Revisiting the muscle spindle in ALS: Implications for spinal motor network dysfunction.

Amyotrophic lateral sclerosis (ALS) is typically characterized by progressive degeneration of upper and lower motor neurons. However, the contribution of afferent sensory pathways to spinal motor network disorganization has received little attention. Converging human histological evidence from multiple independent laboratories demonstrates that fusimotor (gamma) motor neuron innervation of muscle spindles degenerates in ALS while sensory endings remain morphologically intact at the end organ, although the afferent neurons and ascending sensory pathways are also affected. This dissociation crea

Multidimensional Effects of Stress on Neuronal Exosome Levels and Simultaneous Transcriptomic Profiles.

BACKGROUND: An excess of exosomes, nanovesicles released from all cells and key regulators of brain plasticity, is an emerging therapeutic target for stress-related mental illnesses. The effects of chronic stress on exosome levels are unknown; even less is known about molecular drivers of exosome levels in the stress response. METHODS: We used our state-of-the-art protocol with 2 complementary strategies to isolate neuronal exosomes from plasma, ventral dentate gyrus, basolateral amygdala, and olfactory bulbs of male mice to determine the effects of chronic restraint stress (CRS) on exosome l

Targeting STMN2 for neuroprotection and neuromuscular recovery in Spinal Muscular Atrophy: evidence from in vitro and in vivo SMA models.

The development of ground-breaking Survival Motor Neuron (SMN) replacement strategies has revolutionized the field of Spinal Muscular Atrophy (SMA) research. However, the limitations of these therapies have now become evident, highlighting the need for the development of complementary targets beyond SMN replacement. To address these challenges, here we explored, in in vitro and in vivo disease models, Stathmin-2 (STMN2), a neuronal microtubule regulator implicated in neurodegenerative diseases like Amyotrophic Lateral Sclerosis (ALS), as a novel SMN-independent target for SMA therapy. Our find

Amyloid-β mediates intestinal dysfunction and enteric neurons loss in Alzheimer's disease transgenic mouse.

Alzheimer's disease (AD) is traditionally considered as a brain disorder featured by amyloid-β (Aβ) deposition. The current study on whether pathological changes of AD extend to the enteric nervous system (ENS) is still in its infancy. In this study, we found enteric Aβ deposition, intestinal dysfunction, and colonic inflammation in the young APP/PS1 mice. Moreover, these mice exhibited cholinergic and nitrergic signaling pathways damages and enteric neuronal loss. Our data show that Aβ42 treatment remarkably affected the gene expression of cultured myenteric neurons and the spontaneous contra

Neuromyelitis optica MOG-IgG causes reversible lesions in mouse brain.

INTRODUCTION: Antibodies against myelin oligodendrocyte glycoprotein (MOG-IgG) are present in some neuromyelitis optica patients who lack antibodies against aquaporin-4 (AQP4-IgG). The effects of neuromyelitis optica MOG-IgG in the central nervous system have not been investigated in vivo. We microinjected MOG-IgG, obtained from patients with neuromyelitis optica, into mouse brains and compared the results with AQP4-IgG. RESULTS: MOG-IgG caused myelin changes and altered the expression of axonal proteins that are essential for action potential firing, but did not produce inflammation, axonal

Childhood infection and adult disease.

In England and Wales there is a strong geographical relation between current mortality from chronic bronchitis and emphysema in adults and infant mortality from bronchitis and pneumonia 50 years ago. Follow-up studies of infants and children show that certain pulmonary infections cause persisting abnormalities of lung function. This suggests that infection of an organ system during a period of rapid growth may have permanent deleterious effects. Long-term consequences of infection may also depend on age-related differences in the host response. The relationship between age of infection with he

Enhanced alpha-synuclein pathology and exacerbated motor dysfunction in alpha-synuclein transgenic mice with autophagy deficiency.

