Development and validation of a stability-indicating method for determination of free sterols in the Asian medicinal leech Hirudo manillensis.

A rapid, simple, sensitive, selective, precise and robust thin-layer chromatography densitometric method for the determination of free sterols in leech was developed and validated on silica gel layer using carbon tetrachloride-methanol-formic acid (9.5:1.5:0.55, v/v/v). Spectrodensitometric scanning was carried using a Camag TLC scanner III at 366 nm after spraying 2% methanolic sulphuric acid, which gave compact spots for cholesterol (R(F) = 0.35 ± 0.02). The regression analysis data for calibration plot implied a good linear relationship (r(2) = 0.99958) between response and concentration ov

The place of chemical leeching with heparin in digital replantation: subcutaneous calcium heparin for patients not treatable with systemic heparin.

BACKGROUND: In patients with replanted digits, systemic heparinization or other techniques may be used for prevention of thrombosis of the vascular anastomoses or to prevent coagulation at the drainage site (e.g., a fish-mouth incision or partially removed nail). However, systemic heparin may be contraindicated or require cautious administration in certain patients. The authors injected calcium heparin subcutaneously into the congested replanted digits of seven patients with a history of or undergoing treatment for hemorrhoids or gastric ulcer and drained the congestion by means of a fish-mout

Evaluating 3D-Patch Efficacy in Wound Healing Using the Medicinal Leech Hirudo verbana as an In Vivo Model.

Skin injuries are common and can result from surgeries, burns, pressure sores, cuts, and diseases. Proper wound healing is crucial for maintaining homeostasis; wounds can be classified as acute or chronic. Acute wounds heal in four sequential phases: hemostasis, inflammation, proliferation, and remodeling. Chronic wounds arise when this process fails, often due to prolonged inflammation. Existing treatments for chronic wounds are limited, and antibiotic resistance complicates infection control, highlighting the urgent need for new therapies. Biomaterials, particularly gelatin, have gained atte

Coincidental occurrence of endometrial hyperplasia with atypia and vaginal leech infestation in a postmenopausal woman: a rare case report.

BACKGROUND: Postmenopausal bleeding is a common gynecological complaint with multiple etiologies, ranging from benign to malignant conditions. Leech infestation of the female genital tract is a rare cause, particularly in rural regions with exposure to natural pooled water sources. CLINICAL PRESENTATION: We report a 50-year-old postmenopausal woman who presented with a 1-week history of vaginal bleeding. Ultrasound evaluation revealed a thickened endometrium (20 mm), and histopathology confirmed endometrial hyperplasia with atypia. She underwent total hysterectomy with bilateral salpingo-ooph

Total lower lip and chin replantation following a trampoline accident: Rescue surgery with intensive leech therapy.

Lip amputations in children may require urgent microsurgical management. Here we report a salvage procedure with partial survival following lower lip and chin replantation after a trampoline accident. Total amputation was managed with inferior labial artery anastomosis; however, venous drainage was not reestablished, necessitating leech therapy. Despite arterial revisions, 75 % of the lip and 25 % of the chin were lost by post-operative day 14. Reconstruction with a combined right Karapandzic flap and left Camille Bernard flap was performed one month later, supplemented by mesotherapy and nano

A Novel Integrated Strategy for Discovering Absorbable Anticoagulant Bioactive Peptides: A Case Study on Leech Protein Hydrolysates.

Medicinal plants and animal-derived proteins represent valuable natural sources of bioactive components with pharmaceutical potential. Whilst some medicinal plants and animal-derived proteins also offer rich sources of anticoagulant bioactive peptides, their development faces multiple challenges: anticoagulant evaluation relies on single-parameter assays with limited reliability, native proteins demonstrate suboptimal activity without enzymatic treatment, and few researchers investigate bioavailable peptides. Our study establishes an innovative framework using the leech as a case study to over

Oropharyngeal leech infestation as a cause of severe anemia in a 65-year-old male patient: a case report.

BACKGROUND: Leech infestations are rare but potentially serious medical conditions, often leading to hemorrhage and anemia. Leeches, which belong to the phylum Annelida and class Hirudinea, typically infest humans through contact with contaminated water or ingestion of unfiltered water. They attach to the mucosa of the aerodigestive tract, causing bleeding, and in severe cases, complications such as airway obstruction or severe anemia. Leech infestations are more prevalent in rural areas with limited access to safe water, where environmental conditions facilitate exposure. CASE PRESENTATION:

Leech therapy (Hirudo medicinalis) attenuates testicular damages induced by testicular ischemia/reperfusion in an animal model.

BACKGROUND: Testicular torsion/detorsion triggers tissue ischemia/reperfusion, leading to reactive oxygen species overgeneration and apoptosis. The saliva of leeches is full of anti-inflammatory, anticoagulants, antioxidants, and antimicrobial agents. Therefore, this study aimed to assess the protective mechanism of leech therapy on testicular ischemia/reperfusion damage. METHODS: 18 adult male rats were randomly divided into three groups: 1-Sham-operated group (SO). 2-Torsion/detorsion (T.D) group: two hours of testicular torsion with two hours of testicular detorsion was performed. 3-Torsio

Complete penile amputation during ritual neonatal circumcision and successful replantation using postoperative leech therapy.

Circumcision is the most common surgical procedure in males in the United States, and minor complications are not uncommon. Major complications like partial penile amputations have been reported with successful replantation. Complete penile amputations in adult males have been described, and successful replantation has been reported with increasing success. We report a case of complete penile amputation at the penopubic junction using a Mogen clamp in a 7-day-old neonate with replantation using postoperative leech therapy. To our knowledge this is the first time leech therapy has been used pos

Novel genotypes of Trypanosoma binneyi from wild platypuses (Ornithorhynchus anatinus) and identification of a leech as a potential vector.

