Formation and mobilization of methylmercury across natural and experimental sulfur deposition gradients.

We investigated the influence of sulfate (SO42-) deposition and concentrations on the net formation and solubility of methylmercury (MeHg) in peat soils. We used data from a natural sulfate deposition gradient running 300 km across southern Sweden to test the hypothesis posed by results from an experimental field study in northern Sweden: that increased loading of SO42- both increases net MeHg formation and redistributes methylmercury (MeHg) from the peat soil to its porewater. Sulfur concentrations in peat soils correlated positively with MeHg concentrations in peat porewater, along the depos

Increased Methylmercury Accumulation in Rice after Straw Amendment.

Consumption of rice has been shown to be an important route of dietary exposure to methylmercury (MeHg, a neurotoxin) for Asians having a low fish but high rice diet. Therefore, factors that increase MeHg production and bioaccumulation in soil-rice systems, could enhance the risk of MeHg exposure. On the basis of a national-scale survey in China (64 sites in 12 provinces) and rice cultivation experiments, we report that straw amendment, a globally prevalent farming practice, could increase MeHg concentrations in paddy soils (11-1043%) and rice grains (95%). By carrying out a series of batch in

Effects of cadmium, inorganic mercury and methyl-mercury on the physiology and metabolomic profiles of shoots of the macrophyte Elodea nuttallii.

Macrophytes are known to bioaccumulate metals, but a thorough understanding of tolerance strategies and molecular impact of metals in aquatic plants is still lacking. The present study aimed to compare Hg and Cd effects in a representative macrophyte, Elodea nuttallii using physiological endpoints and metabolite profiles in shoots and cytosol. Exposure 24 h to methyl-Hg (30 ng L-1), inorganic Hg (70 ng L-1) and Cd (280 μg L-1) did not affect photosynthesis, or antioxidant enzymes despite the significant accumulation of metals, confirming a sublethal stress level. In shoots, Cd resulted in a hi

Heliobacteria Reveal Fermentation As a Key Pathway for Mercury Reduction in Anoxic Environments.

The accumulation of mercury (Hg) in rice, a dietary staple for over half of the world's population, is rapidly becoming a global food safety issue. Rice paddies support the anaerobic production of toxic methylmercury that accumulates in plant tissue, however the microbial controls of Hg cycling in anoxic environments remain poorly understood. In this study, we reveal a novel reductive Hg metabolism in a representative of the family Heliobacteria ( Heliobacterium modesticaldum Ice1) that we confirm in model chemotrophic anaerobes. Heliobacteria served as our initial model because they are a fam

Long-Term Feeding of Elemental Sulfur Alters Microbial Community Structure and Eliminates Mercury Methylation Potential in Sulfate-Reducing Bacteria Abundant Activated Sludge.

This study reported a novel observation that the long-term cultivation of sulfur-reducing bacteria (S0RB) from a sulfate-reducing bacteria (SRB)-abundant seeding sludge with elemental sulfur feeding significantly shaped the microbial community structure and eliminated the mercury methylation potential in the S0RB-enriched sludge. In this study, the enrichments of SRB and S0RB from activated sludge were obtained through long-term cultivations. Subsequently, the batch tests showed that approximately 5000 μg/L Hg (II) was completely removed from the solution by both the SRB-enriched and S0RB-enri

Unraveling Microbial Communities Associated with Methylmercury Production in Paddy Soils.

Rice consumption is now recognized as an important pathway of human exposure to the neurotoxin methylmercury (MeHg), particularly in countries where rice is a staple food. Although the discovery of a two-gene cluster hgcAB has linked Hg methylation to several phylogenetically diverse groups of anaerobic microorganisms converting inorganic mercury (Hg) to MeHg, the prevalence and diversity of Hg methylators in microbial communities of rice paddy soils remain unclear. We characterized the abundance and distribution of hgcAB genes using third-generation PacBio long-read sequencing and Illumina sh

Methylmercury uptake and degradation by methanotrophs.

