Microplastics (1 μm-5 mm) and nanoplastics (<1 μm) arise from the breakdown of larger plastic materials through thermal, photochemical, and mechanical degradation. Due to the wide use and haphazard disposal of plastic wastes, micro- and nano-plastics (MNPs) are ubiquitously detected in air, water, soil, and food, which leads to growing concern about chronic exposure and potential health impacts. MNPs have been detected across trophic levels and in more than 1000 species, indicating their extensive bioaccumulation. Recent studies have also detected MNPs in multiple human tissues and bodily flui

BACKGROUND: Both environmental exposure and type 2 diabetes (T2D) genetic susceptibility affect fetal growth. However, most previous studies used single exposure rather than an exposome strategy to explore the association between environmental factors and fetal growth, and the interactions of environmental exposures with maternal and fetal genes were often overlooked.
OBJECTIVES: To explore the associations between a broad range of prenatal environmental factors and fetal growth and further evaluate the effect modification of maternal and fetal T2D genetic susceptibility on the identified exp…

Previous studies on prenatal environmental factors and early-life growth generally applied the single-exposure strategy, mostly relying on cross-sectional growth indicators. No attempt has been made to explore the environmental effects on longitudinal growth at the exposome level. Further, the underlying bio-mechanisms and gene-environment interactions are poorly understood. Within 1,944 mother-child pairs from the Shanghai Birth Cohort, neonatal sex-specific z-scores of weight-for-age (WAZ), length-for-age (LAZ), and weight-for-length (WLZ) were used as the fetal growth outcomes. The growth t…

BACKGROUND: The exposome is defined as encompassing all environmental exposures one undergoes from conception onwards. Challenges of the application of this concept to environmental-health association studies include a possibly high false-positive rate.
OBJECTIVES: We aimed to reduce the dimension of the exposome using information from DNA methylation as a way to more efficiently characterize the relation between exposome and child body mass index (BMI).
METHODS: Among 1,173 mother-child pairs from HELIX cohort, 216 exposures ("whole exposome") were characterized. BMI and DNA methylation fro…

The One Exposome framework integrates exposomics, One Health toxicology, New Approach Methodologies (NAMs) and artificial intelligence to advance environmental health from reactive risk assessment towards predictive, preventive, and participatory science. The paper positions One Health as the ethical and conceptual rationale for recognizing the interdependence of human, animal, and environmental health, the Exposome as the framework for measuring cumulative and life-course exposures, and NAMs together with digital and AI-enabled tools as the means to generate, integrate, and interpret mechanis…

INTRODUCTION: The exposome framework provides an integrative perspective to characterize real-life exposures beyond single-chemical assessments. However, evidence on perinatal exposomes in non-urban populations is limited, particularly regarding pollutant mixtures and their contribution to adverse birth outcomes.
METHODS: We conducted a population-based cross-sectional study including 471 neonates from La Palma (Spain). A total of 106 pollutants were quantified in cord blood using validated methods. Exposures encompassed essential elements, toxic metals, prioritized pollutants, emerging eleme…

BACKGROUND: Exposome-wide association studies (ExWAS) systematically evaluate numerous environmental exposures, but typically focus on the mean effect, determining whether the association is statistically significant for the average individual. This approach may overlook environmental chemicals that specifically affect susceptible subpopulations. We conducted an exposome-wide interaction study (ExWIS) to identify environmental chemicals associated with cognitive function in the context of social vulnerability.
METHODS: We examined associations between 147 biomarker-based chemicals and cogniti…

Volatile organic compounds (VOCs) are pervasive environmental pollutants. However, their impact on sarcopenia, a condition characterized by progressive muscle loss and closely linked to numerous adverse health outcomes, remains poorly understood. Using data from the National Health and Nutrition Examination Survey (NHANES, 2011 to 2018), we analyzed 16 urinary metabolites of VOCs, adjusted for creatinine, to explore potential associations between VOC exposure and sarcopenia. Our findings consistently revealed a positive correlation across statistical models, with 2-aminothiazoline-4-carboxylic

