Intergenerational and organ-specific alterations in mitochondrial DNA copy number following preconception irradiation.

Ionizing radiation, a potent inducer of redox stress, perturbs both nuclear and mitochondrial genomes, yet how such stress shapes mitochondrial inheritance across generations remains unclear. In this study, we examined intergenerational and organ-specific mitochondrial responses to parental X-ray irradiation in mice. Eight-week-old male and female C57BL/6N mice were exposed to 2 Gy of single whole-body X-ray irradiation before mating, generating paternal-, maternal-, and dual-irradiated lineages. In the parents, peripheral blood-derived mitochondrial DNA copy number (mtDNAcn) transiently incre

Research Review: Developmental origins of depression - a systematic review and meta-analysis.

BACKGROUND: Many observational studies have found a direct association between adverse in utero, perinatal and postnatal exposures and offspring's depression. These findings are consistent with the 'developmental origins of disease hypothesis'. But no review has comprehensively summarized the roles of these exposures. This review aims to systematically scrutinize the strength of associations between individual prenatal, perinatal, and postnatal exposures and subsequent depression in offspring. METHODS: We conducted a systematic review and meta-analysis to synthesize the literature from the EM

Multi-omics analysis of gestational PM2.5 exposure induces abnormal cardiac metabolism and development in offspring.

Epidemiological evidence suggests that exposure to ambient fine particulate matter (PM2.5) during pregnancy may increase the risk of congenital heart disease; however, the underlying mechanisms remain unclear. This study aimed to investigate the effects of gestational PM2.5 exposure on offspring cardiac development and to elucidate the potential mechanisms involved. Gestational exposure to PM2.5 significantly reduced litter size, birth weight, and offspring cardiac mass. Further analyses revealed inflammatory infiltration, lipid peroxidation, and decreased ATP levels in the offspring hearts, i

Effects of Excessive Activation of N-methyl-D-aspartic Acid Receptors in Neonatal Cardiac Mitochondrial Dysfunction Induced by Intrauterine Hypoxia.

Intrauterine hypoxia is a common complication during pregnancy and could increase the risk of cardiovascular disease in offspring. However, the underlying mechanism is controversial. Memantine, an NMDA receptor antagonist, is reported to be a potential cardio-protective agent. We hypothesized that antenatal memantine treatment could prevent heart injury in neonatal offspring exposed to intrauterine hypoxia. Pregnant rats were exposed to gestational hypoxia or antenatal memantine treatment during late pregnancy. Newborns were then sacrificed to assess multiple parameters. The results revealed t

Effects of maternal nutrition on fetal and neonatal reproductive development and function.

Maternal undernutrition and, under certain circumstances overnutrition, before or during pregnancy or during early postnatal life can alter reproductive function of the offspring. Effects can be exerted at many stages of development, from prior to conception until after birth and may be expressed at the time of the nutritional insult or later. Since patterns of development differ between species, it is probably more appropriate to consider effects in relation to a stage of development rather than relative to the time of birth. Effects exerted at one stage of development may be expressed later,

Maternal Undernutrition before and during Pregnancy and Offspring Health and Development.

Maternal undernutrition remains a critical public health problem. There are large regional and within-country disparities in the burden of underweight, anemia, and micronutrient deficiencies across the globe. Driving these disparities are complex and multifactorial causes, including access to health services, water and sanitation, women's status, and food insecurity as well as the underlying social, economic, and political context. Women's health, nutrition, and wellbeing across the continuum of preconception to pregnancy are critical for ensuring positive pregnancy and long-term outcomes for

A case-control study of paternal smoking and birth defects.

Although the influence of paternal smoking on birth defects is of great public interest, epidemiological evidence concerning this potential relationship is extremely limited. A stratified random sample of 29 hospitals in the Shanghai Municipality, China, was used to select 1012 birth defects cases and controls. Mothers of the cases and controls were interviewed in the hospitals from October 1986 to September 1987. A modest relationship between paternal smoking and overall birth defects in offspring was identified [odds ratio (OR) = 1.21, 95% confidence interval (CI): 1.01-1.45]. More markedly

Early maternal obesity shapes offspring development through immune mechanisms: a systematized review.

In recent years, rates of maternal obesity before conception have continued to rise globally. Maternal obesity in early pregnancy may alter intrauterine immune mechanisms, impact fetal metabolic programming, and increase the risk of later-life obesity. This study aimed to review the literature on changes in immune parameters and weight-related outcomes in offspring reported in human and animal studies of maternal obesity. A systematized literature search was conducted using PubMed and Web of Science for relevant human and animal studies published between 2010 and 2025. Study screening was faci

The childhoods of multiple problem adolescents: a 15-year longitudinal study.

