Nutritional management of chronic renal failure in childhood.

Protein malnutrition is common in children with chronic renal failure (CRF) and is especially severe in young infants. Growth is usually rapid in infants and CRF causes a reduction in cell mass associated with extreme growth retardation in this age group. Poor energy intake and altered energy metabolism are common in CRF and are associated with protein malnutrition and poor growth. Uraemic toxicity may reduce protein synthesis by altering energy metabolism or by a direct effect, low protein high energy diets improve growth and extremely low nitrogen intakes supplemented with essential amino ac

Determination of R- and S-3-methyl-2-oxopentanoate enantiomers in human plasma: suitable method for label enrichment analysis.

A sensitive method for the determination of S- and R-3-methyl-2-oxopentanoate enantiomers (KMV, alpha-keto-beta-methylvalerate) in physiological fluids suitable for isotope enrichment analysis is described: after extraction with acid, 2-oxo acids are separated from interfering amino acids by cation-exchange chromatography. Reductive amination of the branched-chain 2-oxo acids by use of L-leucine dehydrogenase yields the corresponding L-amino acids. L-Isoleucine and L-alloisoleucine which are formed from S- and R-3-methyl-2-oxopentanoate, respectively, are then quantified by amino acid analysis

Ketone body oxidation and susceptibility to ethyl acetoacetate in a novel hemolytic multidrug-resistant strain Leptospira interrogans KeTo originated from sewage water.

Terrestrial and aquatic environments contaminated with animal urine may contribute to the transmission of Leptospira, a causative agent of leptospirosis in humans and wild/domesticated animals. Although enormous amounts of work have been done decoding the ecophysiology, the factors governing the cell growth and virulence in Leptospires derived from environmental samples still remain elusive. Here, we show oxidation of a wide array of organic acids including acetoacetate by a new strain of Leptospira interrogans designated as KeTo, isolated from a sewage sample originating from a wildlife enclo

DNA damage induced by alloisoleucine and other metabolites in maple syrup urine disease and protective effect of l-carnitine.

Maple syrup urine disease (MSUD) is an inherited deficiency of the branched-chain α-keto dehydrogenase complex, characterized by accumulation of the branched-chain amino acids (BCAAs) and their respective branched chain α-keto-acids (BCKAs), as well as by the presence of alloisoleucine (Allo). Studies have shown that oxidative stress is involved in the pathophysiology of MSUD. In this work, we investigated using the comet assay whether Allo, BCAAs and BCKAs could induce in vitro DNA damage, as well as the influence of l-Carnitine (L-Car) upon DNA damage. We also evaluated urinary 8-hydroxydeog

Bile acids and conjugates identified in metabolic disorders by fast atom bombardment and tandem mass spectrometry.

From a study of the collision-activated fragmentation of bile acids, a qualitative analytical method based on negative ion fast atom bombardment tandem mass spectrometry has been developed. The times for sample preparation and analyses are short. Both free and conjugated bile acids are detected as they occur in biological fluids, without derivatization. For identifying bile acids and conjugates, the method offers better specificity and sensitivity than does the fast atom bombardment mass spectrometric technique alone. Specific scan modes have been developed for the selective detection of tauri

Visible light photoredox-catalyzed difunctionalization of electron-deficient alkenes with tert-butyl nitrite: oxocarboxylic acid as an acyl source.

We report a visible-light photoredox-catalyzed acylative oximation of electron-deficient alkenes using tert-butyl nitrite and α-oxocarboxylic acids, providing direct access to β-keto oxime derivatives under mild conditions. The protocol combines tert-butyl nitrite as an NO donorwith acyl radicals generated through photoredox-mediated decarboxylation of α-oxocarboxylic acids. Vinyl sulfones and several other activated alkenes undergo the transforamtion to afford the corresponding products in moderate yields. Preliminary mechanistic studies support the involvement of acyl radical intermediates.

Adding free leucine to a balanced essential amino acid mixture suppresses intracellular transamination of isoleucine and valine in healthy older adults.

