtOH) intake can lower the incidence of various cardiovascular disease but can result in neuropsychiatric issues during adolescence. EtOH acts on GABAA receptor, which can slow down neurotransmission and lead to changes in synaptic functions. These neurological changes due to EtOH can result in transient memory loss and may increase the risk of developing various neurological and psychiatric disorders such as dementia. Therefore, there is a need for strategies to overcome EtOH-induced brain dysfunctions. In this study, we investigated the effects of oleanolic acid…

Punishment of ethanol choice in rhesus monkeys.

A defining characteristic of individuals diagnosed with alcohol use disorder (AUD) is that negative outcomes related to drinking do not lead them to reduce their alcohol use. In rodent models of AUD, this characteristic has been studied by adding the bitter tastant quinine to an ethanol solution. In this study, we extended this approach to a nonhuman primate model in which the ability of quinine to decrease the choice of a 4% ethanol solution vs. water was measured. Five adult female rhesus monkeys with 7.3 years of experience drinking ethanol were given access to a 4% ethanol solution and wat…

The fructose-dependent acceleration of ethanol metabolism.

The aim of the present study was to elucidate how fructose is able to increase the rate of ethanol metabolism in the liver, an observation previously termed the fructose effect. Previous studies suggest that an increase in ATP consumption driven by glucose synthesis from fructose stimulates the oxidation of NADH in the mitochondrial respiratory chain, allowing faster oxidation of ethanol by alcohol dehydrogenase; however, this idea has been frequently challenged. We tested the effects of fructose, sorbose and tagatose both in vitro and in vivo. Both ethanol and each sugar were either added to …

Embryonic ethanol exposure on zebrafish early development.

INTRODUCTION: Embryonic exposure to ethanol leads to a condition of physical, behavioral, and cognitive deficiencies named fetal alcohol spectrum disorders (FASD). The most severe variations are in fetal alcohol syndrome (FAS), which is easier to diagnose and not studied in animal models. On the other side, the pFAS (partial fetal alcohol syndrome) includes cases of alcohol-related congenital disabilities and neurodevelopmental disorder with an inconclusive diagnosis. In recent years, the zebrafish has become a valuable model to study FASD and its variations. METHODS: This study characterizes…
