Prodrugs of hydrogen sulfide and related sulfur species: recent development.

Hydrogen sulfide (H2S) is commonly referred to as the third gasotransmitter with firmly established physiological roles. Prodrug approaches have been broadly applied to deliver H2S for various applications and mechanistic studies. Since S-persulfidation and glutathionylation are known to be important in cellular signaling by sulfur species, there have been interests in developing donors of persulfide and glutathione persulfide as well. In this review, we discuss the recent development in area of prodrugs for various sulfur species.

Hydrogen Sulfide Regulates Macrophage Function in Cardiovascular Diseases.

Significance: Hydrogen sulfide (H2S) is an endogenous gasotransmitter that plays a vital role in immune system regulation. Recently, the regulation of macrophage function by H2S has been extensively and actively recognized. Recent Advances: The mechanisms by which endogenous H2S controls macrophage function have attracted increasing attention. The generation of endogenous H2S from macrophages is mainly catalyzed by cystathionine-γ-lyase. H2S is involved in the macrophage activation and inflammasome formation, which contributes to macrophage apoptosis, adhesion, chemotaxis, and polarization. In

Role of hydrogen sulfide donors in cancer development and progression.

In recent years, a vast number of potential cancer therapeutic targets have emerged. However, developing efficient and effective drugs for the targets is of major concern. Hydrogen sulfide (H2S), one of the three known gasotransmitters, is involved in the regulation of various cellular activities such as autophagy, apoptosis, migration, and proliferation. Low production of H2S has been identified in numerous cancer types. Treating cancer cells with H2S donors is the common experimental technique used to improve H2S levels; however, the outcome depends on the concentration/dose, time, cell type…

Regulation of vascular tone homeostasis by NO and H2S: Implications in hypertension.

Nitric oxide (NO) and hydrogen sulfide (H2S) are two gasotransmitters that are produced in the vasculature and contribute to the regulation of vascular tone. NO and H2S are synthesized in both vascular smooth muscle and endothelial cells; NO functions primarily through the sGC/cGMP pathway, and H2S mainly through activation of the ATP-dependent potassium channels; both leading to relaxation of vascular smooth muscle cells. A deficit in the NO/H2S homeostasis is involved in the pathogenesis of various cardiovascular diseases, especially hypertension. It is now becoming increasingly clear that t…

The role of hydrogen sulfide in cyclic nucleotide signaling.

Hydrogen sulfide (H2S) is recognized as an endogenous gaseous transmitter alongside nitric oxide (NO) and carbon monoxide (CO). By integrating into multiple signaling pathways, H2S elicits biological functions in various mammalian systems. Among these pathways, cyclic nucleotide signaling has gradually gained attention in the past decade. Based on current evidence, it seems that H2S may differentially affect the activity of resting adenylyl cyclases (ACs) and activated ACs, therefore playing a dual role in the regulation of cyclic adenosine monophosphate (cAMP) mediated signaling. However, how

H2S Activated Drug Release from Protein Cages.

We took advantage of gasotransmitter H2S as a chemical reaction-based trigger for controlled release of doxorubicin which is precoordinated by copper ions and enclosed in horse spleen apoferritin. The nanocomposite is stable at physiological pH and temperature before H2S activation. The drug release process avoids disassembly of protein shells and is controllable by the strong affinity of sulfide with copper ions. The in vitro cytotoxicity assay indicates the antitumor effect of doxorubicin toward tumor cells could be achievable by H2S activation.

A biotin-guided near-infrared fluorescent probe for imaging hydrogen sulfide and differentiating cancer cells.

Imaging cancer specific biomarkers with near-infrared (NIR) fluorescent probes can help inaccurate diagnosis. Hydrogen sulfide (H2S) has been reported to be involved in many physiological and pathological processes and is considered as one of the key gasotransmitters during the development of cancer. To achieve specific H2S detection in cancer cells, we reported a biotin-guided NIR fluorescent sensor P1 targeting a cancer cell surface biomarker, based on the H2S-specific thiolysis of the NBD-amine-hemicyanine conjugate. The probe showed a fast turn-on signal at 754 nm upon H2S activation and g…

Simultaneous activation of the hydrogen sulfide biosynthesis genes (CBS and CSE) induces sex-specific geroprotective effects in Drosophila melanogaster.

