The role of N6-methyladenosine modification on diapause in silkworm (Bombyx mori) strains that exhibit different voltinism.

N6-methyladenosine (m6 A) plays a key role in regulating gene expression in myriad organisms. Diapause is an important plastic phenotype that allows insects to survive under specific environmental conditions. However, the diapause molecular mechanism remains unknown. In this study, we analyzed the phylogenetics of genes related to the m6 A modification complex in the silkworm (Bombyx mori) based on identified sequences from other organisms. We detected the expression of these genes during different developmental phases from four strains with different voltinism. We also determined total m6 A c

Sequence and specificity of two antibacterial proteins involved in insect immunity.

Immune responses have been described for many different insect species. However, it is generally acknowledged that immune systems must therefore differ from those of vertebrates. An effective humoral immune response has been found in pupae of the cecropia moth, Hyalophora cecropia. The expression of this multicomponent system requires de novo synthesis of RNA and proteins and its broad antibacterial activity is due to at least three independent mechanisms, the most well known of which is the insect lysozyme. However, this enzyme is bactericidal for only a limited number of Gram-positive bacter

Neuropeptide hormone bursicon mediates female reproduction in the whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae).

Bursicon, a neuropeptide hormone comprising two subunits-bursicon (burs) and partner of burs (pburs), belongs to the cystine-knot protein family. Bursicon heterodimers and homodimers bind to the lucine-rich G-protein coupled receptor (LGR) encoded by rickets to regulate multiple physiological processes in arthropods. Notably, these processes encompass the regulation of female reproduction, a recent revelation in Tribolium castaneum. In this study we investigated the role of burs/pburs/rickets in mediating female vitellogenesis and reproduction in a hemipteran insect, the whitefly, Bemisia taba

Cloning and Characterization of Drosophila melanogaster Juvenile Hormone Epoxide Hydrolases (JHEH) and Their Promoters.

Juvenile hormone epoxide hydrolase (JHEH) plays an important role in the metabolism of JH III in insects. To study the control of JHEH in female Drosophila melanogaster, JHEH 1, 2 and 3 cDNAs were cloned and sequenced. Northern blot analyses showed that the three transcripts are expressed in the head thorax, the gut, the ovaries and the fat body of females. Molecular modeling shows that the enzyme is a homodimer that binds juvenile hormone III acid (JH IIIA) at the catalytic groove better than JH III. Analyses of the three JHEH promoters and expressing short promoter sequences behind a reporte

The role of serotonin in feeding and gut contractions in the honeybee.

Serotonin (5-hydroxytryptamine, 5-HT) is involved in the regulation of feeding and digestion in many animals from worms to mammals. In insects, 5-HT functions both as a neurotransmitter and as a systemic hormone. Here we tested its role as a neurotransmitter in feeding and crop contractions and its role as a systemic hormone that affected feeding in adult foraging honeybees. We found 5-HT immunoreactive processes throughout the gut, including on the surface of the oesophagus, crop, proventriculus, and the midgut, as well as in the ventral nerve cord. mRNA transcripts for all four of the known

The lethal effects of Cyperus iria on Aedes aegypti.

The sedge Cyperus iria, a common weed in rice, contains large amounts of the insect hormone (10R) juvenile hormone III (JH III). Given its widespread distribution in Asia and Africa, we examined the possibility that C. iria could be used as a safe, inexpensive, and readily available mosquito larvicide. Plants of varying ages were harvested and leaves tested for lethal effects on larvae of the yellow fever mosquito, Aedes aegypti. The median lethal doses (LD50s) for frozen leaves from 1- and 2-month-old plants were 267 and 427 mg/100 ml of water, respectively. Leaves from 1-month-old C. iria co

Jasmonic acid signaling in plants: regulatory mechanisms in development and stress responses.

Jasmonic acid (JA), one of the most pivotal plant hormones, has emerged as a central focus in plant biology research due to its extensive and diverse biological functions. This review synthesizes the significant advancements made over the past decade in understanding JA's role in regulating plant development and mediating responses to environmental stresses, areas that lacked systematic review in previous years. We provide a concise overview of JA's biosynthetic and signal transduction pathways, with particular emphasis on the key regulatory complex comprising CORONATINE INSENSITIVE 1 (COI1),

RNAi Targeting MhieJHEH Disrupts Hormonal Homeostasis and Induces Systemic Metabolic Dysregulation in Monolepta Hieroglyphica.

