Research progress of pancreatic cancer based on ferroptosis-cuproptosis interaction (Review).

Pancreatic cancer is a highly aggressive malignancy with a poor prognosis and pancreatic ductal adenocarcinoma (PDAC) constitutes the overwhelming majority of histological subtypes. KRAS mutation-driven metabolic reprogramming renders PDAC highly dependent on metal ion homoeostasis and antioxidant systems, thereby providing a novel rationale for therapeutic strategies targeting ferroptosis and cuproptosis. Ferroptosis is a form of regulated cell death caused by the excessive accumulation of iron-dependent lipid peroxidation, whereas cuproptosis is a newly characterised mode of cell death trigg

Cuproptosis and the bioinorganic basis of copper-induced cell death.

Copper is an essential yet potentially toxic biological trace element, whose functions in cells arise from its unique redox and coordination chemistry. The same chemical properties that make copper indispensable for a variety of fundamental biological processes, including respiration and antioxidant defense, also render it highly toxic when intracellular concentrations exceed homeostatic levels. Recently, a copper-dependent form of regulated cell death (RCD) termed cuproptosis has been identified, in which excess copper triggers cytotoxicity by directly binding the metabolic machinery of mitoc

FASN targeting by G28UCM impairs mitochondrial fatty acid synthesis and reveals a FASN-SDHB synthetic interaction.

Metabolic reprogramming in cancer relies on lipid synthesis and mitochondrial function, yet how these processes, other than citrate flux and β-oxidation, intersect remains unclear. While inhibitors of lipogenic pathways have been developed as potential therapeutic agents in cancer therapy, their impact on oxidative metabolism is underexplored. Here, we identify the fatty acid synthase (FASN) inhibitor G28UCM as a compound that additionally destabilizes mitochondrial fatty acid synthase (mtFAS) and succinate dehydrogenase subunit B (SDHB), thereby targeting cytosolic and mitochondrial metabolis

Synergistic Heterojunction/Cu─N Coordination Engineering Drives Cuproptosis and PANoptosis for Sonodynamic Glioblastoma Therapy.

Glioblastoma (GBM) remains one of the most lethal intracranial malignancies due to its diffuse invasiveness and inevitable recurrence. Sonodynamic therapy (SDT) offers a noninvasive strategy for deep-tissue tumor ablation; however, its efficacy is often limited by rapid electron-hole recombination in conventional sonosensitizers, resulting in insufficient reactive oxygen species (ROS). In this study, a biomimetic nanosonosensitizer is rationally engineered via synergistic type II heterojunction interface construction and Cu─N coordination electronic activation. By anchoring Cu─N coordinated ca

Copper Homeostasis and Cuproptosis in Neurological Disorders.

Neurological disorders such as Alzheimer's disease (AD) and Parkinson's disease (PD) pose a serious global public health threat, with complex etiologies involving genetic, environmental, and metabolic factors. Current data indicate that the prevalence of these disorders is rapidly increasing with the aging population, resulting in a growing economic and healthcare burden worldwide. In recent years, the imbalance of copper homeostasis has been increasingly implicated in the pathogenesis of neurological diseases. Copper overload can aggravate neuronal injury by inducing oxidative stress (OS), mi

Combined use of vitamin C and antifolates induces cuproptosis-like bacterial death.

Combined use of antimicrobial agents is a key strategy to combat antimicrobial resistance. After several decades of approval, clinical application of the classical combination co-trimoxazole has gradually become limited, mainly due to the prevalence of resistance. Vitamin C is a natural antimicrobial and redox agent. While sporadic studies suggest vitamin C might synergize with antimicrobial drugs, results have been inconsistent and the underlying mechanisms are unclear. Here we show that vitamin C increased bacterial sensitivity to antifolates and protein synthesis inhibitors through differen

Induction of primary biliary cirrhosis in guinea pigs following chemical xenobiotic immunization.

Although significant advances have been made in dissecting the effector mechanisms in autoimmunity, the major stumbling block remains defining the etiological events that precede disease. Primary biliary cirrhosis (PBC) illustrates this paradigm because of its high degree of heritability, its female predominance, and its extraordinarily specific and defined immune response and target destruction. In PBC, the major autoantigens belong to E2 components of the 2-oxo-acid dehydrogenase family of mitochondrially located enzymes that share a lipoylated peptide sequence that is the immunodominant tar

Immunogenetic analysis reveals that epitope shifting occurs during B-cell affinity maturation in primary biliary cirrhosis.

Primary biliary cirrhosis (PBC) is a liver disease characterized by serum autoantibodies against the pyruvate dehydrogenase complex (PDC) located in the inner mitochondrial membrane. The predominant target in PDC has previously been localized to the inner lipoyl domain (ILD) of the E2 subunit. The etiology of PBC is unknown, although molecular mimicry with bacterial PDC has been proposed. Here, we have investigated the etiology of PBC and nature of the autoimmune response by analyzing the structure of a human monoclonal antibody with ILD specificity. Mutants of the monoclonal antibody, which w

The peculiar autoimmunity of primary biliary cirrhosis.