Methylmercury (CH3Hg+) is a potent neurotoxin produced by certain anaerobic microorganisms in natural environments. Although numerous studies have characterized the basis of mercury (Hg) methylation, no studies have examined CH3Hg+ degradation by methanotrophs, despite their ubiquitous presence in the environment. We report that some methanotrophs, such as Methylosinus trichosporium OB3b, can take up and degrade CH3Hg+ rapidly, whereas others, such as Methylococcus capsulatus Bath, can take up but not degrade CH3Hg+. Demethylation by M. trichosporium OB3b increases with increasing CH3Hg+ conce

Transcriptomic and Physiological Responses of the Green Microalga Chlamydomonas reinhardtii during Short-Term Exposure to Subnanomolar Methylmercury Concentrations.

The effects of short-term exposure to subnanomolar methyl-mercury (MeHg) concentrations, representative of contaminated environments, on the microalga Chlamydomonas reinhardtii were assessed using both physiological end points and gene expression analysis. MeHg bioaccumulated and induced significant increase of the photosynthesis efficiency, while the algal growth, oxidative stress, and chlorophyll fluorescence were unaffected. At the molecular level, MeHg significantly dysregulated the expression of genes involved in motility, energy metabolism, lipid metabolism, metal transport, and antioxid

[Concentrations and deposition fluxes of different mercury species in precipitation in Jinyun Mountain, Chongqing].

Concentrations of mercury (Hg) species in precipitation were measured during the period from April 2013 to March 2014 in Jinyun Mountain, Chongqing, and corresponding deposition fluxes were also estimated. The result showed that the ranges of concentration of total mercury (THg) , dissolved mercury (DHg), particulate mercury (PHg), reactive mercury (RHg), total methyl mercury (MeHg), dissolved methyl mercury (DMeHg), particulate methyl mercury (PMeHg) were 7.47-120.11, 2.51-43.03, 2.28-77.99, 0.14-15.14, 2.58 x 10(-2)-101.62 x 10(-2), 0.30 x 10(-2)-72.29 x 10(-2), and 1.45 x 10(-2)-63.55 x 10(

Rice methylmercury exposure and mitigation: a comprehensive review.

Rice cultivation practices from field preparation to post-harvest transform rice paddies into hot spots for microbial mercury methylation, converting less-toxic inorganic mercury to more-toxic methylmercury, which is likely translocated to rice grain. This review includes 51 studies reporting rice total mercury and/or methylmercury concentrations, based on rice (Orzya sativa) cultivated or purchased in 15 countries. Not surprisingly, both rice total mercury and methylmercury levels were significantly higher in polluted sites compared to non-polluted sites (Wilcoxon rank sum, p<0.001). However,

Detailed assessment of the kinetics of Hg-cell association, Hg methylation, and methylmercury degradation in several Desulfovibrio species.

The kinetics of inorganic Hg [Hg(II)(i)] association, methylation, and methylmercury (MeHg) demethylation were examined for a group of Desulfovibrio species with and without MeHg production capability. We employed a detailed method for assessing MeHg production in cultures, including careful control of medium chemistry, cell density, and growth phase, plus mass balance of Hg(II)(i) and MeHg during the assays. We tested the hypothesis that differences in Hg(II)(i) sorption and/or uptake rates drive observed differences in methylation rates among Desulfovibrio species. Hg(II)(i) associated rapid

Simultaneous determination of mercury methylation and demethylation capacities of various sulfate-reducing bacteria using species-specific isotopic tracers.

The use of species-specific isotopic tracers for inorganic and methyl mercury has allowed the simultaneous determination of the methylation and demethylation potentials of pure culture of isolated sulfate-reducing (SR) bacterial strains using low Hg species concentration levels (7 µg/L (199)Hg(II), 1 µg/L Me(201)Hg). A major advantage of the method reported here is that it can be used to follow simultaneously both the degradation of the species added but also the formation of their degradation products and thus the determination during the same incubation of the specific methylation/demethylat

Direct absorption of methyl mercury by lymph.

