Quantitative study of testis histology and plasma androgens at onset of spermatogenesis in the prepuberal laboratory-born macaque (Macaca fascicularis).

Laboratory-born Macaca fascicularis underwent successive testicular biopsies and peripheral blood sampling during the peripuberal age. Testicular fragments were studied quantitatively on seminiferous cord or tubule histological cross sections. Plasma testosterone, androstenedione, and dihydrotestosterone were quantified by RIA. Enlargement of the diameter of seminiferous cords or tubules resulted from the increasing number of spermatocytes and the formation of the lumen. The number of Sertoli cells per cord or tubule cross section decreased abruptly at the onset of full spermatogenesis, and, a…

Age-related decline of lymphoid-tissue components in the oral mucosa of the lip, cheek and soft palate of Macaca fascicularis.

In three mature monkeys (Macaca fascicularis; 3.5 +/- 0.3 kg in weight), the labial, buccal and soft-palate mucosae were examined morphologically and stereologically. Using fixation by perfusion, standardized methods of tissue preparation and morphometric analysis at the light-microscopic level, the gross dimensions (i.e., epithelial thickness, width of combined lamina propria/submucosa) and the volumetric composition of the oral mucosae were estimated and compared with those of young animals examined previously. The data show (1) an age-related decline in the volume and prevalence of organize…

Anticancer Activity of Extracellular Vesicles Derived From Adipose Mesenchymal Stem Cells of Macaca fascicularis on WiDr Colon Cancer Cells.

BACKGROUND: Colon cancer death toll due to metastasis is expected to rise. Extracellular vesicles are signalling molecules which can regulate communication between cells and either inhibit or promote cancer metastasis. METHODS: Extracellular vesicles from adipose mesenchymal stem cells of Macaca fascicularis were obtained from previous research. Various concentrations of extracellular vesicles (5 μg/mL, 10 μg/mL, and 20 μg/mL) were applied to colon cancer cell line WiDr in a transwell invasion assay. CXCR3 gene expression analysis was done using RT-qPCR. RESULTS: Application of extracellular…

Differential roles of coding and non-coding transcripts in obesity: insights from RNA-seq analysis of Macaca fascicularis hepatocytes.

Long non-coding RNAs (lncRNAs) can regulate metabolism-associated genes and cooperate to activate obesity-related pathways. However, the role of lncRNAs in obesity-related diabetes is not clear. To address this, we analyzed the hepatic transcriptomes of diabetic and non-diabetic Cynomolgus monkeys (Macaca fascicularis) using next-generation sequencing (NGS). Our findings demonstrate that coding and non-coding RNAs exhibit distinct patterns of expression, with coding mRNAs notably enriched for metabolic pathways and particularly lipid transport. At the same time, the expression of genes related…

Evaluation of Selected Markers in Kidneys of Cynomolgus Monkey (Macaca fascicularis) with Induced Diabetes during Renal Ischemia-reperfusion Injury.

We previously reported that induced type 1 diabetes mellitus (DM) increases the susceptibility of acute kidney injury in- duced by ischemia-reperfusion injury (IRI) in cynomolgus monkeys. In this follow-up study, we compared the expression of selected markers in the renal tissues of monkeys subjected to bilateral renal IRI with and without diabetes. All tissues were obtained from the original study. Renal biopsies were obtained before and 24 and 48 h after ischemia and were examined for expression of KI-67 (tubular proliferation), Na+ /K+ ATPase (sodium-potassium pump), TNF-α(tumor necrosis fa

Effect of treatment for 6 months with human parathyroid hormone (1-34) peptide in ovariectomized cynomolgus monkeys (Macaca fascicularis).

A potential negative side effect of intermittent parathyroid hormone (PTH) therapy to treat osteoporosis is the loss of cortical bone concomitant with increased cancellous bone mass. We addressed this issue by studying the effects of PTH on whole-body, axial, and appendicular bone mass in an animal model with haversian cortical bone remodeling. Ovariectomized, young adult female cynomolgus monkeys were assigned to placebo (n = 9) or PTH groups (n = 10). The PTH group received 10 microg/kg synthetic human PTH(1-34) peptide by subcutaneous injection, 3 days/week for 6 months, and the placebo gro

Architecture of minor salivary gland duct/lymphoid follicle associations and possible antigen-recognition sites in the monkey Macaca fascicularis.

In Macaca fascicularis, lymphoid follicles with germinal centres related to minor salivary gland ducts are frequently found in the mucosa of lips, cheeks and the soft palate. Three semi-three-dimensional reconstructions, each based on 3 sets of 230-300 serial Epon sections, of such duct/follicle-assemblies (2 in the soft palate and 1 in the lip) and electron microscopic observations were made. These revealed that (1) these structures were about 0.05-0.1 mm3 in size, (2) they were found preferentially at sites where small interlobular ducts fused to form a pelvis-like basin from which blind duc…

Increases in the number of reactive glia in the visual cortex of Macaca fascicularis following subclinical long-term methyl mercury exposure.

The number of neurons, astrocytes, reactive glia, oligodendrocytes, endothelia, and pericytes in the cortex of the calcarine sulcus of adult female Macaca fascicularis following long-term subclinical exposure to methyl mercury (MeHg) and mercuric chloride (inorganic mercury; IHg) has been estimated by use of the optical volume fractionator stereology technique. Four groups of monkeys were exposed to MeHg (50 micrograms Hg/kg body wt/day) by mouth for 6, 12, 18, and 12 months followed by 6 months without exposure (clearance group). A fifth group of monkeys was administered IHg (as HgCl2; 200 mi…

Bilateral projections from precentral motor cortex to the putamen and other parts of the basal ganglia. An autoradiographic study in Macaca fascicularis.