We aimed to test the hypothesis that determinants of the perinatal clinical exposome related to the underlying etiology of premature birth (PTB) impact differently on select neonatal outcomes. We conducted a prospective longitudinal study of 377 singleton preterm neonates [gestational age (GA) at birth: 23-34 weeks] separated into three distinct contemporaneous newborn cohorts: i) spontaneous PTB in the setting of intra-amniotic infection/inflammation (yes-IAI, n = 116); ii) spontaneous PTB in the absence of IAI (no-IAI, n = 130), and iii) iatrogenic PTB for preeclampsia (iPTB-PE, n = 131). Per- and polyfluoroalkyl substances (PFAS) comprise thousands of persistent and structurally diverse chemicals that contribute to chronic human exposure. Yet, routine biomonitoring targets only a limited set of regulated legacy compounds, underestimating overall exposure. We present a retrospective, wide-scope suspect screening analysis framework to extract PFAS information from archived LC-HRMS exposomics datasets. A curated list of 1,666 PFAS, filtered for reversed-phase LC compatibility and enriched with structural descriptors, predicted retention indices, and MS2 fragments, was used to rep

Environmental noise is a well-recognized health risk and part of the external exposome-the World Health Organization estimates that 1 million healthy life years are lost annually in Western Europe alone due to noise-related complications, including increased incidence of hypertension, heart failure, myocardial infarction, and stroke. Previous data suggest that noise works through two paired pathways in a proposed reaction model for noise exposure. As a nonspecific stressor, chronic low-level noise exposure can cause a disruption of sleep and communication leading to annoyance and subsequent sy…

The physical environment is an important yet underexplored domain in Alzheimer's disease and Alzheimer's disease and related dementias (AD/ADRD), shaping exposures and health-related behaviors across the life course. We identified key gaps and priorities for incorporating physical environment measures into population-based research, with an emphasis on geospatial data that can be applied in large cohorts and administrative datasets. The Gateway Exposome Coordinating Center (GECC) synthesized input from six town halls, a 4-day expert workshop, and the literature to build consensus on priority p…

Pyrethroid insecticides are widely used indoors in residential, commercial, and occupational pest-control settings and are generally regarded as safe because of their relatively low acute toxicity. This perception is largely based on short-term exposure data and application practices conducted under well-ventilated conditions. However, indoor pesticide application does not always occur in environments that allow for rapid residue clearance. In enclosed workplaces-particularly those with centralized heating, ventilation, and air-conditioning systems, limited fresh-air exchange, and dust-retaini

While multiple factors are associated with cardiovascular disease (CVD), many environmental exposures that may contribute to CVD have not been examined. To understand environmental effects on cardiovascular health, we performed an exposome-wide association study (ExWAS), a hypothesis-free approach, using survey data on endogenous and exogenous exposures at home and work and data from health and medical histories from the North Carolina-based Personalized Environment and Genes Study (PEGS) (n = 5015). We performed ExWAS analyses separately on six cardiovascular outcomes (cardiac arrhythmia, con…

Background: The exposome (e.g., totality of environmental exposures) and its role in Alzheimer's Disease and Alzheimer's Disease and Related Dementias (AD/ADRD) are increasingly critical areas of study. However, little is known about how interventions on the exposome, including personal behavioral modification or policy-level interventions, may impact AD/ADRD disease burden at the population level in real-world settings and the cost-effectiveness of interventions. Methods: We performed a critical review to discuss the challenges in modeling exposome interventions on population-level AD/ADRD bu…

INTRODUCTION: Air pollution presents a major public health threat to India, affecting more than three quarters of the country's population. In the current project, GEOHealth Health Effects of Selected Environmental Exposomes Across the Life CourSe-India, we aim to study the effect of environmental exposomes-fine particulate matter (PM2.5), nitrogen dioxide (NO2), ozone (O3) and extremes of temperature-on multiple health outcomes using a modified life course approach. The associated training grant aims to build capacity in India to address the unique environmental health problems.

PURPOSE: Socioeconomically disadvantaged areas are more resource poor, impacting adherence to swallowing care recommendations. Neighborhood-level disadvantage metrics, such as the Area Deprivation Index (ADI), allow for examination of social determinants of health (SDOH) in a precise region. We examined ADI in a cohort of persons living with dementia (PLWD) to determine representation of those residing in areas of socioeconomic disadvantage (high ADI), distribution of swallowing care provided, and frequency of SDOH-related counseling or resource linking prior to discharge.
