Monitoring residues of pesticides in food in Brazil: A multiscale analysis of the main contaminants, dietary cancer risk estimative and mechanisms associated.

IntroductionPesticides pose a risk for cancer development and progression. People are continuously exposed to such substances by several routes, including daily intake of contaminated food and water, especially in countries that are highly pesticide consumers and have very permissive legislation about pesticide contamination as Brazil. This work investigated the relationship among pesticides, food contamination, and dietary cancer risk.MethodsAnalyzed two social reports from the Brazilian Government: the Program for Analysis of Residues of Pesticides in Food (PARA) and The National Program for…

Identification of Signaling Pathways Targeted by the Food Contaminant FB1: Transcriptome and Kinome Analysis of Samples from Pig Liver and Intestine.

SCOPE: Fumonisin B1 (FB1) is a mycotoxin produced by Fusarium species. In mammals, this toxin causes widespread organ-specific damage; it promotes hepatotoxicity, is immunotoxic, alters intestinal functions etc. Despite its inhibitory effect on de novo ceramide synthesis, its molecular mechanism of action and toxicity is not totally elucidated. METHODS AND RESULTS: To explore the mechanism of FB1 toxicity, we analyzed the transcriptome and the kinome of two organs targeted by FB1: the liver and the jejunum. Pigs were fed for 4 weeks a control diet or a FB1-contaminated diet (10 mg/kg). As exp

Impact of traditional practices on food safety: a case of acute toxoplasmosis related to the consumption of contaminated raw pork sausage in Italy.

A case of acute toxoplasmosis in an adolescent girl, almost certainly related to the consumption of raw sausage, is described. The girl suffered of fever and weakness and presented a swollen lymph node in the submandibular region. Serology analysis was positive for Toxoplasma gondii and excluded other infections. Further analysis, with avidity test and immunoblot, confirmed the acute toxoplasmosis. She reported that about a month before the appearance of the symptoms, she had eaten a piece of raw sausage while it was being prepared by her father. We analyzed sausage samples prepared from this

Potentially toxic element contamination in Ghana's vegetable value chain: a systematic review of mining-related soil-to-food transfer and human health risk.

In Ghana, artisanal and small-scale mining (ASM) and smallholder vegetable farming occupy the same landscapes, and in many communities the same households combine both livelihoods, yet no systematic account has previously quantified how much potentially toxic element (PTE) contamination travels from mine waste into food. This review addresses that gap for the pre-harvest and post-harvest stages of the value chain for the four vegetables that define Ghanaian diets: tomato (Solanum lycopersicum), pepper (Capsicum annuum), garden egg (Solanum aethiopicum), and onion (Allium cepa). Adhering to PRI

Phthalate acid esters in water and edible oils: a comprehensive review of contamination sources, migration mechanisms, and emerging removal technologies of food matrices.

Phthalate acid esters (PAEs), a group of plasticizers widely used to improve the flexibility and processability of plastic materials, have been frequently detected in food matrices. This review provides a comparative framework for understanding PAEs contamination in two representative dietary matrices, water and edible oils, with emphasis on direct inputs from food packaging and processing, as well as indirect pathways associated with microplastic migration, atmospheric deposition, and recycled plastics. A key difference dependent on the matrix is highlighted: in water, the migration and redis

Tracking Listeria monocytogenes contamination and virulence-associated characteristics in the ready-to-eat meat-based food products industry according to the hygiene level.

Listeria monocytogenes isolates collected from final products and food contact surfaces of 10 ready-to-eat meat-based food products (RTEMP) producing industries were analyzed to relate their virulence-associated characteristics and genetic profiles with the hygiene assessment of those industries. Together with sample collection, an audit was performed to evaluate the implemented food safety management system and to investigate the specific audit requisites more associated to the occurrence of those L. monocytogenes serogroups frequently related with human disease. L. monocytogenes was present

Risk-based assessment and criteria specification of the microbial safety of wastewater reuse in food processing: Managing Listeria monocytogenes contamination in pasteurized fluid milk.