Hydrogen sulfide (H2S) is one of the most important gasotransmitters that affect lifespan and provide resistance to adverse environmental conditions. Here we investigated geroprotective effects of the individual and simultaneous overexpression of genes encoding key enzymes of H2S biosynthesis - cystathionine-β-synthase (CBS) and cystathionine-γ-lyase (CSE) on D. melanogaster model. Simultaneous overexpression of CBS and CSE resulted in additive (in males) and synergistic (in females) beneficial effects on median lifespan. Individual overexpression of CBS was associated with increased thermotol…

Signal pathways involved in the biological effects of sulfur dioxide.

Gasotransmitters, such as nitric oxide, carbon monoxide and hydrogen sulfide, play important roles in life and have attracted great interest in scientists. In recent years, sulfur dioxide (SO2) has also been found to play important roles in mammals. The redox pathway is involved in the biological effects of SO2, such as the protective effect on myocardial ischemia reperfusion, myocardial injury, pulmonary hypertension and atherosclerosis. Ion channels, such as L-type calcium and adenosine triphosphate-sensitive potassium channels, as well as 3'-5'-cyclic guanosine monophosphate and 3'-5'-cycli…

Hydrogen Sulfide Signaling in Neurodegenerative Movement Disorders.

Hydrogen sulfide (H2S) is a gaseous signaling molecule, also known as a gasotransmitter, present in nearly all mammalian organs. It plays crucial roles in regulating various physiological processes in both the brain and peripheral systems. The body maintains tight control over H2S levels, as both excessive and deficient levels can disrupt normal physiological functions and lead to disease. H2S has a significant impact on cognitive and motor functions, which are often compromised in neurodegenerative disorders. It modulates signaling and metabolism primarily by post-translationally modifying re

Molecular crosstalk between gas transmitters and catecholamine biosynthesis pathways: a narrative review.

Catecholamines, particularly the hormone and neurotransmitter epinephrine, are important regulators of blood pressure and cardiovascular function. The expression of genes encoding catecholamine biosynthetic enzymes is subject to strict transcriptional regulation through both hormonal and neuronal mechanisms. These genes are also regulated by hypoxia and reactive oxygen species. A class of small gaseous transmitters, known as gas transmitters, serve as potent regulators of blood pressure and hypoxic signaling; however, their roles in catecholamine biosynthesis remain poorly understood. This man

Hydrogen Sulfide Promotes Nodulation and Nitrogen Fixation in Soybean-Rhizobia Symbiotic System.

The rhizobium-legume symbiotic system is crucial for nitrogen cycle balance in agriculture. Hydrogen sulfide (H2S), a gaseous signaling molecule, may regulate various physiological processes in plants. However, whether H2S has regulatory effect in this symbiotic system remains unknown. Herein, we investigated the possible role of H2S in the symbiosis between soybean (Glycine max) and rhizobium (Sinorhizobium fredii). Our results demonstrated that an exogenous H2S donor (sodium hydrosulfide [NaHS]) treatment promoted soybean growth, nodulation, and nitrogenase (Nase) activity. Western blotting

Protective Effect of Triphala Mediated by H2S Signaling Pathway in LPS-Induced RAW Macrophages.

INTRODUCTION: MacrophagesHydrogen sulfide (H2 S), an endogenously produced gasotransmitter, is a wellknown inflammatory agent production successfully controlled with triphala in macrophages. MATERIALS AND METHOD: The ELISA analysis revealed that triphala extracts suppress the protein expression of NFκB (p65) and downregulation of the CSE gene expression. Moreover, in the presence of propargylglycine (PAG) the expression of NFκB and CSE gene was further reduced. RESULTS: These results suggest that triphala could be a potent antiinflammatory effect that could be used for the treatment of many

Near-infrared fluorescent chemodosimeter for real-time in vivo evaluation of H2S-release efficiency of prodrug.

Hydrogen sulfide (H2S) is a significant gasotransmitter. A deficiency in H2S might contribute to some serious diseases. The development of H2S drugs has received a great deal of attention. There is a pressing need for an effective method to evaluate H2S release efficiency, especially in in vivo evaluation. In this study, a highly sensitive and selective near-infrared (NIR) fluorescent chemodosimeter (NRh-N3) was synthesized for detecting H2S. Importantly, NRh-N3 was shown to be capable of visually monitoring H2S-release from the prodrug in vitro and in vivo.
