Postmortem observations on rumen wall histology and gene expression and ruminal and caecal content of beef cattle fattened on barley-based rations.

Sub-acute ruminal acidosis (SARA) can reduce the production efficiency and impair the welfare of cattle, potentially in all production systems. The aim of this study was to characterise measurable postmortem observations from divergently managed intensive beef finishing farms with high rates of concentrate feeding. At the time of slaughter, we obtained samples from 19 to 20 animals on each of 6 beef finishing units (119 animals in total) with diverse feeding practices, which had been subjectively classified as being high risk (three farms) or low risk (three farms) for SARA on the basis of the

Interaction of soil pH and phosphorus efficacy: Long-term effects of P fertilizer and lime applications on wheat, barley, and sugar beet.

Phosphorus (P), a plant macronutrient, must be adequately supplied for crop growth. In Germany, many soils are high in plant-available P; specifically in arable farming, P fertilizer application has been reduced or even omitted in the last decade. Therefore, it is important to understand how long these soils can support sustainable crop production, and what concentrations of soil P are required for it. We analyzed a 36-year long-term field experiment regarding the effects of different P application and liming rates on plant growth and soil P concentrations with a crop rotation of sugar beet, w

The Celiac Patient Antibody Response to Conventional and Gluten-Removed Beer.

Enzymatic digestion, or hydrolysis, has been proposed for treating gluten-containing foods and beverages to make them safe for persons with celiac disease (CD). There are no validated testing methods that allow the quantitation of all the hydrolyzed or fermented gluten peptides in foods and beverages that might be harmful to CD patients, making it difficult to assess the safety of hydrolyzed products. This study examines an ELISA-based method to determine whether serum antibody binding of residual peptides in a fermented barley-based product is greater among active-CD patients than a normal co

Anti-bacterial and anti-biofilm activity of probiotic bacteria against oral pathogens.

In this study, three lactic acid bacteria (LAB), isolated from barley, traditional dried meat and fermented olive were characterized and tested for their anti-bacterial and anti-biofilm activities against oral bacteria. Our results revealed that the tested LAB were γ-hemolytic and were susceptible to four antibiotics. All the strains were resistant to low pH, bile salt, pepsin and pancreatin. Furthermore, FB2 displayed a high aut-oaggregative phenotype (99.54%) while FF2 exhibited the best co-aggregation rate. Concerning the microbial adhesion to solvent, FB2 was the most hydrophobic strain (d

[Celiac disease--the chameleon among the food intolerances].

Celiac disease is an autoimmune disorder resulting from gluten intolerance and is based on a genetically predisposition. Symptoms occur upon exposure to prolamin from wheat, rye, barley and related grain. The pathogenesis of celiac disease has not yet been sufficiently elucidated but is being considered as an autoimmune process. At its core are the deamidation of prolamin fragments, the building of specific antibodies and the activation of cytotoxic T-cells. The immunological inflammatory process is accompanied by structural damages of the enterocytes (villous atrophy, colonization and crypt h

myo-Inositol-1,2,3,4,5,6-hexakisphosphate.

myo-Inositol-1,2,3,4,5,6-hexakisphosphate (Ins P(6)) was first described as an abundant form of phosphorus in plant seeds and other plant tissues and dubbed "phytic acid". Subsequently it was found to be a common constituent in eukaryotic cells, its metabolism a basic component of cellular housekeeping. In addition to phosphate, myo-inositol (Ins) and mineral storage and retrieval in plant organs and tissues, other roles for Ins P(6) include service as a major metabolic pool in Ins phosphate and pyrophosphate pathways involved in signaling and regulation; possibly as an effector or ligand in t

The neurology of gluten sensitivity: separating the wheat from the chaff.

Coeliac disease is the prototypical gluten-sensitive disease. Clinico-pathological features heal on a gluten-free diet and relapse when gluten is reintroduced. An immunopathology is suspected. A number of neurological syndromes may be associated with coeliac disease but it is unclear whether these are directly or indirectly caused by gluten ingestion. It has been proposed that idiopathic ataxias and central nervous system white matter disease are gluten-sensitive syndromes. This is an exciting hypothesis because it offers new therapeutic possibilities including simple exclusion diets. However,

Rhizosphere competitiveness of trichloroethylene-degrading, poplar-colonizing recombinant bacteria.

Indigenous bacteria from poplar tree (Populus canadensis var. eugenei 'Imperial Carolina') and southern California shrub rhizospheres, as well as two tree-colonizing Rhizobium strains (ATCC 10320 and ATCC 35645), were engineered to express constitutively and stably toluene o-monooxygenase (TOM) from Burkholderia cepacia G4 by integrating the tom locus into the chromosome. The poplar and Rhizobium recombinant bacteria degraded trichloroethylene at a rate of 0.8 to 2.1 nmol/min/mg of protein and were competitive against the unengineered hosts in wheat and barley rhizospheres for 1 month (coloniz

Glucose and L-lactate metabolism in pregnant and in lactating ewes fed barley- or ground maize-based diets.