By tracing radioactively labeled proteins transported by axonal flow, projections from area 4 to the ipsi- and contralateral neostriatum and claustrum were demonstrated in 7 monkeys. A reversed topographic organization was found on both sides for the corticoneostriatal and corticoclaustral projections. The most extensive terminal field could be observed in the putamen. In contrast, very few area 4 efferents seemed to terminate in the caudate nucleus. This suggests differential functions for the two striatal components in sensorimotor mechanisms. These unexpected results give further evidence f

A Novel Pre-Clinical Modeling of Massive Hemorrhagic Shock in Non-Human Primate: A Safe and Reproducible Method (Macaca fascicularis).

BACKGROUND: Although non-human primates are the closest animals to humans to simulate physiological and metabolic responses, there is a paucity of primate hemorrhagic shock models that are standardized and reproducible. Herein, we describe a model that is a clinical replica of extreme class IV hemorrhagic shock with a step-by-step description of the procedure in cynomolgus macaque monkeys. METHODS: The physiological changes that occurred during the process were evaluated using hemodynamic parameters, echocardiogram, and laboratory values. Five female monkeys were subjected to trauma laparotom

Identification and characterization of the tyrosinase gene (TYR) and its transcript variants (TYR_1 and TYR_2) in the crab-eating macaque (Macaca fascicularis).

Tyrosinase is a copper-containing enzyme that regulates melanin biosynthesis and is encoded by the tyrosinase (TYR) gene. Previous studies demonstrated that mutations in TYR could lead to oculocutaneous albinism type 1 (OCA1) owing to the failure of melanin formation. Although a previous study found that albinism in the rhesus monkey was derived from a mutation in TYR, the identification and characterization of this gene in non-human primates has not been achieved thus far. Thus, using the rapid amplification of cDNA ends (RACE) and internal reverse transcription PCR (RT-PCR) we identified the

Sex differences in the stone tool-use behavior of a wild population of burmese long-tailed macaques (Macaca fascicularis aurea).

We investigated sex differences in how Burmese long-tailed macaques (Macaca fascicularis aurea) used stone tools to open shelled food items along the shores of two islands in Laemson National Park, Thailand. Over a 2-week period in December 2009, we collected scan and focal samples on macaques when they were visible along the shores and mangroves. We found females used stones more often while feeding and used smaller tools than males. Females also processed sessile oysters more than males, whereas males processed unattached foods more than females. It was unclear which sex was overall more pro…

Plasmodium knowlesi infections in a small number of non-immune natural hosts (Macaca fascicularis) and in rhesus monkeys (M. mulatta).

The natural host of Plasmodium knowlesi is the kra monkey, Macaca fascicularis, but this parasite, initially mistaken for P. malariae, is now infecting humans in some areas of Southeast Asia. Here we present data from experiments performed in the 1970s in which sera from a few naive M. fascicularis, taken in the course of a first infection, exhibited rapidly rising inhibition of in vitro replication of P. knowlesi. The results were compared with sera from P. knowlesi-infected rhesus monkeys that usually die if left untreated.

Measurement of the flux of lead from bone to blood in a nonhuman primate (Macaca fascicularis) by sequential administration of stable lead isotopes.

To better understand the kinetics of the transfer of lead from bone to blood, we have developed and tested a method in which sequential doses of lead, each enriched with a different stable isotope, were administered in a nonhuman primate Macaca fascicularis whose skeleton had been previously labeled with lead of known isotopic composition. Lead isotopic ratios of blood and bone samples, analyzed by thermal ionization mass spectrometry (TIMS), were unmixed by isotope dilution techniques. The first label administered allows the contribution from historical bone stores to be measured. Subsequent …

Suppression of plasma gonadotropins and testosterone in adult male monkeys (Macaca fascicularis) by a potent inhibitory analog of gonadotropin-releasing hormone.

The reduction of circulating levels of gonadotropins and testosterone is of value for the treatment of steroid-dependent neoplasms and the control of fertility. We tested the ability of single and multiple doses of the GnRH antagonist [N-Ac-D-Nal(2)1, D-pCl-Phe2, D-Trp3, D-hArg(Et2)6, D-Ala10]GnRH (GnRH-Ant) to suppress levels of these hormones in intact and castrate adult male cynomolgus monkeys. In Exp I, single injections of the antagonist at doses of 0 (vehicle), 5, 50, 250, and 500 micrograms/kg BW were given to castrate and intact animals. In Exp II, daily injections of antagonist at dos

Connections of the ventroposterior nucleus of the thalamus with the body surface representations in cortical areas 3b and 1 of the cynomolgus macaque, (Macaca fascicularis).

The somatotopic pattern of the connections of the ventroposterior nucleus of the thalamus with the representations of the body surface in somatic cortical Areas 3b and 1 were investigated in macaque monkeys using the anatomical tracers horseradish peroxidase (HRP), and 3H-proline. These tracers, in combination or separately, were injected into one or more cortical sites, after the skin surface capable of activating neurons at each site had been defined with microelectrode mapping methods. These injection sites were later related to the overall organizations of the two cutaneous representations

Single-nucleus RNA sequencing reveals molecular determinants of functional and physiological differences between extraocular and quadriceps muscles in Macaca fascicularis.

Extraocular muscles (EOM) display distinct physiological and pathological characteristics compared with limb skeletal muscles. To define the molecular basis of these differences, we performed single-nucleus RNA sequencing on EOM and quadriceps femoris muscle (QFM) from Macaca fascicularis. Transcriptomic profiling revealed distinct cell type compositions, with EOM enriched in fast-twitch fibers (2 A/2×), fibro-adipogenic progenitors, and neural-associated cells. EOM myonuclei expressed genes involved in synaptic transmission and axon development, whereas QFM myofibers were enriched for oxidati…