Little is known about the prevalence and pathogenesis of trypanosomes in Australian monotremes, and few genetic characterisation studies have been conducted with these haemoparasites. During the present investigation, molecular and microscopic methods were used to screen peripheral blood (n=28) and ectoparasites (n=10 adult ticks; n=5 tick nymphs; n=1 leech; and n>500 tick eggs) collected from wild Tasmanian platypuses (Ornithorhynchus anatinus), for the presence of trypanosomatid-specific DNA and/or trypomastigotes. The genes for the small ribosomal subunit RNA (18S rDNA) and glycosomal glyce

Mg2+-malate co-transport, a mechanism for Na+-independent Mg2+ transport in neurons of the leech Hirudo medicinalis.

Mg2+-extrusion from Mg2+-loaded neurons of the leech, Hirudo medicinalis, is mediated mainly by Na+/Mg2+ antiport. However, in a number of leech neurons, Mg2+ is extruded in the nominal absence of extracellular Na+, indicating the existence of an additional, Na+-independent Mg2+ transport mechanism. This mechanism was investigated using electrophysiological and microfluorimetrical techniques. The rate of Na+-independent Mg2+ extrusion from Mg2+-loaded leech neurons was found to be independent of extracellular Ca2+, K+, NO3-, HCO3-, SO4(2-), HPO4(2-), and of intra- and extracellular pH. Decorsin. A potent glycoprotein IIb-IIIa antagonist and platelet aggregation inhibitor from the leech Macrobdella decora.

The discovery, purification, and characterization of decorsin, a protein isolated from the North American leech Macrobdella decora, are described. Decorsin acts as an antagonist of platelet glycoprotein IIb-IIIa (GPIIb-IIIa), and is a potent inhibitor of platelet aggregation. The protein was purified to apparent homogeneity from crude whole leech extracts by treatment with trifluoroacetic acid followed by GPIIb-IIIa affinity chromatography and C18 reverse-phase high performance liquid chromatography. Decorsin was also isolated from a solution of leech ingestate by treatment with trifluoroaceti

Leech Diversity in the Maghreb (North Africa): A Checklist and a Case Report of Parasitism on a Berber Toad (Sclerophys mauritanica) in Algeria.

Leeches (Hirudinea) are ecologically important annelids that interact with a wide range of aquatic vertebrates, yet their diversity, distribution, and epidemiological relevance remain poorly documented in North Africa. Here, we provide a comprehensive synthesis of freshwater and marine leech species reported from the Maghreb (Algeria, Tunisia, and Morocco), based on an extensive review of the available literature. In total, 21 species belonging to 13 genera and four families (Glossiphoniidae, Erpobdellidae, Hirudinidae, and Piscicolidae) are documented, with updated information on their ecolog

Leech Extract Enhances the Pro-Angiogenic Effects of Endothelial Cell-Derived Exosomes in a Mouse Model of Ischemic Stroke.

BACKGROUND: Intercellular communication, facilitated by exosomes (Exos) derived from endothelial cells (ECs), significantly influences the regulation of angiogenesis. Leech extract significantly reduces ischemia-reperfusion injury, promotes angiogenesis, and improves neurological function in mice with stroke. However, further investigation is required to determine whether leech promotes angiogenesis through EC-Exo. OBJECTIVE: This study aims to further explore whether leech regulates Exos to promote the establishment of collateral circulation in mice with ischemic stroke (IS) and the specific

Optimized deep belief network based on an improved Blood-sucking Leech Optimizer algorithm for wastewater quality forecasting.

To address the issues of limited exploration capability and premature convergence in the optimization process of the Blood-Sucking Leech Optimizer (BSLO) algorithm, we propose an Improved BSLO (IBSLO) algorithm. Initially, a directional leeches switching mechanism based on an inverted S-shaped nonlinear perceived distance to strike a balance between exploitative and exploratory capabilities of the algorithm. Subsequently, a dynamic perception signal was designed to simulate dynamic stimulus signals, guiding leeches to search and optimize more accurately. Finally, the memory sharing mechanism i

Acute cadmium toxicity in the medicinal leech Hirudo verbana Carena, 1820 with a novel biomarker for aquatic risk assessment.

Cadmium (Cd) is a common aquatic contaminant that poses significant ecological risks by disrupting organismal functions and ecosystem balance. This study presents the first detailed evaluation of acute Cd toxicity in the medicinal leech Hirudo verbana Carena, 1820, a species of ecological, biomedical, and conservation importance listed under CITES. In a 96-hour semi-static bioassay with graded Cd exposures (0–32 mg/L), we determined an LC50 of 4.02 mg/L and observed a range of concentration-dependent behavioral and pathological responses, including erratic locomotion, excessive mucus secretion

Manifestation of leech infestation as severe gastrointestinal bleeding in a 3-year-old boy: a case report and review of the literature.

BACKGROUND: Leeches are a class of hermaphroditic parasites that can attach to various body parts and start sucking blood. Gastrointestinal (GI) bleeding due to leeches is a rare phenomenon that is more common in less developed countries. Common symptoms include melena, hematemesis, pallor, weakness, and fatigue. Due to the similar symptoms of this issue to the main differential diagnoses of GI bleeding in pediatrics, such as diarrhea, constipation, diverticulitis, esophagitis, and anal fissures, it is challenging to differentiate it from the rest. CASE PRESENTATION: We present a three-year-o
