Cardiopulmonary toxicity of adoptive immunotherapy.

Adoptive immunotherapy, the administration of interleukin-2 (IL-2) and interleukin-2 activated cells, leads to tumor regression in some patients with advanced cancer. Although this new therapeutic modality offers hope for the future, at present, a multitude of toxicities limit the total dose and duration of therapy. Among the toxic side effects a purported third space or vascular leak syndrome is the most serious. In this review, we detail the evidence for a third space syndrome (peripheral edema, ascites, oliguria, elevated serum creatinine levels) and cardiopulmonary dysfunction (hypotension…

Immunotherapy of renal cell carcinoma.

The clinical trials of immunotherapy of renal carcinoma have been based on the theory that manipulation of the host-immune response can influence the growth of malignant cells in the host. The results cited above do not demonstrate a clear therapeutic role of immunotherapy but they do provide evidence to encourage further studies. This is especially pertinent in view of the current absence of effective systemic agents. The stimulation of the host response appears to be finite and cannot be expected to control massive tumor burden. The clinical trials analyzed above must be interpreted with thi…

Adjuvant immunotherapy in bronchogenic carcinoma.

Ten patients with locally far advanced bronchogenic carcinoma were treated with autologous tumor vaccine after a radical surgical resection. Three of 10 patients treated with tumor vaccine are alive (median survival of 26 months) whereas all nine patients in the control group are dead (median survival of 6.5 months). Active immunotherapy adjunct to radical surgery appeared to improve cellular immunity of the treated patients in the postoperative period, and the migration inhibition test indicated sensitization of treated patients against their tumor.

Immunotherapies and their potential to preserve beta cells in type 1 diabetes: a review of current immunotherapy.

Type 1 diabetes is a chronic immune-mediated disease that leads to the loss of beta cells in the pancreas, clinically characterized by polyd … een developed based on diverse mechanisms in the disease's pathogenesis. This review focuses on new immunotherapy approaches for type 1 diabetes, including cell-targeted therapies, cytokines, and cell transplants to preserve beta-cell function.

DNA Damage Repair Status Predicts Opposite Clinical Prognosis Immunotherapy and Non-Immunotherapy in Hepatocellular Carcinoma.

Immune checkpoint inhibitors(ICIs) that activate tumor-specific immune responses bring new hope for the treatment of hepatocellular carcinoma(HCC). However, there are still some problems, such as uncertain curative effects and low objective response rates, which limit the curative effect of immunotherapy. Therefore, it is an urgent problem to guide the use of ICIs in HCC based on molecular typing. We downloaded the The Cancer Genome Atlas-Liver hepatocellular carcinoma(TCGA-LIHC) and Mongolian-LIHC cohort. Unsupervised clustering was applied to the highly variable data regarding expression of …

Perspectives in immunotherapy: meeting report from the Immunotherapy Bridge (29-30 November, 2017, Naples, Italy).

Immunotherapy represents the third important wave in the history of the systemic treatment of cancer after chemotherapy and targeted therapy and is now established as a potent and effective treatment option across several cancer types. The clinical success of anti-cytotoxic T-lymphocyte-associated antigen (CTLA)-4, first, and anti-programmed death (PD)-1/PD-ligand (L)1 agents in melanoma and other cancers a few years later, has encouraged increasing focus on the development of other immunotherapies (e.g. monoclonal antibodies with other immune targets, adoptive cell transfer, and vaccines), wi…

Cancer immunotherapy out of the gate: the 22nd annual Cancer Research Institute International Immunotherapy Symposium.

The 22nd annual Cancer Research Institute (CRI) International Immunotherapy Symposium was held from October 5-8, 2014, in New York City. Titled "Cancer Immunotherapy: Out of the Gate," the symposium began with a Cancer Immunotherapy Consortium satellite meeting focused on issues in immunotherapy drug development, followed by five speaker sessions and a poster session devoted to basic and clinical cancer immunology research. The second annual William B. Coley lecture was delivered by Lieping Chen, one of the four recipients of the 2014 William B. Coley Award for Distinguished Research in Tumor …

Immunotherapy in Allergic and Mixed Rhinitis.

Allergen immunotherapy is the only treatment modality that can modify severity of allergic rhinitis. Subcutaneous immunotherapy (SCIT) and sublingual immunotherapy (SLIT) are both effective treatment options. SLIT has a better safety profile with rare reports of systemic reaction events. In this article, we will review evidence pertaining to efficacy of individual treatment allergens and assess relative efficacy and safety of SLIT versus SCIT.

Advances in Immunotherapy for Breast Cancer.

Immunotherapy has transformed the treatment of a variety of malignancies, including breast cancer. Given the unmet clinical need, immunotherapy was first studied in advanced-stage, triple-negative breast cancer (TNBC). The anti-PD-1 agent pembrolizumab plus chemotherapy is approved for both advanced and early TNBC, and data from randomized phase 3 trials in high-risk, early-stage, hormone receptor-positive breast cancer have demonstrated promising efficacy. Ongoing trials aim to identify the most appropriate patients for treatment, amount and timing of immunotherapy needed for optimal outcomes…

Immunotherapy in Head and Neck Cancer.

Head and neck cancer (HNC) is the seventh most common cancer worldwide, with incidence growing every year. While the standard regimen of surgery, chemotherapy, radiotherapy, targeted anti-EGFR therapy, and immune checkpoint inhibition are effective in a subset of patients, therapeutic resistance is a persistent challenge for managing this disease. The use of immunotherapeutic agents to treat HNC has risen over recent years, with advances in immune checkpoint inhibition, targeted therapy with bispecific antibodies, modified cytokine delivery, adoptive cell therapy, and virus-based therapeutics

New developments in immunotherapy for SCLC.

Small cell lung cancer (SCLC) is an aggressive form of neuroendocrine neoplasm known for its striking initial response to treatment, followed by fast relapse and refractoriness in response to additional lines of therapy. New advances in immunotherapy are paving the way for more effective treatment strategies and have promising results with early clinical trial data. While SCLC rarely harbors actionable mutations, the receptor DLL3 is extensively present in SCLC, making it a potential target for immunotherapy. Three emerging therapeutic options include bispecific T cell engagers targeting DLL3,…

Immunobiology and immunotherapy of COVID-19.

The SARS-CoV-2 outbreak in late 2019 triggered a major increase in activities related to immunobiology and immunotherapy to cope with and find solutions to end the COVID-19 pandemic. The unprecedented approach to research and development of drugs and vaccines against SARS-CoV-2 has substantially improved the understanding of immunobiology for COVID-19, which can also be applied to other infectious diseases. Major efforts were dedicated to the repurposing of existing antiviral drugs and the development of novel ones. For this reason, numerous approaches to evaluating interferons, immunoglobulin…

Cellular and immunotherapies for myelodysplastic syndromes.

In this review article, we outline the current landscape of immune and cell therapy-based approaches for patients with myelodysplastic syndromes (MDS). Given the well characterized graft-versus-leukemia (GVL) effect observed with allogeneic hematopoietic cell transplantation, and the known immune escape mechanisms observed in MDS cells, significant interest exists in developing immune-based approaches to treat MDS. These attempts have included antibody-based drugs that block immune escape molecules, such as inhibitors of the PD-1/PD-L1 and TIM-3/galectin-9 axes that mediate interactions betwee
