Iron regulatory proteins secure iron availability in skeletal muscle to preserve exercise tolerance in heart failure.

AIMS: Iron deficiency (ID) is a frequent comorbidity in heart failure (HF) and contributes to exercise intolerance. Tissue iron levels are maintained by cellular iron uptake, sequestration, and release, processes that are tightly controlled by iron regulatory proteins (IRP). Our aim was to explore the role of IRP activity in skeletal muscle function and exercise capacity during HF. METHODS AND RESULTS: We observed that skeletal muscle ID is associated with IRP1 and 2 inactivation 12 weeks after transverse aortic constriction (TAC) in mice with left ventricular (LV) dysfunction and cachexia. Metabolism-Disrupting Chemicals and the Constitutive Androstane Receptor CAR.

During the last two decades, the constitutive androstane receptor (CAR; NR1I3) has emerged as a master activator of drug- and xenobiotic-metabolizing enzymes and transporters that govern the clearance of both exogenous and endogenous small molecules. Recent studies indicate that CAR participates, together with other nuclear receptors (NRs) and transcription factors, in regulation of hepatic glucose and lipid metabolism, hepatocyte communication, proliferation and toxicity, and liver tumor development in rodents. Endocrine-disrupting chemicals (EDCs) constitute a wide range of persistent organi

Folliculin (Flcn) inactivation leads to murine cardiac hypertrophy through mTORC1 deregulation.

Cardiac hypertrophy, an adaptive process that responds to increased wall stress, is characterized by the enlargement of cardiomyocytes and structural remodeling. It is stimulated by various growth signals, of which the mTORC1 pathway is a well-recognized source. Here, we show that loss of Flcn, a novel AMPK-mTOR interacting molecule, causes severe cardiac hypertrophy with deregulated energy homeostasis leading to dilated cardiomyopathy in mice. We found that mTORC1 activity was upregulated in Flcn-deficient hearts, and that rapamycin treatment significantly reduced heart mass and ameliorated c

mTORC1 Upregulation Leads to Accumulation of the Oncometabolite Fumarate in a Mouse Model of Renal Cell Carcinoma.

Renal cell carcinomas (RCCs) are common cancers diagnosed in more than 350,000 people each year worldwide. Several pathways are de-regulated in RCCs, including mTORC1. However, how mTOR drives tumorigenesis in this context is unknown. The lack of faithful animal models has limited progress in understanding and targeting RCCs. Here, we generated a mouse model harboring the kidney-specific inactivation of Tsc1. These animals develop cysts that evolve into papillae, cystadenomas, and papillary carcinomas. Global profiling confirmed several metabolic derangements previously attributed to mTORC1. N

Effects of Bordetella avium toxin on turkey tracheal organ cultures as measured with a tetrazolium-reduction assay.

Turkey tracheal organ cultures (TOCs) were exposed to one of the following Bordetella avium fractions or controls: live B. avium, formalin-killed B. avium, B. avium sonicate, heat-inactivated sonicate, culture supernatant, heat-inactivated culture supernatant, phosphate-buffered saline, or brain-heart infusion broth. After the TOCs were incubated for 2 hours with the bacterial fractions, the cellular metabolism of each TOC was evaluated using a tetrazolium chloride reduction assay, and cellular morphology was determined by light microscopy. Additionally, bacterial fractions and controls were i

Rett phenotype with X/autosome translocation: possible mapping to the short arm of chromosome X.

Rett syndrome (RS) was diagnosed in a girl with a t(X;22) (p11.22;p11). This translocation was also present in her unaffected mother and her sister affected by a neurological disorder compatible with a "forme fruste" of RS. Different etiological mechanisms are considered: gene disruption, X inactivation disturbance, metabolic interference. Whatever this may be, the localization of a RS related gene to the short arm of chromosome X is likely.

Inactivation of low-Km rat liver mitochondrial aldehyde dehydrogenase by cyanamide in vitro. A catalase-mediated reaction.

