Impact resistance emerges from the coupling of strong load-bearing networks and dynamic interfacial interactions that enable effective stress transfer and energy dissipation. Although hydrogels are promising candidates for impact-resistant soft materials, it remains challenging to reinforce both networks and interfaces simultaneously in hydrogels, which limits their performance under high strain-rate loading. To overcome this limitation, we develop a composite hydrogel comprised of a poly(vinyl alcohol) (PVA) matrix reinforced with chitosan-sodium alginate nanofibers (CSNFs), using sodium citr…

Most organoids are cultured in Matrigel, a complex and poorly defined matrix that limits our mechanistic understanding. Synthetic hydrogels offer a versatile alternative, providing precise control over mechanical and biochemical cues. Using two topologically different types of hydrogel precursors, branched poly-(ethylene glycol) (PEG) and PEG bisdendrons, we have obtained a library of hydrogels via thiol-ene cross-linking with branched PEG-thiol. Their chemical conversion was monitored by Raman spectroscopy, while swelling and mechanical properties, including elastic, viscoelastic, and relaxat…

Macrophages play a pivotal role in tissue regeneration and immune regulation through efferocytosis and other mechanisms, with their intracellular communication largely dependent on second messengers (e.g., cyclic adenosine monophosphate, cAMP). However, the broad-spectrum nature of these second messengers renders related interventions susceptible to uncontrollable systemic and off-target effects. Here, we developed an injectable apoptosis-mimicking hybrid hydrogel (PAS@Gel) that enables specific and coordinated macrophage intervention of both first and second messengers involved in efferocytos

Hydrogel electrolytes are promising for aqueous energy storage, yet their compatibility with kosmotropic salts remains fundamentally limited by the salting-out effect. In conventional hydrophilic polymer networks, strongly hydrated kosmotropic anions preferentially retain their hydration shells, leading to polymer dehydration and structural instability. Here, we show that this incompatibility can be overcome by reversing the Hofmeister response of the hydrogel through anion-polymer affinity chemistry. Guided by the law of matching water affinities, we introduce chaotropic quaternary ammonium g

Pancreatic ductal organoids (PDOs) generated from human induced pluripotent stem cells (iPSCs) can be used to model pancreatic diseases and to conduct drug screening/testing. However, current protocols for generating PDOs rely heavily on tumor-derived Matrigel, which has been shown to upregulate oncogenes. Furthermore, Matrigel has undefined composition and weak mechanical properties that hamper mechanistic studies of cell-material interactions. In this study, we explore photo-clickable decellularized small intestine submucosa-norbornene (dSIS-NB) hydrogels as a Matrigel replacement for genera…

Oncolytic viruses act as in situ cancer vaccines by inducing antigen release to boost antitumor immunity. Specific intratumoral bacteria exacerbate immunosuppression; however, their influence on oncolytic immunotherapy remains unknown. Here, we report a hybrid bacteriophage hydrogel composed of engineered oncolytic M13 phage (eM13) and Escherichia coli (E. coli) phage Tequatrovirus YZ2, which suppresses breast cancer progression and metastasis by boosting antitumor immunity through selective elimination of intratumoral E. coli. Particularly, YZ2 is conjugated with photosensitizer CyI to produc

This study presents a novel approach to modulate peptide self-assembly, solubility, and functional properties by utilizing an electric field as an external stimulus. Three short, heterochiral tripeptides, P1, P2, and P3, were designed based on their ability to form hydrogels. We demonstrate that the structural and functional properties of peptide-based hydrogels can be modulated upon exposure to an electric field. Our findings indicate that the external electric field does not alter the secondary structure of the designed peptides, however, the electric field plays a significant role in regula…

Conventional therapies still provide only limited true tissue regeneration for periodontal diseases, particularly periodontitis, which are highly prevalent chronic inflammatory conditions associated with irreversible loss of tooth-supporting tissues and relevant systemic consequences. In this context, hydrogels based on natural polymers have been widely explored in periodontal tissue engineering as biomimetic matrices capable of modulating the inflammatory response, enabling localized delivery of bioactive agents, and offering temporary structural support. This review summarizes recent advance…

INTRODUCTION: This study investigates, for the first time, the effects of plasma-treated alginate hydrogels (PTH) on porcine leukocyte activity in vitro. Cold atmospheric plasma (CAP) generates reactive oxygen and nitrogen species (RONS), which can modulate immune cell behavior and may promote tissue repair.
RESULTS: Our findings demonstrate that PTH induce a controlled increase in nitrite and superoxide production in both neutrophil and monocyte cultures, particularly at higher treatment levels, without evidence of lytic cell death or significant loss of viability. PTH application resulted i…