This paper examines the life history of a small group of adolescents (3%) who were identified during the course of a longitudinal study of a birth cohort of New Zealand children as displaying multiple problem behaviours at the age of 15 years. This group was characterised by conduct disorder, police contact, substance abuse behaviours, early onset sexual activity, suicidal ideation, mood disorders and lowered self esteem. Statistical biographies of this group of young people showed that many were the offspring of seriously disadvantaged, dysfunctional and disorganised home environments. The im

Maternal Chlamydia infection during pregnancy among younger mothers and risk of gastroschisis in singleton offspring.

BACKGROUND: Prevalence of gastroschisis has been increasing in the United States (US) since 1980s. Genital infection with Chlamydia has also been increasing in prevalence, particularly among younger women. The etiology of gastroschisis is unknown. Ongoing research suggests multiple causal factors may be involved, including Chlamydia infection in mothers during pregnancy. METHODS: Using birth certificate data from 2014 US vital records, we conducted a population-based case-control study to examine the association between prenatal Chlamydia exposure in young women (16-25 years of age) and gastr

Effect of gestational diabetes and intrauterine growth restriction on the offspring's circulating galanin at birth.

CONTEXT: Experimental studies linked gestational diabetes mellitus (GDM) and intrauterine growth restriction (IUGR) with altered expression of the offspring's hypothalamic galanin mRNA, possibly contributing to the development of obesity and metabolic syndrome in later life. We hypothesized that plasma galanin levels at birth would reflect presumably altered hypothalamic galanin expression and production that cannot be assessed in the human offspring. OBJECTIVE: Our objective was to investigate whether neonates born to GDM mothers or being IUGR differ from healthy ones in circulating galanin

Maternal immunoglobulins are distributed in the offspring's brain to support the maintenance of cortical interneurons in the postnatal period.

It is known that maternal immunoglobulins (Igs) are transferred to the offspring across the placenta. However, receiving maternal Igs, especially before the blood-brain barrier (BBB) is formed in the offspring's brain, carries the risk of transferring some brain-reactive Igs. It is thus hypothesized that there may be some unknown benefit to the offspring's brain that overweighs this risk. In this study, we show that the Ig detected in the embryonic/perinatal mouse brain is IgG not produced by the pups themselves, but is basically transferred from the mother across the placenta using the neonat

The Pathophysiology of Spontaneous Preterm Birth: Emerging Mechanisms Reviewed by the Preterm Birth International Collaborative.

Around 10% of global births are preterm (before 37 weeks of gestation), posing a significant challenge to maternal and neonatal health. Preterm infants face an increased risk of mortality and long-term health complications, impacting their survival and development across all life stages. Despite decades (~ 80 years) of research, effective methods to predict and prevent idiopathic or spontaneous preterm birth remain limited. Therefore, a deeper understanding of the pathophysiology of spontaneous preterm birth is warranted. This review explores some aspects of recent progress in unravelling the

A population-based study of birth defects in Malaysia.

Birth defects are one of the leading causes of paediatric disability and mortality in developed and developing countries. Data on birth defects from population-based studies originating from developing countries are lacking. One of the objectives of this study was to determine the epidemiology of major birth defects in births during the perinatal period in Kinta district, Perak, Malaysia over a 14-month period, using a population-based birth defect register. There were 253 babies with major birth defects in 17,720 births, giving an incidence of 14.3/1000 births, a birth prevalence of 1 in 70.

Long-term neuropsychiatric outcomes in offspring of mothers with multiple sclerosis and neuromyelitis optica spectrum disorder: A nationwide cohort study.

BACKGROUND: Multiple sclerosis (MS) and neuromyelitis optica spectrum disorder (NMOSD) predominantly affect women of childbearing age, yet the impact of maternal disease status on long-term offspring outcomes remains unclear. METHODS: Using the Korean National Health Insurance Service database (2005-2019), we conducted a nationwide cohort study. Live-born offspring of mothers diagnosed with MS or NMOSD before childbirth were identified and matched 1:4 to controls using propensity scores. Neuropsychiatric outcomes were defined using ICD-10 codes. RESULTS: Among 3,524,987 live births, 257 were

A case-control study of infant, maternal and perinatal characteristics associated with deformational plagiocephaly.

Deformational plagiocephaly, an abnormal asymmetric flattening of infants' heads, is diagnosed in approximately 10% of infants. The prevalence of plagiocephaly has increased dramatically since 1992 when it was first recommended that infants be placed to sleep in a non-prone position to reduce the risk of sudden infant death syndrome. The authors conducted a case-control study to evaluate associations between plagiocephaly and perinatal characteristics. The authors assessed whether risk factors for plagiocephaly have changed since 1992. Cases were born 1987-2002 in Washington State and diagnose