The inactivation of the affinity chromatography purified low-Km rat liver mitochondrial aldehyde dehydrogenase (ALDH)--free of catalase activity--by the alcohol sensitizing agent cyanamide was studied in vitro. This ALDH-purified preparation was not susceptible to cyanamide inactivation at concentrations up to 2.5 mM. On the other hand, ALDH activity appears to be irreversibly inhibited when the incubation mixture contained ALDH, catalase, NAD+ and cyanamide. Influence of catalase, NAD+ and cyanamide concentrations in the incubation mixtures on the ALDH activity were also established. The time

Glucokinase Inactivation Reduces Lysophosphatidylcholine Accumulation to Alleviate High-Fat Diet-Induced Hepatic Steatosis.

Partial inactivation of glucokinase (GCK) is typically characterized by mild hyperglycemia and a favorable lipid profile compared to type 2 diabetes. Previous studies have shown that GCK activity influences serum lipid profiles in a diet-dependent manner; however, its role in hepatic lipid metabolism in the context of metabolic dysfunction-associated steatotic liver disease (MASLD) remains unclear. To address this, we utilized a newly established heterozygous GCK mutation knock-in mouse model (GCKMut) fed either a normal diet (ND) or a high-fat diet (HFD). Under ND conditions, GCKMut mice deve

Integrated inactivation of Microcystis aeruginosa and degradation of microcystin-LR by direct current glow discharge plasma in liquid-phase: Mechanisms and cell deactivation process.

The frequent occurrence of blooms of Microcystis aeruginosa (M. aeruginosa) and the subsequent release of microcystin-LR (MC-LR) in eutrophic waters pose a serious threat to aquatic ecosystems. This study investigated the optimal conditions for inactivating M. aeruginosa and the degrading MC-LR using direct current glow discharge plasma in liquid phase (DC-LGDP), analyzed the potential inactivation mechanisms and the cell deactivation process of M. aeruginosa. The results showed that DC-LGDP generated reactive species (i.e., •OH, 1O2, and H2O2), active Cl and electroporation effect collectivel

CDKN2A alterations as possible molecular link between environmental pollutants and pathogenesis of liver cancer: Epidemiology, mechanisms, and insights.

Hepatocellular carcinoma (HCC) represents a global public health burden of increasing magnitude, driven by risk factors including viral hepatitis, metabolic dysfunction-associated fatty liver disease and various environmental carcinogens, which may inactivate key tumor suppressor genes or activates oncogenes. Recent studies have linked various environmental exposures with inactivation of tumor suppressor gene CDKN2A (cyclin dependent kinase inhibitor 2A) and its encoded proteins p16INK4a and p14ARF to various molecular pathways leading to hepatocarcinogenesis. Current studies suggest p16INK4a

Malnutrition with hypoaminoacidemia in a 22-year-old pregnant patient masking a likely ornithine transcarbamylase deficiency.

BACKGROUND: Symptoms and clinical presentations of OTC deficiency vary widely according to the remaining activity of the enzyme. Three factors determine the residual enzyme activity. First, as the OTC gene is carried on the X chromosome, a complete inactivation of this enzyme in a newborn boy results an acute ammonia intoxication. Second, the female mosaicism due to lyonization (differential randomized X-inactivation) leads to differential OTC expression in hepatocytes. Third, the degree of severity depends on the mutation and the level of remaining activity it leaves to the protein. Published

Metabolic perspectives on in vitro toxicity tests.

1. The toxic effects of chemicals arise either from the chemical itself or from metabolites. In either event, the concentration of the molecular species responsible for toxicity is under the control of metabolism. This metabolism is highly variable between animals and humans, and in human populations may exhibit substantial inter-individual differences. The range of human variability can be covered by resorting to a number of animal models. 2. For many purposes, it is desirable to have in vitro procedures to allow detailed examination of mechanisms, but for a long time it was commonly the case

[Microbe-inspired system enzymology of vitamin B₁₂ metabolism].

Vitamin B₁₂ is produced only by prokaryotes and utilized by animals as an essential micronutrient. Genetic complementation analysis of cell lines from patients indicated that at least eight gene products are involved in intracellular B₁₂ metabolism and utilization. We have investigated bacterial adenosylcobalamin-dependent enzymes and elucidated their structure-based fine mechanisms. They tend to undergo mechanism-based inactivation during catalysis, because they use highly reactive radicals for catalyzing chemically difficult reactions. We have discovered molecular chaperone-like reactivating

Liver-specific deletion of histone deacetylase 3 disrupts metabolic transcriptional networks.