Intense pressure on water resources has led to efforts to reuse reclaimed processing wastewater for cleaning purposes in food processing plants. The milk industry produces considerable amounts of wastewater, which can be used for cleaning of equipment after appropriate treatment. However, due to naturally occurring microbiological contamination in raw milk, the wastewater is often contaminated, and therefore the reuse of reclaimed wastewater is perceived as risky. This study aims to quantify the risks of Listeria monocytogenes infection and associated disease burden when wastewater reclaimed f

Low concentrations of the food contaminant Deoxynivalenol trigger apoptosis and impair GnRH-induced LH secretion in pituitary gonadotrope-like cells.

The Fusarium mycotoxin deoxynivalenol (DON) is one of the most frequently occurring food contaminants. Nearly all individuals are exposed to DON, due to it widespread presence in grains and grain-based products. Chronic DON poisoning is associated with growth retardation, immunotoxicity as well as impaired reproduction and fetal development. At the molecular level, DON alters intracellular signaling by activating mitogen-activated protein kinases (MAPKs) that modulate cell growth, differentiation, and apoptosis. Of note, these MAPKs are also critical mediators of gonadotrophin-releasing hormon

The role of PGC-1α and metabolic signaling pathway in kidney injury following chronic administration with 3-MCPD as a food processing contaminant.

3-Monochloropropane-1,2-diol (3-MCPD) as a byproduct of food processing and a carcinogenic agent has attracted much attention in the last decades. Kidney is the main target organ that is sensitive to the toxicity of 3-MCPD. Due to limited evidence about possible 3-MCPD toxicity, we design an investigation to determine the role of mitochondrial biogenesis following chronic oral administration of 3-MCPD (2, 4, 8 and 32 mg/kg) for 2 months in male C57 mice. The present study evaluated the affects of 3-MCPD in modulating metabolic signalling which is associated with Il-18, PGC-1α, Nrf-2 and Sir3 w

Mycotoxin contamination in food and feed: Occurrence, co-occurrence, combined impacts on human and animal health, climate change implications, risk assessment, and mitigation strategies.

Mycotoxins are toxic secondary metabolites produced by numerous fungi that infect crops during pre- and post-harvest stages. Aflatoxins, ochratoxins, fumonisins, zearalenone, and trichothecenes are of greatest concern due to their widespread occurrence and high toxicity. Consumption of mycotoxin-contaminated food and feed poses serious public health risks, as these compounds are linked to carcinogenic, neurotoxic, nephrotoxic, hepatotoxic, and immunotoxic effects. The frequent co-occurrence of multiple mycotoxins further amplifies health risks through additive or synergistic interactions and c

Outbreaks where food workers have been implicated in the spread of foodborne disease. Part 5. Sources of contamination and pathogen excretion from infected persons.

In this article, the fifth in a series reviewing the role of food workers in foodborne outbreaks, background information on the routes of infection for food workers is considered. Contamination most frequently occurs via the fecal-oral route, when pathogens are present in the feces of ill, convalescent, or otherwise colonized persons. It is difficult for managers of food operations to identify food workers who may be excreting pathogens, even when these workers report their illnesses, because workers can shed pathogens during the prodrome phase of illness or can be long-term excretors or asymp

Non-Targeted LC-Q-Orbitrap HRMS Screening of Veterinary Drugs, Pesticides, and Emerging Contaminants in Animal-Derived Foods and Honey.

BACKGROUND: Veterinary drug residues, pesticides, and chemical contaminants in animal-derived foods pose public health risks, including antimicrobial resistance development and consumer exposure to banned or withdrawn substances. Conventional targeted methods are inherently limited to predefined compounds and may overlook emerging or unexpected contaminants. Many developing countries still lack comprehensive multi-residue surveillance across diverse food matrices, highlighting the need for analytical frameworks adaptable to regulatory laboratories. OBJECTIVE: To evaluate an LC-Q-Orbitrap HRMS

Rapid and Green Extraction of Food Contaminants: Integrating Advanced Porous Materials and Miniaturized Functional Platforms for Chromatographic and Mass Spectrometric Analysis.