Alpha-synuclein (α-synuclein), a key component of Lewy body pathology, is a hallmark of Parkinson's disease. In previous studies, we examined dopaminergic neuron-specific Atg7 autophagy-deficient mice and observed α-synuclein aggregation in vivo. Notably, p62 accumulation preceded synuclein deposition, resulting in the formation of inclusions containing both α-synuclein and p62. This pathological process led to dopamine neuron loss and age-related motor impairments, such as hindlimb defects in 120-week-old mice. In this study, we developed a mouse model by crossing human α-synuclein bacterial

LRRK2 knockdown in zebrafish causes developmental defects, neuronal loss, and synuclein aggregation.

Although mutations in the leucine-rich repeat kinase 2 (LRRK2) gene are the most common cause of genetic Parkinson's disease, their function is largely unknown. LRRK2 is pleiotropic in nature, shown to be involved in neurodegeneration and in more peripheral processes, including kidney functions, in rats and mice. Recent studies in zebrafish have shown conflicting evidence that removal of the LRRK2 WD40 domain may or may not affect dopaminergic neurons and/or locomotion. This study shows that ∼50% LRRK2 knockdown in zebrafish causes not only neuronal loss but also developmental perturbations su

Sensorimotor Expectations Bias Motor Resonance during Observation of Object Lifting: The Causal Role of pSTS.

Transcranial magnetic stimulation studies have highlighted that corticospinal excitability is increased during observation of object lifting, an effect termed "motor resonance." This facilitation is driven by movement features indicative of object weight, such as object size or observed movement kinematics. Here, we investigated in 35 humans (23 females) how motor resonance is altered when the observer's weight expectations, based on visual information, do not match the actual object weight as revealed by the observed movement kinematics. Our results highlight that motor resonance is not robus

Morphine or U-50,488 suppresses Fos protein-like immunoreactivity in the spinal cord and nucleus tractus solitarii evoked by a noxious visceral stimulus in the rat.

Immunohistochemical visualization of Fos protein, the nuclear phosphoprotein product of the early-immediate gene c-fos, permits identification of populations of neurons that are activated in response to a variety of stimuli. This study examined the distribution of Fos-like immunoreactive (FLI) neurons in the spinal cord and the nucleus tractus solitarii (NTS) of the caudal medulla evoked by a noxious visceral stimulus in the unanesthetized rat. It also compared the inhibition of pain behavior and Fos expression by a mu-selective opioid agonist, morphine, and a kappa-selective opioid agonist, U

Evolution of cognitive and neural solutions enabling numerosity judgements: lessons from primates and corvids.

Brains that are capable of representing numerosity, the number of items in a set, have arisen repeatedly and independently in different animal taxa. This review compares the cognitive and physiological mechanisms found in a nonhuman primate, the rhesus macaque, and a corvid songbird, the carrion crow, in order to elucidate the evolutionary adaptations underlying numerical competence. Monkeys and corvids are known for their advanced cognitive competence, despite them both having independently and distinctly evolved endbrains that resulted from a long history of parallel evolution. In both speci

Nociceptor-immune interactomes reveal insult-specific immune signatures of pain.

Inflammatory pain results from the heightened sensitivity and reduced threshold of nociceptor sensory neurons due to exposure to inflammatory mediators. However, the cellular and transcriptional diversity of immune cell and sensory neuron types makes it challenging to decipher the immune mechanisms underlying pain. Here we used single-cell transcriptomics to determine the immune gene signatures associated with pain development in three skin inflammatory pain models in mice: zymosan injection, skin incision and ultraviolet burn. We found that macrophage and neutrophil recruitment closely mirror

[Autism. Neurobiological aspects].

Autism is a neurodevelopmental disorder with a neurobiological basis, characterized by a qualitative disturbance in social interaction and communication, associated with restricted interests and stereotyped behaviors. The genesis of autism cannot be interpreted through a single theory, and we can't compartmentalize brain areas as the only ones responsible for it. Among the neurobiological bases we can include: deficit in the social reward system, which generates poor social initiative; dysfunctions and disorders of the amygdala and the mirror neuron system, related to compromised empathy and s

CXCL10 activates STAT3 signaling to promote ferroptosis in the spinal dorsal horn during neuropathic pain.