Histone deacetylase 3 (Hdac3) is an enzymatic component of transcriptional repression complexes recruited by the nuclear hormone receptors. Inactivation of Hdac3 in cancer cell lines triggered apoptosis, and removal of Hdac3 in the germ line of mice caused embryonic lethality. Therefore, we deleted Hdac3 in the postnatal mouse liver. These mice developed hepatomegaly, which was the result of hepatocyte hypertrophy, and these morphological changes coincided with significant imbalances between carbohydrate and lipid metabolism. Loss of Hdac3 triggered changes in gene expression consistent with i

Stressful preconditioning and HSP70 overexpression attenuate proteotoxicity of cellular ATP depletion.

Rat H9c2 myoblasts were preconditioned by heat or metabolic stress followed by recovery under normal conditions. Cells were then subjected to severe ATP depletion, and stress-associated proteotoxicity was assessed on 1) the increase in a Triton X-100-insoluble component of total cellular protein and 2) the rate of inactivation and insolubilization of transfected luciferase with cytoplasmic or nuclear localization. Both heat and metabolic preconditioning elevated the intracellular heat shock protein 70 (HSP70) level and reduced cell death after sustained ATP depletion without affecting the rate

Mechanistic Investigation of the Time-Dependent Aldehyde Oxidase Inhibitor Hydralazine.

The anti-hypertensive agent hydralazine is a time-dependent inhibitor of the cytosolic drug-metabolizing enzyme aldehyde oxidase (AO). Glutathione (GSH) was found to suppress the inhibition of AO by hydralazine in multiple enzyme sources (human liver and kidney cytosol, human liver S9, rat liver S9, and recombinant human AO) and with different AO substrates (zoniporide, O6 -benzylguanine, and dantrolene). Hydralazine-induced AO inactivation was unaffected when GSH was added to the incubation mixture after pre-incubation of hydralazine with AO (rather than during the pre-incubation), suggesting

Formation of Both Heme and Apoprotein Adducts Contributes to the Mechanism-Based Inactivation of Human CYP2J2 by 17α-Ethynylestradiol.

17α-Ethynylestradiol (EE), a major component of many oral contraceptives, affects the activities of a number of the human cytochrome P450 (P450) enzymes. Here, we characterized the effect of EE on CYP2J2, a major human P450 isoform that participates in metabolism of arachidonic acid. EE inactivated the hydroxyebastine carboxylation activity of CYP2J2 in a reconstituted system. The loss of activity is time and concentration dependent and requires NADPH. The KI and kinact values for the inactivation were 3.6 μM and 0.08 minute-1, respectively. Inactivation of CYP2J2 by EE was due to formation of

IKKβ signaling mediates metabolic changes in the hypothalamus of a Huntington disease mouse model.

Huntington disease (HD) is a neurodegenerative disorder caused by a CAG repeat expansion in the huntingtin (HTT) gene. Metabolic changes are associated with HD progression, but underlying mechanisms are not fully known. As the IKKβ/NF-κB pathway is an essential regulator of metabolism, we investigated the involvement of IKKβ, the upstream activator of NF-κB in hypothalamus-specific HD metabolic changes. We expressed amyloidogenic N-terminal fragments of mutant HTT (mHTT) in the hypothalamus of mice with brain-specific ablation of IKKβ (Nestin/IKKβlox/lox) and control mice (IKKβlox/lox). We ass

The effects of biocides (antiseptics and disinfectants) on the endospores of Rhinosporidium seeberi.

No data exists on the activity of biocides (antiseptics and disinfectants) on Rhinosporidium seeberi that causes rhinosporidiosis in humans and animals. On account of the inability to culture R. seeberi, in vitro, dyes were used to assess the morphological integrity and viability of biocide-treated endospores that are considered to be the infective stage of this pathogen. Evan's Blue (EvB) identifies the morphological integrity of the endospores while MTT (3-[4, 5-dimethylthiazol-2yl]-2, 5-diphenyl tetrazolium bromide) identifies metabolic activity through its reduction by cellular dehydrogena
