Mechanisms of host cell exit by the intracellular bacterium Chlamydia.

The mechanisms that mediate the release of intracellular bacteria from cells are poorly understood, particularly for those that live within a cellular vacuole. The release pathway of the obligate intracellular bacterium Chlamydia from cells is unknown. Using a GFP-based approach to visualize chlamydial inclusions within cells by live fluorescence videomicroscopy, we identified that Chlamydia release occurred by two mutually exclusive pathways. The first, lysis, consisted of an ordered sequence of membrane permeabilizations: inclusion, nucleus and plasma membrane rupture. Treatment with proteas

Cholesterol-binding toxins and anti-cholesterol antibodies as structural probes for cholesterol localization.

Cholesterol is one of the major constituents of mammalian cell membranes. It plays an indispensable role in regulating the structure and fun … esterol-enriched membranes, and have been used in many studies to detect microdomains in plasma and intracellular membranes. Anti-cholesterol antibodies that recognize cholesterol in domain structures have been developed in recent years. In this chapter, we describe the characteristics of these cholesterol-binding proteins and their applications to studies on membrane ch …

Relaxin and the human fetal membranes.

The human fetal membranes are complex tissues that perform many important functions during gestation. The extracellular matrix provides their strength to withstand the forces directed from the fetus and myometrium. Relaxin is a collagenolytic hormone that causes increased production of the matrix metalloproteinases. Its expression from the decidua is increased in patients with preterm premature rupture of the membranes, and its leucine-rich G receptor 7 is upregulated at preterm. The authors previously showed that relaxin is not involved in the infection-mediated cytokine response, but in the

Regulation of intracellular proliferation of Salmonella typhimurium by CD63.

BACKGROUND: Autophagy is an intracellular degradation process involving the lysosomal breakdown of unnecessary or abnormal cellular components. Targets of autophagy include pathogens, altered organelles, and protein aggregates, which are sequestered within double-membrane-limited vesicles known as autophagosomes. A variety of lysosomal membrane proteins play essential roles in numerous cellular processes, including lysosomal acidification, metabolite transport, and interaction with other membrane systems. Cluster of differentiation 63 (CD63), a member of the transmembrane 4 superfamily, is pre

Bimodal regulation of adenylate cyclase by prostaglandin E2 receptors in porcine ciliary epithelium.

Prostaglandin E2 (PGE2) binding revealed that porcine ciliary epithelial membranes possess two subclasses of binding sites or receptors. Ciliary nonpigmented epithelial (NPE) membranes have two binding sites (Kd,1 = 35 x 10(-9) M, Bmax,1 = 485 x 10(-12) mol/mg protein; Kd,2 = 0.723 x 10(-6) M, Bmax,2 = 1473 x 10(-12) mol/mg protein), while pigmented epithelial (PE) membranes have one binding site (Kd = 82 x 10(-9) M, Bmax = 377 x 10(-12) mol/mg protein). The three sites of NPE/PE membranes were found to show the highest affinity for PGE2 by competitive binding assay; affinity for PGF2 alpha an

Effect of rufloxacin upon non-specific immune defences: in-vitro, ex-vivo and in-vivo results.

This study investigated in-vitro, ex-vivo and in-vivo the immunomodulatory effects of rufloxacin. 0.5 MIC of rufloxacin significantly enhanced human macrophage phagocytosis and increased intracellular killing of Klebsiella pneumoniae in vitro. Pre-incubation of K. pneumoniae with rufloxacin made the bacteria more susceptible to both phagocytosis and intracellular killing by human macrophages than control organisms. Following pre-exposure of macrophages to 0.5 MIC of rufloxacin, there was a significant increase in the intracellular killing of K. pneumoniae compared with the controls, indicating

Cellular organization of the halophilic, phototrophic bacterium strain WS 68.

