Protein Corona-Tailored Nanoplatform Boosts Tumor Accumulation and Reprograms Macrophages for Osteosarcoma Therapy.

Osteosarcoma is featured with an immunosuppressive tumor microenvironment (TME) and limited therapeutic efficacy. Herein, we report a pH-responsive nanoplatform based on calcium phosphate (CaP)-coated Prussian blue nanoparticles (PB@CaP NPs) for optimized systemic delivery and antitumor immune response. The CaP interface inhibits the protein corona formation in blood, reducing complement-mediated clearance and prolonging circulation, and enhancing tumor accumulation of the nanoparticles. The pH-responsive dissolution of PB@CaP NPs enables the sequential release of calcium ions in the acidic TM

A multi-mode vertical flow immunofiltration detection system based on multifunctional PB@Au NPs for highly sensitive and diversified detection of brucellosis antibodies.

A multi-mode immunofiltration detection assay is introduced, which uses Prussian blue-gold nanocomposites (PB@Au NPs) as nanoprobes and integrates four signal output modes: colorimetric (CM), enzymatic catalysis (CL), photothermal (PT), and surface-enhanced Raman scattering (SERS). Rapid vertical flow technology (RVFT) serves as the detection platform, achieving highly sensitive, qualitative, and diversified quantitative detection of brucellosis antibodies. First, PB@Au NPs were successfully synthesized using the wet chemical method, exhibiting excellent catalytic activity, a PT conversion eff

Mechanical Biomimetic Nanocomposites for Multidimensional Treatment of Arterial Thrombosis.

Arterial thrombosis is a severe cardiovascular condition associated with high mortality and disability rates. Traditional pharmacological thrombolytic therapies, however, are limited by a narrow therapeutic window, short drug half-lives, restricted therapeutic effectiveness, and the risk of hemorrhagic complications. Moreover, the unique biological microenvironment of arterial thrombosis is characterized by high shear stress and an inflammatory milieu rich in reactive oxygen species (ROS), resulting in a relatively high recurrence rate. To address these challenges, a mechanical biomimetic nano

Concurrent antibiosis and anti-inflammation against bacterial pneumonia by zinc hexacyanoferrate nanocatalysts.

The bacterial pneumonia has been demonstrated to cause acute and severe pathological lung injury as well as the uncontrollable oxidative cytokine storms. The effective therapeutics against bacterial pneumonia demands highly efficient pathogen elimination, oxidative stress alleviation and anti-inflammation. Nevertheless, current therapeutics fail to achieve these goals by a single medicine with satisfactory performance. Herein, we report a self-assembled zinc-doped prussian blue-analogue - zinc hexacyanoferrate nanocatalysts (ZnPBA NCs) possessing excellent broad-spectrum anti-bacterial activit

Prussian Blue: A Nanozyme with Versatile Catalytic Properties.

Nanozymes, nanomaterials with enzyme-like activities, are becoming powerful competitors and potential substitutes for natural enzymes because of their excellent performance. Nanozymes offer better structural stability over their respective natural enzymes. In consequence, nanozymes exhibit promising applications in different fields such as the biomedical sector (in vivo diagnostics/and therapeutics) and the environmental sector (detection and remediation of inorganic and organic pollutants). Prussian blue nanoparticles and their analogues are metal-organic frameworks (MOF) composed of alternat

More Efficient Prussian Blue Nanoparticles for an Improved Caesium Decontamination from Aqueous Solutions and Biological Fluids.

Any release of radioactive cesium-137, due to unintentional accidents in nuclear plants, represents a dangerous threat for human health and the environment. Prussian blue has been widely studied and used as an antidote for humans exposed to acute internal contamination by Cs-137, due to its ability to act as a selective adsorption agent and to its negligible toxicity. In the present work, the synthesis protocol has been revisited avoiding the use of organic solvents to obtain Prussian blue nanoparticles with morphological and textural properties, which positively influence its Cs+ binding capa

The application of in situ mid-FTIR fibre-optic reflectance spectroscopy and GC-MS analysis to monitor and evaluate painting cleaning.

The development of non-invasive methodologies and portable instrumentation for in situ studies has been subject to great research and development in recent years in the field of conservation science. Despite such interest, very few reported studies employ these versatile techniques in the monitoring of cleaning treatments. This paper describes the application of mid-FTIR fibre-optic reflectance spectroscopy to monitor and evaluate the cleaning treatment of an oil painting using the chelating agent, triammonium citrate, a task undertaken in close collaboration with the painting conservator. A smartphone-integrated Pt@Cu-HCF nanozyme-based paper sensor for on-site determination of total antioxidant capacity in marine oils.