The rapid and reliable monitoring of trace contaminants through chromatographic and mass spectrometric analysis is a very important but challenging task in food safety monitoring, in which efficient sample pretreatment is critical. Integrating advanced porous materials (e.g., metal-organic frameworks, covalent organic frameworks, and molecularly imprinted polymers) with miniaturized functional platforms (e.g., metal probes, wooden tips, and filter paper) offers a promising solution. This "porous material to miniaturized platform" integration enables green and efficient sample pretreatment for

The influence of a submerged meadow on uptake and trophic transfer of legacy mercury from contaminated sediment in the food web in a brackish Norwegian fjord.

There are contradicting results on the importance of legacy mercury (Hg) contaminated sediments to Hg fish tissue concentrations. Still, sediment remediation actions often aim at minimizing ecosystem exposure and human risk caused by the consumption of fish and seafood. The aim of this study was to investigate the possible influence of a permanently submerged meadow on the availability and transfer of Hg from sediment to biota, three decades after the Hg discharges was halted and the previous biota survey was carried out, in the severely contaminated brackish fjord Gunneklevfjorden in southern

Endocrine disruptors found in food contaminants enhance allergic sensitization through an oxidative stress that promotes the development of allergic airway inflammation.

In the past few decades, there has been a significant increase in incidence of allergic diseases. The hygiene hypothesis may provide some clues to explain this rising trend, but it may also be attributable to other environmental factors that exert a proallergic adjuvant effects. However, there is limited information on the risks of developing allergic asthma and related diseases through the ingestion of environmental chemicals found in food contaminants. In the present study, we have shown that oral administration of tributyltin, used as a model environmental chemical, induced oxidative-stress

Gestational Exposure to the Food Processing Contaminant 3-MCPD Impairs Fetal Growth by Disrupting Placental Taurine Metabolism to Drive Vascular Senescence.

3-Monochloro-1,2-propanediol (3-MCPD), a common food contaminant, induces fetal growth restriction in mice by triggering senescence in the placental vasculature. Transcriptomic analysis reveals suppression of the taurine-hypotaurine pathway through downregulation of Fmo1. Mechanistically, 3-MCPD upregulates Ezh2, which deposits repressive H3K27me3 marks at the Fmo1 promoter, silencing its transcription and depleting placental taurine. Taurine supplementation or Ezh2 inhibition rescues 3-MCPD-induced vascular senescence in human endothelial cells. These findings identify an Ezh2-taurine axis es

Enzyme-mediated "one to more" PAM-independent and amplification free CRISPR/Cas12a for one-pot rapid detection of food contaminated bacteria.

The requirement of protospacer-adjacent motif (PAM) and nucleic acid amplification have restricted the application of CRISPR/Cas12a system in bacteria detection. Herein, we proposed a restriction endonuclease mediated, "one to more", PAM-independent and amplification-free CRISPR/Cas12a strategy for bacteria detection. The bacterial genomic DNA can be cleaved by specific endonuclease, exposing a large number of sticky ends. The combination of these exposed sticky ends with a short ssDNA could effectively activate the trans-cleavage activity of Cas12a to break a FQ reporter probe, achieving the

Nanotechnology-enabled aptamer/G-quadruplex DNAzyme biosensors for food contaminant detection: nanoscale engineering, signal amplification strategies, and analytical applications.

Despite several advances in biosensor technology, reliable quantification in complex matrices remains challenging due to limitations in signal amplification, selectivity, and operational stability. The development of nanotechnology has played an important role in improving the performance of biosensors. Considerable attention has been paid to the integration of various nanomaterials and bioreceptors in the structure of sensing devices. DNA-based biosensors have provided promising alternatives to protein-based systems due to excellent chemical stability, ease of modification, and selectivity. A
