Polymorphisms in genetics of vitamin D metabolism confer susceptibility to ocular Behçet disease in a Chinese Han population.

PURPOSE: To test whether single nucleotide polymorphisms (SNPs) of the 4 vitamin D family genes (DHCR7, CYP2R1, CYP27B1, and CYP24A1) previously associated with several autoimmune diseases are associated with ocular Behçet disease, Vogt-Koyanagi-Harada (VKH) syndrome, acute anterior uveitis (AAU) with ankylosing spondylitis, or pediatric uveitis in the Chinese Han population. DESIGN: Prospective case-control study. METHODS: Genotyping was performed by polymerase chain reaction-restriction fragment length polymorphism, and the genotypes were verified with direct sequencing. The first-stage st

Differences in structural color and population genetic structure of Western and Central Palearctic Polyommatus icarus populations.

The blue structural coloration of male Polyommatus icarus butterflies functions as a sexual signaling trait and exhibits remarkable spectral stability within populations despite being generated by highly complex photonic nanoarchitectures. The correlation of the blue sexual signaling color and population genetic variation of the butterflies was investigated across the Western and Central Palearctic regions. Dorsal wing reflectance spectra was measured for 95 male specimens and compared with the population genetic structure revealed in 99 specimens by 18 recently developed microsatellites. Refl

Population genetic analysis of Aedes aegypti reveals evidence of emerging admixture populations in coastal Kenya.

BACKGROUND: The Aedes aegypti mosquito is widespread in tropical and subtropical regions. There are two recognized subspecies; the invasive Aedes aegypti aegypti (Aaa) and the ancestral Aedes aegypti formosus (Aaf). Aaf is common throughout Kenya whereas Aaa, which was historically confined to coastal regions, has undergone a range expansion. In areas of sympatry, gene flow may lead to admixed populations with potential differences in vectorial capacity. We hypothesize that coastal Ae. aegypti populations have a higher proportion of Aaa ancestry than those from inland locations of Kenya, influ

Attitudes towards disclosure of familial genetic risk in a Mediterranean island population - A survey of the Maltese population.

Germline genetic testing has implications that extend beyond the individual patient to relatives, particularly for high-penetrance variants implicated in hereditary cancer or neurodegenerative syndromes. Many countries encourage patient-led communication to inform at-risk relatives, although the efficacy and uptake of this approach varies. Alternative scenarios envisage direct contact mediated by clinicians. The familial disclosure of sensitive genetic information is also determined by complex socio-ethnic factors. To date, no study has explored whether relatives would want to be informed of f

Population-specific facial traits and diagnosis accuracy of genetic and rare diseases in an admixed Colombian population.

Up to 40% of rare disorders (RD) present facial dysmorphologies, and visual assessment is commonly used for clinical diagnosis. Quantitative approaches are more objective, but mostly rely on European descent populations, disregarding diverse population ancestry. Here, we assessed the facial phenotypes of Down (DS), Morquio (MS), Noonan (NS) and Neurofibromatosis type 1 (NF1) syndromes in a Latino-American population, recording the coordinates of 18 landmarks in 2D images from 79 controls and 51 patients. We quantified facial differences using Euclidean Distance Matrix Analysis, and assessed th

Clinical, Radiographic, and Genetic Analyses in a Population-Based Cohort of Adult Spinal Deformity in the Older Population.

OBJECTIVE: This study aimed to identify the sagittal parameters associated with health-related quality of life and genetic variations that increase the risk of adult spinal deformity (ASD) onset in the older population. METHODS: We recruited 120 participants who had a sagittal vertical axis > 50 mm in a sagittal imbalance study. Sagittal radiographic parameters, cross-sectional area, and intramuscular fatty infiltration using the Goutallier classification in the paraspinal lumbar muscles were evaluated. Functional scales included the self-reported Oswestry Disability Index (ODI), 36-item Shor

The effect of sample size on estimates of genetic differentiation and effective population size for Schistosoma mansoni populations.

Eradication or local extinction of the human parasite Schistosoma mansoni is a goal for many control programs. Population genetic analyses are helping to evaluate and guide these efforts, yet what to sample, how to sample and how densely to sample is not well established. We determined the S. mansoni allele frequency profile of nearly all infected inhabitants in two small Brazilian communities and created sub-samples representing 5-50% of all detected human infections (infrapopulations). Samples were selected at random with replacement, and each size class was replicated 100 times. Mean pairwi

Extensive genetic diversity among populations of the malaria mosquito Anopheles moucheti revealed by population genomics.

Malaria vectors are exposed to intense selective pressures due to large-scale intervention programs that are underway in most African countries. One of the current priorities is therefore to clearly assess the adaptive potential of Anopheline populations, which is critical to understand and anticipate the response mosquitoes can elicit against such adaptive challenges. The development of genomic resources that will empower robust examinations of evolutionary changes in all vectors including currently understudied species is an inevitable step toward this goal. Here we constructed double-digest

Mitochondrial genome-derived microsatellites reveal genetic diversity and population structure in Callery pear populations.

Callery pear (Pyrus calleryana Decne.; PC) possesses many desirable characteristics valued in managed landscapes. This has driven the release of numerous cultivars, including both hybrids and selections derived from native populations. The extensive planting of PC cultivars in managed areas has contributed to the widespread occurrence of invasive individuals across a broad range of habitats in the eastern United States (US). Self-incompatibility, tolerance to various environmental conditions, pathogen and pest resistance, intraspecific hybridization among the cultivars, possible interspecific

The population frequency of predicted pathogenic genetic variants in commonly affected CAKUT genes in the general population.

