HbA2' is a haematologically silent delta chain variant that elutes in the S-region on high performance liquid chromatography. The major clinical significance of HbA2' is that failure to detect it might lead to failure to recognize beta-thalassaemia minor. Co-inheritance of HbA2' and beta-thalassaemia in cis has been described only once in a family from Suriname. We describe a new case of co-inheritance in cis of HbA2' and beta-thalassaemia in a family from Ghanaian origin with a HbA2' of more than 3%. Molecular investigation revealed the same mutations as in the family from Suriname, suggestin

One hundred apparently healthy individuals were used in this study. 71% of these had normal haemoglobin (HbAA) while 29% had sickle cell trait (HbAS). Serum cholesterol and phospholipids were investigated. The mean lipid and lipoprotein concentrations (cholesterol, high density lipoprotein cholesterol and phospholipid) were higher in the sickle cell trait than in the subjects with normal haemoglobin. When separated into blood groups, 24% belonged to the type A+, 11% to the type B+, 2% to the type AB+ and 63% to the type O+. In relation to sexes, a greater percentage of female population had hi

We developed a binary gradient high-performance liquid chromatography (HPLC) method for measuring HbA2 in whole blood samples using a Pharmacia Mono Q column (1 mL) and measurement at 415 nm. The assay requires a simple lysis and centrifugation step before injection onto the column. We found good agreement of results between the HPLC method and the Helena column chromatography method. The within batch precision was 2-6% and between batch precision was 4-6%. We found that using 30 mM Tris buffers (pH 7-8) with a sodium chloride gradient resulted in short analysis times and good chromatographic

BACKGROUND/AIMS: β-Thalassemia is a common hemoglobin disorder distributed worldwide and caused by mutations in the HBB gene that reduce or abolish β-globin chain synthesis, resulting in chronic hemolytic anemia, ineffective erythropoiesis, and variable clinical severity. Although the molecular spectrum of β-thalassemia has been extensively investigated, data correlating β-globin mutations with hematological characteristics in Iraqi patients remain limited. This study aimed to evaluate genotype-phenotype correlations by integrating molecular mutation analysis with hematological and biochemical

Homozygosity for the hemoglobin (Hb) S mutation (HbSS, sickle cell anemia) results in hemoglobin polymerization under hypoxic conditions leading to vaso-occlusion and hemolysis. Sickle cell anemia affects 1:500 African Americans and is a strong risk factor for kidney disease, although the mechanisms are not well understood. Heterozygous inheritance (HbAS; sickle cell trait) affects 1:10 African Americans and is associated with an increased risk for kidney disease in some reports. Using transgenic sickle mice, we investigated the histopathologic, ultrastructural, and gene expression differences

CONTEXT AND OBJECTIVE: Hemoglobin (Hb) D hemoglobinopathies are widespread diseases in northwestern India and usually present with mild hemolytic anemia and mild to moderate splenomegaly. The heterozygous form of Hb D is clinically silent, but coinheritance of Hb D with Hb S or beta-thalassemia produces clinically significant conditions like thalassemia intermedia of moderate severity. Under heterozygous conditions with coinheritance of alpha and beta-thalassemia, patients show a degree of clinical variability. Thus, our aim was to molecularly characterize the Hb D trait among individuals who

β-thalassemia is a quantitative hemoglobin (Hb) disorder resulting in reduced production of Hb A and increased levels of Hb A2. Diagnosis of β-thalassemia can be problematic when combined with other structural Hb variants, so that the separation approaches in routine clinical centers are not sufficiently decisive to obtain accurate results. Here, we separate the intact Hb subunits by high-performance liquid chromatography, followed by top-down tandem mass spectrometry of intact subunits to distinguish Hb variants. Proton transfer reaction-parallel ion parking (PTR-PIP), in which a radical anio

Iran is a country with high prevalence of about 5-10% of beta-thalassemia trait. The prevalence of Cooly's anemia has declined from 11.6 in 10000 population to 7.2 in 10000 in a five-year period due to screening program of beta-thalassemia trait before marriage. This study was conducted to compare the sensitivity of mean corpuscular hemoglobin (MCH) < 27 pg and mean corpuscular volume (MCV) < 80 fl as a screening test in first step of screening of beta-thalassemia trait. From 2449 couples (4898 cases) participating in the premarital screening to our clinic, 902 cases with either MCH < 27 pg, M