Total antioxidant capacity (TAC) serves as a key indicator for evaluating the nutritional quality of foods. In this study, we designed a platinum-embedded copper hexacyanoferrate (denoted as Pt@Cu-HCF) nanozyme that exhibits high oxidase-like activity, efficiently catalyzing the oxidation of chromogenic substrates to generate robust colorimetric signals. Antioxidants quench hydroxyl radicals (∙OH) produced during the catalytic process, leading to a concentration-dependent suppression of the color signal. Leveraging this mechanism, a smartphone-integrated, colorimetric paper sensor for on-site

Citrate-coated Prussian blue nanozyme hitchhikes neutrophils to ameliorate calcium oxalate crystal-induced kidney injury via inhibiting pyroptosis and NETosis.

Kidney stones are among the most common renal diseases with high global incidence and recurrence rates, with calcium oxalate (CaOx) stones constituting 65.9% to 80% of all cases. Minimally invasive surgery remains the primary approach for kidney stones, while limited progress has been made in the drug therapy for CaOx kidney stones over the recent years. The limitations of traditional drug therapy, such as renal toxicity and poor targeting, along with an elusive pathogenesis of CaOx kidney stones, have prevented its widespread clinical application. Renal tubular epithelial cells injury has bee

Neutrophil-Mimicking Prussian blue nanozymes for synergistic targeted immuno-metabolic therapy of hepatic ischemia-reperfusion injury.

Liver ischemia-reperfusion injury (IRI), an unavoidable complication during liver resection and transplantation, leads to significant postoperative liver failure and graft dysfunction. This injury is driven by severe oxidative stress, inflammation, and metabolic disruption, with clinical management hindered by the absence of effective multi-target therapies. This study introduces a novel therapeutic platform: neutrophil membrane-coated Prussian blue nanozymes (PBN) delivering metformin (Met@PBN@Neu-CVs). The neutrophil membrane coating enables active targeting of the nanoparticles to the infla

Do-It-Yourself: An Ultralow-Cost Sensing Platform with Integrated High-Entropy Nanozyme for the Rapid Detection of Exhaled H2S.

Herein, a noninvasive platform for rapid, low-cost detection of exhaled H2S was developed using nano-high-entropy Prussian blue frames (nanoPBFs) based on nanozyme poisoning. Synthesized via two-step coprecipitation and ammonia etching, nanoPBFs showed uniform element distribution, 4.1-fold higher catalytic activity than high-entropy Prussian blue nanocubes, and strong substrate affinity (Km = 0.016 mM for H2O2). Molecular dynamics simulations confirmed enhanced H2O2 diffusion on nanoPBFs (mean square displacement = 34.56 Å2 ps-1). Integrated into a polyethylene microtube with homemade modules

Designed synthesis of prussian blue@Cu-doped zinc phosphate nanocomposites for chemo/chemodynamic/photothermal combined cancer therapy.

Designing a multifunctional nanoplatform that combines multiple treatments has emerged as an innovative cancer treatment strategy. A simple and clear route is put forward to develop Cu2+-doped zinc phosphate coated prussian blue nanoparticles (designated as PB@Cu2+/ZnP NPs) integrating tri-modal therapy (chemo, chemodynamic and photothermal therapy) for maximizing anti-tumor efficacy. The obtained PB@Cu2+/ZnP NPs possess drug loading capacity due to the mesoporous structure present in the Cu2+-doped ZnP shell. In addition, the Cu2+-doped ZnP shell can gradually degrade in response to the mildl

Determination of ribavirin by molecularly imprinted electrochemical sensors using pyrro-1-propionyl-alaninoyl-chitooligosaccharide and pyrrole as bifunctional monomers on Prussian blue-gold nanocomposite films.