BACKGROUND: Adding leucine (LEU) to protein meals enhances protein anabolism, but excess LEU reduces valine (VAL) and isoleucine (ILE) plasma concentrations. We examined in older adults whether additional LEU added to an essential amino acid (EAA) mixture reduces the intracellular catabolism of VAL and ILE, and their keto acids ɑ-ketoisovaleric acid (KIV)and 2-keto-3-methyl-pentanoate (KMV). METHODS: Using a randomized placebo-controlled crossover design, 11 older adults (60-80 years) consumed 20g EAA, 20g EAA+3g LEU (EAA + LEU), and water (baseline) on separate study days. A pulse of stable

Genetic and metabolic drivers of membrane remodeling in Clostridium thermocellum under alcohol stress.

UNLABELLED: Clostridium thermocellum is a leading candidate for consolidated bioprocessing of lignocellulosic biomass into biofuels due to its native cellulolytic capabilities. Beyond ethanol, C. thermocellum is being developed as a platform for producing higher-chain alcohols such as isobutanol and n-butanol. However, its physiological adaptations to alcohol stress remain poorly understood. Here, we investigate how C. thermocellum remodels its membrane lipid composition in response to exogenous ethanol, n-butanol, isobutanol, and butyrate. Exposure to linear alcohols such as n-butanol or to o

Delta13-reductase dependent metabolism of prostaglandins in the mammalian brain and eye.

This paper describes the distribution of NAD+-dependent 15-hydroxy prostaglandin dehydrogenase (delta13-reductase) in the mammalian brain and eye tissues. In addition, an NADH-dependent 15-ketoprostaglandin delta13-reductase (15-PGDH) activity was determined in the brain and eye tissue of some species. The rabbit brain and eye tissues were obtained after arterial perfusion with PBS buffer while the monkey, bovine and porcine tissues were obtained without any treatment. [3H]-PGF2alpha or [3H]-15-keto-17-phenyl-18,19,20-trinor-PGF2alpha1-1-isopropy l ester was incubated with different eye tissue

Water-Derived Radical Species Initiate Six-Electron Oxidation of Cyclic Ketones in Microdroplets.

Direct six-electron oxidation of cyclohexanone and other cyclic ketones and associated C-C bond cleavage can be achieved in microdroplets without the need for external oxidants, metal catalysts, or thermal activation. Mechanistic studies indicate that presence of trace amounts of water initiate the oxidative transformation by generating reactive radical species. Isotopic labeling experiments employing D2O and H2 18O provide evidence for involvement of water radical cation (H2O•+) as the active oxidant. Experiments using different nebulizing gases suggest that both water radical cation and mole

Porcine aldo-keto reductase 1C subfamily members AKR1C1 and AKR1C4: Substrate specificity, inhibitor sensitivity and activators.

Most members of the aldo-keto reductase (AKR) 1 C subfamily are hydroxysteroid dehydrogenases (HSDs). Similarly to humans, four genes for AKR1C proteins (AKR1C1-AKR1C4) have been identified in the pig, which is a suitable species for biomedical research model of human diseases and optimal organ donor for xenotransplantation. Previous study suggested that, among the porcine AKR1Cs, AKR1C1 and AKR1C4 play important roles in steroid hormone metabolism in the reproductive tissues; however, their biological functions are still unknown. Herein, we report the biochemical properties of the two recombi

Anodic oxidation triggered divergent 1,2- and 1,4-group transfer reactions of β-hydroxycarboxylic acids enabled by electrochemical regulation.

We report a set of electrochemically regulated protocols for the divergent synthesis of ketones and β-keto esters from the same β-hydroxycarboxylic acid starting materials. Enabled by electrochemical control, the anodic oxidation of carboxylic acids proceeded in either a one-electron or a two-electron pathway, leading to a 1,4-aryl transfer or a semipinacol-type 1,2-group transfer product with excellent chemoselectivity. The 1,4-aryl transfer represents an unprecedented example of carbon-to-oxygen group transfer proceeding via a radical mechanism. In contrast to previously reported radical gro

Enhancement of hydroxyl radical generation in the Fenton reaction by alpha-hydroxy acid.