Autoantibodies to mitochondria (AMA, anti-M2) are a serologic hallmark of primary biliary cirrhosis (PBC). These react with three structurally and functionally related multienzymic complexes, the 2-oxoacid dehydrogenase complexes, but chiefly with the E2 subunit of pyruvate dehydrogenase complex (PDC-E2). Their very dose (95%) and specific association with PBC underpins the autoimmune concept of pathogenesis of that disease, notwithstanding several non-congruent features. Detailed studies, including structural analysis of epitopes, do not disclose how these autoantibodies originate. Their ubiq

Palmitoylation-Mediated Ubiquitination of SRPK1 Regulates Ferroptosis in High-Fat-Induced Erectile Dysfunction.

Serine-arginine protein kinase 1 (SRPK1) is a major protein kinase involved in mRNA splicing, cell cycle, and endothelial function. Recent studies have highlighted a close relationship between palmitic acid (PA) and endothelial cell ferroptosis. Here, we demonstrate that PA promotes the ubiquitination-dependent degradation of SRPK1 mediated by the E3 ubiquitin ligase mindbomb 1 (MIB1) at lysine 494. Moreover, SRPK1 S-palmitoylation is catalyzed by zinc-finger DHHC S-acyltransferase 24 (ZDHHC24) at cysteines 188/502/647 and deacylated by acyl protein thioesterase 1 (APT1). The dynamic S-palmito

The Causal Relationship Between Palmitoylation Genes and Parkinson's Disease: A Mendelian Randomization Study.

This work examined the causal link between palmitoylation-related genes and Parkinson's disease (PD) by Mendelian randomization (MR). We conducted two-sample Mendelian randomization utilizing genome-wide association study (GWAS) summary statistics. Expression quantitative trait loci (eQTL) data for palmitoylation genes were sourced from eQTLGen, whilst PD data were derived from the GWAS Catalogue (15,056 cases, 12,637 controls) and FinnGen (4,681 cases, 407,500 controls). Instrumental variables (p < 5 × 10⁻⁶, r2 < 0.1) were examined using inverse-variance weighted (IVW) regression, along with

Initiation of human cytomegalovirus secondary envelopment requires the gM/gN glycoprotein complex and involves palmitoylation.

Glycoprotein M (gM) of human cytomegalovirus (HCMV) forms a conserved protein complex with glycoprotein N (gN), whose precise function in viral morphogenesis is poorly understood. To elucidate the function of the gM/gN complex in secondary envelopment, we employed a combination of viral mutants, siRNA knockdown, and ultrastructural analyses. Ultrastructural examination of a mutant virus with a cysteine-to-serine mutation in the cytoplasmic tail of gN (TB-gN-C123S) showed a defect in the initiation of secondary envelopment as most capsids in TB-gN-C123S-infected cells were either not in contact

Palmitoyl acyltransferase Aph2 in cardiac function and the development of cardiomyopathy.

Protein palmitoylation regulates many aspects of cell function and is carried out by acyl transferases that contain zf-DHHC motifs. The in vivo physiological function of protein palmitoylation is largely unknown. Here we generated mice deficient in the acyl transferase Aph2 (Ablphilin 2 or zf-DHHC16) and demonstrated an essential role for Aph2 in embryonic/postnatal survival, eye development, and heart development. Aph2(-/-) embryos and pups showed cardiomyopathy and cardiac defects including bradycardia. We identified phospholamban, a protein often associated with human cardiomyopathy, as an

Protein Palmitoylation as a Molecular Switch Linking Regulated Cell Death and Disease.

Regulated cell death is essential for tissue homeostasis, immune defense, and disease progression, yet the lipid-based regulatory mechanisms that coordinate cell death signaling remain incompletely understood. Protein palmitoylation is a dynamic and reversible lipid post-translational modification that controls protein membrane association, trafficking, stability, and signaling complex assembly. This review summarizes the regulatory roles of palmitoylation and depalmitoylation in major forms of regulated cell death, including apoptosis, necroptosis, pyroptosis, ferroptosis, and autophagy-relat

Co-dispersion-mediated stabilization of a poorly soluble palmitoylation inhibitor within an alginate hydrogel for localized ferroptosis therapy.

Stabilizing high-loadings of hydrophobic small molecules in aqueous systems remains challenging due to crystallization and phase separation, which limit their therapeutic application. Here, we report a co-dispersion-mediated interfacial stabilization strategy that enables high-loading delivery of the poorly soluble palmitoylation inhibitor 2-bromopalmitic acid (2-BP) within an injectable alginate hydrogel system. Through high-shear co-dispersion with Fe₃O₄ nanoparticles, 2-BP undergoes amorphization and is stabilized in a uniformly dispersed state via interfacial interactions and spatial confi

Palmitoylation Induced Activation of SMAD3 Exacerbates Colitis by Promoting Tissue-resident Memory T Cells Differentiation.

Tissue-resident memory T cells (TRM cells) have been shown to play an instrumental role in driving the onset and relapse of inflammatory bowel diseases (IBD). However, the underlying mechanism of TRM cells differentiation and its regulation in intestines remain to be unveiled. Mothers against decapentaplegic homolog 3 (SMAD3) is translocated from nucleus to membrane and activated in response to transforming growth factor beta (TGF-β), which is a key cytokine in the process of TRM cells polarization.