Juvenile hormone epoxide hydrolase (JHEH) plays a crucial role in insect development by degrading juvenile hormone (JH). While functionally characterized in some leaf beetle species (e.g., Leptinotarsa), its molecular mechanisms and physiological roles in many agriculturally important Chrysomelids remain poorly understood. This study provides the first comprehensive functional analysis of MhieJHEH in the maize pest Monolepta hieroglyphica. We cloned and sequenced the gene, which encodes a 454-amino acid protein classified within the Coleoptera JHEH clade. Structural modeling revealed a binding

Identification of ETH receptor and its possible roles in the mud crab Scylla paramamosain.

Ecdysis-triggering hormone (ETH) is a neuropeptide hormone characterized by a conserved KxxKxxPRx amide structure widely identified in arthropods. While its involvement in the regulation of molting and reproduction in insects is well-established, its role in crustaceans has been overlooked. This study aimed to de-orphanise a receptor for ETH in the mud crab Scylla paramamosain and explore its potential impact on ovarian development. A 513-amino-acid G protein-coupled receptor for ETH (SpETHR) was identified in S. paramamosain, exhibiting a dose-dependent activation by SpETH with an EC50 value

Systemic and local effect of the Drosophila headcase gene and its role in stress protection of Adult Progenitor Cells.

During the development of a holometabolous insect such as Drosophila, specific group of cells in the larva survive during metamorphosis, unlike the other larval cells, and finally give rise to the differentiated adult structures. These cells, also known as Adult Progenitor Cells (APCs), maintain their multipotent capacity, differentially respond to hormonal and nutritional signals, survive the intrinsic and environmental stress and respond to the final differentiation cues. However, not much is known about the specific molecular mechanisms that account for their unique characteristics. Here we

Vitamin C and the abscisic acid-insensitive 4 transcription factor are important determinants of aphid resistance in Arabidopsis.

AIMS: Aphids, like other insects, are probably unable to synthesize vitamin C (ascorbic acid), which is therefore an essential dietary nutrient that has to be obtained from the host plant. Plant responses to aphids involve hormones such as salicylic acid (SA), jasmonic acid (JA), and abscisic acid (ABA), but hormone/redox interactions remain poorly characterized. We therefore investigated hormone/redox signaling in the response of Arabidopsis thaliana to infestation by the aphid Myzus persicae, focusing on the interactions between ascorbic acid and ABA, together with the influence of altered a

Expression and RNAi characterization of neuropeptides underlying developmental regulation in the Colorado potato beetle.

Neuropeptides are evolutionarily ancient signaling molecules that act through diverse G protein-coupled receptor (GPCR) pathways to coordinate essential physiological and behavioral processes in insects. Despite their broad functional importance, the specific roles, receptor interactions, and regulatory mechanisms of individual neuropeptides remain underexplored, limiting our understanding of how these signaling systems operate across developmental and physiological contexts. In this study, we characterized transcript expression of five functionally diverse neuropeptides and one GPCR across Le

Evaluation of juvenile hormone analogues as rodent feed-through insecticides for control of immature phlebotomine sandflies.

The juvenile hormone analogues methoprene and pyriproxyfen were evaluated as rodent feed-through insecticides for control of immature stages of the sandfly Phlebotomus papatasi Scopoli (Diptera: Psychodidae). The development and survival of P. papatasi second-instar larvae fed faeces from Syrian hamsters, Mesocricetus auratus, that had been fed a diet containing methoprene (0, 9.788, 97.88 or 978.8 p.p.m.) or pyriproxyfen (0, 9.82, 98.2 or 982 p.p.m.) were evaluated. The faeces of methoprene-treated hamsters greatly reduced the percentage of larvae that pupated at all concentrations tested and

In vivo structure-activity studies on the dark-color-inducing neurohormone of locusts.

