Treatment of Nocardia infections continues to be difficult, especially for central nervous system infections or disseminated diseases and when sustained by highly drug-resistant species such as Nocardia farcinica. Linezolid, the first oxazolidinone approved for clinical use, shows an excellent in vitro activity against all species of Nocardia, and achieves good central nervous system and lung concentrations. We describe the case of a patient with pulmonary nocardiosis and intolerance to trimethoprim/sulfamethoxazole successfully treated with linezolid, and review all the published cases of sys…

Linezolid is an oxazolidinone derivative which is active mostly against Gram-positive bacteria. In this work its activity against the facultatively intracellular bacterium Listeria monocytogenes was examined in vitro, in tissue culture and in animal models of systemic and intracerebral infection and compared with ampicillin which is the antibiotic of choice for treatment of listeriosis. All strains of L. monocytogenes were susceptible to the substance, with minimal inhibitory concentrations (MICs) determined by E-test ranging from 0.38 to 1.5 mg/l which is below the preliminary breakpoint of t…

Exposure to D-galactose (D-gal) induces psychiatric problems and cognitive decline characterized by oxidative stress and neuronal dysfunction. In this study, linezolid (Lnz) was used to evaluate its mitigating effects on D-gal-induced neurological deficits. Lnz has been reported as an antidepressant and anxiolytic agent that can alleviate neurological and behavioral alterations1. Thirty-six animals were separated into 6 sets (n = 6): [Veh + Veh], [Veh + Lnz] (10 mg/kg), [Veh + Lnz] (20 mg/kg), [Veh + D-gal] (100 mg/kg), [D-gal + Lnz] (10 mg/kg), and [D-gal + Lnz] (20 mg/kg). Both D-gal and Lnz…

INTRODUCTION: Post influenza pneumonia is a leading cause of mortality and morbidity, with mortality rates approaching 60% when bacterial infections are secondary to multi-drug resistant (MDR) pathogens. Staphylococcus aureus, in particular community acquired MRSA (cMRSA), has emerged as a leading cause of post influenza pneumonia.
HYPOTHESIS: Linezolid (LZD) prevents acute lung injury in murine model of post influenza bacterial pneumonia.
METHODS: Mice were infected with HINI strain of influenza and then challenged with cMRSA at day 7, treated with antibiotics (LZD or Vanco) or vehicle 6 ho…

OBJECTIVE: Beyond its role as a ribosome-targeting antibiotic, linezolid was recently shown to modulate immune responses by inhibiting mitochondrial translation. Because mitochondrial dysfunction is implicated in various fibrotic diseases, including systemic sclerosis (SSc), this study aimed to evaluate the antifibrotic potential of linezolid and delineate its underlying mechanisms in SSc.
METHODS: The effects of linezolid on fibrotic tissue remodeling were assessed using multiple experimental systems: human dermal fibroblasts, human macrophages, three-dimensional SSc skin equivalents (SScSE)…

A woman in her 30s was administered linezolid treatment for pneumonia. Without continuous medical prescriptions, the patient self-administered linezolid for a total of 9 months, which subsequently led to symptoms of bilateral visual impairment and central scotomas. Fundus examination revealed blurred disc margins, accompanied with mild edema, flame-shaped hemorrhages around the optic disc, and mild tortuosity and dilation of retinal veins in both eyes. Visual field testing indicated the presence of central scotomas in both eyes. Visual evoked potentials testing showed reduced amplitudes in bot…

BACKGROUND AND OBJECTIVES: Elderly patients are at an increased risk of supratherapeutic linezolid exposure, and elevated trough concentrations of linezolid and its metabolite (PNU-142300) are associated with the development of linezolid-induced thrombocytopenia. Clarifying the population pharmacokinetic (PPK) characteristics of linezolid and PNU-142300 in this population is critical for optimizing therapeutic strategies. This study aimed to develop a PPK model for linezolid and PNU-142300 in elderly patients to guide dose adjustments and mitigate thrombocytopenia risk.
METHODS: Patients aged…

Multidrug-resistant tuberculosis (MDR-TB) is characterized by resistance to at least isoniazid and rifampicin. Linezolid is an antibiotic used for drug-resistant Gram-positive bacteria and is a treatment option for MDR-TB. However, its use is associated with optic neuropathy, presenting as acute worsening and bilateral vision loss, typically within 4 months of therapy. A 47-year-old male with MDR-TB relapsed during the sixth month of an individualized treatment regimen at Dr. Soetomo General Academic Hospital, Surabaya. The patient presented with weakness and anemia, receiving a regimen includ…