Fetal hemoglobin may be slightly or significantly elevated in post-natal life due to a number of causes. We report two novel mutations found on the promoter of the Aγ gene and summarize all common and rare determinants associated with hereditary persistence of fetal hemoglobin (HPFH) described thus far. Hematological and molecular analysis of the Aγ globin gene in two cases of HPFH. Comparison of the novel cases with all those described in the literature. We have found two novel mutations in three Italian patients with HbF values between 5.9% and 6.5% without an elevated HbA(2) and with normal

OBJECTIVE: Hemoglobin Quong Sze (Hb QS) is one of the most common non-deletional α-thalassemia (α-thal), which is prevalent in the Southern Chinese population. However, there are still few comprehensive researches on the molecular characterization of Hb QS. So it is important to find out appropriate diagnosis and characterization of Hb QS carrier for genetic counseling.
MATERIALS AND METHODS: A hematological screening including hematological indices and hemoglobin analysis was performed in 113,400 individuals from Huizhou city, Southern China. Then, suspected thalassemia carriers were detecte

INTRODUCTION: Glycated hemoglobin (HbA1c) is used as a cumulative estimate of mean blood glucose levels from the preceding 5-12 weeks. This is the gold standard in assessing glycemic control in patients with diabetes. The ADA criteria for the diagnosis of diabetes, including HbA1c level, contribute to the importance of recognizing any variation pertaining to the HbA1c measurement. HbA1c is often used as a primary endpoint in the interventional studies among patients with diabetes. Thus, knowledge about factors independently to glycemia, affecting HbA1c is clinically useful.

Haemoglobinopathies are among the most common inherited monogenic disorders worldwide. Thalassaemia screening for carrier status is recommended for adults of reproductive age if suspected of being at risk. Conventional laboratory methods for screening include the assessment of haematological indices, and high-performance liquid chromatography, capillary electrophoresis or isoelectric focusing to measure the levels of HbA2 and HbF, and to identify haemoglobin variants. Each screening method has its advantages and disadvantages, the main disadvantage being that none can fully resolve all variant

BACKGROUND: With the widespread application of third-generation sequencing (TGS) technology, an increasing number of rare or previously unreported β-globin gene variants have been discovered. This study aims to characterize two rare complex β-globin gene variants identified in the Chinese population using TGS, aiming to enrich the spectrum of globin gene variations in this ethnic group. METHODS: Two unrelated Chinese families residing in Fujian Province were recruited for this study. Hematological screening was conducted via routine blood tests and capillary hemoglobin (Hb) electrophoresis. To

Intravascular hemolysis has been described in envenomings by the Eastern coral snake, Micrurus fulvius, in dogs. An experimental model of intravascular hemolysis was developed in mice after intravenous (i.v.) injection of M. fulvius venom. Within one hr, there was prominent hemolysis, associated with a drastic drop in hematocrit, morphological alterations of erythrocytes, hemoglobinemia, and hemoglobinuria. Hemoglobin was identified in urine by mass spectrometry. Histological sections of kidney revealed abundant hyaline casts, probably corresponding to hemoglobin. This effect was abrogated by

Present study demonstrated an existence of anti-aggregation factor(s) in water-soluble extract of a gas-phase of cigarette smoke, and studied chemical characteristics of the factor(s) evaluating its inhibitory potency on platelet aggregation of human and rabbit platelets. The water-soluble extract was prepared by passing mainstream smoke of one cigarette through a Cambridge glassfiber filter and then bubbling it through 1 ml of water. The inhibitory effects were similar in both human and rabbit platelets with the final concentrations of 1 to 5% of the aqueous extract, and were also non-specifi

OBJECTIVE: Per-, sub-, and intertrochanteric femur fractures are common in older adults and elderly patients. They frequently lead to functional decline and increased dependency. Despite the routine use of cephalomedullary nails (CMN) for stabilization, detailed data on perioperative outcomes in this vulnerable population are scarce.
RESULTS: In this retrospective study, we analyzed 401 patients aged ≥ 50 years who underwent CMN fixation for per-, sub-, or intertrochanteric femur fractures at a university medical center between 2019 and 2024. Pre-, intra-, and postoperative parameters, includ