Methyl mercury is contained in fish and seafood products and is taken up into the body in food. While the central nervous system is known as a target organ, methyl mercury also induces autoimmunity and acts as a potent immunosuppressor. The aim of the present study is to know whether methyl mercury is directly absorbed by lymph. Conscious rats were infused with methyl mercury (4 mg/kg) via duodenal tubing as a single pulse infusion, followed by the continuous infusion of saline, and lymphatic fluids were continuously collected from the thoracic lymph duct every 30 min until 360 min after infus

Reducing total mercury and methylmercury accumulation in rice grains through water management and deliberate selection of rice cultivars.

Rice consumption has been identified as a major route of methylmercury (MeHg) exposure in some areas of inland China. We investigated two potential mitigation methods (water management and deliberate selection of rice cultivars) to reduce the amount of total mercury (Hg) and MeHg within the grain. Rice grown aerobically had markedly reduced total Hg and MeHg concentrations as well as a much lower proportion of MeHg in the grain. Remarkably, there were considerable variations in the total Hg and MeHg concentrations as well as the proportion of MeHg in the grain among the 24 cultivars grown in t

Distribution patterns of inorganic mercury and methylmercury in tissues of rice (Oryza sativa L.) plants and possible bioaccumulation pathways.

Whole rice plants (Oryza sativa L.) were collected at different typical mercury (Hg) contaminated sites during regular harvest periods to investigate the distribution of inorganic mercury (IHg) and methylmercury (MeHg) in tissues. The whole rice plants were divided into rice seed (brown rice), hull, root, stalk and leaf. Elevated IHg and MeHg concentrations were observed in rice plants cultivated in Hg mining area compared to those obtained from the control site, which attributed to the Hg contamination of soil compartments by the historical large-scale Hg mining/smelting and ongoing artisanal

Mercury in the blood and eggs of American kestrels fed methylmercury chloride.

American kestrels (Falco sparverius) were fed diets containing methylmercury chloride (MeHg) at 0, 0.6, 1.7, 2.8, 3.9, or 5.0 µg/g (dry wt) starting approximately eight weeks before the onset of egg laying. Dietary treatment was terminated after 12 to 14 weeks, and unhatched eggs were collected for Hg analysis. Blood samples were collected after four weeks of treatment and the termination of the study (i.e., 12-14 weeks of treatment). Clutch size decreased at dietary concentrations above 2.8 µg/g. The average total mercury concentration in clutches of eggs and in the second egg laid (i.e., egg

Preparation and characterization of a soil reference material from a mercury contaminated site for comparability studies.

The preparation and characterization of a soil reference material (SOIL-1) from a site polluted with mercury due to the past mercury mining in Idrija, Slovenia is reported. Homogeneity tests and intercomparison exercises for total (T-Hg) and methylmercury (MeHg) were performed. In addition, selective sequential extraction was applied for Hg fractionation, and multielemental analyses were performed by k(0) standardization neutron activation analysis (k(0)-INAA) and inductively coupled mass spectrometry (ICP-MS) for other trace elements. Comparison of different analytical methods, as well as the

Methylmercury in freshwater fish linked to atmospheric mercury deposition.

A connection between accumulation of methylmercury (MeHg) in wild fish populations and atmospheric mercury deposition has not been made. Large databases for both MeHg in fish and atmospheric mercury deposition have been assimilated from monitoring efforts spanning the contiguous United States. Here, we compare results of these data sets and show that state-wide average concentrations of MeHg in a cosmopolitan freshwater fish, the largemouth bass Micropterus salmoides, are related positively to wet atmospheric Hg fluxes among most of the 25 states that are analyzed, which span a 5-fold range in