The obligatory halophilic, phototrophic bacterium, strain WS 68, grows under anaerobic conditions in the light in a mineral medium with acetate as carbon source and 6 to 20% sodium chloride, or in the same medium semiaerobically in the dark. The cells are vibrio- or spiral-shaped and show a bipolar polytrichous flagellation. The absorption spectrum of membranes, isolated from phototrophically grown cells, shows absorption maxima at 375, 490, 520, 555, 590, 800, 840-845 and 870-875 nm indicating the presence of bacteriochlorophyll a and carotenoids as pigments of the photosynthetic apparatus. Sphingolipid trafficking--sorted out?

Studies of intracellular membrane traffic have traditionally focused on the protein components of membranes, but what about lipids? Recent findings have drawn attention to the transport of one type of lipid, the sphingolipids. Their unique physical properties may allow them to aggregate into microdomains in membranes that concentrate sphingolipids into specific transport pathways. Gerrit van Meer and Koert Burger consider here the routes of sphingolipid biosynthesis and transport, and the role of proteins in their targeting. The following article by Deborah Brown turns the tables to review the

Intracellular "fibrous long spacing" collagen in morbus dupuytren (Dupuytren's contracture).

Fine structural analysis of 21 surgically removed specimens of Morbus Dupuytren (Dupuytren's contracture) in the cells of the proliferative phase revealed spindle-shaped or round, periodic banded structures of 1--1.5 micron size. These intracellular structures are surrounded by trilaminar membranes of 45 A continuous with membranes of the endoplasmic reticulum. The periodic banded structures display a light-dark pattern of 750--900 A and resemble with respect to periodicity and general morphology "fibrous long spacing" collagen. To date, "fibrous long spacing" collagen in human tissue has only

Effect of calcium antagonist compound nisoldipine on transepithelial electrical parameters in the isolated frog cornea.

The new calcium antagonist, nisoldipine, inhibited short circuit current and transcorneal potential difference in the isolated frog cornea (a chloride transporting epithelium). Transepithelial resistance increased. The effects of nisoldipine were dose-dependent. Nisoldipine did not change the stimulation of short circuit current induced by epinephrine. Intracellular membrane potential across both the apical and basolateral cell membrane were decreased by nisoldipine. The data indicate that intracellular calcium seemed to modulate chloride transport of the cornea by changing cell membrane perme

The β-cell assassin: IAPP cytotoxicity.

Islet amyloid polypeptide (IAPP) forms cytotoxic oligomers and amyloid fibrils in islets in type 2 diabetes (T2DM). The causal factors for amyloid formation are largely unknown. Mechanisms of molecular folding and assembly of human IAPP (hIAPP) into β-sheets, oligomers and fibrils have been assessed by detailed biophysical studies of hIAPP and non-fibrillogenic, rodent IAPP (rIAPP); cytotoxicity is associated with the early phases (oligomers/multimers) of fibrillogenesis. Interaction with synthetic membranes promotes β-sheet assembly possibly via a transient α-helical molecular conformation. C

Proliferative effect of ammodytin L from the venom of Vipera ammodytes on 208F rat fibroblasts in culture.

Ammodytin L, purified from the venom of Vipera ammodytes, triggers a rapid and dramatic lytic process in myotubes in vitro, as well as in differentiated muscle cells in vivo, through a mechanism that is not well understood. Despite its great sequence similarity to phospholipase A2, it is devoid of any enzyme activity. Data on artificial membranes demonstrating a direct interaction between this toxin and the hydrophobic core of the lipid bilayer suggest that the toxin also acts on the lipid microenvironment in cell membranes. Recent experiments on living cells do not confirm this hypothesis, an

[Intrauterine defensive mechanism of amniotic fluid and fetal membranes].

To determine the intrauterine defensive role of urinary trypsin inhibitor (UTI), we studied the effects of UTI in amniotic fluid, fetal membranes and myometrium. The level of UTI was 94 +/- 34 U/ml in neonatal urine (compared to adult urine 8.0 +/- 6.0 U/ml) and 88 +/- 37 U/ml in amniotic fluid. This may indicate that the main source of UTI in the amniotic fluid is the fetal urine. UTI was found to be concentrated in vernix, fetal intestine, amniotic membranes and uterine myometrium. Immunostaining of term amnion revealed a dark staining for UTI, whereas in premature deliveries UTI staining wa

Enhanced ferroptosis by light-triggered biomimetic nano-erythrocyte membranes for tumor synergistic therapy.