The contribution of glucose absorbed from the small intestine to whole-body glucose metabolism was examined in ewes during late pregnancy and early lactation, using diets based on ground barley or ground maize. Glucose and L-lactate turnover in the whole body and the mesenteric-drained viscera were investigated in these ewes using isotope dilution techniques. The net absorption of glucose by the mesenteric-drained viscera and whole-body glucose turnover were unaffected by diet or reproductive status. Arterial and mesenteric venous blood glucose concentrations, arterial and mesenteric venous L-

Transformation patterns of masked fusarium mycotoxins during barley malting: Mechanistic evidence and multimodal detection in industrial products.

Malt, a key food industry ingredient, faces safety risks from Fusarium mycotoxins and masked derivatives. While malt's agricultural role is clear, biotransformation pathways of masked mycotoxins during barley malting remain poorly understood. This study assesses mycotoxin contamination in commercial malt and links fungal communities to mycotoxin metabolism. Deoxynivalenol dominated, detected in 55% beer and 70% malt samples, with low adult exposure risk. Using ultra performance liquid chromatography-tandem mass spectrometry and internal transcribed spacer sequencing, natural malting showed bip

A four-dimensional spatial transcriptome atlas of barley caryopsis development and germination.

A 4-dimensional spatial gene expression atlas of Hordeum vulgare (barley) grain development and germination was generated using spatial transcriptomic analysis of serial sections to reconstruct transcript abundance in 3 physical dimensions and with temporal kinetics. We investigated the subtissue localizations of specific biological activities, using energy biology as an example, including genes encoding proteins involved in starch synthesis and degradation, sugar transport, and mitochondrial and chloroplast activity. This atlas revealed different patterns in gene expression across tissues and…

Improving the Antioxidant Activity and Consumer Acceptance of Rice Snacks Fortified With Barley.

Although ricebased snacks are popular, they are primarily composed of refined starch and often lack dietary fiber and bioactive compounds. Barley is rich in these compounds; however, its incorporation into rice-based products often results in undesirable changes in physicochemical and sensory properties. Therefore, this study aimed to evaluate the effects of barley fortification on physicochemical properties and antioxidant activities, as well as consumer acceptance of rice snacks to identify an appropriate barley percentage. Rice snacks containing different barley percentages (0%, 20%, 40%, 6

DNA-based assessment of root lesion nematode infections in cereal roots.

Root lesion nematodes (RLN) of the genus Pratylenchus are causing significant damage in cereal production worldwide. Due to climate change and without efficient and environment-friendly treatments, the damages through RLNs are predicted to increase. Microscopic assessments of RLNs in the field and the greenhouses are time-consuming and laborious. As a result, cereal breeders have mostly ignored this pest. We present a method measuring RLN in infected cereal roots using a standardized PCR approach. Publicly available Pratylenchus neglectus primer combinations were evaluated. An optimal primer c

A single administration of barley β-glucan and arabinoxylan extracts reduces blood glucose levels at the second meal via intestinal fermentation.

Diet with barley may suppress the glycemic response after consuming the next meal ("second meal effect"). This study aimed to investigate the second meal effect and its mechanism. Mice were given a single dose of β-glucan or arabinoxylan, the primary sources of soluble fiber in barley. A single dose of β-glucan or arabinoxylan extract, followed 6 h later by a 20% glucose solution (second meal), suppressed blood glucose elevation. Arabinoxylan and β-glucan increased the levels of short-chain fatty acids (SCFAs) in the ileum and cecum, respectively. Total GLP-1 secretion in the blood increased w

Barley Ror1 encodes a class XI myosin required for mlo-based broad-spectrum resistance to the fungal powdery mildew pathogen.

Loss-of-function alleles of plant MLO genes confer broad-spectrum resistance to powdery mildews in many eudicot and monocot species. Although barley (Hordeum vulgare) mlo mutants have been used in agriculture for more than 40 years, understanding of the molecular principles underlying this type of disease resistance remains fragmentary. Forward genetic screens in barley have revealed mutations in two Required for mlo resistance (Ror) genes that partially impair immunity conferred by mlo mutants. While Ror2 encodes a soluble N-ethylmaleimide-sensitive factor-attached protein receptor (SNARE), t…

Agrobacterium-Mediated Transformation of Barley Immature Embryos.

Barley transformation is an essential tool for a range of functional genomics studies as well as for future crop improvement. The demand for efficient crop transformation systems continues to grow, with new genome editing technologies adding to that demand. Here we describe an efficient and routine transformation protocol for the spring barley Golden Promise, based on Agrobacterium-mediated inoculation of immature embryos. This protocol has been widely used for overexpression and RNAi applications and more recently for CRISPR/Cas9 mediated genome editing. Average transformation efficiencies of

Components of rhizospheric bacterial communities of barley and their potential for plant growth promotion and biocontrol of Fusarium wilt of watermelon.

This work aimed to characterize antagonistic bacteria from the field-grown barley rhizosphere, and evaluate their potential for growth promotion and biocontrol of Fusarium wilt on watermelon caused by Fusarium oxysporum f. sp. Niveum (FON). Seven bacteria were isolated and screened for plant growth promoting and antagonistic traits. Based on the results of phenotypic characterization and 16S rRNA gene sequencing, the isolates were identified to be related to Bacillus methylotrophicus (DMK-1), Bacillus amyloliquefaciens subsp. plantarum (DMK-7-2), Bacillus cereus (DMK-12), Pseudomonas brassicac