Hydrogel/electrospun polymer nanofiber composites (HENC) integrate the advantages of both components. Hydrogels provide high water content, biocompatibility, and tunable drug release, while electrospun nanofibers offer a high surface area, loading capacity, customizable morphology, and opportunities for functionalization. Nanofibers can also be incorporated into hydrogels as 3D-printable inks. Together, these features create biomimetic composites that modulate drug release and mimic native tissues. This article reviews electrospinning fundamentals, limitations, preparation methods for HENC, an…

The bioartificial pancreas, composed of a semi-permeable hydrogel encapsulating insulin-secreting cells, has attracted attention as a treatment for type 1 diabetes. In this study, we developed phospholipid polymer-modified alginate hydrogel beads that encapsulated spheroids of the pancreatic beta cell line MIN6. The hydrogel beads were composed of methacrylated alginic acid, which enabled both ionic and covalent cross-linking, resulting in a hydrogel that was more stable than conventional alginate hydrogels. Furthermore, modification of biocompatible 2-methacryloyloxyethyl phosphorylcholine (M…

Chemotherapy using anticancer agents and radiotherapy of cancers frequently induce the development of stomatitis as a side effect. In the present study, hydrogels for effective stomatitis healing under anticancer drug administration were developed using three components, namely proline, carboxyvinyl polymer, and water (denoted proline gels). Characterization including tilting, Fourier transform infrared spectra, and viscoelasticity measurements indicated that proline gels with proline concentrations over 300 μmol/g could retain water on the tongue of mice. The degradation and release behavior …

The study presents novel hydrogels obtained by crosslinking chitosan with both furfural and glutaraldehyde via dynamic imine bonds. Scanning electron microscopy confirmed the formation of porous networks with a mean diameter of the pores between 15 and 35 μm, while the supramolecular characterization by polarized optical microscopy and wide-angle X-ray diffraction proved the gelation mechanism. The hydrogels presented great rheological properties, along with an anti-creep behavior. The resulting materials were highly adhesive and had great antioxidant activity, leading to an inhibition of 78 %…

Extracellular vesicles (EVs) have been actively explored for therapeutic applications in the context of cancer and other diseases. However, the poor tissue retention of EVs has limited the development of EV-based therapies. Here we report a facile approach to fabricating injectable EV hydrogels with tunable viscoelasticity and gelation temperature, by metabolically tagging EVs with azido groups and further crosslinking them with dibenzocyclooctyne-bearing polyethylene glycol via efficient click chemistry. One such EV gel has a gelation temperature of 39.4 °C, enabling in situ gelation of solut…

Typically, externally triggered initiators are required for norbornene-modified polysaccharides to undergo thiol-ene gelation into a hydrogel. However, anecdotal and reported evidence exists of these prehydrogel solutions undergoing spontaneous thiol-ene gelation without the trigger. Understanding the origin of this spontaneous process would improve the storage of prehydrogel solutions. We found that polysaccharides with carboxylic acids (e.g., carboxymethyl cellulose) can overcome the buffering capacity of phosphate-buffered saline (PBS), creating an acidic solution that is required for spont

Nanocomposite hydrogels, i.e.; a combination of hydrogels and nanoparticles, have emerged as a promising platform for drug delivery owing to the high degree of design flexibility. Despite the wealth of literature on nanocomposite hydrogels, there is scant experimental data on the release of drug-loaded nanoparticles from hydrogels. Here, we report on the incorporation and subsequent release of dye-loaded nanoparticles from alginate-based hydrogels. We investigated the rheological properties of alginate and alginate-poly(ethylene oxide) hydrogels as a function of nanoparticle loading, as well a…

Microorganisms can induce biomineralization of inorganic ions to form a lipophobic layer on the surface of cave rocks. Mimicking this, we developed and propose a protocol that produces a highly lipophobic hybrid layer of CaCO3 nanoparticles on hydrogel surfaces. This lipophobic layer endows hydrogels with an oil contact angle of 162°, causing oil droplets placed on the surface to bead up and roll off immediately. The lipophobic surface effectively resists staining from lipophilic dyes, and does not adhere to double-sided tapes. A lipophobic layer on hydrogel tubes effectively prevents the adhe…