In the 11-residue long dark-color-inducing neurohormone (DCIN = [His7]-corazonin), of locusts, from residue 2 to residue 11, one amino acid at each time was substituted by D-phenylalanine (D-Phe). The dark-color-inducing effect of these peptides was investigated in comparison with unaltered DCIN by a bioassay based on nymphs of a DCIN-deficient albino mutant of the migratory locust, Locusta migratoria. Substitution of any single amino acid by D-Phe always reduced the activity, but did not abolish it completely. Maximum inactivation was obtained after substitution of Gln4, Ser6, or Trp9. The la

Drought, herbivory and combined stress caused treatment-specific changes in phytohormones, but species-specific changes in secondary metabolite profiles in thyme.

Thyme species are important medicinal herbs predominantly cultivated in arid regions. They increasingly experience the adverse effects of climate change, particularly drought. Although abiotic stress effects on thyme have been examined, limited information exists on insect herbivory or combined stress response. This study investigated the effects of drought stress, herbivory and their combination on secondary metabolism, phytohormone regulation and insect performance in three thyme species: Thymus serpyllum (drought-sensitive), T. kotschyanus (drought-tolerant), and T. vulgaris. Plants were ex

Low-lethal and intergenerational effects of broflanilide on the diamondback moth, Plutella xylostella (L.).

The diamondback moth (DBM), Plutella xylostella (L.) is an important agricultural pest worldwide that causes substantial damage to cruciferous crops. Broflanilide is a novel insecticide with high activity against DBM. This study demonstrated that exposure of DBM larvae to low concentrations (LC5, LC10, and LC30) of broflanilide significantly affected their development and fecundity. These treatments prolonged larval and pupal durations by 13.77% - 23.34% (14.95 h - 25.34 h) and 16.15% - 19.06% (13.46 h - 15.89 h), respectively, and reduced fecundity by 10.59% - 17.10%. Adipokinetic hormone signaling in the malaria vector Anopheles gambiae facilitates Plasmodium falciparum sporogony.

An obligatory step in the complex life cycle of the malaria parasite is sporogony, which occurs during the oocyst stage in adult female Anopheles mosquitoes. Sporogony is metabolically demanding, and successful oocyst maturation is dependent on host lipids. In insects, lipid energy reserves are mobilized by adipokinetic hormones (AKHs). We hypothesized that Plasmodium falciparum infection activates Anopheles gambiae AKH signaling and lipid mobilization. We profiled the expression patterns of AKH pathway genes and AgAkh1 peptide levels in An. gambiae during starvation, after blood feeding, and

Comparative Transcriptomics Reveals the Regulatory Mechanisms Underlying the Feeding Preference of Spodoptera frugiperda for Different Host Plants.

This study investigated the physiological responses and molecular mechanisms underlying the host preference of the polyphagous fall armyworm (FAW) for maize (preferred), rice (alternative), and cotton (nonpreferred). Life-table analysis confirmed that feeding on cotton plants significantly prolonged larval development and reduced survival rates compared with maize. Integrated midgut transcriptomics and physiological analyses revealed that a cotton diet severely impaired larval development by suppressing crucial detoxification pathways (cytochrome P450, glutathione metabolism), reducing juvenil

Juvenile hormone analog, fenoxycarb induced apoptotic effects in the developing ovary of Daphnia magna.

Juvenile hormone analogs (JHAs) are widely used insect growth regulators that disrupt endocrine signaling and impair reproduction in arthropods. Although their effects on non-target crustaceans such as Daphnia magna are well documented, the cellular mechanisms underlying JHA-induced reproductive toxicity remain unclear. In this study, we performed time-course exposure experiments to identify the sensitive window during which fenoxycarb reduces fecundity and investigated the associated ovarian changes. We generated a germline-specific VASA:H2B-GFP knock-in line to visualize ovarian nuclei in vi

Pigment-dispersing factor neuropeptides act as multifunctional hormones and modulators in tardigrades.

Pigment-dispersing factors (PDFs) are neuropeptides that play key roles in controlling the circadian rhythms in various insects, whereas their function remains elusive in other protostomes including tardigrades (water bears). Here we show that the three PDFs of the tardigrade Hypsibius exemplaris are co-localized in two pairs of inner lobe cells in the brain, whereas only one PDF occurs in four additional cerebral and two extracerebral cells. The axons of the inner lobe cells pass through the contralateral brain hemisphere, descend to the ventral nerve cord and terminate in two pairs of potent