The effect of various organic acids on hydroxyl radical (.OH) generation in the Fenton reaction were examined by the ESR spin trapping technique, where 5,5-dimethyl-1-pyroline-N-nitroxide (DMPO) and alpha-phenyl-tert-butyl nitrone (PBN) were used as the spin trapping reagents. alpha-Hydroxy acids such as lactic acid, glycolic acid and 2-hydroxy isobutyric acid were found to markedly enhance .OH generation in the reaction. In contrast, beta-hydroxy acid, alpha-keto acid, esters of alpha-hydroxy acids, aldehydes and other straight chain organic acids had no such enhancing activity. alpha-Amino a

Beyond the solvent role: discovery of DMSO as a versatile radical reagent for organic transformations.

DMSO is well-known as a solvent but here we have explored DMSO as a sole reagent for dehydrogenation of tetrahydrocarbazoles, oxidation of alcohols to carbonyl compounds, decarboxylation of keto-acids to aldehydes, and C-C/C-N bond formations via cyclization, self-coupling, the Povarov reaction and Friedlander synthesis via a radical pathway. This methodology involves simple, metal-free and catalyst-free conditions.

Cytoskeleton as a potential target in the neuropathology of maple syrup urine disease: insight from animal studies.

In this short review we provide evidence that the branched-chain keto acids accumulating in the neurometabolic disorder maple syrup urine disease disturb rat cerebral cytoskeleton in a developmentally regulated manner. Alterations of protein phosphorylation leading to brain cytoskeletal misregulation and neural cell death caused by these metabolites are associated with energy deprivation, oxidative stress and excitotoxicity that may ultimately disrupt normal cell function and viability.

Control of Selectivity in FeCl3 -Catalyzed Aerobic Oxidation of Cycloketones.

The FeCl3 -catalyzed aerobic oxidation of ketones always gives rise to the α-C-C cleavage product, having challenges to afford hydroxyl keto compounds. Here we report an effective control of the main product from keto acid to α-hydroxyl ketone, by reducing the concentration of FeCl3 catalyst, together with the use of DMSO as the solvent. In addition, mechanistic investigations suggested the same FeCl3 -coordinated peroxide intermediate for both hydroxylation and C-C cleavage routes, and emphasize the role of DMSO as both ligand and reductant. This work provides a new approach for selective aer

[Maple Syrup Urine Disease in a newborn infant].

Maple Syrup Urine Disease (MSUD) is a severe metabolic disorder secondary to an enzyme defect in the catabolic pathway of the branched chain amino acids: leucine, isoleucine and valine. Accumulation of these amino acids and derived from them alpha-keto-acids leads to encephalopathy and progressive degeneration of the nervous system in undiagnosed and hence untreated patients. Early diagnosis and elimination diet prevent complications and therefore create a possibility of both normal intellectual and physical progress. In consequence, in a few countries MSUD has been added to newborn screening

Acute Administration of Branched-Chain Amino Acids Increases the Pro-BDNF/Total-BDNF Ratio in the Rat Brain.

Maple syrup urine disease (MSUD) is caused by an inborn error in metabolism resulting from a deficiency in the branched-chain α-keto acid dehydrogenase complex activity. This blockage leads to accumulation of the branched-chain amino acids (BCAA) leucine, isoleucine and valine, as well as their corresponding α-keto acids and α-hydroxy acids. High levels of BCAAs are associated with neurological dysfunction and the role of pro- and mature brain-derived neurotrophic factor (BDNF) in the neurological dysfunction of MSUD is still unclear. Thus, in the present study we investigated the effect of an

Neurological damage in MSUD: the role of oxidative stress.

Maple syrup urine disease (MSUD) is a metabolic disease caused by a deficiency in the branched-chain α-keto acid dehydrogenase complex, leading to the accumulation of branched-chain keto acids and their corresponding branched-chain amino acids (BCAA) in patients. Treatment involves protein-restricted diet and the supplementation with a specific formula containing essential amino acids (except BCAA) and micronutrients, in order to avoid the appearance of neurological symptoms. Although the accumulation of toxic metabolites is associated to appearance of symptoms, the mechanisms underlying the b