Neuropathic pain (NP) remains a clinical challenge due to the limited efficacy and significant side effects of current treatments, highlighting the need for a deeper understanding of its pathogenesis. While dysfunction of GABAergic inhibitory neurons in the spinal dorsal horn is a known key driver of central sensitization, the mechanisms precipitating this neuronal injury remain obscure. In this study, using a spinal nerve ligation (SNL) model, we aimed to investigate whether the chemokine CXCL10 drives NP progression by promoting ferroptosis, a process that is associated with and ultimately l

Modeling disrupted synapse formation in wolfram syndrome using hESCs-derived neural cells and cerebral organoids identifies Riluzole as a therapeutic molecule.

Dysregulated neurite outgrowth and synapse formation underlie many psychiatric disorders, which are also manifested by wolfram syndrome (WS). Whether and how the causative gene WFS1 deficiency affects synapse formation remain elusive. By mirroring human brain development with cerebral organoids, WFS1-deficient cerebral organoids not only recapitulate the neuronal loss in WS patients, but also exhibit significantly impaired synapse formation and function associated with reduced astrocytes. WFS1 deficiency in neurons autonomously delays neuronal differentiation with altered expressions of genes

Neural mechanisms of self-processing in autism: An ALE-based meta-analysis.

Self-processing in autism affects social interactions. By choosing 8 fMRI studies of autism spectrum disorder (ASD) self-processing, this study used ALE meta-analysis to investigate the brain underpinnings of self-processing in ASD. The findings revealed the following: (1) The capacity to make self-other distinctions and mirroring in social interactions is impacted by inadequate activation of brain areas of the mirror neuron system in ASD, which in turn impairs social interactions. (2) ASD activated more brain regions in self-processing than typically developing (TD) individuals. TD individual

Neuroplasticity and Rhinoplasty: Bridging Neuroscience and Aesthetic Surgery.

Neuroplasticity, the brain's ability to reorganize its structure and function, plays a pivotal role in adapting to anatomical changes introduced by surgical interventions such as rhinoplasty. This review explores the interplay between functional and aesthetic outcomes of rhinoplasty and their implications on cognitive and emotional processes. Key findings highlight the involvement of specialized neural circuits, including the fusiform face area, somatosensory cortex, and mirror neuron systems, in adapting to altered facial features. Furthermore, the importance of preoperative expectations, emo

Normal movement reading in Asperger subjects.

Patients with autism and Asperger syndrome (AS) are impaired in mindreading and imitation skills. One possibility would be that their 'mirror neuron' system, which matches action execution and observation, does not function properly. To test this hypothesis we compared action-viewing related motor cortex functions in an AS group (one autistic and four AS subjects) and eight control subjects. In both groups viewing hand actions modified the neuromagnetic approximately 20 Hz oscillatory activity in the primary motor cortex to the same extent. Thus impaired mindreading and imitation skills found

Spatiotemporal Phase Synchronization in Adaptive Reconfiguration from Action Observation Network to Mentalizing Network for Understanding Other's Action Intention.

In action intention understanding, the mirror system is involved in perception-action matching process and the mentalizing system underlies higher-level intention inference. By analyzing the dynamic functional connectivity in α (8-12 Hz) and β (12-30 Hz) frequency bands over a "hand-cup interaction" observation task, this study investigates the topological transition from the action observation network (AON) to the mentalizing network (MZN), and estimates their functional relevance for intention identification from other's different action kinematics. Sequential brain microstates were extracte

Activity of Corticotropin-Releasing Factor Neurons in the Bed Nucleus of the Stria Terminalis Reduces Anxiety-Potentiated Startle in Female Rats in an Estrous Phase-Dependent Manner.

BACKGROUND: The prevalence of posttraumatic stress disorder (PTSD) and anxiety disorders is higher in women than men. The severity of hallmark symptoms including hypervigilance and fear reactivity to unpredictable threats varies with sex and reproductive cycle, but the underlying mechanisms remain unclear. Here, we investigated corticotropin-releasing factor (CRF) neurons in the dorsolateral bed nucleus of the stria terminalis (BNSTDL) as a potential nexus for the influence of biological sex and reproductive cycle on fear- and anxiety-related behaviors. METHODS: A total of 125 male and 156 cy