Breast cancer is the most prevalent fatal cancer among women worldwide and the leading cause of death for women. Ferroptosis is a form of programmed cell death that relies on iron and is non-apoptotic, triggered by the inhibition of the cellular antioxidant system. Photodynamic therapy (PDT) employs photosensitizers to produce reactive oxygen species (ROS), increasing oxidative stress in tumor cells. When combined with ferroptosis, PDT can work synergistically to regulate intracellular redox balance. In this study, we designed engineered nano-erythrocyte membranes for targeted delivery of Chlo

Regulation of hepatic Na(+)-HCO3- cotransport and pH by membrane potential difference.

Hepatocytes possess several mechanisms for membrane acid-base transport, which work in concert to maintain intracellular pH (pHi) in a narrow physiological range, despite metabolic processes that produce and consume substantial quantities of H+ and HCO3-.Na(+)-H+ and Cl(-)-HCO3- exchangers contribute to recovery from intracellular acidosis and alkalosis, respectively, but are largely inoperative at physiological values of pHi. Recent studies indicate that hepatocytes also possess a mechanism for coupled transport of Na+ and HCO3- across the basolateral membrane. This appears to be the dominant

Electrical membrane properties of a cell clone derived from human nonpigmented ciliary epithelium.

Intracellular potentials were measured in a SV-40 virus-transformed cell clone derived from human nonpigmented ciliary epithelium using the microelectrode technique. (1) Membrane potential averaged -50.2 mV (+/- 0.6, n = 207). (2) Increasing the extracellular K+ concentration depolarized the membrane voltage. The amplitude of this potential response was reduced in the presence of 1 mM Ba2+. (3) Superfusing the cells with a Ca2+-free solution containing 1 mM EGTA depolarized the intracellular potential and diminished the voltage response upon increasing extracellular K+. (4) Extracellular alkal

New and notable ion-channels in the sarcoplasmic/endoplasmic reticulum: do they support the process of intracellular Ca²⁺ release?

Intracellular Ca(2+) release through ryanodine receptor (RyR) and inositol trisphosphate receptor (IP3 R) channels is supported by a complex network of additional proteins that are located in or near the Ca(2+) release sites. In this review, we focus, not on RyR/IP3 R, but on other ion-channels that are known to be present in the sarcoplasmic/endoplasmic reticulum (ER/SR) membranes. We review their putative physiological roles and the evidence suggesting that they may support the process of intracellular Ca(2+) release, either indirectly by manipulating ionic fluxes across the ER/SR membrane o

Antimicrobial Activity of Ferulic Acid Against Cronobacter sakazakii and Possible Mechanism of Action.

Cronobacter sakazakii is an opportunistic pathogen transmitted by food that affects mainly newborns, infants, and immune-compromised adults. In this study, the antibacterial activity of ferulic acid was tested against C. sakazakii strains. Minimum inhibitory concentration of ferulic acid against C. sakazakii strains was determined using the agar dilution method. Changes in intracellular pH, membrane potential and intracellular ATP concentration were measured to elucidate the possible antibacterial mechanism. Moreover, SYTO 9 nucleic acid staining was used to assess the effect of ferulic acid o

Membrane-Targeted Consequences of Acetaminophen Toxicity and Off-Target Effects of Antimicrobial Peptides on Host Cell Membranes.

Acetaminophen (paracetamol, APAP) is a widely used analgesic and antipyretic drug. Under normal physiological conditions, it does not directly interact with or disrupt cellular membranes. However, in cases of acetaminophen overdose or toxicity, severe cellular damage has been described, involving a broad spectrum of effects at different cellular levels. These toxic effects may involve membrane structures, including mitochondrial, plasma, and other intracellular membranes. Antimicrobial peptides (AMPs) are short, usually cationic and amphipathic peptides produced by both microorganisms and mult…