Herein, we developed a highly sensitive imprinted electrochemical sensor for the trace detection of ribavirin (RBV) using pyrrole (PYR) and pyrro-1-propionyl-alaninoyl-chitooligosaccharides (PPACO) as bifunctional monomers on Prussian blue-gold nanocomposite films. PPACO had strong molecular effect on RBV molecule and was selected by quantitative calculations. After the deposition of the Prussian blue-gold nanocomposite on a glassy carbon electrode (GCE) surface, a 4-aminothiophenol layer successfully self-assembled on the surface. Subsequently, the molecularly imprinted membrane (MIM) was sub

Reactive oxygen species scavenging nanofibers with chitosan-stabilized Prussian blue nanoparticles for enhanced wound healing efficacy.

Chronic inflammatory wounds pose therapeutic challenges in the biomedical field. Polymeric nanofibrous matrices provide extracellular-matrix-like structures to facilitate wound healing; however, wound infection and the subsequent accumulation of reactive oxygen species (ROS) delay healing. Therefore, we herein developed electrospun nanofibers (NFs), composed of chitosan-stabilized Prussian blue (PBChi) nanoparticles (NPs) and poly(vinyl alcohol) (PVA), with ROS scavenging activity to impart antioxidant and wound healing properties. The PBChi NPs were prepared using chitosan with different mole

Label-free Electrochemical Detection of CGG Repeats on Inkjet Printable 2D Layers of MoS2.

Flexible and ultrasensitive biosensing platforms capable of detecting a large number of trinucleotide repeats (TNRs) are crucial for future technology development needed to combat a variety of genetic disorders. For example, trinucleotide CGG repeat expansions in the FMR1 gene can cause Fragile X syndrome (FXS) and Fragile X-associated tremor/ataxia syndrome (FXTAS). Current state-of-the-art technologies to detect repeat sequences are expensive, while relying on complicated procedures, and prone to false negatives. We reasoned that two-dimensional (2D) molybdenum sulfide (MoS2) surfaces may be

Atomic matching catalysis to realize a highly selective and sensitive biomimetic uric acid sensor.

A main challenge for biomimetic non-enzyme biosensors is to achieve high selectivity. Herein, an innovative biomimetic non-enzyme sensor for electrochemical detection of uric acid (UA) with high selectivity and sensitivity is realized by growing Prussian blue (PB) nanoparticles on nitrogen-doped carbon nanotubes (N-doped CNTs). The enhancement mechanism of the biomimetic UA sensor is proposed to be atomically matched active sites between two reaction sites (oxygen atoms of 2, 8-trione, 6.9 Å) of UA molecule and two redox centers (FeII on the diagonal, 7.2 Å) of PB. Such an atomically matching

Additive effect of DL-penicillamine plus Prussian blue for the antidotal treatment of thallotoxicosis in rats.

DL-penicillamine (DL-P) and Prussian blue (PB) given alone or in combination were tested as possible treatments against acute thallium toxicity. Rats were intoxicated by i.p. injection of thallium (I) acetate at LD(50) (32 mg/kg). A day later, pharmacological treatment was administered until day 4 as follows: (1) vehicles, (2) PB 50mg/kg, by oral route, twice a day, (3) DL-P 25mg/kg i.p. route, twice daily and (4) PB+DL-P. The Estimated Probability Survival (EPS) was recorded during the experiment for each treatment. DL-P alone did not show a significant effect on survival. However, when it wa

Validation of an in vitro method for the determination of cyanide release from ferric-hexacyanoferrate: Prussian blue.

Prussian blue (PB), ferric hexacyanoferrate, Fe(4)[Fe(CN)(6)](3) is indicated for the treatment of known or suspected internal contamination with radioactive cesium, radioactive thallium, or non-radioactive thallium. Owing to the molecular properties, cyanide is likely dissociated from PB under physiologically relevant pH conditions, thus raising a concern for the safety of the product. The objective of this study was to calibrate and validate a cyanide assay over a wide pH range (from 0.5 to 12) on the basis of Spectroquant cyanide test method (Merck). Merck's photometric method requires that

Dimercaptosuccinic acid and Prussian Blue in the treatment of acute thallium poisoning in rats.

INTRODUCTION: Despite being banned as a pesticide, thallium still results in human and animal poisonings. Current recommended treatments include the use of the chemical Prussian Blue. Limitations in its availability may result in Prussian Blue not being obtainable in the thallium-poisoned patient. The chelator 2,3-Dimercaptosuccinic acid (DMSA) is currently FDA-approved for use in childhood lead poisoning and has been reported to be beneficial in treating other heavy metal poisonings. The objective of this study was to determine the efficacy of DMSA as a treatment for thallium poisoning by stu
