BACKGROUND: Human fibrinogen gamma chain variants, termed gamma' chains, contain a unique 20-residue sequence after gamma chain residue 407 that ends at gamma'427, and is designated gamma'(427L). Full-length (FL) gamma'(427L) chains are constituents of a fibrin-dependent thrombin inhibitory system known as antithrombin I, whereas a gamma' chain processed in vivo, termed gamma'(423P), lacks the C-terminal tetrapeptide EDDL, and does not bind thrombin. Screening of 200 blood donors for the presence of polymorphisms in three fibrinogen genes (FGA, FGB, FGG), revealed two individuals with a heterozygous missense mutation (c.323C > G, gammaAla82Gly) in the FGG gene. The value of the 125I-labelled fibrinogen uptake test as a screening method for deep vein thrombosis was estimated in 49 patients (97 extremities) undergoing elective hip operations. The legs were monitored preoperatively, on the 1st, 2nd and 4th postoperative days. Contrast venography, as a reference method, was performed on the 5th postoperative day. Fibrinogen and fibrinogen-like proteins (FLP) were isolated from plasma and serum of cattle acutely infected with Babesia bovis. The sizes and chain structures of these proteins were examined and clotting assays performed. The results indicated that the blood was in a hypercoagulable state due mainly to enhanced production of hydrogen bonded fibrin and offset partly by slight inhibition of chain cross-linking. Several techniques are used to detect rejection episodes in renal transplants; complexity renders these tests impractical for clinical use in most cases. This paper reports the results obtained in the study of urinary FDP, by counterimmunoelectrophoresis, in 50 healthy subjects and in 360 consecutive samples from 18 renal transplant recipients; urine specimens were examined daily for periods of time of up to 60 days post transplant. In-vivo studies of fibrinogen metabolism were carried out using an I-120 labelled preparation of fibrinogen supplied commercially by the Radiochemical Centre, Amerhsham for use in the diagnosis of deep venous thrombosis. These studies show that the material is suitable for obtaining the main metabolic parameters of clinical interest, though degradation products may be present initially or appear during storage. If suitable corrections are applied the results agree well with those of other authors.