BACKGROUND: Renal imaging suggests that congenital anomalies of the kidney and urinary tract (CAKUT) affect one in 200 of the population, and 20% have a monogenic cause. This study determined the population frequency of predicted pathogenic variants in six commonly affected CAKUT genes. METHODS: HNF1B, SALL1, EYA1, PBX1, GATA3, and PAX2 variants were downloaded from gnomAD v.2.1.1 (n = 141,456) and the population frequency of predicted disease-causing variants calculated from the sum of structural, null, and predicted pathogenic missense changes in the overall cohort or from variants shared w

Population-Specific Carrier Frequencies in an Underrepresented Genetically Heterogeneous Population: Implications for Expanded Carrier Screening.

Background/Objectives: Expanded carrier screening (ECS) panels often rely on pan-ethnic databases, such as the ACMG Tier 3 panel, but their ability to capture population-specific reproductive risk is uncertain. We characterized pathogenic/likely pathogenic (P/LP) variants and at-risk-couple (ARC) probabilities in an underrepresented population from North Macedonia to inform ECS gene prioritization. Methods: NGS data from 1438 subjects of Macedonian, Albanian, and other local ancestry, referred for rare-disease diagnostics, were analyzed across 1795 candidate autosomal recessive/X-linked genes.

Genetic associations of eosinophilic granulomatosis with polyangiitis in the Japanese population: exploring similarities and differences with European populations.

OBJECTIVES: Eosinophilic granulomatosis with polyangiitis (EGPA) is classified as a subtype of anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV). A recent genome-wide association study (GWAS) of EGPA in populations of European ancestry reported several susceptibility loci. In view of population differences in genetic factors of AAV, we made an attempt to examine the contribution of these loci to EGPA in a Japanese population. METHODS: The EGPA-associated variants in LPP, TSLP, IRF1/IL5, HLA, CDK6, 10p14 and 12q21 in European populations (EUR) were analysed in a total of

Phylogeography, genetic diversity, and population structure of Nile crocodile populations at the fringes of the southern African distribution.

Nile crocodiles are apex predators widely distributed in sub-Saharan Africa that have been viewed and managed as a single species. A complex picture of broad and fine-scale phylogeographic patterns that includes the recognition of two species (Crocodylus niloticus and Crocodylus suchus), and the structuring of populations according to river basins has started to emerge. However, previous studies surveyed a limited number of samples and geographical regions, and large areas of the continent remained unstudied. This work aimed at a fine scale portrait of Nile crocodile populations at the fringes

Population genetics of Aedes aegypti (Diptera: Culicidae) in India reveals high connectivity and weak geographic and climatic structuring.

Understanding the genetic structure and diversity of Aedes aegypti (Linnaeus, 1762) populations is critical for predicting transmission dynamics and implementing effective vector control strategies. This study assessed the genetic diversity, population structure, and influence of geography and climate on Ae. aegypti populations across India. Of 308 genotyped individuals, 257 high-quality genotypes from 13 collection sites across 6 climatic regions were retained for analysis using 10 microsatellite loci. High genetic diversity was observed across all populations, consistent with substantial eff

Comparative Population Genetics and Fludioxonil Sensitivity of Rhizoctonia solani AG-3 Subgroups from Potato and Tobacco in China.

Rhizoctonia solani AG-3 is a major pathogen of solanaceous crops, with subgroups PT, TB, and TM causing potato black scurf and tobacco target spot in China, yet their population structures remain unexplored. Here, we used microsatellite markers to genotype 160 PT isolates from Northeast China, 189 TB and 124 TM isolates from tobacco-growing areas nationwide, and assessed their baseline sensitivity to fludioxonil. The three subgroups exhibited contrasting structures. PT showed high gene diversity (Hexp = 0.634) but significant linkage disequilibrium and low differentiation (FST = 0.008-0.033) w

Taxonomy of Strongyloides in humans, dogs and cats: a comprehensive review from morphology to molecular and population genetics.

The genus Strongyloides (Nematoda; Strongyloididae) comprises over 50 species of nematodes parasitic in terrestrial vertebrates, including humans (Homo sapiens), dogs (Canis lupus familiaris) and cats (Felis catus). Taxonomy of the genus has been shaped by over a century of morphological research, with the most widely adopted framework established in the late 1980s. Advances in molecular genetics have increasingly revealed cryptic diversity and yielded new insights into interspecific and intraspecific relationships within the genus. Despite the rapid expansion of molecular genetic data over th

Genetics by genetic algorithm: Defining an ideal platelet donor population to support patients with HLA-mediated alloimmune refractoriness.

BACKGROUND: Immune factors underlie approximately 20% of patients exhibiting platelet transfusion refractoriness (PTR). PTR can be mitigated by supplying HLA-A and B compatible platelets to these patients. Since 2019, Canadian Blood Services has managed a program to provide compatible platelets to alloimmunized patients. This study evaluates donor pool size and ethnic diversity options to serve Canada's population. STUDY DESIGN AND METHODS: We adapted simulation-based stem cell matching methods by generating simulated patients and matching them to existing and simulated donors using a Python
